Effects of temperature, salinity and light intensity on growth rates of Gracilaria lichenoides and G. tenuistipitata var. liui Zhang et Xia were tested. Eight to ten levels of each factor were first tested separately. The best growth rate was obtained under the conditions of 32°C, 30 and 240 μmol/(m2·s) for G. lichenoides, and 24°C, 20 and 200 μmol/(m2·s) for G. tenuistipitata, respectively. Then a uniform design was used to evaluate the optimal combinations of the three factors. The best conditions for the highest daily specific growth rates (% increase in wet weight) are determined to be 31.30°C, 32.10, and 287.23 μmol/(m2·s) for G. lichenoides (16.26%/d), and 25.38°C, 21.10, and 229.07 μmol/(m2·s) for G. tenuistipitata (14.83%/d), respectively. A 4.34 liter two-stage air-lift photobioreactor incorporatingAnabaena variabilis ATCC29413 mutant PK84 was used to study H2 production. Results showed that H2 production increased with increasing light intensity from 47 μE/(m2·s) up to 190 μE/(m2·s), but that further increase of light intensity decreased the H2 production because of the inhibition due to the high pO2. The data also indicated that longer argon gas charge resulted in more H2 produced due to the increase of nitrogenase activities and heterocyst frequency, and that more than 1.3 L net H2 was produced from this computer controlled photobioreactor.  相似文献

During a survey for marine microalgal resources, we isolated a rare marine euglenoid from the coastal waters of Qingdao, China in 2009, and established a pure culture. Electron microscopic and molecular phylogenetic (18S rDNA and 16S rDNA sequences) analyses revealed a close affinity with Eutreptiella gymnastica, a bloom-forming species. Different culture conditions were monitored to understand optimal E. gymnastica growth characteristics. The optimal growth conditions in a batch culture of this isolate were 20°C, 160 μmol photons/(m2×s) of white light, and a salinity of 10-31. Nutrient experiments demonstrated that growth increased dramatically with a phosphorus concentration greater than 72 μmol/L. Understanding the effect of culture conditions on E. gymnastica may help understanding the blooming mechanism of this alga in its natural environment.  相似文献

Polysiphonia urceolata is one type of potential commercial red seaweeds used for breeding and cultivation, because of its significant biochemical and biomedical application. However, the information of breeding and seedling incubation for cultivation is limited, especially the early development. In this study, tetrasporohyte and gametophyte of P. urceolata were taken as the study materials in Huiquan Bay, Qingdao, China. The cleaned and sterilized tetrasporophytes and gametophytes were pre-cultured in sterilized seawater, then nurtured at 18°C, 25 μmol photons m−2 s−1 in 12:12 h (light:dark) photoperiod. Continuous observation under microscope showed that the early development consists of bipolar division stage and seedling stage. In the division stage, tetraspores germinate into bipolar sporelings that further differentiate into a colorless rhizoidal portion and a lightly pigmented upright shoot. The lightly pigmented rhizoidal cell develops to a rhizoid and the larger pigmented cell transforms to an erect axis. In the seedling stage, several quasi-protuberances appear on the erect axis and form juvenile seedlings. The results demonstrate the culture of P. urceolata from tetraspores under laboratory conditions. Acrochaete leptochaete (Huber) Nielsen (Chaetophoraceae, Chlorophyta) was isolated from the macroalgae Chaetomorpha collected from intertidal pools in Rongcheng, Shandong, China. 18S rDNA combined with ITS regions were used to ascertain the morphological identification of the isolated material. Based on the unialgal culture, asexual reproduction and growth characteristics of A. leptochaete were investigated over wide ranges of temperature and irradiance. Results revealed that asexual reproduction of A. leptochaete could be realized by biflagellate zoospores. The zoospores germinated directly to give self- replicating generations. Zoospore germination was bipolar. A temperature range from 13-21°C and a lower irradiance of 36 μmol/(m2·s) were most favorable for the growth of A. leptochaete. Thallus organization, an important taxonomic criterion for the genus Acrochaete, was affected markedly by temperature and irradiance. Our results extend the knowledge about the species’ general biology and its morphological plasticity. For classification and identification of a simple microphytic algae like A. leptochaete, which are traditionally placed in the class Chaetophoraceae, we propose that molecular tools associated with culture observations are applied.  相似文献

Gracilaria lemaneiformis Bory is an economically important alga that is primarily used for agar production. Although tetraspores are ideal seeds for the cultivation of G. lemaneiformis, the most popular culture method is currently based on vegetative fragments, which is labor-intensive and time-consuming. In this study, we optimized the conditions for tetraspore release and evaluated the photosynthetic activities of different colonies formed from the branches of G. lemaneiformis using a PAM (pulse-amplitude-modulated) measuring system. The results showed that variations in temperature and salinityhad significant effects on tetraspore yield. However, variations in the photon flux density (from 15 μmol m−2 s−1 to 480 μmol m−2 s−1) had no apparent effect on tetraspore yield. Moreover, the PAM-parameters Y(I), Y(II), ETR(I), ETR(II) and F v/F m of colonies formed from different branches showed the same trend: parameter values of first generation branches>second generation branches>third generation branches. These results suggest that the photosynthetic activities of different colonies of branches changed with the same trend. Furthermore, photosynthesis in G. lemaneiformis was found to be involved in vegetative reproduction and tetraspore formation. Finally, the first generation branches grew slowly, but accumulated organic compounds to form large numbers of tetraspores. Taken together, these results showed that the first generation branches are ideal materials for the release of tetraspores.  相似文献

The light demands of seaweeds is an interesting and rather complex phenomenon because they depend not only on the species but also on their different development stages. Even different parts of the same plant sometimes have different light demands. Light control is an important procedure at large scaleLaminaria nursery stations in China. Technicians and scientists have different viewpoints on the best method to regulate light. A culture study onLaminaria japonica starting from zoospores to several centimeter sporophytes to find the optimal and critical irradiance ranges for juvenileLaminaria at different development stages added more knowledge on this aspect. Experiment results show gametophytes can not tolerate irradiance of more than 150 μE m−2s−1 while sporophytes can tolerate more than 519 μE m−2s−1. This big difference starts from the very early stage of 1-to 2-celled sporophytes. The biological basis and mechanism of this phenomenon need further research. When cultured under certain environmental conditions (25°C, light intensity 80 μmol/m2·s, LD 12/12, in enriched seawater medium with 7×10−4 mol/L NO3−N, 1.56×10−4 mol/L, PO4−P and supplements of other elements like Mn, Fe, I, etc.), male and female gametophytes ofU. pinnatifida kept growing vegetatively and propagated fast at average daily fresh weight increase rate of about 20%. The empirical formulaG m=G o·3m was established to estimate the output of vegetative gametophytes. Vigorous vegetative gametophyte cells began to form reproductive structures (oogonium and spermatangium, when the temperature was lower than 25°C and other environmental factors were kept optimal. The sufficient supply of gametophyte cells provided enough seeds for raisingUndaria sporelings on production scale. Controlled cross-breeding experiments using selected male and female gametophyte clones which increase their cell number by mitosis instead of meiosis were also carried out in vitro. Juvenile sporophytes from the cross-breeding had almost the same length and width increase rates as those of the control. The fact that vegetative gametophytes can be purposely selected, propagated quickly, cross-bred, with the resulting sporophytes having almost the same characteristics leads to a new way to select desiredUndaria strains for long time use without losing the desired economic characteristics. Effect of temperature and irradiance on growth and reproduction of Enteromorpha prolifera that bloomed offshore along the Qingdao coast in summer 2008, was studied. It was showed that E. prolifera propagated mainly asexually with specific growth rate (SGR) of 10.47 at 25℃/40 μmol m^-2s^-1. Under this condition, gametes with two flagellate formed and released in 5 days. At the beginning of the development, the unicell gamete divided into two cells with heteropolarity, and then the apical cell developed into thalli primordial cells, whereas the basal cell developed into rhizoid primordial cells. In 8-day culture, the monoplast gamete developed into juvenile germling of 240 μm in length. Unreleased gametes can develop directly within the alga body. E. prolifera could either reproduce through lateral branching or fragmenting except apomixis revealed by Microscopic observation. On aged tissue of E. prolifera, although the degraded pigments partially remained in faded algal filaments, numerous vegetative cells could still divide actively in the algal tissues.  相似文献

The changes in arachidonic acid (AA) and fatty acids profiles along the growth curve ofParietochloris incisa, a coccoid snow green alga, were studied in a 2.8 cm light-path flat photobiorcactor, exposed to strong photon flux density [PFD, 2400 μEmol/(m2·s)]. Sixteen fatty acids were identified by gas chromatography showing that AA was the dominant fatty acid (33%–41%) followed by linoleic acid (17%–21%). AA content was closely investigated with respect to total fatty acids (TFA), ash free dry weight (AFDW) of cell mass as well as total culture content. These parameters were influenced significantly in a similar manner by culture growth phase, i.e., slightly decreasing in the lag period, gradually increasing in the logarithmic phase, becoming maximal at the early stationary phase, starting to decrease at the late stationary phase, sharply dropping at the decline phase. The increase in AA per culture volume during the logarithmic phase was not only associated with the increase in AFDW but also connected with a corresponding increase in AA/TFA, TFA/AFDW as well as AA/AFDW. The sharp decrease in AA content of the culture during the decline phase was mainly due to the decrease in AA/TFA, TFA/AFDW and AA/AFDW, although AFDW declined only a small extent. Maximal AA concentration, obtained at the early stationary phase, was 900 mg/L culture volume, and the average daily net increase of AA during 9 days logarithmic growth was 1.7 g/(m2·day). Therefore, harvesting prior to the decline phase in a batch culture, or at steady state in continuous culture mode seems best for high AA production. Acrochaete leptochaete,a species in Chaetophoraceae(Chlorophyta),was observed during our recent laboratory culture of the macroalgae Chaetomorpha that was originally collected from an intertidal pool in Rongcheng,Shandong Province,China.This is the first record of this species in China.Its morphology,taxonomy,and distribution were introduced and discussed in detail.Isolated culture experiments at different temperatures(9-29℃)and light intensities(36-108μmmol/m 2·s)were also carried out.The culture-based observations have extended our knowledge of growth morphology and general biology of the species.  相似文献

The energy budget ofBellamya earuginosa in a shallow algal lake, Houhu Lake (Wuhan, China) was investigated by the measurement of flesh production (32.8 kJ/(m2·a)), egestion (337.7 kJ/(m2·a)), metabolism (246.7 kJ/(m2·a)), and estimation of excretion (21.4kJ/(m2·a)). The net growth efficiency of the species is about 10.9%, which accords with the generally reported value for gastropods. In addition, the relationships between starvation respiration (R, mgO2/(Ind·d)), body weight (Wd, mg in dry wt) and temperature (T, °C) were also determined. The regression equationR=0.044Wd 0.537 e 0.061T was obtained by the least square method, The measured SDA of the species is 26.51% of its gross metabolism. Vertical profiles of chl-a and primary productivity in the middle continental shelf area and eddy area of the East China Sea were studied using data from a cruise in the East China Sea in February to March, 1997 and a cruise in July, 1998. The results showed that chl-a vertical distribution closely related to in situ hydrological and nutrient conditions. Chla-a concentration ranged from 0.22 to 0.35 mg/m3 and 0.93–1.09 mg/m3 in the eddy area and in the middle continental shelf area, respectively. In both areas, chl-a concentrations in deep layers were slightly higher than those in shallow layers, but was of the same order of magnitude. In summer, when a thermocline existed in the water column, highest chl-a concentrations appeared at the base of the thermocline layers in both areas. In the eddy area, chl-a concentration maximized at 31.743 mg/m3, and averaged 1.143 mg/m3 below 30 m depth. In the middle continental shelf area, the highest chl-a concentration was 2.120 mg/m3, the average was 1.168 mg/m3. The primary productivity reached 1418.76 mgC/(m2·d) in summer and 1360.69 mgC/(m2·d) in winter. In the eddy area, the primary productivity was 787.50 mgC/(m2·d) in summer and 159.04 mgC/(m2·d) in winter. Estimating carbon sequestration and nutrient accumulation rates in Northeast China are important to assess wetlands function as carbon sink buffering greenhouse gas increasing in North Asia. The objectives of this study were to estimate accreting rates of carbon and nutrients in typical temperate wetlands. Results indicated that average soil organic carbon(SOC), total nitrogen(TN) and total phosphorus(TP) contents were 37.81%, 1.59% and 0.08% in peatlands, 5.33%, 0.25% and 0.05% in marshes, 2.92%, 0.27% and 0.10% in marshy meadows, respectively. Chronologies reconstructed by 210 Pb in the present work were acceptable and reliable, and the average time to yield 0–40 cm depth sediment cores was 150 years. Average carbon sequestration rate(Carbonsq), nitrogen and phosphorus accumulation rates were 219.4 g C/(m~2·yr), 9.16 g N/(m~2·yr) and 0.46 g P/(m~2·yr) for peatland; 57.13 g C/(m~2·yr), 5.42 g N/(m~2·yr) and 2.16 g P/(m~2·yr) for marshy meadow; 78.35 g C/(m~2·yr), 8.70 g N/(m~2·yr) and 0.71 g P/(m2·yr) for marshy; respectively. Positive relations existed between Carbonsq with nitrogen and precipitations, indicating that Carbonsq might be strengthened in future climate scenarios.  相似文献

The adsorption/desorption of phosphorus across the sediment-water interface is the main mechanism that controls the buffering capacity of DRP in coastal and estuarine water. Determination by three processes revealed that: 1) DRP flux from early diagenesis in surface sediment isF p Dia =31.9μg/(m2·d), which is only 1/1600 of the ammonia released; 2) at normal temperature and in the long term, the TDP released by adsorption/desorption across the sediment-water interface is a function of the major environmental factors; and 3) based on the determined SODR and assumed average water depthH(m) and residence timet R (d), the maximum flux of DRP can be estimated. Chondrus is a type of commercially produced red seaweed that widely used for food and carrageen extraction. Although the natural life history of the alga had been well understood, the factors influencing development of the tetraspore and carpospore remain poorly understood. In the perspective of seedling resources, the regulation of early development is crucial for the seedling nursing; therefore, it is necessary to understand the physiological influences during its early development. In this study, we studied the effects of temperature and irradiance on the early development of Chondrus ocellatus Holm under laboratory conditions. The released tetraspores and carpospores were cultivated at different temperatures (10–28°C) and irradiances (10, 60 μmol photons m−2s−1) with a photoperiod of 12L:12D. The results indicate that both tetraspores and carpospores are tolerant to temperatures of 10–25°C, and have the highest relative growth rate at 20°C. Irradiance variances influenced the growth of the discoid crusts, and the influence was more significant with increasing temperature; 60 μmol photons m−2s−1 was more suitable than 10 μmol photons m−2s−1. The optimum temperature and irradiance for the development of seedlings was 20°C and 60 μmol photons m−2s−1, respectively.  相似文献

Costaria costata (C. Agardh) Saunders is one of common kelps distributed in many coastal areas worldwide; however, in China, no reports have been made on cultivation of the genus. To investigate potential cultivation of the species in the northern part of China, trials on isolation and preservation of the gametophytes were conducted using C. costata from Korea; growth and development of the gametophytes were observed. We showed that at 10±1°C, 60 μmol m−2s−1 and 12:12 h (L:D), freshly released zoospores settled down within 1 hour, and then developed into the primary cell during the following 2 days. After a vegetative growth phase lasting 6–8 days, female gametophytes became 3–4 times larger in diameter than that of the primary cell, but still remained at a unicellular stage, while male gametophytes divided into 4–10 cells with only a slight change in size. Fertilization occurred within 10 days after the zoospores were released from the sporangia, and the apical and basal tissues of the juvenile sporophyte divided and differentiated into the blade and stipe. Temperature and irradiance influenced gametophytic vegetative growth and developmental patterns. Generally, low irradiance (15 μmol m−2s−1 and 30 μmol m−2s−1) was unfavorable to the induction of fertility, but it enhanced female gametophyte division. The optimal conditions for vegetative growth were 15°C and 30 μmol m−2s−1. After transplantation of the juvenile seedlings and after eight months cultivation, the harvested mature blade reached 194 cm in length and 32.7 cm in width. Our study proves that it is feasible to implement propagation and large scale cultivation of C. costata in northern China.  相似文献
