山东近海褐牙鲆自然群体活样本共 79尾 ,分别于 1 996年 5月、1 997年 1月和 1 998年 4月采自青岛近海 ;养殖群体活样本 5 2尾于 1 997年 1 2月采自山东荣成寻山养鱼场。采用水平淀粉胶和垂直聚丙烯酰胺凝胶电泳方法及 3种缓冲系统 (TC、EBT和TG)分别对自然和养殖群体的 1 5种同工酶进行生化遗传分析。结果表明 ,山东近海牙鲆自然群体的多态基因座位比例 ( 31 .0 % )和群体平均杂合度 ( 0 .0 80 2 )都明显高于养殖群体 ( 2 4 .1 % ,0 .0 788) ;在自然群体的 9个多态基因座位、养殖群体的 7个多态基因座位中 ,除了Cat(P <0 .0 5 )和Idhp - 1(P <0 .0 5 ,养殖群体中 )有显著差异、Ldh -C(P <0 .0 1 )完全偏离Hardy -Weinberg定律外 ,其余多态座位基因频率均符合Hardy-Weinberg遗传平衡定律  相似文献

Measurements of sub-surface light attenuation (Kd), Secchi depth and suspended particulate material (SPM) were made at 382 locations in transitional, coastal and offshore waters around the United Kingdom (hereafter UK) between August 2004 and December 2005. Data were analysed statistically in relation to a marine water typology characterised by differences in tidal range, mixing and salinity. There was a strong statistically significant linear relationship between SPM and Kd for the full data set. We show that slightly better results are obtained by fitting separate models to data from transitional waters and coastal and offshore waters combined. These linear models were used to predict Kd from SPM. Using a statistic (D) to quantify the error of prediction of Kd from SPM, we found an overall prediction error rate of 23.1%. Statistically significant linear relationships were also evident between the log of Secchi depth and the log of Kd in waters around the UK. Again, statistically significant improvements were obtained by fitting separate models to estuarine and combined coastal/offshore data – however, the prediction error was improved only marginally, from 31.6% to 29.7%. Prediction was poor in transitional waters (D = 39.5%) but relatively good in coastal/offshore waters (D = 26.9%).SPM data were extracted from long term monitoring data sites held by the UK Environment Agency. The appropriate linear models (estuarine or combined coastal/offshore) were applied to the SPM data to obtain representative Kd values from estuarine, coastal and offshore sites. Estuarine waters typically had higher concentrations of SPM (8.2–73.8 mg l−1) compared to coastal waters (3.0–24.1 mg l−1) and offshore waters (9.3 mg l−1). The higher SPM values in estuarine waters corresponded to higher values of Kd (0.8–5.6 m−1). Water types that were identified by large tidal ranges and exposure typically had the highest Kd ranges in both estuarine and coastal waters. In terms of susceptibility to eutrophication, large macrotidal, well mixed estuarine waters, such as the Thames embayment and the Humber estuary were identified at least risk from eutrophic conditions due to light-limiting conditions of the water type.  相似文献

中国沿海不同地区泥蚶(Tegillarca granosa)的遗传多样性分析   总被引:1,自引:0,他引:1  
以采自海南海口、广西防城港、广西北海、广东湛江、福建漳州、山东荣成、山东威海7个地理群体62个泥蚶个体为材料,获取592bp线粒体COI基因片段序列,并进行遗传多样性及分化分析。多态性遗传参数统计显示:62个个体共检测出103个多态性位点,定义了26个单倍型;总群体单倍型多样性指数为0.834,核苷酸多样性指数为0.01665,平均核苷酸差异数为9.85772。7个群体均显示出较丰富的遗传多样性,群体内遗传距离及群体内遗传多样性参数显示中国沿海泥蚶遗传多样性由北向南呈升高趋势,而群体内遗传分化系数也呈现升高的趋势。基于26个单倍型COI序列构建的NJ树和UPGMA树以及基于群体间遗传距离构建的UPGMA树显示,荣成群体和威海群体亲缘关系较近,聚成一小支,而后与漳州群体相聚,然而南方类群并没有聚为独立的一支。  相似文献

1996年5月,1997年1月、12月和1998年4月在山东青荣成近海采集了131尾活牙鲆共710个生化样品,采用水平淀粉胶和垂直聚丙烯酰胺凝胶电泳的方法,对牙鲆的25种同工酶进行了分析。结果检测出LDH、MDH、MEP、IDHP、G3PDH、AK、CK、ACP、PGM、SDH、ADH、CAT、SOD、GDH和GPI等15处同工酶在牙鲆眼睛、肌肉、心脏、肝脏、胸鳍和肾脏等6种组织或器官听表达情况,  相似文献

根据2014年8月和10月、2015年2月和5月使用单船底拖网进行的4个航次调查数据,对山东半岛近岸海区的莱州湾及渤海南部、山东半岛北部和山东半岛南部等3个海域蟹类种类组成、时空分布、优势种、群落结构稳定性等群落结构特征进行了研究。结果显示,4次调查共捕获蟹类20种,隶属于10科17属,其中日本蟳(Charybdis japonica)和三疣梭子蟹(Portunus trituberculatus)是主要经济种类,其余均为小型饵料型蟹类;海区内主要的优势种为双斑蟳(Charybdis bimaculata)、日本蟳(Charybdis japonica)和三疣梭子蟹(Portunus trituberculatus),3个海域优势种季节更替均十分明显;莱州湾及渤海南部和山东半岛南部2个海域生物量季节变化明显,春季较低,在夏季达到峰值,夏季到冬季呈降低趋势,山东半岛北部海域生物量季节变化不明显。3个海域蟹类群落物种多样性指数(H')、丰富度指数(D)及均匀度指数(J')均处于较低水平,并有一定的季节变化;水深、水温和盐度是影响蟹类群落结构的重要因子,渤莱沿岸流、黄海暖流和黄海冷水团通过影响海域内水温与盐度影响蟹类群落结构。研究表明山东半岛近岸海区蟹类以小型饵料型蟹类为主,蟹类多样性水平较低,优势种更替明显,群落季节更替指数较高,季节间迁移频繁,蟹类群落不稳定。  相似文献

Phyllosoma larvae collected to date in Japanese and Taiwanese waters have been classified into two genera (Linuparus, Panulirus) of the Palinuridae, four genera (Ibacus, Parribacus, Scyllarides, Scyllarus) of the Scyllaridae, and one genus (Palinurellus) of the Synaxidae. However, phyllosoma larvae of three Scyllarus species (S. bicuspidatus, S. cultrifer, S. kitanoviriosus) are absolutely dominant among the larvae collected in the waters. Scyllarus larvae are abundant in coastal waters while those of Panulirus are often collected in offshore/oceanic waters. Based on previous and ongoing studies dealing with spatial distributions of phyllosoma larvae in Japanese and Taiwanese waters, it appears that phyllosoma and nisto larvae of the Scyllarus are retained within coastal waters north of the Kuroshio Current. On the other hand, the life history of the Panulirus (particularly P. japonicus) may be completed within the Kuroshio Subgyre: their phyllosoma larvae may be flushed out from coastal waters into the Kuroshio, then transported through the Counter Current south of the Kuroshio into the water east of Ryukyu Archipelago and Taiwan where they attain the subfinal/final phyllosoma or puerulus stages, once again entering the Kuroshio and dispersing into coastal waters. Lichia amia is an important coastal recreational fishery species with a cosmopolitan distribution in the eastern Atlantic. In southern Africa, it is distributed from southern Angola to northern KwaZulu-Natal in South Africa. A recent biological survey revealed differences between Angolan and South African individuals suggesting that they may represent separate stocks. As fishery management decisions should be based on accurate knowledge of population structure, an initial survey of the genetic substructuring of L. amia was conducted on samples collected in southern Angola and South Africa. Assessment of genetic diversity and population structure was conducted using a fragment of the mitochondrial Control Region. Obtained genetic diversity levels were within the expected range for marine teleosts (h = 0.867, π = 0.007), and two deeply divergent (southern Angolan and South African) populations were identified (average φ ST = 0.78) across the Benguela Current system. These results suggest that Angolan and South African L. amia should be managed as two independent stocks. This work represents the first assessment of population genetic substructuring across the Benguela Current of a valuable coastal fishery resource, and has application for future fisheries management.  相似文献

筛选5时可分析引物对斜带石斑鱼(Epinephelus coioides)台湾群体和广东群体的DNA多态性进行分析.根据电泳检测结果,平均每个引物获得4.2个等位基因,未发现群体的特异位点.获得2个群体平均观测杂合度(H_o)分别为0.672 1和0.682 6,平均期望杂合度(H_e)分别为0.647 9和0.544 4,两群体间的遗传分化系数(F_(ST))为0.011 0.统计结果表明,2个群体有着较高的遗传变异性,群体间的遗传分化程度处于多数海水鱼类的平均水平.  相似文献

应用聚丙烯酰胺凝胶电泳技术研究了牙鲆养殖群体(F1代)的遗传结构.结果表明,牙鲆养殖群体具有较高的遗传多样性水平.牙鲆群体平均每位点等位基因数的变化为1.19(0.11)~1.24(0.10).多态位点百分数的变化为14.29%~23.81%.期望杂合度的变化为0.056(0.036)~0.057(0.033).观察杂合度的变化为0.099(0.065)~0.104(0.065).牙鲆群体间遗传分化较低,群体间分化度为0.005,表明总的遗传变异中有99.5%来自群体内,有0.5%来自群体间.群体间遗传距离和遗传一致度均值分别为0.001和0.999.群体间基因流很大,等于49.75.  相似文献

采用微卫星DNA分子标记方法对中国南部沿海多鳞鱚(Sillago sihama)广西北海、广东湛江、汕尾及海南三亚4个野生地理群体进行了遗传多样性及遗传分化等研究。结果表明:(1)用于研究的34对微卫星引物在4个群体中具有丰富的多态性,有效等位基因数介于1.71—20.46之间,多态信息含量PIC介于0.40—0.95之间。4个群体总的平均期望杂合度(He)为0.598,观测杂合度(Ho)为0.572,SY群体的杂合度最大(He=0.606,Ho=0.623),SW群体的最小(He=0.586,Ho=0.508)。(2)群体间F-统计量分析表明,群体遗传分化处于高度分化水平(Fst平均值为0.306);基于奈氏遗传距离(DA)构建的NJ和UPGMA聚类树均显示地理位置相邻的群体聚在一起。  相似文献

The extreme environmental variability of coastal lagoons suggests that physical and ecological factors could contribute to the genetic divergence among populations occurring in lagoon and open‐coast environments. In this study we analysed the genetic variability of lagoon and marine samples of the sand goby, Pomatoschistus marmoratus (Risso, 1810) (Pisces: Gobiidae), on the SW Spain coast. A fragment of mitochondrial DNA control region (570 bp) was sequenced for 196 individuals collected in five localities: Lo Pagan, Los Urrutias and Playa Honda (Mar Menor coastal lagoon), and Veneziola and Mazarrón (Mediterranean Sea). The total haplotype diversity was h = 0.9424 ± 0.0229, and the total nucleotide diversity was π = 0.0108 ± 0.0058. Among‐sample genetic differentiation was not significant and small‐scale patterns in the distribution of haplotypes were not apparent. Gene flow and dispersal‐related life history traits may account for low genetic structure at a small spatial scale. The high genetic diversity found in P. marmoratus increases its potential to adapt to changing conditions of the Mar Menor coastal lagoon.  相似文献

Kingklip Genypterus capensis inhabit deep continental shelf and slope waters off southern Africa and are now largely harvested as a bycatch in trawl and longline fisheries for Cape hakes Merluccius spp. Regional differences in growth, vertebral count and otolith morphology indicate that kingklip may consist of three stocks. In this study, allozyme markers are used to test whether the two southern stocks, the putative Cape and South Coast stocks, are genetically distinguishable. Standard electrophoretic methods detected Mendelian polymorphisms at six (23%) of 26 enzyme-encoding loci in five samples extending from about 29°S on the West Coast to about 25°E on the South Coast. No deviations from Hardy-Weinberg proportions were observed in the samples or in the pooled sample. Gene diversities (heterozygosity) ranged from HS = 0.039 to 0.042 among samples, and averaged 0.041. Genetic differentiation among localities was remarkably small, FST = 0.003, and did not differ significantly from zero. A Bayesian analysis of population structure, based on detecting departures from Hardy-Weinberg proportions and gametic phase equilibria between loci, indicated with a high likelihood that the five samples were drawn from a single genetic population. In combination, these results are consistent with the occurrence of a single population off South Africa. However, the most conservative management strategy is to set harvest quotas individually on the Cape and South Coast stocks.  相似文献

利用18对微卫星引物分析了荣成湾群体1、荣成湾群体2、即墨群体和龙口群体共4个许氏平鲉(Sebastes schlegeli)野生群体的遗传多样性。结果表明:4个群体的平均有效等位基因数分别为3.1、2.7、2.8和2.7,平均期望杂合度分别为0.590、0.540、0.552和0.491,平均多态信息含量分别为0.577、0.495、0.538和0.528。4个许氏平鲉群体均表现出较高的遗传多样性水平,但也都存在杂合子缺失现象。Χ2检验显示,4个群体中大部分微卫星位点偏离Hardy-Weinberg平衡(P<0.05)。群体间Fst值为0.04,说明遗传变异主要存在于群体内,群体间的遗传分化程度较低。该研究结果将为许氏平鲉种质资源保护和合理利用以及人工增殖提供理论参考。  相似文献

Genetic studies on South African marine fishes have shown that many species exist as single, well-mixed stocks throughout their core distribution. The black musselcracker or poenskop Cymatoceps nasutus is a slow-growing, late-maturing and long-lived sparid (Perciformes: Sparidae) that is endemic to South African coastal waters. Conventional tagging studies suggest that it is resident, with limited connectivity between different regions along the coast. Mitochondrial and nuclear DNA were used to investigate the genetic structure of C. nasutus along the South African coastline and showed evidence of at least two populations. Samples collected from the Tsitsikamma Marine Protected Area on the South Coast to KwaZulu-Natal on the East Coast showed no significant population structure over much of the distribution. Gene flow for C. nasutus within this region is not limited by geographic distance and appears to be unaffected by various oceanographic barriers and biogeographical boundaries. Samples from the Western Cape to the west of Tsitsikamma, however, showed moderate to substantial differentiation for both markers, which may be influenced by a temperature barrier – a coldwater ridge – along the South Coast. These data provide the first genetic assessment of this South African sparid, and suggest that a two-stock management strategy would be appropriate for the species.  相似文献

麻痹性贝类毒素在我国近海污染问题已十分突出,基本呈现逐年加剧的趋势。20世纪90年代,南海麻痹性贝类毒素污染较重; 21世纪初,北黄海麻痹性贝类毒素污染较重;近几年,渤海和福建近海麻痹性贝类毒素污染较重;可产生麻痹性贝类毒素的微藻有亚历山大藻和裸甲藻等。采用小鼠生物法检测我国近海腹泻性贝类毒素超标率32%左右,采用液相色谱/质谱法检测,仅有3起超标的研究报道;现行小鼠生物法检测腹泻性贝类毒素假阳性问题十分突出,应尽快废除;腹泻性贝类毒素均是脂溶性的,脂溶性海洋生物毒素在我国近海常年可检出,偶有虾夷扇贝毒素和鳍藻毒素超标现象。可产生脂溶性毒素微藻有鳍藻和原甲藻及网状原角藻等。失忆性贝类毒素在我国近海常有检出,但无超标现象;产毒微藻有拟菱形藻等。西加鱼毒素在我国南海污染较重,但毒素标准物质的匮乏,限制了西加鱼毒素的调查研究;至今尚未确定产生西加鱼毒素的微藻。酶联免疫吸附法和液相色谱串联质谱法相结合已成为藻毒素快速准确检测成熟的技术,逐渐代替小鼠生物法和液相色谱法。  相似文献

The diatom genus Pseudo-nitzschia blooms throughout New Zealand's coastal waters. More than a decade of monitoring data were analysed in this study and seasonality was a significant factor in the appearance of Pseudo-nitzschia blooms. Austral spring and summer were the main bloom periods, although there were regional differences. Between January 2000 and August 2011, 8.4% of 29,000 seawater samples analysed for Pseudo-nitzschia exceeded the voluntary trigger level for biotoxin testing (10×104 cells l?1), and these were analysed by fluorescent in situ hybridisation (FISH) assays. Pseudo-nitzschia fraudulenta and P. pseudodelicatissima (low domoic acid [DA] producers) were the dominant bloom formers throughout New Zealand (each contributing 25%) followed by the highly toxic P. australis (10%). Shellfish flesh testing for DA was triggered on 8477 occasions between 2000 and 2011, but no samples exceeded the regulatory limit (20 mg/kg); the maximum concentration was 13 mg/kg (Marlborough Sounds, mid-winter 2010).  相似文献

Dissolved proteins in seawater samples collected from a coastal area of Tokyo Bay, Sagami Bay and a location off the Kuroshio Current were investigated by one-dimensional sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and high resolution two-dimensional electrophoresis (2-DE). Four to nine protein bands were detected in SDS-PAGE in the apparent molecular weight (MW) range from 12 kilo Dalton (kDa) to 49 kDa. The 2-DE technique distinguished 10 to 46 protein spots exhibiting isoelectric point (pI)/MW ranging 4.3–9.2/12–63 kDa. The elecrophoretic patterns were similar between the coastal and pelagic samples, as well as previously reported patterns from various pelagic areas. The close similarity of electrophoretic mobility on both SDS-PAGE and 2-DE gels indicates the compositional homogeneity of dissolved proteins in seawater throughout a broad range of marine environments. Proteinaceous dissolved organic matter (DOM) that was unresolved and smeared staining characteristics on both SDS-PAGE and 2-DE gels was first observed in Tokyo Bay waters in the present study and its possible sources are discussed. Although the two protein species, 48 kDa and 39 kDa proteins, have been identified as homologues of Porin P and low molecular weight-alkaline phosphatase of Pseudomonas aeruginosa PAO1, respectively, four strains of P. aeruginosa and two species of Pseudomonas spp. have been newly identified as the source organisms of these proteins using the N-terminal amino acid sequence data determined in previous studies.  相似文献
