The purpose of this study was to compare enzymatic biomarker activities in fish caged at two sites, Masan Bay (contaminated) and Haeguemgang (reference). In the present study, ethoxyresorufin O-deethylase (EROD), brain acetyl cholinesterase (bAChE), muscle acetyl cholinesterase (mAChE) and butyryl cholinesterase (mBChE) in caged rockfish (Sebastes schlegeli) were measured 0, 1, 3, 7, 14, 21 and 30 days after caging. The level of CYP1A mRNA and Protein expression was induced higher in Masan Bay at 1, 3, 7, 14 and 30 days after caging. EROD activity in the caged fish was significantly higher in Masan Bay than in Haeguemgang 3 and 7 days after caging, but not at 14 and 30 days after caging. bAChE activity was significantly inhibited at 7 and 14 days after caging in Masan Bay. However, mBChE activity was not significantly inhibited during the experiment. Taken together, the data suggest that the caged fish were exposed, at least transiently, to CYP1A inducers and ChE inhibitors, which is consistent with our previous observations.  相似文献

Biochemical markers and ovarian histology were investigated in prespawning females of grass goby (Zosterisessor ophiocephalus) and grey mullet (Mugil cephalus) collected, respectively, in late spring and summer 2000 in four sites of a highly eutrophic brackish ecosystem of central Italy, the Orbetello Lagoon. Exposure to chlorinated and aromatic hydrocarbons was evaluated in fish livers by the somatic liver index (SLI) and by measuring 7-ethoxyresorufin-O-deethylase (EROD) and benzo(a)pyrene monooxygenase (BaPMO) activities. Acetylcholinesterase (AChE) activity was measured in brain and gills to evaluate exposure to organophosphates (OPs) and carbamates (CBs). The gonad somatic index (GSI) was used to confirm ovarian maturation and ovarian histology was investigated as a potential biomarker for environmental effects. Samples from the Western Basin, near a sewage treatment plant (STP) off the town of Orbetello, showed higher SLI values and higher EROD and BaPMO activities than those collected from the Ansedonia Canal (AC) in the Eastern Basin (p<0.05) and respect to those from reference sites: the Albegna River (AR) Delta for grass goby and the Nassa Canal (NC), connected with the sea, for grey mullet both located in the Western Basin as well. Low brain AChE activity was observed in both species from the reference sites (AR and NC) in association with the presence of anomalies in developing oocytes: unexpectedly small in grass goby and irregular disintegrated cytoplasm in grey mullet. The results indicate that the Western Basin is more polluted than the Eastern Basin particularly in the Orbetello where the sewage treatment plant may be a source of aromatic and chlorinated compounds while the Albegna River and the Nassa Canal may be sources of OPs and CBs.  相似文献

A battery of biochemical biomarkers and the SigmaPCB concentration in adult female perch (Perca fluviatilis) verified an aquatic pollution gradient with the city of Stockholm (Sweden) as a point source of anthropogenic substances. The investigation included both an upstream gradient, 46 km westwards through Lake M?laren, and a downstream gradient, 84 km eastwards through the Stockholm archipelago. Besides the main gradient from Stockholm, there were strong indications of pollution coming from the Baltic Sea. The results indicated a severe pollution situation in central Stockholm, with poor health status of the perch, characterised by increased specific EROD activity in the liver, increased liver EROD somatic index, decreased AChE activity in the muscle, increased amount of DNA adducts in the liver, and a high concentration of biliary 1-pyrenol. In addition, laboratory exposure to common EROD inducers elicited an abnormal response, suggestive of chronic intoxication.  相似文献

The current study was designed to validate the biomarkers of sewage pollution in Mozambique Tilapia (Tilapia mossambica, Peters) reared in sewage treatment plant (STP) effluent in Ras Al Khaimah, United Arab Emerates, before and following depuration/detoxification. Cellular biomarkers, cholinesterase activity using acetylcholine as a substrate (acetylcholinesterase AChE) and reduced glutathione (GSH) and hepatosomatic index (HSI) were investigated in fresh water fish, Tilapia, raised in a fish farm (Group I/Clean, as Control), treated sewage water/TSW (Group II/Sewage) and thereafter exposed to fresh water in an aquarium for 6 weeks (Group III/Depurated) for depuration. The results showed significantly lower levels of AChE activities in liver (26% p < 0.01) and muscle (30% p < 0.01) of the fish reared in the STP water (Group II/Sewage) as compared to those recorded in the fish from fish farm (Group I/Clean). The depressed AChE level was fully restored in the muscle but partially in the liver after depuration (Group III/Depurated). In contrast, GSH levels were significantly raised in both liver (1.3-fold p < 0.01) and muscle (4-fold) of Group II fish as compared to Group I (control) fish raised in fish farm and following depuration in fresh water (Group III/Depurated) elevated GSH level in liver restored to control values, while remained unchanged in muscle. The average hepatosomatic index (HSI = weight of liver × 100/total fish weight), an indicator of hepatomegaly, in the Group II fish reared in TSW was also significantly higher than that in the reference Group I fish, but decreased to control level in Group III fish following depuration. This study suggests the importance of cellular biomarkers, AChE, GSH and hepatosomatic index in monitoring the impact of sewage water pollution on fish caused by a complex mixture of chemico-biological contaminants and its mitigation following depuration, an effective mean of fish detoxification.  相似文献

In order to evaluate the health status of fish inhabiting Port Phillip Bay, Australia, southern sand flathead (Platycephalus hassensis, N = 133) were collected at six stations throughout the Bay. Fish had a similar serum sorbitol dehydrogenase (SDH) activity level (p = 0.12), indicating that they were not experiencing hepatocellular injuries. Ethoxyresorufin-O-deethylase (EROD) activity was generally lower in the non-urbanized and non-industrialized southern part of the Bay. The highest EROD activity was observed in Hobson Bay, the closest station from Melbourne city. Naphthalene-type biliary metabolites were also highest in Hobson Bay with intermediate levels found in Corio Bay where refineries are present. An opposite trend was observed with the pyrene-type bile metabolites, the highest levels being observed in Corio Bay while intermediate levels were found in Hobson Bay. The ratio of naphthalene-type to benzo(a)pyrene (B(a)P)-type metabolites indicate that relatively to other sites sampled in Port Phillip Bay, Corio Bay is subjected to enriched petroleum hydrocarbons of pyrolytic origin. Temporal trends indicate that the availability of xenobiotics to fish remained unchanged over the 1990s.  相似文献

In the heavily industrialized Masan Bay of southern coast, Korea, the potential harmful effects of heavy metals (Cd, Co, Cu, Ni, Pb, Sn, Zn, and Hg) were evaluated in terms of the pollution load index (PLI) and ecological risk assessment index (ERI) methods, and the results obtained were considered alongside the health of the macrobenthic fauna communities. The results revealed that the bay sediments, especially in the inner bay and the outfall area of a sewage treatment plant, are exposed to moderate to serious levels of metal pollution. Hg and Cd contributed the most to the potential toxicity response indices in sediments recently deposited in the bay. The potential ecological risk assessment of heavy metals in the bay was highlighted by the use of the benthic biological pollution index (BPI), suggesting that the ERI is a useful toxicity response index, which can quantify the overall ecological risk level to a target environment.  相似文献

Baseline information is presented on embryo malformation rate and biomarkers in fish as indicators of sub-lethal stress caused by pollution in coastal waters of Xiamen, PR China. Fish and eggs were sampled from several areas in Xiamen coastal waters (Xiamen Harbour, Maluan and Tongan Bays and East Channel), where varying levels of pollutant input have been documented. Comparative sampling was done at a "cleaner" reference site at Dongshan Island. Embryonic malformation rates, which indicate general water quality, varied with location and species of fish, and exceeded background levels for unpolluted waters (assumed approximately 5%) by up to eightfold at some sites. Generally, sites around Xiamen Harbour show signs of poor water quality having highest mean levels of embryo deformity (20-30%) and these decreased towards open waters (Tongan Bay, Eastern Channel) where abnormalities approached background levels. An indication that toxic contaminants may be having a localised effect in the region, particularly in the harbour was reinforced by the biomarker assays. However, activities of the biomarkers ethoxyresorufin O-deethylase (EROD) and glutathione S-transferase in fish livers indicate no clear pattern, and there is no evidence that fish from the four sampling areas have been more or less exposed to PAHs and other compounds that induce these biomarkers. Antioxidant biomarkers (glutathione peroxidase, catalase, superoxide dismutase, and reduced glutathione) suggest that exposure to xenobiotics appears to be lowest in Dongshan and Maluan and highest in the harbour and Tongan. Inhibition of acetylcholinesterase in fish muscle indicated possible effects by organophosphate and carbamate pesticides in Xiamen waters and these effects may be greatest in the area of the harbour.  相似文献

A suite of biomarkers were measured in barramundi (Lates calcarifer) from five North Queensland estuaries along a perceived pollution gradient. The biomarkers selected were 7-ethoxyresorufin-O-deethylase (EROD), cytochrome P450, fluorescent aromatic compounds (FACs), DNA integrity, RNA:DNA ratio, cholinesterase activity (ChE), condition factor and hepatosomatic index. The resulting database was subjected to uni- and multi-variate analyses in order to assess the most suitable biomarkers to assess pollution in North Queensland estuaries and to classify the environmental quality of the sites. Principal components analysis (PCA) on the biochemical markers revealed that EROD, EROD/P450, DNA damage and to a lesser extent ChE and FACs were found to be responsive to contaminants in the environment while cytochrome P450, condition factor and the hepatosomatic index were found to be less responsive biomarkers. This study has demonstrated the utility of applying a multibiomarker approach in conjunction with traditional analysis of contaminants in providing valuable information in environmental risk assessment.  相似文献

Horizontal and vertical distributions of organochlorine compounds (OCs) were determined in sediments from Masan Bay. The concentrations of polychlorinated biphenyls (PCBs), dichlorodiphenyltrichloroethanes (DDTs), HCB, hexachlorocyclohexanes (HCHs) and chlordane related compounds (CHLs) in sediments were in the range of 1.24-41.4, 0.28-89.2, 0.02-0.59, nd-1.03, and nd-2.56 ng/g, respectively. The spatial distribution of OCs showed a negative gradient from the inner of the bay to outer part of the bay, indicating that the source of OCs was probably located inside the bay. Compositional pattern of PCB congeners showed a relatively high concentration of high-chlorinated congeners in the inner part of the bay and a relatively low concentration of low-chlorinated congeners in the outer part. In sediment core from Masan Bay maximum concentrations of PCBs and DDTs are observed in the subsurface samples and correspond to an age of early 1980s and late 1960s. The concentration profiles of PCBs and DDTs in sediments of Masan Bay appear to correspond to use of PCBs and DDTs in Korea.  相似文献

The aims of this study were to evaluate levels and effects of trace metals (Cd, Cr, Cu, Fe, Mn, Pb, Zn), PAHs and PCBs in mussels and fish native to the Lagoon of Venice. To this end, bioaccumulation and various biomarkers of generic stress and exposure (malondialdehyde, MDA and metallothioneins, MT; ethoxyresorufin O-deethylase activity, EROD; fluorescent bile metabolites and somatic indices in fish; lysosomal membrane stability, LMS; lipofuscins, LF; neutral lipids, NL and survival-in-air, SOS in mussels) were measured in organisms seasonally collected from two differently influenced areas.In mussels, metal bioaccumulation levels at the two sites were low and fluctuated similarly, exhibiting decreasing levels in summer, like MDA and MT. Micro-organic pollutants and the probably related biological response of LMS did not show either significant site differences or seasonal trends. LF and NL revealed inconclusive patterns. The results of the survival-in-air test were quite erratic, showing that they were associated with the high variability of both natural and physiological parameters temperature, food, reproduction, and body reserve cycle.In fish, both inorganic and organic micropollutant levels were low, and differences were detected between sites only for PCBs. The pattern exhibited by micro-organic contaminants was clearly related to the reproductive cycle at both sites; a relationship with PCBs, EROD and MDA was established at the site where their levels were generally higher. There is a lack of information on monitoring neurotoxicity in offshore commercial species. To help fill this gap, we sampled hake (Merluccius merluccius) and Norway lobster (Nephrops norvegicus) in fishing grounds off the coast of l’Ametlla de Mar (NW Mediterranean) in June 2005 at a depth of 100 m and 400 m. Additionally, at 400 m depth, two other fish species, Micromesistius poutassou and Phycis blennoides were included. Neurotoxicity markers such as Colinesterases (ChEs), namely acethyl- (AChE), butyryl- (BChE), propionyl- (PrChE) and carboxilesterase (CbE) were measured in muscle. Lipid peroxidation (LP), a marker of oxidative damage, was also included. The results are discussed in relation to the animal’s sex, size and fishing depth. A comparison of esterases and LP levels between muscle and liver of hake and between muscle and hepatopancreas of Norway Lobster was made.

AChE was dominant in muscle and CbE in hepatopancreas. No differences between fish species were seen for AChE. However, N. norvegicus, presented lower levels of ChEs and LP. A size-dependence in ChEs was seen for M. merluccius, with larger animals showing significantly lower activities (p < 0.05). Sex-dependence was seen in N. norvegicus for most esterases, except AChE, with males displaying higher activities (p < 0.05). A sampling-depth effect was also seen in the crustacea, with animals from 100 m generally presenting lower esterase activities and higher LP levels.  相似文献

Hepatic microsomal 7-ethoxyresorufin O-deethylase (EROD) activities (indicative of exposure to polycyclic aromatic hydrocarbons (PAHs) and polychlorobiphenyls (PCBs)) were measured in eel Anguilla anguilla from the Thames Estuary. Fish were collected from up to 13 sites during November 1997, May and August 1998 and October 1999. Throughout this period no clear seasonal variation could be identified at every site along the Thames. However, during the summer months, fish sampled from sites in the middle to the upper estuary (Woolwich, Greenhithe and West Thurrock) reported up to 3-fold higher EROD activities compared to sites either at the upper reaches (Richmond and Brentford) at the same time of the year, or fish sampled in winter, along the entire length of the estuary. A laboratory exposure experiment demonstrated a 3-fold elevation of EROD activity 2 days after injection with β-naphthoflavone (β-NF). However, higher levels of activity could be determined in fish sampled from the Weston canal near the Merseyside. The lowest levels of A. anguilla EROD activity were observed in fish sampled from the upper reaches of the River Tamar, Devon, and were comparable to activities determined in fish from the Wear and Humber estuaries. A. anguilla sampled along the Thames, Tyne and Tees estuaries reported between 2.5- and 7-fold higher EROD activities compared to fish collected from the Tamar. These results indicate that a low to moderate induction of A. anguilla CYP1A had occurred (indicative of low to moderate exposure to PAHs and planar PCBs) in fish collected from the Thames, Tyne, Wear, Tees, Humber and Tamar estuaries. However, the highest level of EROD activity was observed in fish from the Weston Canal (Merseyside).  相似文献

Habitat Suitability (HS) models have been extensively used by conservation planners to estimate the spatial distribution of threatened species and of species of commercial interest. In this work we compare three HS models for the estimation of commercial yield potential and the identification of suitable sites for Tapes philippinarum rearing in the Sacca di Goro lagoon (Italy) on the basis of six environmental factors. The habitat suitability index (HSI) is based on expert opinion while the habitat suitability conditional (HSC) is calibrated on observational data. The habitat suitability mixed (HSM) model is a two-part model combining expert knowledge and regression analysis: the first component of the model uses logistic regression to identify the areas in which clams are likely to be present; the second part applies the same parameter-specific suitability functions of the HSI model only in the areas previously identified as productive by the logistic component.The HS models were validated on an independent data set and estimates of potential yield of the Goro lagoon were compared. The effectiveness of the three approaches is then discussed in terms of predicted yield and identification of suitable sites for farming.  相似文献

A variety of sources of organic contaminants to the Great Barrier Reef lagoon and near-shore environment exist including boating activity, agriculture and urban run-off. Cytochrome P-450 1A activity as measured by ethoxy-resorufin O-deethylase (EROD) activity has been widely used as an indicator of the exposure of fish to organic contaminants such as polychlorinated biphenyls (PCB), polycyclic aromatic hydrocarbons (PAH) and some organochlorine pesticides. This study demonstrates the successful application of EROD measurements in a common Australian tropical estuarine fish species, Acanthopagrus berda (Pikey Bream), to identify areas under potential stress from organic contaminants. Fish were captured from four creeks draining agricultural land, a creek draining urban land and two creeks with less disturbed catchments. Significant induction of cytochrome P450-1A was observed in fish captured from Ross Creek (urban catchment, 7.4-fold) and Cromarty Creek (agricultural catchment, 6.4-fold). Increased activity was also observed in fish captured from other creeks draining agricultural land (Plantation Creek, Victoria Creek, Seymour River, 1.9–2.6-fold) as compared to those captured from creeks in undisturbed catchments (Baldy Creek, Fisher Creek, 67–114 pmol/min/mg protein).  相似文献
