用基因枪将GUS基因导入褐藻细胞中表达

于１９９３年１１月－１９９４年２月，用高压氦气式基因枪，将ＰＨ１２２１质粒装有ＧａＭＶ ３５Ｓ启动子，ＧＵＳ基因以及ｎｏｓ的３＇调控区导入海带和裙带菜组织切块中。４８ｈ后，在海带假根细胞和裙带菜中肋部叶片细胞中检测到ＧＵＳ基因的表达。实验表明，微粒子轰击法是外源基因导入大型褐藻的一个有效途径。ＧａＭＶ ３５Ｓ启动子能够驱动外源基因在海洋藻类中的表达，可作为藻类基因工程的启动元件。     

New and little‐known arietellidae (Copepoda: Calanoida) mainly from the south‐west pacific

Three species of Arietellidae are described from the south‐west Pacific: a new species of Paraugaptilus, a species which can not be separated from P. buchani, and Arietellus aculeatus. The identity of A. aculeatus is clarified by comparison of type material and material from the U.S. National Museum of Natural History, and an unidentified Atlantic species of Arietellus, evident from the comparison, is described.     

黄海绿潮浒苔提取物的化感效应及化感物质的分离鉴定

化感作用是浒苔绿潮暴发的重要原因之一,目前,对于浒苔产生化感物质的结构、种类以及产量的研究鲜有报道。本文针对黄海浒苔绿潮致灾藻种,利用毒性鉴别程序（TIE）对其体内的化感物质进行分离提取及结构鉴定,明确了浒苔产生化感物质的主要成分。首先用石油醚、乙酸乙酯和甲醇3种有机溶剂分别对新鲜浒苔进行提取,得到3种不同溶剂粗提物,其中乙酸乙酯粗提物的抑藻活性最强,对测试物种中肋骨条藻的半数有效浓度（96h-EC₅₀）值为22.25 mg/L;其次对乙酸乙酯粗提物进行萃取分离得到初步提纯物,其对中肋骨条藻的96h-EC₅₀值为21.12 mg/L;然后用硅胶层析柱对上述初步提纯物进行分离,共得到6种组分,其中抑藻活性最强的组分对中肋骨条藻的96h-EC₅₀值为10.57 mg/L;最后,对新鲜浒苔提取物用红外光谱和气相色谱-质谱联用仪进行检测,实验结果表明主要化感物质为:4,7,10,13,16,19-二十二碳六烯酸、2-十六碳烯酸、棕榈酸、5,8,11,14,17-二十碳五烯酸、9,12-十八碳二烯酸、9,12,15-十六碳三烯酸、花生四烯酸、13-二十二碳烯酸等8种脂肪酸。研究结果对于系统阐释浒苔绿潮暴发机制具有重要意义。

抗生素对浒苔形态及共附生细菌多样性的影响

本文研究了3种抗生素联合处理黄海绿潮浒苔(Ulva prolifera)形态及共附生细菌多样性的影响。将浒苔放散的配子加入到含有3种广谱抗生素(青霉素G、硫酸新霉素、多黏菌素B)的培养液中进行培养, 与对照组相比, 发现浒苔幼苗生长速度缓慢, 且细胞变小, 藻体无法形成正常形态。进一步采用16S rDNA高通量测序分析抗生素处理藻体前后的共附生细菌多样性, 发现抗生素处理后浒苔共附生细菌丰富度和多样性降低, 其中Maribacter丰度降低。将藻体与形态发生化合物thallusin(Maribacter spp. 提取物)共培养, 发现thallusin可促进藻体形态恢复和生长, 推测抗生素可通过抑制Maribacter等细菌从而影响浒苔形态和生长, 这将为浒苔共附生细菌功能的深入研究奠定一定基础。
