The purpose of these studies was to evaluate if there were relationships between lysosomal destabilization or glutathione concentrations and gamete viability of oysters, Crassostrea virginica. Oysters were collected from field sites during the peak spawning period (May-June) during 2001 and 2002. Lysosomal destabilization rates and glutathione concentrations of hepatopancreas tissues (e.g. digestive gland) were determined. Eggs and sperm from the same adults were also used to conduct embryo development assays with reference seawater collected from a clean site, site water, and also a range of Cd concentrations (the Cd exposures were used to determine if there were differences in susceptibility to pollutants). Baseline embryo development success (e.g. percent normal development when the assays were conducted with reference seawater or site water) was related to lysosomal destabilization, but not glutathione status. However, the susceptibility of embryos to metal toxicity was related to glutathione status, i.e., sensitivity to Cd exposures increased with decreasing glutathione levels. These studies support the hypotheses that there are linkages between embryo development success and susceptibility to pollutant stress and cellular biomarker responses. These kinds of effects on reproductive success could lead to subtle but significant long-term effects on recruitment and viability of oyster populations.  相似文献

Nanoparticles may be introduced into aquatic environments during production processes and also as a result of release following their use in various commercial formulations and biologic applications. Filter-feeding bivalve mollusks such as oysters are valuable model species for characterizing nanoparticle bioavailability and interactions with basic cellular processes. The adults release their gametes into the environment, so their embryos and larvae are also likely targets of nanoparticles. The purpose of these studies was to characterize the toxicity of metal nanoparticles on embryonic development of oysters, Crassostrea virginica and to compare the relative sensitivity of embryos to adults. Newly-fertilized oyster embryos were exposed to silver nanoparticles (AgNP) and then the percent normal development after 48 h was assessed. Studies were conducted with adult oysters in which they were also exposed to AgNP for 48 h, and the effects on lysosomal destabilization were determined. The expression of metallothionein (MT) gene expression was also assessed in both embryos and adults. Adverse effects on embryonic development were observed at concentrations similar to those that caused both statistically and biologically significant effects on lysosomal destabilization of adults. Significant increases in MT mRNA levels were observed in both embryos and adult oysters, and MT levels were highly induced in embryos. While we do not know whether the toxicity and gene expression responses observed in this study were due to the nanoparticles themselves or the Ag ions that dissociated from the nanoparticles, these kinds of basic studies are essential for addressing the potential impacts of nanoengineered particles on fundamental cellular processes as well as aquatic organisms.  相似文献

Elevation of metal concentrations in coastal environments associated with anthropogenic enrichment pose a significant threat to estuarine organisms. The purpose of this study was to evaluate the relationships between cellular responses that may be potentially valuable as indicators of chronic stress and metal-contaminated sediments. For these studies, hatchery-reared juvenile oysters were deployed in situ at 15 sites for approximately 1 month around Charleston Harbor, SC. The effects on lysosomal destabilization and glutathione concentrations were determined; and the relationships between the cellular responses and sediment metal concentrations were described. Both single metal and multiple metal parameters (based on total metal concentrations, aluminum normalizations, and summed sediment quality guidelines) were considered. Generally, significant correlations were observed for individual metal analytes and multiple metal parameters. Since many of the individual metal analytes covary, the responses may reflect overall contaminant loading rather than responses to individual metals. Methods for estimating overall contaminant loading based on multiple analytes provide a more realistic estimate of potential adverse effects.  相似文献

In order to study Zn and Cd accumulation and depuration, a set of oysters, Crassostrea rhizophorae, were transplanted to a metal contaminated coastal lagoon and another one was harvested there and transplanted to a non-polluted site. C. rhizophorae oysters and Perna perna mussels native from both sites were collected in order to monitor variability of metal concentration in resident populations. After three months exposure, oysters transplanted to the polluted site accumulated fourfold Zn (307-1319 microgg(-1)) without reaching the concentration level of resident oysters (9770 microgg(-1)). Cadmium concentrations had a slight but significant decrease during the same period (1.25-0.54 microgg(-1)). Oysters transplanted to the non-polluted site, showed threefold Zn depuration (6727-2404 microgg(-1)), while Cd had no significant variation (0.90-1.45 microgg(-1)). Results showed that transplanted oysters do not reach heavy metal concentrations in indigenous populations suggesting transplanted organisms would be better used to evaluate bioavailability instead of environmental concentrations.  相似文献

The effect of climate change on natural oyster recruitment has the potential to disrupt many of the ecosystem services oysters provide. Due to the temperature‐sensitivity of reproduction, oyster recruitment may shift as water temperatures rise. A biological imprint of climate change was revealed in a multi‐decadal time series of recruitment of non‐native Pacific oysters (Crassostrea gigas) in the main stem of Hood Canal, Washington, USA, extracted from historic fishery documents. Water in July and August warmed significantly from 1945 to 1995 (0.028 ± 0.004°C per year [±SE]) and accounted for an increase in Pacific oyster recruitment (7% per year, 0.028 ± 0.006 spat per year on log scale [±SE]); recruitment also strongly tracked inter‐annual variability in summer water temperature. Methods used to collect historical data were repeated in 2013–2015 when recruitment of both Pacific oysters and native Olympia oysters (Ostrea lurida) were recorded in main stem and lower Hood Canal. Both historic and modern data show large variation within and between years for temperature as well as recruitment. The modern data add information regarding spatial variation, in that recruitment patterns in the two regions of Hood Canal were decoupled. As temperatures continue to increase, non‐native Pacific oysters are likely to be favored over Olympia oysters, which recruit earlier at lower temperatures and presently contribute less than half of total oyster recruits. Future recruitment, however, may be limited by environmental factors other than temperature, a point indicated particularly in Hood Canal where many subtidal species already respond strongly to gradients in dissolved oxygen.  相似文献

Phytoplankton acclimate to low irradiance by increasing their cellular demand for Fe, to allow synthesis of additional light-harvesting pigments and Fe-containing redox proteins involved in photosynthesis. In the open NE subarctic Pacific, Fe concentrations limit primary productivity and irradiances may be suboptimal, particularly during winter. Phytoplankton thus may be unable to fulfill their extra Fe requirements for growth under these low-light conditions and become effectively co-limited. We tested this hypothesis by manipulating Fe and light in in vitro experiments at OSP (Ocean Station PAPA, 50°N 145°W) during winter 1997. The results show that metabolic rates, growth, and photosynthetic parameters of phytoplankton are enhanced in winter by increasing either irradiance or Fe. The greatest response occurs when Fe and light are amended concomitantly, confirming that the community is indeed co-limited by both resources. Analysis of environmental conditions (i.e. incident irradiance, mixed layer depth and Fe concentrations) in winter at OSP reveals that they are similar to those observed in the austral spring and fall at three sites in the Southern Ocean. Extrapolating our experimental field results to the Southern Ocean illustrates that co-limitation by light and Fe also may play an important role in regulating phytoplankton growth in this region.  相似文献

Temporal patterns of larval brooding and settlement were investigated in a flat oyster (Ostrea chilensis) population in Tasman Bay, central New Zealand. The proportion of the population brooding larvae and larval settlement rates were monitored over 26 months. A peak period of brooding activity began in late spring and continued through summer. Maximum rates of 17% and 23% of adult oysters brooding larvae occurred in November and December, and an estimated 55–78% of adult oysters incubated larvae over the entire summer breeding period. These proportions of brooders are higher than those previously reported for Tasman Bay. A very low level of brooding activity (1%) occurred during winter. Temporal trends in larval settlement closely tracked brooding patterns. Settlement was greatest between November and January, and there were very low rates in winter. The seasonal breeding pattern in the population was intermediate between northern and southern populations, confirming a latitudinal gradient of reproductive behaviour for O. chilensis in New Zealand. Results are useful in optimising the timing of substrate deployment in an enhancement programme for the oyster fishery.  相似文献

Culture of pearl oysters is rapidly increasing worldwide, including the western Indian Ocean. The oyster Pinctada margaritifera L., which produces the most highly valued black pearls, occurs in East Africa, a nd as been exploited there for the shell for many decades. The growth patterns of P. margaritifera from a natural population in the sheltered back-reef, and fro oysters translocated to a tidal current-swept site, both sites within Gazi Bay, Kenya, are described. The growth rate in the natural population ranged from 31.3mm year?1 (60–65mm size-class) to 7.6mm year?1 (105–110mm size-class). The von Bertalanffy growth coefficient (K), calculated with a fixed L of 127.2mm, was 0.30 for the natural population and 0.38 for the translocated oysters. The mean growth rate during the north-east monsoon season was approximately double that for during the south-east monsoon season. The dailyrate of nacre deposition ranged from 1.3μm to 5.9μm (mean 3.45μm); it declined with the size of oysters and was marginally higher at the high-energy current site. At that rate, it would take approximately two years to produce a marketable cultured half pearl with a 2.5mm layer of nacre. The results of the study are relevant to the understanding of the influence of the environment on growth, and are applicable to the optimisation of growth rate of pearl oysters in the inshore region along the east coast of Africa.  相似文献

Pacific oysters (Crassostrea gigas Thunberg 1793) have been introduced into the Wadden Sea (North Sea), where they settle on native mussel beds (Mytilus edulis L.), which represent the only extensive insular hard substrata in this soft-sediment environment. As abundances of C. gigas rose, some mussel beds became increasingly overgrown with oysters, whereas others did not. Field experiments revealed that recruitment of C. gigas was higher in the lower intertidal than in the upper subtidal zone, that it was higher on conspecifics than on mussels, and that it was not affected by barnacle epigrowth except when settling on mussels. Mussel recruitment is known from inter- and subtidal zones. It occurred equally on oyster and mussel shells but showed a clear preference for barnacle epigrowth over clean shells. Assuming that settlement and recruitment are key processes for species abundances on the North Sea coast, it is predicted that the positive feedback in oyster settlement will lead to rapid reef formation of this invader at the expense of mussel beds. Mussels, however, may escape competitive exclusion by settling between or on the larger oysters especially when barnacles are abundant. Experimental patches with mussels were more often covered by fucoid algae (Fucus vesiculosus forma mytili Nienburg) than patches with oysters, and oyster recruitment was poor underneath such algal canopies. Thus, fucoids may provide the native mussels with a refuge from the invading oysters and the two bivalves may coexist, provided food is not limiting.  相似文献

We collected polychaete diversity and abundance data at a range of impacted and reference sites near an alumina refinery in Melville Bay, northern Australia. The aims were to measure the impact of sediment modified by the alumina refinery discharge on polychaete communities and secondly to gather baseline data from which to measure future changes. Polychaete communities in both soft-bottom habitats and subtidal areas adjacent to mangrove forests were studied. We also developed and deployed an artificial substratum device to sample polychaetes associated with hard-substrate habitats. For each habitat, polychaete community composition was different between impacted and reference sites and at multiple time points. The impact of future changes either from bioremediation or management practices can be measured against these baseline data. Indicator species analysis was used to identify polychaete species that were significantly different at the locations tested, and we discuss their potential as indicator species.  相似文献

Expanding industrial activities in the Arctic require an urgent assessment of the toxicity of chemicals at low temperatures. Organisms acclimatized to low temperature exhibit specific adaptations. For example, the amount of unsaturated lipids is increased to maintain the fluidity of the cell membranes. It has been hypothesized that such temperature-induced alterations in membrane lipid composition may affect the stability of lysosomal and cell membranes in the common mussel, Mytilus edulis, an organism exposed to seasonal temperature extremes. As mussels may be exposed to petroleum compounds along industrialized coastlines, we tested the combined effects of exposure to low temperature and the petroleum compound, phenanthrene, on haemocyte membrane stability. Test animals, acclimated to either 0 or 10 degrees C, were exposed to phenanthrene (0 = control or 500 micrograms l-1) and haemocytes were examined using the neutral red retention assay (lysosomal stability) and a fluorescence assay (cell membrane stability). At 0 degree C, lysosomal and cell membranes from uncontaminated mussels were destabilized compared with 10 degrees C (P = 0.0005). No significant effects (P > 0.05) of phenanthrene were detected at either temperature. Possible mechanisms underlying membrane destabilization include a weaker physical resistance of the membrane due to a higher amount of unsaturated lipids, a potentially higher level of reactive oxygen radicals at low temperature and the higher susceptibility of unsaturated lipids to oxidative stress. More work is required to better understand the consequences of this membrane destabilization at low temperature on the susceptibility of the organism to pollutants.  相似文献

A field study was carried out in the Lagoon of Venice (north-east Italy) with the aim of evaluating the potential use of lysosomal destabilisation as a biomarker of anthropogenic stress in the autochthonous mussel Mytilus galloprovincialis. Two different approaches were adopted in biomonitoring six sites in the Lagoon, evaluating indigenous populations of mussels and organisms transplanted from a reference site and checked at several points in time. Lysosomal membrane stability was investigated by means of two tests: neutral red retention assay (NRRA) for evaluating haemocyte lysosomes and lysosomal latency test (LLT) for digestive cell lysosomes. Results indicate that the lysosomal response measured in haemocytes according to NRRA is a more valuable biomarker of anthropogenic stress in the framework both of passive and active biomonitoring in marine coastal environments.  相似文献

The MAST II WAVEMOD Project aimed at improving stochastic modelling of ocean waves and currents in coastal waters. In order to supplement existing data, two measurement campaigns were carried out, one on the Atlantic coast of Portugal, and, secondly, on the northern coast of Crete in the Mediterranean. The Portuguese measurements covered 7 months during winter 1993–1994, while off Crete, measurements started early in February 1994 and continued for 10 months. In both sites, two directional Waveriders and one non-directional Waverider were deployed along a transect out from the coast in water depths between 10 and 100 m. In addition, the Mediterranean site also incorporated measurements of mean current velocities. Available satellite data were analysed to supplement the buoy data. In this paper, both measurement campaigns are described together with a presentation of some of the data collected.  相似文献

A study of the main oyster areas polluted by the Amoco Cadiz oil spill (Abers, Baie de Morlaix) was carried out between 1978 and 1985 by chemical analyses (mainly aromatic hydrocarbon determinations) and histopathological observations of lesions in the digestive tracts and gonads of oysters. The hydrocarbon content in oyster tissues was monitored and analyzed by UV spectrofluorimetry, and gas and liquid chromatographic techniques.Seven years after the Amoco Cadiz wreck, the oysters sampled in the AberBenoit and in the Baie de Morlaix (Carantec) are still polluted with residual aromatic hydrocarbons, corresponding, respectively, to about five and two times the values from an unpolluted site (Baie de Saint-Brieuc). Since petroleum residues are buried in anaerobic sediments, especially in the Abers areas, they are expected to persist and accordingly may still contaminate oysters for several more years.Depuration kinetics of contaminated flat and Pacific oysters from the heavily polluted zones (Abers, Carantec) were studied and compared with that of healthy oysters transplanted in the same areas. The decontamination rates of oysters after transfer to a clean environment were heavily dependent on the duration of oil exposure.Necrosis and inflammation were the most significant and noticeable changes occurring during the first months after the pollution. Atrophy of the gonadal cells was detected in the oysters from the Abers only during the first six months of the study. No neoplastic and parasitic diseases have been related to this oil spill.  相似文献

A time series of surface geostrophic velocity is developed using satellite altimetry data during 1992–2010 for a track across the Kuroshio southeast of Kyushu, Japan. The temporal mean geostrophic velocity is estimated by combining the along-track sea level anomaly and shipboard ADCP data. This approximately 6-km resolution dataset is successful in representing the Kuroshio cross-current structure and temporal variation of the Kuroshio current-axis position during 2000–2010. The authors use this dataset to examine the winter Kuroshio path destabilization phenomenon. Its seasonal features are characterized as follows: the velocity shear on the inshore side of the Kuroshio becomes stronger and the Kuroshio path state becomes unstable from the summer to winter. This evidence is consistent with the hypothetical mechanism governing the destabilization phenomenon discussed in a previous study. Furthermore, the interannual amplitude modulation of the seasonality is examined in relation to interannual variations in the winter northerly wind over the northern Okinawa Trough and the Pacific Decadal Oscillation (PDO) index. The destabilization phenomenon appears 15 times in the period 2000–2010. Ten cases are related to local wind effects, and 7 of these are also connected with the PDO index. This is probably because the winter northerly wind over the northern Okinawa Trough is regulated by the PDO signal in interannual time-scales. Only 4 cases are related to the PDO index, but their driving mechanism remains uncertain.  相似文献

We measured tissue phosphorus content of high and low intertidal Hormosira banksii to test the hypothesis that tissue phosphorus content would be greater in individuals from the low intertidal because of greater total uptake associated with longer immersion in seawater. Moreover, we predicted that tissue phosphorus would be greater at sites where the seawater contained higher phosphate concentrations. There was a positive, linear relationship between local seawater phosphate concentrations and tissue phosphorus content of H. banksii from high and low intertidal zones at six different sites in winter. However, there were no comparable relationships in summer, even though the range of seawater phosphate concentration was similar in both seasons. The phosphorus contents of low intertidal H. banksii were significantly greater than high intertidal H. banksii in winter, but not in summer. Reasons for these differences may be related to greater access to seawater phosphate in low intertidal algae (than those in the high intertidal) in winter, followed by greater utilisation of the internal phosphorus between winter and summer, due to faster growth rates in the low intertidal.  相似文献

ABSTRACT

A contemporary approach to the assessment of metal pollutants in aquatic environments has been to measure contaminant concentrations in biological indicator species (mussels, oysters). However, for environments in which such indicator species do not occur naturally and cannot be deployed, alternative approaches for monitoring trace-metal pollution are required. Three years after the 2011 grounding of the MV Rena, diffusive gradients in thin films (DGT) samplers were deployed at Otaiti (the offshore reef where the Rena grounded) to assess levels of waterborne trace-metal contamination. Although the probability of detecting waterborne contaminants within a dynamic open ocean reef environment would seem low, due to mixing and dilution, our analyses confirmed the presence of copper (≤0.43 vs ≤0.27?nmol?kg?1), iron (≤6.3 vs ≤4.2?nmol?kg?1), aluminium (≤8.9 vs ≤1.3?nmol?kg?1), zinc (≤2.8 vs ≤2.0?nmol?kg?1) and manganese (≤0.11 vs ≤0.09?nmol?kg?1) at elevated concentrations in the Rena debris field relative to reference sites. The results demonstrate the utility of DGTs as a tool for measuring waterborne contaminants and suggest further research is required to characterise the contaminant plume and determine what effect it might have on the recovery of Otaiti's impacted biological communities.  相似文献
