目的:基于网络药理学技术挖掘及佐证巴戟天-牛膝药对切合骨关节炎(OA)治则的效应机制。方法:在TCMSP数据库中筛选巴戟天-牛膝药对的所有成分,借助Uniprot数据库将药对中显效物质成分所靶向关联的效用点进行筛选,对OA的疾病靶点进行联合检索、收集与分析。将筛选出的显效靶标与OA靶点进行交联分析,将其共有靶标作为巴戟天-牛膝药对治疗OA的预测靶基因。将预测靶基因通过蛋白质-蛋白质互作关系(PPI)网络显示,筛选核心基因,构建“药物-成分-疾病-靶点”网络。联合基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析,探究巴戟天-牛膝药对切合OA治则的药效基础及效应机制。结果:经筛选分析得出槲皮素可能在巴戟天-牛膝药对治疗OA中发挥主导作用,观察预测靶基因之间的PPI关系,发现网络中转录因子AP-1(JUN)、丝氨酸/苏氨酸蛋白激酶(AKT1)、丝裂原活化蛋白激酶1(MAPK1)、V-Rel网状内皮增生病毒癌基因同源物A(RELA)、白细胞介素-6(IL-6)5个核心基因度值较高。GO功能和KEGG通路富集分析提示巴戟天-牛膝药对治疗OA主要涉及抗炎、抗氧化以及白细胞介素-17(IL-17)、肿瘤坏死因子(TNF)信号通路、细胞凋亡途径。在大鼠体外实验中,模型组与正常组比较,RELA、IL-17、IL-6蛋白表达升高(P<0.01);经过槲皮素治疗后RELA、IL-17、IL-6蛋白表达降低(P<0.01)。结论:巴戟天-牛膝药对治疗OA的潜在机制是通过槲皮素降低RELA、IL-17、IL-6蛋白表达,抑制炎症反应,从而延缓关节软骨退变。  相似文献

目的:基于生物信息学探讨骨质疏松症与 2 型糖尿病的关系。方法:通过基因表达汇编(GEO)数据库检索骨质疏松症和2型糖尿病的相关数据集,并筛选骨质疏松症-2型糖尿病共同差异表达基因,将数据导入STRING数据库找寻关键基因,以一个单细胞测序数据集作为检验集验证关键基因细胞层面的表达水平,对筛选结果进行基因本体(GO)功能及京都基因与基因组百科全书(KEGG)通路富集分析,并对关键靶点、相关中药进行频数统计及功效分类。结果:骨质疏松症与 2 型糖尿病相关通路集中在 ErbB信号通路、丝活化蛋白激酶(MAPK)信号通路、神经营养素信号通路、NOD样受体信号通路。频数统计结果显示,与骨质疏松症-2型糖尿病关键基因相关的中药有51味,以功效分类,可分为清热解毒药、止痛药、祛风药、补虚药、止呕药。结论:骨质疏松症和2型糖尿病都是复杂的代谢性疾病,涉及众多基因表达网络和信号通路。尽管两种疾病存在复杂的基因网络背景,但是分析结果表明两种疾病之间仍存在高度重合的差异基因表达和信号通路。这表明两种疾病的分子机制密切相关,可能存在共同调控的靶点,因此这些靶点可能成为药物同时干预两种疾病的治疗靶点。  相似文献

目的:运用网络药理学和分子对接技术探讨罗汉果治疗糖尿病的作用机制。方法:通过TCMSP平台和Swiss Target Prediction数据库筛选出罗汉果的活性成分及其靶点;利用GeneCard、TTD、OMIM数据库筛选出糖尿病潜在靶点;采用Cytoscape 3.7.2软件构建药物-活性成分-疾病-靶点的可视化网络;通过STRING数据库对关键靶点构建蛋白质-蛋白质相互作用(PPI)网络;利用Metascape数据库进行基因本体(GO)功能以及京都基因与基因组百科全书(KEGG)通路富集分析;利用Autodock4和Pymol软件对关键活性成分和关键靶点进行分子对接分析和可视化。结果:筛选出有效成分18种,糖尿病靶点146个。GO功能富集分析得到GO条目572条,KEGG通路富集分析得到信号通路167条。分子对接结果显示β-谷甾醇、山柰酚、罗汉果醇等关键活性成分与原癌基因酪氨酸蛋白激酶(SRC)等关键靶点的亲和力较高。结论:罗汉果通过多靶点、多通路发挥抗糖尿病的作用,其关键活性成分通过与SRC、丝氨酸/苏氨酸蛋白激酶1(AKT1)、丝裂原活化蛋白激酶3(MAPK-3)等靶点调节晚期糖基化终末产物-糖基化终末产物受体(AGE-RAGE)、缺氧诱导因子1(HIF-1)、磷脂酰肌醇-3-激酶/蛋白激酶(PI3K-Akt)等信号通路起到治疗糖尿病的作用。  相似文献

目的:基于网络药理学探讨六味地黄丸合缩泉丸治疗尿崩症(DI)的作用机制。方法:利用TCMSP和 UniProt 数据库获取六味地黄丸合缩泉丸的有效成分和作用靶点,通过GeneCards、OMIM、DrugBank数据库预测疾病靶点,将有效成分靶点与疾病靶点取交集,构建韦恩图获取共同靶点,运用STRING数据库和Cytoscape 3.8.2软件绘制交集靶点的蛋白质-蛋白质相互作用(PPI)网络,经拓扑分析得到关键靶点,并进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析。结果:共获得六味地黄丸合缩泉丸有效成分 31 个,作用于 185 个靶点,DI靶点共 897 个,六味地黄丸合缩泉丸与DI交集靶点 39 个,主要有效成分有槲皮素、山柰酚、β-谷甾醇、薯蓣皂苷元等,关键靶点包括丝氨酸/苏氨酸蛋白激酶1(AKT1)、白细胞介素-1β(IL-1β)、基质金属蛋白酶(MMP9)、过氧化物酶体增生激活受体(PPARG)、趋化因子(CCL2)、肿瘤蛋白TP53(p53)等。GO功能富集得到1667个条目,包括生物过程1582个、分子功能78个、细胞组分7个;KEGG通路富集得到110条通路,主要涉及动脉粥样硬化信号通路、糖尿病并发症中的AGE-RAGE信号通路、疟疾、肿瘤坏死因子信号通路等。结论:本研究初步揭示了六味地黄丸合缩泉丸治疗DI的作用机制,并通过数据分析侧面印证了中医辨证论治理论体系的正确性,可为进一步研究提供借鉴。  相似文献

目的:通过网络药理学和分子对接技术,探讨黄芪-白花蛇舌草抗非小细胞肺癌(NSCLC)的潜在活性成分和作用机制。方法:利用TCMSP和Uniprot数据库获取黄芪-白花蛇舌草的主要活性成分及靶点基因;GeneCards、OMIM数据库获取NSCLC靶点基因;绘制韦恩图得到疾病和活性成分共同靶点;STRING数据库构建中药成分靶蛋白-疾病靶蛋白的蛋白质-蛋白质相互作用(PPI)网络;Cytoscape 3.9.1软件绘制成分-靶点网络图,通过网络拓扑分析筛选出关键靶点及成分;DAVID数据库及Bioinformatics平台进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析;PyMol软件和AotoDock软件绘制相应的分子对接图。结果:经筛选共得到653个黄芪-白花蛇舌草活性成分靶点和6178个疾病靶点,两者交集靶点154个。GO功能富集到747个生物过程、83个分子功能和150个细胞组分,KEGG通路富集分析得到167条信号通路。分子对接结果显示,PTGS1蛋白与活性成分槲皮素、山柰酚、芒柄花黄素均结合稳定。结论:黄芪-白花蛇舌草可能通过槲皮素、山柰酚、芒柄花黄素等成分,PTGS1、丝氨酸/苏氨酸蛋白激酶(AKT1)、白细胞介素-6(IL-6)等靶点,丝裂原激活的蛋白激酶(MAPK)、磷脂酰肌醇3 激酶-蛋白激酶 B(PI3K-Akt)等信号通路治疗NSCLC,初步预测了黄芪-白花蛇舌草对NSCLC的作用机制,为深入揭示其治疗NSCLC的分子机制提供了理论基础。  相似文献

目的:利用网络药理学和分子对接技术探讨抑痰丸治疗医院获得性肺炎(HAP)的作用机制。方法:利用TCMSP、BATMAN-TCM数据库筛选抑痰丸的有效成分及作用靶点,从GeneCards、PharmGkb、DrugBank数据库获取HAP相关靶点,使用Venny 2.1.0平台筛选药物与疾病作用的交集靶点,采用Cytoscape软件绘制化学成分-靶点-通路-疾病交互网络,利用 STRING数据库构建蛋白质-蛋白质相互作用(PPI)网络;将所获得的交集靶点进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路分析,利用AutoDockTools软件对抑痰丸核心活性成分及HAP关键靶点进行分子对接。结果:获得抑痰丸化学成分146种,抑痰丸药物靶点1824个,HAP疾病相关靶点132个,抑痰丸-HAP共同靶点44个。抑痰丸主要活性成分包括麻黄碱、贝壳杉烯、胸腺嘧啶、羟基芜花素、泻根醇酸、贝母素等。治疗HAP的关键靶点包括白蛋白、肿瘤坏死因子(TNF)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、重组人趋化因子8、基质金属肽酶9(MMP9)、Toll样受体4、C反应蛋白(CRP)等,主要涉及调控细胞凋亡及增殖、抗炎和免疫等生物过程及炎症及免疫等通路。分子对接结果显示抑痰丸中麻黄碱与白细胞介素-10、白细胞介素-2、TNF、IL-6,胸腺嘧啶与前列腺素内过氧化物合成酶2(PTGS2)、CRP、IL-1β、TNF,黄芩素与MMP9、PTGS2,β谷甾醇与PTGS2均具有较好的结合能力。结论:抑痰丸通过多成分-多靶点-多通路治疗HAP,为更加深入研究抑痰丸的药效基础和实验研究提供理论依据。  相似文献

目的:运用网络药理学和分子对接技术探析养心活血通脉汤治疗稳定性冠心病的作用机制。方法:应用TCMSP、Herb、TCMID平台检索养心活血通脉汤成分及作用靶点;经GeneCards、OMIM、DisGeNET平台获取稳定性冠心病靶点;运用Cytoscape 3.9.1建立“药物-活性成分-靶点”网络及蛋白质-蛋白质相互作用(PPI)网络,筛选核心靶点。由Metascape平台进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析。利用Auto Dock Tools软件进行有效成分与和核心靶点分子对接。结果:养心活血通脉汤治疗稳定性冠心病主要成分为槲皮素、山柰酚、β-谷甾醇、芒柄花黄素、黄芩素、丹参酮ⅡA,关键靶点为肿瘤坏死因子(TNF)、白细胞介素-6(IL-6)、丝氨酸/苏氨酸激酶1(AKT1)、白细胞介素-1β(IL-1β)、血管内皮生长因子(VEGFA),关键通路为脂质和动脉粥样硬化(Lipid and atherosclerosis)、磷脂酰肌醇3激酶-丝氨酸/苏氨酸蛋白激酶1信号通路(PI3K-Akt信号通路)、肿瘤坏死因子信号通路(TNF信号通路)、丝裂原活化蛋白激酶信号通路(MAPK信号通路)、糖基化终产物/受体信号通路(AGE-RAGE信号通路)等。分子对接结果显示各有效成分与各核心靶点结合活性较好。结论:养心活血通脉汤治疗稳定性冠心病作用机制主要为降脂、抗动脉粥样硬化、抗炎、抗心肌损伤。  相似文献

目的:通过网络药理学方法结合分子对接技术研究半夏泻心汤治疗反流性食管炎(RE)的潜在有效成分及分子作用机制。方法:利用中药系统药理学数据库和分析平台(TCMSP)查找半夏泻心汤活性成分和药物潜在靶点。基于GeneCards、OMIM、DrugBank和TTD数据库获取RE疾病靶点,使用R语言4.0.4得到药物和疾病的交集靶点。采用Cytoscape 3.8.0分析软件筛选并构建“中药-活性成分-靶点”网络,结合STRING数据库对交集靶点进行蛋白互作网络分析,使用R软件进行GO功能富集分析和KEGG通路富集分析。利用AutoDock Vina 1.1.2对关键活性成分与作用靶点进行分子对接验证。结果:半夏泻心汤治疗RE共收集到162个活性化学成分和151个靶点,Cytoscape 3.8.0软件筛选出Jun激酶(JUN)、信号传导与转录激活因子3(STAT3)、MYC基因(MYC)、丝裂原活化蛋白激酶3(MAPK3)、肿瘤蛋白p53(TP53)、丝裂原活化蛋白激酶14(MAPK14)、蛋白激酶(AKT1)、丝裂原活化蛋白激酶8(MAPK8)、核转录因子-κB/p65(RELA)、丝裂原活化蛋白激酶1(MAPK1)、肿瘤坏死因子(TNF)、细胞周期蛋白D1(CCND1)等核心靶点和槲皮素、山柰酚、汉黄芩素、柚皮素、黄芩素等关键活性成分。GO富集条目2740个,包括2470个生物过程、84个细胞组成和186个分子功能;KEGG通路富集得到糖尿病并发症中的晚期糖基化终产物-晚期糖基化终产物受体、白细胞介素17、TNF、p53和缺氧诱导因子-1等信号通路。分子对接结果显示半夏泻心汤的关键活性成分可以对JUN、STAT3、MYC、MAPK3、TP53、MAPK14、AKT1、MAPK8等核心靶点产生调控作用。结论:半夏泻心汤可以通过多成分、多靶点、多通路治疗RE,为半夏泻心汤治疗RE作用机制的进一步阐明提供了指引。  相似文献

目的:基于网络药理学方法分析中医名方泻白散治疗小儿喘息性支气管炎的作用机制。方法:通过TCMSP数据库获取泻白散的有效成分及其作用蛋白-基因靶点,通过GeneCards、Drug Bank等数据库获取主要疾病靶点。利用STRING数据平台进行蛋白质-蛋白质相互作用(PPI)网络分析,运用Metascape数据平台进行基因本体(GO)功能与京都基因与基因组百科全书(KEGG)通路富集分析。采用Cytoscape 3.8.0软件建立“中药-共同靶点-主要通路”网络。结果:获得泻白散治疗小儿喘息性支气管炎的主要活性成分为山柰酚、槲皮素、甘草查尔酮A、柚皮素、18α-羟基甘草酸等;其核心靶点为丝裂原活化蛋白激酶 1(MAPK1)、丝裂原活化蛋白激酶 3(MAPK3)、核因子κ-B激酶亚基β抑制剂(IKBKB)、白细胞介素-6(IL-6)、肿瘤坏死因子(TNF)、丝裂原活化蛋白激酶8(MAPK8)、半胱天冬酶-3(CASP3)和丝裂原活化蛋白激酶14(MAPK14)等。通过分子对接验证,大部分靶点与化合物都有较好的结合效果。靶点富集通路包括白细胞介素-17信号通路(IL-17 signaling pathway)、肿瘤坏死因子信号通路(TNF signaling pathway)和缺氧诱导因子-1信号通路(HIF-1 signaling pathway)等诸多炎症通路。结论:泻白散可通过多靶点、多通路的互相作用,降低喘息性支气管炎的炎症风暴,并缓解气道痉挛。本研究结果为泻白散治疗小儿喘息性支气管炎提供了理论基础。  相似文献

目的:利用网络药理学探讨前痛定方治疗慢性前列腺炎(CP)的潜在作用机制。方法:从中药系统药理学分析平台(TCMSP)和中药化学数据库(Neosuite TCMID)收集前痛定方中的有效成分并预测作用靶点,与Genecards、OMIM、DrugBank网站所收集的CP潜在靶点进行相互交集后,筛选出前痛定方治疗CP的潜在靶点,然后利用Cytoscape分析软件(v3.7.2)及其插件Cyto NCA绘制“中药-有效成分-靶点”网络图并分析其核心成分,同时利用R语言进行GO分子功能和KEGG通路富集分析,最后在蛋白质互作网络分析平台STRING数据库构建靶蛋白质互作网络并分析其核心靶点。结果:前痛定方治疗CP的潜在靶点共124个,其核心成分为川陈皮素、黄柏酮酸、黄柏内酯、诺米林、壬基醇-10,核心靶点为磷脂酰肌醇-3-激酶催化亚基α、鸡肉瘤基因、丝裂原活化蛋白激酶1、Harvery鼠肉瘤基因、丝裂原活化蛋白激酶8等,相关通路涉及生长、增殖、血管生成、炎症、免疫防御和耐药性等,其中磷脂酰肌醇3激酶、白细胞介素-17、丝裂原活化蛋白激酶途径可能是其治疗的关键通路。结论:前痛定方治疗CP的作用机制具有多成分、多靶点、多通路的特点,为今后进一步探析前痛定方有效成分与体内靶点之间的作用机制提供了新的思路和理论基础。  相似文献

The New Zealand Waters Pollution Regulations 1963, referring to recreational water to which the public can gain access, states that the “coliform bacteria content shall not consistently exceed 1,000/100 ml”. This standard is now under revision, and the suggested new standard is a median faecal coliform index of 200/100 ml with a maximum value not exceeding 400/100 ml.

Recent literature supports the use of faecal coliform sub‐group bacteria as the best indicator of faecal pollution and reference is made to the membership of this sub‐group. Inherent inaccuracies of the Most Probable Number (MPN) method of assessing bacterial populations are pointed out, such as experimental variations with different cultural media, and possible errors in interpretation of results. In North America, there is evidence that an MPN for the total coliform group of 1000/100 ml indicates a faecal coliform sub‐group MPN of 200/100 ml, and epidemiological experience supports these values as standards.

We tabulate and discuss evidence that the usual ratio of faecal coliforms : coliform group bacteria is typically 1 : 1 in fresh and marine waters of the Wellington region, and that a more attainable standard would be 500 faecal coliforms/100 ml. In ratios quoted in this paper, ‘coliform group’ means the remainder of the coliform group excluding faecal coliforms. The World Health Organisation (1963) definition, of the entire group of coliforms, is not intended. The ratio is different in other parts of New Zealand; in Auckland, for example, the ratio is close to that found in the United States.  相似文献

Using the logarithmic wind profile and typical values of dimensionless sea roughness for different sea states, simple power expressions are derived for the wind profile over water. The power expressions are useful for correcting measured wind speed data at a given elevation (up to an elevation of 60 m) to the standard elevation of 10 m commonly used in coastal engineering studies. The analysis shows that the 1/7 power expression (suggested in [U.S. Army Corps of Engineers, 1984. Shore Protection Manual, Vol. 1. Coastal Engineering Research Center, Vicksburg, USA.] and commonly used today) is a good approximation (<3% error) of the logarithmic profile for various sea states up to an elevation of 20 m. Above this elevation, the 1/7 power expression underestimates the wind speed for moderate to fully developed sea states.  相似文献

This paper systematically reviews and synthesizes peer-reviewed, English-language scientific publications (n=212) to identify relevant research about how Ecosystem-Based Adaptation (EBA) is integrated with coastal planning. Preferred Reporting Items for Systematic reviews and Meta-Analysis (PRISMA) methodology is applied in this study. Attention was given to studies concerning human-environment interactions as opposed to physical or biological climate change issues alone because the coastal planning and EBA approach addresses the management of human actions in nature. The literature references include the issue of climate change (77%); however, limited evidence of EBA in coastal areas are reported (18%), and it is evident that the issues have become relevant in the scientific literature published in recent years. Broad texts demonstrate that SLR is one of the major long-term impacts (68%), and all of these papers recognize the most affected ecosystems in the tropics would be mangroves. EBA is an emerging option that can offset anticipated ecosystem losses and improve coastal planning to cope with SLR because it provides benefits beyond climate change stressors. There is a need to synthesize a road map for incorporation of mangrove regulations into local planning instruments and for building capacity for their implementation. Application of PRISMA in marine science will enhance future reviews, facilitate the systematic search and adequately document any theme, and also be useful in determining research gaps or information needs.  相似文献

This paper summarises research and options for sustainable sea transport in Oceania with a focus on domestic shipping. This debate is situated initially within the context of the current Pacific domestic shipping scenario, a region of minute economies connected by some of the longest sea transport routes in the world. All current options are fossil fuel powered and increasingly uneconomic and unsustainable. Many routes are marginal or unviable and a vicious cycle of old ships replaced with old ships prevails. Although a central and essential issue of many Pacific communities, the option of pursuing sustainable sea transport is currently invisible within the policy space at all levels. Various renewable energy options are possible and increasingly available. Recent research finds that these have strong potential for providing benefits across multiple wellbeings. The barriers to pursing this agenda are complex and poorly understood but are perceptual and institutional more than technological. A small number of critical experiments during the last oil crisis provide critical lessons and direction.  相似文献

Marine conservation actions are promoted to conserve natural values and support human wellbeing. Yet the quality of governance processes and the social consequences of some marine conservation initiatives have been the subject of critique and even human rights complaints. These types of governance and social issues may jeopardize the legitimacy of, support for and long-term effectiveness of marine conservation. Thus, we argue that a clearly articulated and comprehensive set of social standards - a code of conduct - is needed to guide marine conservation. In this paper, we draw on the results of an expert meeting and scoping review to present key principles that might be taken into account in a code of conduct, to propose a draft set of foundational elements for inclusion in a code of conduct, to discuss the benefits and challenges of such a document, and to propose next steps to develop and facilitate the uptake of a broadly applicable code of conduct within the marine conservation community. The objectives of developing such a code of conduct are to promote fair conservation governance and decision-making, socially just conservation actions and outcomes, and accountable conservation practitioners and organizations. The uptake and implementation of a code of conduct would enable marine conservation to be both socially acceptable and ecologically effective, thereby contributing to a truly sustainable ocean.  相似文献

计算避风锚地的面积是渔港规划设计的重要内容之一.<渔港总体设计规范>(SC/T 9010-2000)推荐采用确定性方法计算避风锚地面积.由于没有规定输入参数代表值的确定方法,实际应用时参数取值带有很大的随意性和经验性,从而导致计算结果的不确定性.引入Monte Carle方法建立了渔港避风锚地面积计算的随机模拟模型,采用不同的概率分布刻划各相关参数,从而获得避风锚地面积的概率分布.实例计算表明,无论采用何种概率分布假定,得到的避风锚地面积均近似地服从正态分布,且不同概率分布假定对95%保证率的避风锚地面积值影响不大;当船长、船宽均取各分组船型的最大值,水深取相应区域水深的平均值,每组渔船之间安全距离及每组渔船数量取规范给定的平均值时,规范方法计算的避风锚地面积保证率在95%左右,从而为规范方法输入参数的确定提供了依据.  相似文献
