Flow cytometry (FCM) is now becoming a routine tool for the enumeration and optical characterization of bacteria in marine environments. We investigated the effects of sample fixation and storage upon flow cytometric determination of marine bacteria. Fixed and unfixed seawater samples were analyzed by FCM immediately aboard ship and/or later in the laboratory, and the appearances of the fluorescence signals and bacterial counts of these samples were compared. Fixation and storage led to the formation of multiple peaks in fluorescence histograms; this was also seen in 22 out of 36 samples frozen in liquid nitrogen. Fixation did not, but storage did induce a decrease of bacterial counts: a rapid decrease during the first 3 days followed by a slower decline. The decline of cell numbers in stored samples was expressed by a regression model. Our studies indicate that precaution is necessary when interpreting the data from fixed and/or stored marine bacterial samples analyzed by FCM. The possibility that the procedure of fixation and storage leads to the appearance of high DNA and low DNA bacterial groups should be considered.  相似文献

Abstract. Benthic samples were taken seasonally at 32 stations in five coastal lagoons of Central Italy (Fogliano, Monaci, Caprolace, Fondi, and Lungo) during a two-year period (1982–1984). Composition and distribution of benthic populations in each lagoon as well as seasonal trends of species richness, density, diversity, and redundancy are presented and discussed.
The faunal differentiation appeared to be mainly due to the different degree of water exchange, both with marine and continental systems.  相似文献

Abstract. Distribution patterns of epiphytic hydroids of seagrass ( Posidonia occanica and Cymodocea nodosa ) beds were studied in the 'Stagnone', a moderately hypcrhaline lagoon in Sicily. The role of water movement in determining the spatial and temporal zonation of epiphytic hydroids and their morpho-physiological variations is evaluated. Abstract. Seasonal variation of temperature effects on photosynthesis and respiration of Cymodorea nodosa (Ucria) Ascherson was investigated in a 1 m depth meadow in the Mar Menor lagoon (SE Spain). Light-saturated net photosynthesis (Pmax)and dark respiration (R) rates were linearly correlated with water temperature between 10 and 30°C. and 10 and 35°C, respectively, during the year. Water temperature had a similar effect on R rates in August, November, and February, but increased Pmax rates in August vs. those in February, thus promoting a shift of the maximum Pmax/R ratio values from 15°C in February to 30°C in August. P-I curves had higher Pmax values in May and August than in November or February, while Ic values remained relatively constant throughout the year. Whole-plant carbon balances suggested that C. nodosa might not be able to survive in the central basin of the lagoon and that the spatial distribution of this seagrass in the Mar Menor might be controlled by light availability.  相似文献

Abstract. Dark respiration rates, dissolved inorganic nitrogen (DIN) fluxes and nitrification rates were measured at two sites in the microtidal Sacca di Goro lagoon in September 2000. DIN fluxes correlated with the biomass of the dominant macrofauna species (the amphipod Corophium spp. at station Giralda and the polychaete Neanthes spp. at station Faro). Respiration (> 6 mmol O2 m−2h−1) and ammonium fluxes (> 80μmol N m−2h−1) were higher at station Giralda despite the lower organic matter content (4.5 %) and lower macrofauna biomass (4 g AFDW m−2). At both sites ammonium fluxes were significantly correlated with the biomass of the benthic infauna, but Corophium stimulated ammonium NH4+ fluxes 3-fold compared to Neanthes. The amphipod also enhanced nitrification rates (> 300 μmol N m−2h−1) due to the high density of its burrows, the higher NH4+ regeneration rates and the enhanced oxygen supply to the bacteria.  相似文献

Abstract. ω13C and ω15N of organic matter sources and consumers were employed to analyse trophic differentiation between a benthic consumer, Gobius niger (L., 1758) (Pisces, Osteichthyes), and a pelagic consumer, Atherina boyeri (Risso, 1810) (Pisces, Osteichthyes) in a Mediterranean coastal lagoon (Lake of Sabaudia) in winter and summer 1999. Trophic differences between the two species throughout the two sampling periods were related to the environmental trophic condition (i. e. nutrient and phytopigment concentrations). Although these two fish have different habitats, they both exploited benthic organisms, above all in summer. When the nutrient and phytopigment concentrations were higher (summer), the most abundant benthic organisms were the main food sources for both species. In winter A. boyeri apparently shifted its feeding preferences to include zooplankton, confirming its opportunistic feeding strategy. Par-ticulate organic matter and algae seemed to play the main trophic role in the food web structure. Benthic invertebrates used both sources, while zooplankton seemed to rely exclusively on the particulate fraction. The overlapping and very depleted signatures of sedimentary and particulate organic matter highlights a strong link between sediments and water column, rarely observed in other aquatic ecosystems using stable isotopes. Such a finding further substantiates the importance of particulate organic matter as a food source in the study area.  相似文献
