The effects of various salinity fluctuation amplitudes (2, 4, 6 and 8) on the growth, osmolarity, Na+-K+-ATPase activity and Hsp70 of juvenile Fenneropenaeus chinensis cultured in seawater with a salinity of 20 were studied. The results show that weight gain in the salinity fluctuation treatments was better than that in control; in particular, the weight gain of treatments S4 and S0, at 231.8% and 196.3%, respectively, was significantly different (P<0.05). The hemolymph osmolarity of treatments S0, S2, S4, S6 and S8 was 635.4, 630.8, 623.6, 614.4 and 600.3 mOsm/kg, respectively, and decreased with increasing salinity fluctuation amplitude. The level of Na+-K+-ATPase activity in gills of F. chinensis was higher than that in hepatopancreas, but there were no significant differences among all treatments, either in gills or hepatopancreas (P>0.05). The relative level of Hsp70 in treatment S4 was 48.4% and 40.4% higher than control in muscle and eyestalks, respectively, with the highest values being recorded under a salinity fluctuation amplitude of 4.  相似文献

The effects of environmental Na+/K+ ratio on the gill ion-transport enzyme activity, plasma osmolality and growth of Cynoglossus semilaevis juveniles were investigated. The results showed that, plasma osmolality was similar among flounder adapted to different Na+/K+ ratios of saline groundwaters (P>0.05), while the growth, gill Na+, K+-ATPase and HCO3 -ATPase activities were affected by Na+/K+ ratio significantly (P<0.05). The gill Na+, K+-ATPase activity reached its maximum on day 3, then decreased gradually from day 3 to day 9 and remained constant from day 9 to day 15. The peak values of gill Na+, K+-ATPase activity were detected on day 3 for all Na+/K+ ratios of saline groundwaters, then the enzyme activities descended, and on day 9 the enzyme activities achieved steady state, and the gill HCO3 -ATPase activity increased rapidly and achieved steady state after one day. During steady state, the gill Na+, K+-ATPase and HCO3 -ATPase activity of Na+/K+ ratios 20 and 40 treatments were significantly higher than those in the control group (Na+/K+ ratio 27.5), while there were no significant differences between the Na+/K+ ratio 30 treatment and the control group; the gill Na+, K+-ATPase activity of Na+/K+ ratio 20 and 40 treatments were significantly higher than that for ratio 30 treatment, but there were no significant differences of gill HCO3 -ATPase activity among these treatments. At the end of the 15-day experiment, the weight gain (%) and specific growth rate (SGR) of flounders maintained in seawater were significantly higher than those in groundwaters; significant differences also occurred among the treatments; Na+/K+ ratio 30 treatment had the highest values (33.7% and 1.94 respectively), which were significantly higher than those under Na+/K+ ratios 20 and 40 treatments. Therefore, for the saline groundwater used in this experiment, it is suggested that the Na+/K+ ratio be adjusted to approximately 30, i.e., as close to that of natural seawater as possible in the culture of flounder.  相似文献

This experiment was conducted to examine the effects of salinity fluctuation frequency on the growth, molting rate and hemolymph 20-hydroxyecdysone (20-HE) concentration for juvenile Fenneropenaeus chinensis with an initial wet body weight of 1.115 g±0.012 g. The salinity of the control treatment (represented by D0) was 28 throughout the experiment, whereas treatments D2, D4, D6 and D8 were subjected to different salinity fluctuation frequencies of 2, 4, 6 and 8 d, respectively. The range of salinity fluctuation was 4. The salinity in treatments D2, D4, D6 and D8 was kept at 28 for 2, 4, 6 and 8 d, respectively; each original salinity was decreased abruptly to salinity 24, which first lasted for another 2 d and was then raised to its initial value 28. This constituted a salinity fluctuation cycle and afterwards the cycle repeated. It was found that: 1) The weight gain of shrimp in treatment D4 was 202.9% and significantly higher than that in treatment D0 (P<0.05). The molting rate in treatment D2 was the lowest, while that in treatment D6 was the highest and significantly higher than those in treatments D0 and D2 (P<0.05). 2) The hemolymph 20-HE concentration of shrimps in all treatments was at low levels and increased first gradually during the inter-molt period and then increased sharply. It reached a peak value during the pre-molt stage, and then decreased abruptly during the post-molt stage when the lowest level occurred. 3) During the post-molt stage, the hemolymph 20-HE concentration in treatment D2 was 7.47 pg μL−1 and significantly higher than that in treatment D0 (P<0.05). There were, however, no significant differences in hemolymph 20-HE concentration among all five treatments during the inter-molt stage (P>0.05). During the pre-molt stage, the hemolymph 20-HE concentration in treatment D6 was significantly higher than that in treatment D0 (P<0.05), whereas the hemolymph 20-HE concentration in treatment D2 was significantly lower than that in other treatments (P<0.05). Adequate salinity fluctuation promoted the molting rate through increasing hemolymph 20-HE concentration at the pre-molt stage.  相似文献

The ion-transport enzyme activities were studied in the nauplii 1–2, zoea 1–3, mysis 1–3, and postlarva 1–7 of the shrimps Marsupenaeus japonicus and Fenneropenaeus chinensis. The results showed that total ATPase, Na+-K+-ATPase, and V-ATPase (V-type H+-ATPase) activities increased during the early development for both the species, from zero in nauplii to stable levels after zoea stage. The enzyme activities of the latter species were significantly higher than those of the former species after zoea stage (F>F 0.05). The contributions of Na+-K+-ATPase, V-ATPase and HCO3 -ATPase to the total ATPase activity of the two species varied in different developmental stages and accounted for 40%–70%, 22%–46% and 2%–13% in M. japonicus from zoea to postlarva stage, whilst the shares of them were 42%–69%, 28%–44% and 2.5%–22%, respectively in F.chinensis. These findings suggest the possible culture of the two species with varying water quality, especially the salinity and pH.  相似文献

Improvement in the osmoregulation capacity via nutritional supplies is vitally important in shrimp aquaculture.The effects of dietary protein levels on the osmoregulation capacity of the Pacific white shrimp(L.vannamei) were investigated.This involved an examination of growth performance,glutamate dehydrogenase(GDH) and Na+-K+ ATPase mRNA expression,,and GDH activity in muscles and gills.Three experimental diets were formulated,containing 25%,40%,and 50% dietary protein,and fed to the shrimp at a salinity of 25.After 20 days,no significant difference was observed in weight gain,though GDH and Na+-K+ ATPase gene expression and GDH activity increased with higher dietary protein levels.Subsequently,shrimp fed diets with 25% and 50% dietary protein were transferred into tanks with salinities of 38 and 5,respectively,and sampled at weeks 1 and 2.Shrimp fed with 40% protein at 25 in salinity(optimal conditions) were used as a control.Regardless of the salinities,shrimp fed with 50% dietary protein had significantly higher growth performance than other diets;no significant differences were found in comparison with the control.Shrimp fed with 25% dietary protein and maintained at salinities of 38 and 5 had significantly lower weight gain values after 2 weeks.Ambient salinity change also stimulated the hepatosomatic index,which increased in the first week and then recovered to a relatively normal level,as in the control,after 2 weeks.These findings indicate that in white shrimp,the specific protein nutrient and energy demands related to ambient salinity change are associated with protein metabolism.Increased dietary protein level could improve the osmoregulation capacity of L.vannamei with more energy resources allocated to GDH activity and expression.  相似文献

The capability ofScenedesmus obliquus to remove cations (K^+, Na^+, Ca^2+, Mg^2+) from saline- alkaline water was investigated at different salinities (0, 5, 10, 15, 20, 25) and carbonate alkalinities (0, 5, 10, 15, 20, 25, 30, 35 mmol/L). K^+, Na^+, Ca^2+, and Mg^2+ in saline-alkaline water were efficiently removed by S. obliquus. The maximum removal of the cations (29.37 mg for K^+, 185.85 mg for Na^+, 23.07 mg for Ca^2+, 66.14 mg for Mg^2+) occurred at salinity 25. The maximum removal of K^+ (2.28 mg), Na+ (6.62 mg), Ca^2+ (1.01 mg), and Mg2+ (0.62 mg) occurred at carbonate alkalinities of 25 mmol/L for K*, 35 mmol/L for Na+, 20 mmol/L for Ca2+, and 25 mmol/L for Mf+, respectively. Under a salinity stress, the concentration of Na' in S. obliquus increased significantly, while that of K~ decreased significantly. The concentrations of Ca^2+ and Mg2+ decreased as well. The ratios of K+/Na~, Ca2+/Na^+, and Mg^2+/Na^+ were significantly lower in all salinity treatments than those of the control. Under alkaline stress, the concentrations of Nan and K+ in S. obliquus decreased significantly and the ratios of K^+/Na^+, Ca2+/Na^+, and Mg^2+/Na^+ were significantly higher in all treatments than in the control. Moreover, the concentrations of Ca2+ and Mg2+ in S. obliquus at alkalinities of 5-10 mmol/L were significantly higher than those of the other treatments. The removal of Na+ by S. obliquus mainly occurs through biosorption, and Mg^2+ and Ca^2 + were removed through both biosorption and bioaccumulation.  相似文献

We evaluated the ability of juvenile Amur sturgeon (Acipenser schrenckii) to osmoregulate and grow in saltwater. Hatchery-reared juveniles (mean weight 106.8 g, 5-month old) were transferred from freshwater to 10, 20, and 25 salinity saltwater over a period of 20 d. We measured the growth, serum osmolality, ion concentrations, and Na+/K+-ATPase activity. In addition, we prepared samples of gill tissue to quantify morphological changes in gill ultrastructure. Rearing in up to 25 saltwater for 30 d had no significant effect on growth. Similarly, serum osmolality and ion concentrations were similar to levels reported in other teleosts following acclimation to saltwater. Serum osmolality and Na+, Cl concentrations increased significantly with the initial increase in salinity. Afterwards, levels tended to stabilize and then decrease. Serum K+ levels did not change during acclimation to saltwater. Gill Na+/K+-ATPase activity increased initially as salinity was increased. However, the activity later decreased and, finally stabilized at 3.7±0.1 μmol Pi/mg·prot·h in 25 saltwater (1.6 times higher than the level in those in freshwater). In fish that were held only in freshwater, the chloride cells were located in the interlamellar regions of the filament and at the base of the lamella. Following acclimation to 25 saltwater for 30 d, the number and size of chloride cells increased significantly. Our results suggest that juvenile Amur sturgeon is able to tolerate, and grow in, relatively high concentrations of saltwater.  相似文献

To improve the expression efficiency of exogenous genes in Chlamydomonas reinhardtii, a high efficient expression vector was constructed. Green fluorescent protein (GFP) was expressed in C. reinhardtii under the control of promoters: RBCS2 and HSP70A-RBCS2. Efficiency of transformation and expression were compared between two transgenic algae: RBCS2 mediated strain Tran-Ⅰ and HSP70A-RBCS2 mediated strain Tran-Ⅱ. Results show that HSP70A-RBCS2 could improve greatly the transformation efficiency by approximately eightfold of RBCS2, and the expression efficiency of GFP in Tran-Ⅱ was at least double of that in Tran-Ⅰ. In addition, a threefold increase of GFP in Tran-Ⅱ was induced by heat shock at 40℃. All of the results demonstrated that HSP70A-RBCS2 was more efficient than RBCS2 in expressing exogenous gene in C. reinhardtii.  相似文献

The effects of several nutritional factors on the growth and eicosapentaenoic acid (EPA) production of diatom Nitzschia laevis were studied. 4 LDM (quadrupled concentration of the nutrient salt) was the optimal concentration of nutrient salt for the growth and EPA production of N. laevis. The growth of N. laevis was inhibited when the glucose concentration was either lower than 10 gL−1 or higher than 15 gL−1. Both sodium nitrate and urea were good nitrogen sources for the growth and EPA production, while ammonium chloride seriously decreased the dry cell weight (DW) and the EPA content. Silicate seriously influenced the growth of N. laevis. The maximum DW of 2.34 gL−1 was obtained in the presence of 150 mgL−1 Na2SiO3·9H2O. The EPA content remained almost the same when the silicate concentration was lower than 150 mgL−1; however, higher silicate concentrations resulted in a steady decrease of EPA content. Low medium salinity (⩽29) did not seem to influence the DW of N. laevis, and high salinity resulted in a decrease of DW. The highest EPA content (4.08%) and yield (110 mgL−1) were observed at the salinity of 36 and 29, respectively.  相似文献

Heavy metal pollution can affect the immune capability of organisms.We evaluated the effect of cadmium(Cd) on the defense responses of the Pacific oyster Crassostrea gigas to Listonella anguillarum challenge.The activities of several important defensive enzymes,including superoxide dismutase(SOD),glutathione peroxidase(GPx),acid phosphatase(ACP),Na+,K+-ATPase in gills and hepatopancreas,and phenoloxidase-like(POL) enzyme in hemolymph were assayed.In addition,the expression levels of several genes,including heat shock protein 90(HSP90),metallothionein(MT),and bactericidal/permeability increasing(BPI) protein were quantified by fluorescent quantitative PCR.The enzyme activities of SOD,ACP,POL,and GPx in hepatopancreas,and the expression of HSP90 were down-regulated,whereas GPx activity in the gill,Na+,K+-ATPase activities in both tissues,and MT expression was increased in Cdexposed oysters post L.anguillarum challenge.However,BPI expression was not significantly altered by co-stress of L.anguillarum infection and cadmium exposure.Our results suggest that cadmium exposure alters the oysters’ immune responses and energy metabolism following vibrio infection.  相似文献

The unicellular cyanobacterium Synechocystis sp. PCC 6803, a model organism known for its unique combination of highly desirable molecular genetic, physiological and morphological characteristics, was employed in the present study. The species was cultured in BG11 liquid medium contained various initial concentrations of Pb2+ and Cd2+ (0, 0.5, 1, 2, 4, 6 and 8 mg/L). The experiment was conducted for six days and the metal induced alterations in the ultrastructure, growth and pigment contents were assessed. Alterations in the ultrastructure of the Synechocystis sp. PCC 6803 cells became evident with the increased (>4 mg/L Pb2+) metal concentration. The photosynthetic apparatus (thylakoid membranes) were found to be the worst affected. Deteriorated or completely destroyed thylakoid membranes have made large empty spaces in the cell interior. In addition, at the highest concentration (8 mg/L Pb2+), the polyphosphate granules became more prominent both in size and number. Despite the initial slight stimulations (0.2, 3.8 and 6.5% respectively at 0.5, 1 and 2 mg/L Pb2+), both metals inhibited the growth in a dose-dependent manner as incubation progressed. Pigment contents (chlorophyll α, β carotene and phycocyanin) were also decreased with increasing metal concentration. Cells exposed to 6 mg/L Pb2+, resulted in 36.56, 37.39 and 29.34% reductions of chlorophyll α, β carotene and phycocyanin respectively over the control. Corresponding reductions for the same Cd2+concentrations were 57.83, 48.94 and 56.90%. Lethal concentration (96 h LC50) values (3.47 mg/L Cd2+ and 12.11 mg/L Pb2+) indicated that Synechocystis sp. With increasing demand in China for the mitten crab larvae, understanding its survival mechanism gets more important. This research focused on the effects of temperature and Ca^2 on the larval growth and development. Eriocheir sinensis larvae were reared in laboratory under 21 different combinations of temperature (15, 20, 25℃) and Ca^2 content (120, 130, 140, 150, 160, 170, 180mg/l) and constant salinity (20) and pH (8), The results suggested that the survival rate increases with temperature and Ca^2 content. These combinations of temperature and Ca2 content maximized survival rate inour study and it may be the optimum water environmental conditions for culturing the larvae. To predict surviving larvae number under different water environmental conditions, 21 dynamic mathematical models were developed. This for the first time observation of the zoeal Ⅵ larvae of the Changjiang River E.sinensis population showed that they occurred under stressed water environmental conditions: temperature of 15℃ and Ca^2 content of 120,130mg/l.  相似文献

A laboratory experiment was conducted to assess the bioaccumulation of Pb2+ and its effects on growth, morphology and pigment contents of Spirulina (Arthrospira) platensis. The specimen cultured in Zarrouk liquid medium was treated with various initial metal concentrations (0, 5, 10, 30, 50 and 100 μg mL−1). The growth of S. platensis was adversely affected by Pb2+ at high concentrations (30, 50 and 100 μg mL−1). However, at low concentrations (5 μg mL−1), Pb2+ could stimulate its growth slightly. The pigment contents (chlorophyll α and β carotene) were decreased in a dose-dependent manner. The highest reductions (67% and 53% respectively in chlorophyll α and β carotene) were observed in 100 μg mL−1 treatment group. The LC50 (96 h) of Pb2+ was measured as 75.34 μg mL−1. Apart from a few cases of filament breakages at elevated concentrations (50 and 100 μg mL−1), morphological abnormalities are not specific. Metal bioaccumulation increased with Pb2+ concentrations, but decreased with exposure time. The maximum accumulated amount was 188 mg g−1 dry weight. The bioconcentration factor (BCF) reached to a peak at day 2, followed by a gradual reduction for all the exposure concentrations. S. platensis is able to tolerate considerably high Pb2+ concentrations. Consequently it can be used as a potential species to remove heavy metal from contaminated waters.  相似文献

Fe^2+ acted as an accessorial factor for many cellular enzymatic reactions is very important for seaweed growth and development, but the Fe^2+ requirement in nori had not been seen. Porphyra yezoensis cells were separated enzymatically and cultured in a series of sterilized seawater media containing various concentrations of Fe^2+. The growth development and cell were investigated in this work. Through this experiment, two biologically-meant concentration scales were found, one is low concentrations, 12.1-102.1μg/L, 10-100 times than that in seawater, favoring the development of isolated cells of Porphyra and the other was high concentrations, more than 10mg/L inhibiting the cell growth, leading to the deformity and shrinkage of the cells. At the concentration of 50 mg/L, the cells stopped growing and died eventually.  相似文献

3-factor experiment was used to study the combined effects of temperature, irradiance and salinity on the growth of an HAB species diatomSkeletonema costatum (Grev.) Cleve. The results showed that temperature (12, 19, 25, 32 °C), irradiance (0.02, 0.08, 0.3, 1.6)×1016 quanta/(s·cm2)) and salinity (10, 18, 25, 30, 35) significantly influenced the growth of this species. There were interactive effects between any two of and among all three physical factors on the growth. In the experiment, the most optimal growth condition forS. costatum was temperature of 25°C, salinity of 18–35 and irradiance of 1.6×1016 quanta/(s·cm2). The results indicatedS. costatum could divide at higher rate and were more likely to bloom under high temperature and high illumination from spring to fall. It was able to distribute widely in ocean and estuary due to its adaptation to a wide range of salinities. This study on dynamic changes of culture color,astaxanthin and chlorophylls,inorganic N including N-NO3^-,N-NO2^- and N-NH4^ in batch culture of Haematococcus pluvialis exposed to different additive nitrate concentration showed(1)ast/chl ratio was over 0.8 for brown and red algae,but was usually less than 0.5 for green and yellow algae;(2)N-NO3^-,in general,was unstable and decreased,except for a small unexpected increase in nitrate enriched treatment groups;(3)measurable amounts of N-NO2^- and N-NH4^ were observed respectively with three change modes although no extemal nitrite and ammonia were added into the culture;(4)a non-linear correlation between ast/chl ratio(or color)changes and the levels of N-NO3^-,N-NO2^-,N-NH4^ in H.pluvialis culture;(5)up and down variation of the ast/chl ratio occurred simultaneously with a perceptible color change from yellow to brown(or red)when N-NO3^-,N-NO2^- and N-NH4^ fluctuated around 30,5,5μmol/L respectively;(6)existence of three dynamic modes of N-NO3^-,N-NO2^- and N-NH4^ changes,obviously associated with initial extemal nitrate;(7)the key level of total inorganic N concentration regulating the above physiological changes during indoor cultivation was about 50 μmol/L;and(8)0.5-10mmol/L of nitrate was theoretically conducive to cell growth in batch culture.  相似文献

This paper reports the approprite ranges of Mg^2 ,Ca^2 and their ratio Mg^2 /Ca^2 in mixed seawater for rearing of Penaeus japonicus larvae,The ranges for the above three indices are 1150-1450mg/L,360-440mg/L and 2.8-3.4,respectively,the proper salinity range of mixed seawater is 22.1-33.9 obtained by mixing estuarine water and concentrated seawater.  相似文献
