Coastal zones are among the most productive and vulnerable areas on the planet. An example of impact on these fragile environments was shown in the case of the "Sea Empress" oil tanker, which ran aground in the Bristol Channel in 1996, spilling 72,000 tons of "Forties" crude oil. The objective was to investigate the sub-lethal cellular pathology and tissue hydrocarbon contamination in marine mussel populations, 4 months after the initial spill, using the neutral red retention (NRR) assay for lysosomal stability in blood cells. NRR was reduced in mussels, and indicative of cell injury, from the two sites closest to the spill in comparison with more distant and reference sites. Lysosomal stability was inversely correlated with polycyclic aromatic hydrocarbon concentrations in mussel tissues. Reduced lysosomal stability has previously been shown to contribute to impaired immunocompetence and to autophagic loss of body tissues. The use of this type of technique is discussed in the context of cost-effective, ecotoxicological tools for Integrated Coastal Zone Management.  相似文献

The Baltic Sea is a semi-enclosed sea with a steady salinity gradient (3‰–30‰). Organisms have adapted to such low salinities, but are suspected to be more susceptible to stress. Within the frame of the integrated environmental monitoring BONUS + project “BEAST” the applicability of immune responses of the blue mussel was investigated in Danish coastal waters. The sampling sites were characterised by a salinity range (11–19‰) and different mixtures of contaminants (metals, PAHs and POPs), according to chemical analysis of mussel tissues. Variation partitioning (redundancy analysis) was applied to decompose salinity and contamination effects. The results indicated that cellular immune responses (total and differential haemocyte count, phagocytic activity and apoptosis) were mainly influenced by contaminants, whereas humoral factors (haemolytic activity) were mainly impacted by salinity. Hence, cellular immune functions may be suitable as biomarkers in monitoring programmes for the Baltic Sea and other geographic regions with salinity variances of the studied range.  相似文献

Responses of the neutral red retention (NRR) assay as test for lysosomal stability and the comet assay as test for DNA integrity were measured in the water flea, Daphnia magna, and compared with mortality and effects on population growth rate during short- or long-term exposure to seven different toxicants. The NRR test and the comet assay were performed with fresh preparations of pieces of tissue from the digestive tract or with cell preparations from whole daphnias. Five toxicants caused responses of the NRR test or the comet assay after short-term exposure at concentrations below the acute toxicity level. Preliminary results of long-term exposure experiments suggest that these biomarker responses can be related to chronic effects on survival and/or reproduction of D. magna. This type of research should provide the basis for future use of the NRR test and the comet assay as early warning biomarkers for effects of toxicants on Daphnia populations.  相似文献

A field study was carried out in the Lagoon of Venice (north-east Italy) with the aim of evaluating the potential use of lysosomal destabilisation as a biomarker of anthropogenic stress in the autochthonous mussel Mytilus galloprovincialis. Two different approaches were adopted in biomonitoring six sites in the Lagoon, evaluating indigenous populations of mussels and organisms transplanted from a reference site and checked at several points in time. Lysosomal membrane stability was investigated by means of two tests: neutral red retention assay (NRRA) for evaluating haemocyte lysosomes and lysosomal latency test (LLT) for digestive cell lysosomes. Results indicate that the lysosomal response measured in haemocytes according to NRRA is a more valuable biomarker of anthropogenic stress in the framework both of passive and active biomonitoring in marine coastal environments.  相似文献

Harbours can be considered as model environments for developing and validating field monitoring procedures and to investigate mechanistic relationships between different biological responses. In this study several biomarkers were investigated in marine mussels caged for 4 weeks (June-July 2001) into an industrialized harbour of NW Italy. Organisms were collected at different time-intervals to better characterize the sensitivity, temporal variations and interactions of analysed responses. Besides single antioxidants the total oxyradical scavenging capacity (TOSC) assay was used to analyse the capability of the whole antioxidant system to neutralize specific forms of radicals: these data were further integrated by measurement of DNA integrity, oxidized bases and the impairment of lysosomal membrane stability in haemocytes. Results showed a biphasic trend for single antioxidants and TOSC, with an increase during the first 2 weeks of exposure to the polluted site followed by a progressive decrease up to a severe depletion in the final part of the experiment.  相似文献

The aim of this study was to examine whether a combination of biochemical, histopathological and toxicogenomic data could be used as a valuable tool for the assessment of biological risk associated with pollutants within the Tamar River and Estuary, S.W. England, U.K. Accordingly, biochemical and histopathological biomarkers (protein carbonyls, lipofuscin, neutral lipids, lysosomal stability [N-acetyl-β-hexosaminidase and neutral red], lysosomal volume, ferric reducing antioxidant power [FRAP] and malonaldehyde [MDA]) and gene expression profiles were assessed in 5 sites from the Tamar River and Estuary (Neal Point, Town Quay, Wilcove, Cremyll Ferry and Whitsand; and a reference site, Trebarwith Strand, N. Cornwall). PAHs were measured in mussel tissue and sediment and metals were measured in mussel tissue only. Data from the biomarkers was integrated into a Mussel Expert System (MES) model to produce a simple assessment of mussel stress. Clear gradients of mussel toxicity were identified by the biomarkers (with the exception of neutral lipids) with the highest impacted animals found furthest up the Tamar, whilst the MES was unable to identify a gradient of effect. Gene expression profiles also indicated a gradient of stress with the greatest number of significantly up- or down- regulated genes found at the uppermost 2 sites. The MES did, however, determine that mussels from all sites, except the reference site, were highly stressed; a conclusion that could not be inferred from the biomarker data alone. It is concluded that the MES is a valuable tool that permits integration and interpretation of complex sets of biomarker data by identifying the biological meaning of biomarker changes.  相似文献

Lysosomal sequestration of polynuclear aromatic hydrocarbons (PNAHs), a major class of environmental contaminant, is a well-established phenomenon;1 considerably less is known about their pathological effects on lysosomes. Marine molluscs contain a number of lysosome-rich tissues and PNAHs are known to induce deleterious alterations in lysosomal structure and latency of lysosomal enzymes.2 The latter are presumed to involve destabilisation of the lysosomal membrane, resulting in increased permeability and reduced enzyme latency. If lysosomal injury involves derangement of membrane-lipid structure due to the interaction of PNAHs then it would be expected that membrane damage would be closely linked to the structural characteristics of the intruding molecule. Our results show that the effects of the isomeric PNAHs phenanthrene and anthracene on digestive cell lysosomal stability were markedly different in the marine mussel (Mytilus edulis) over the same range of tissue concentrations. Lysosomal membrane stability was determined using a cytochemical test for enzyme latency.3  相似文献

Although the spatial distribution of hydrothermal vent assemblages in relation to environmental conditions has been assessed in several studies, there is little documented data on the temporal variation of the fauna and corresponding abiotic factors in a vent community. Here, we present one of the longest integrated (faunal and environmental data) time series ever obtained in a hydrothermal ecosystem. The data were acquired using the TEMPO ecological module that was deployed between 2006 and 2008 on the Mid-Atlantic Ridge, providing the first insights into the day-to-day variations in a Bathymodiolus azoricus mussel assemblage from the Lucky Strike vent field for the 48 days during which the video camera operated. The time-series yielded additional valuable information on longer-term variation in faunal distribution (comparing ~2 years), temperature (11.7 months) and iron concentrations (3.8 months).Results from daily observations showed that the vent mussel assemblage was quite stable over the 48 days of the study, reflecting the relative stability of environmental conditions during this period. B. azoricus mussels appeared to thrive in areas of very limited hydrothermal fluid input in habitats that are, as in other deep-sea ecosystems, significantly influenced by ocean tidal signals. Variation in species abundance was observed but, with the exception of Mirocaris fortunata shrimp, no links could be established with measured environmental variables. Although we did not observe any clear tidal influence on vent fauna, it is likely that physiological processes and species’ activities are influenced by these periodic variations. Longer time series are currently being acquired by different experiments deployed on the EMSO-Açores MoMAR observatory (2010–2013 and still recording). They should further improve our knowledge of the dynamics of hydrothermal systems and their associated faunal communities.  相似文献

An ecotoxicological protocol with caged mussels, Mytilus galloprovincialis, was developed to evaluate the potential impact of an offshore gas platform in the central Adriatic Sea. Reference organisms were collected on a seasonal basis from an unpolluted site and transplanted for four weeks in both the sampling area and to the investigated platform. Chemical analyses of trace metals in mussel tissues were integrated with a multi-biomarker approach for the early detection of biological responses at several cellular targets. Induction of metallothioneins, peroxisomal proliferation and activity of acetylcholinesterase were measured as markers for specific classes of chemicals. Special attention was given to oxyradical metabolism and appearance of oxidative-mediated toxicity to reveal a more general onset of cellular disturbance. In addition to individual antioxidants (superoxide dismutase, catalase, glutathione S-transferases, glutathione reductase, Se-dependent and Se-independent glutathione peroxidases, and levels of total glutathione), the total oxyradical scavenging capacity (TOSC) allowed a quantification of the overall capability to neutralize specific forms of intracellular reactive oxygen species (ROS; i.e. peroxyl and hydroxyl radicals). Cellular damages were evaluated as lysosomal destabilization (membrane stability, accumulation of lipofuscin and neutral lipids), lipid peroxidation products (malondialdehyde) and DNA integrity (strand breaks and micronuclei); the air survival test was finally applied to evaluate the overall physiological condition of mussels. Concentration of trace metals (As, Ba, Cd, Cr, Cu, Fe, Hg, Mn, Ni, Pb, Zn) revealed only limited variations in transplanted mussels during various experimental periods and such changes appeared partly related to natural fluctuations. Among biological responses, variations of antioxidants and lysosomal stability were confirmed as sensitive early warning signals for biological disturbance of both natural and anthropogenic origin. The presented protocol with caged mussels allowed marked biological effects caused by the investigated platform to be excluded, and represented a useful approach that is easy to extend for monitoring the impact of offshore activities in the Adriatic sea.  相似文献

The potential prognostic use of lysosomal reactions to environmental pollutants is explored in relation to predicting animal health in marine mussels, based on diagnostic biomarker data. Cellular lysosomes are already known to accumulate many metals and organic xenobiotics and the lysosomal accumulation of the carcinogenic polycyclic aromatic hydrocarbon 3-methylcholanthrene (3-MC) is demonstrated here in the hepatopancreatic digestive cells and ovarian oocytes of the blue mussel. Lysosomal membrane integrity or stability appears to be a generic indicator of cellular well-being in eukaryotes; and in bivalve molluscs it is correlated with total oxygen and nitrogen radical scavenging capacity (TOSC), protein synthesis, scope for growth and larval viability; and inversely correlated with DNA damage (micronuclei), as well as lysosomal swelling (volume density), lipidosis and lipofuscinosis, which are all characteristic of failed or incomplete autophagy. Integration of multiple biomarker data is achieved using multivariate statistics and then mapped onto "health status space" by using lysosomal membrane stability as a measure of cellular well-being. This is viewed as a crucial step towards the derivation of explanatory frameworks for prediction of pollutant impact on animal health; and has facilitated the development of a conceptual mechanistic model linking lysosomal damage and autophagic dysfunction with injury to cells, tissues and the whole animal. This model has also complemented the creation and use of a cell-based bioenergetic computational model of molluscan hepatopancreatic cells that simulates lysosomal and cellular reactions to pollutants. More speculatively, the use of coupled empirical measurements of biomarker reactions and modelling is proposed as a practical approach to the development of an operational toolbox for predicting the health of the environment.  相似文献

Certain specific aspects of cellular structure-linked functions can be used as rapid and sensitive indicators of cellular responses to environmental stimuli such as chemical contaminants.1 It is possible to observe structural-functional alterations in lysosomal membrane stability in the cells of some marine organisms at an early stage of such a response to cell injury,1,2 thus providing an early-warning signal of a potentially deleterious environmental situation. In order to further investigate lysosomal membrane disturbances, mechanisms of injury and capacity for cellular regeneration, the marine snail Littorina littorea was exposed to phenanthrene, which was used as an environmentally widespread and representative polynuclear aromatic hydrocarbon. Lysosomal membrane stability was measured using cytochemical determination of hydrolase latency,2 and lysosomal lipofuscin was measured as a possible indicator of enhanced lipid peroxidative damage.3  相似文献

There is a need for sensitive biological effect methods by which to detect impacts of chronic exposure to low concentrations of contaminants. Two methods shown to be potentially useful for monitoring purposes in fish include lysosomal membrane stability and peroxisome proliferation. These biological endpoints were assessed in Atlantic cod (Gadus morhua) head kidney following exposure to a mixture of produced water components including polycyclic aromatic hydrocarbons, phenol, and alkylphenols. Lysosomal damage of head kidney cells occurred within the first two weeks and did not recover during the entire exposure period (32 weeks). Lysosomal membrane stability was not affected by gender and was responsive at low concentrations of contamination, indicating that lysosomal membrane stability measured in the head kidney could be a useful biomarker for effects of offshore pollution. Peroxisome proliferation, measured as acyl-CoA oxidase activity in the head kidney, appeared to be a potential biomarker in male cod exposed less than 16 weeks.  相似文献

The neutral red lysosomal retention assay (NRR) of the haemocytes, and the acetylcholinesterase activity (AChE) in the haemolymph, the digestive gland, the gills and the mantle/gonad complex have been evaluated on mussels Mytilus galloprovincialis collected from Thermaikos and Strymonikos gulfs (northern Greece) in June and October 2001. The validity of performing the above core biomarkers is supported, firstly by their ability to respond to different pollution levels and, secondly, by the significant linear correlation among them. The evaluation of the micronuclei frequency (MN) has been performed in gill tissue and haemocytes of the same mussels and, according to the results, it needs more research in order its use as stress indices to be validated. In addition, the first results on cAMP levels in the gills, the mantle/gonad complex and the digestive gland, whose concentrations correlated to both, NRR and AChE introduce this signal transduction molecule as a new, promising biomarker.  相似文献

To evaluate the impact of land-based contaminants on benthic faunas, as part of the Land-Ocean Contamination Study (LOCS), a study transect was established in Jakarta Bay, Indonesia during the 1996 southeasterly monsoon. The transect extended 72 km between the city's main port, Tanjung Priok, and the Pilau Seribu Island chain in the Java Sea. The dissolved concentrations of Pb, Cu, Zn, Cr and Ni in seawater, in suspended particulate matter (SPM) and surficial sea bed sediments were measured along the transect at nine sites. In addition, metal concentrations were measured in tissues of the corals Goniopora lobata and Lobophyllia hemprichii at five sites, and of the green mussel, Perna viridis at six sites. An assessment of the impact of contaminants on the faunas was made using a biomarker approach, employing coral community analysis and lysosomal stability with Perna viridis. The results of the study showed two main trends. The distributions of metals dissolved in seawater, in SPM, and in the coral and mussel tissues were similar, and failed to show a consistent graded response from inshore to offshore sites. This suggests that the concentration of metals in waters is the primary route for metal uptake by the coral and mussel tissues. By way of contrast, a clear offshore increase in coral generic diversity, coral cover, coral colony numbers and neutral-red retention time (lysosomal stability) was observed, suggesting increasing nearshore stress. Whilst the coral community stucture may reflect the seasonally-averaged metal distributions in the bay (shown by the metal content of sea bed sediments which increase shorewards), it is more likely that the coral community structure and lysosomal biomarker are responding principally to other nearshore stresses, such as sediment and nutrient loading of water (sewage) or, more likely, organic contaminants such as oils and other hydrocarbons.  相似文献

Protein expression and lysosomal stability were observed in Mytilus edulis exposed to 0, 20, 40, 60, and 80 ppb copper sulfate for 24 h in a static exposure system. Haemolymph was removed from each mussel and analyzed for lysosomal damage using the Neutral Red retention assay expressed as the ratio of lysosomal size to cell area, the lysosomal destabilization ratio. Gill tissue from the same individuals was dissected, homogenized and the proteins extracted and then separated using two-dimensional gel electrophoresis. Two-dimensional protein maps from each concentration were analyzed using MELANIE II 2D gel analysis software (Biorad). Lysosomal damage increased with each dosage, with LDRs ranging from 0.18 +/- 0.01 at 0 ppb copper sulfate to 0.32 +/- 0.11 at 80 ppb copper sulfate. Orthogonal comparisons showed that the control, 20 and 40 ppb classes together differed significantly (P < 0.05) from the 60 and 80 ppb classes together. Specific protein expression signatures were identified at each dosage.  相似文献

The clam Meretrix meretrix was used as a biomonitor to implement an environmental monitoring program along the coast of Beibu Gulf in October 2011. This program not only analyzed biomarkers including acetylcholinesterase, glutathione peroxidase, glutathione S-transferase, catalase and superoxide dismutase activities, total glutathione content and lipid peroxidation level in M. meretrix but also adopted a multi-biomarker approach – integrated biomarker response (IBR) to assess the environmental quality in this ecosystem. In addition, the metal (Hg, As, Cu, Pb, Zn, Cd and Cr) and polychlorinated biphenyls (PCBs) content in the surface sediment at the study area were also measured. The results showed that IBR index was able to distinguish a space trend between sampling sites with different degrees of anthropogenic environmental stress. Integrated contamination degree were displayed in the form of star plots and compared to IBR plots. There was a visual consistency between the pollution level and IBR variation. Based on the results, it was proved that the IBR method coupled with chemical analysis was quite useful for the assessment of environmental pollution in the coastal system.  相似文献

Our intent has been to develop rapid and sensitive bioassay procedures for laboratory and field applications based on hemolymph factors in animals such as the bay mussel, Mytilus edulis. We found that mussel hemolymph could be easily and repeatedly sampled from individual animals in sufficient amounts to allow analyses for lysozyme, proteolytic activity, total protein and hemocyte counts. In static tests there seemed to be graded responses indicating release of increasing amounts of the above factors into the hemolymph in roughly direct proportion to Cu2+ concentration at lethal and sublethal levels. We interpreted this to be evidence for increased lysosomal and cellular disruption as cupric ion exposure was increased or prolonged. At the same time, Cu2+ concentrations in the mussel hemolymph increased up to an order of magnitude above ambient in the range of 26 to 267 ppb. We felt that this was evidence for the release of Cu-binding proteins into the mussel hemolymph. Simultaneously with the above events the serum granulocytes increased in numbers above control values by factors of 3 or 4 in response to copper challenge while macrophage (terminology of Moore & Lowe, 1977) counts, including controls, declined precipitously.  相似文献

溶酶体生物标志物是环境污染引起生物效应的早期预警信号。石油类污染物对菲律宾蛤仔(Ruditapes philippinarum)溶酶体完整性的影响尚未研究。本研究以0号柴油的水溶性成分(WSF)为污染物,以养殖的菲律宾蛤仔为受试生物,测试3种溶酶体生物标志物对柴油WSF胁迫应答的剂量-效应关系和时间-效应关系。采用室内模拟暴露实验,将菲律宾蛤仔置于添加不同浓度(0.1~1.6 mg/L)柴油WSF的海水中5 d,结果显示:不同浓度的暴露组血细胞溶酶体中性红保留时间(NRRT)值比对照组低23.1%~48.5%(p<0.05或p<0.01)。而各浓度暴露组的鳃和内脏团酸性磷酸酶(ACP)活性均与对照组无显著差异。选取柴油WSF为0.8 mg/L进行污染的时间-效应实验,于第0、1、3、7、11天取样测定,结果显示:暴露组NRRT值始终低于对照组,且在第1、3天差异显著(p<0.05)。暴露组鳃和内脏团ACP活性都是先逐步升高到第7天达最大值后迅速下降,鳃ACP活性从第3天到第11天与对照组差异显著(p<0.01),内脏团ACP活性从第7天到第11天与对照组差异显著(p<0.05)。本研究未能获得基于N-乙酰-β-氨基己糖苷酶的组织化学染色反应上的溶酶体膜稳定性(LMS)应答。研究表明,菲律宾蛤仔血细胞NRRT和鳃ACP活性可作为海洋污染生物效应监测的潜在生物标志物。  相似文献

The Ria Formosa lagoon is an ecosystem whose water quality reflects the anthropogenic influence upon the surrounding areas. In this lagoon, the clam Ruditapes decussatus has a great economical importance and has been widely used as a biomonitor. A multibiomarker approach (δ-aminolevulinic acid dehydratase, metallothionein, lipid peroxidation, acetylcholinesterase, alkali-labile phosphates, DNA damage) was applied to assess the environmental quality of this ecosystem and the accumulation of contaminants and their potential adverse effects on clams. Clams were sampled in different shellfish beds in the period between July 2007 and December 2008 and abiotic parameters (temperature, salinity, pH and dissolved oxygen of seawater and organic matter in the sediment), condition index, metals (Cd, Cu, Zn, Ni, Pb), TBTs and PAHs concentrations were measured in clam tissues. Data was integrated using Principal Component Analyses and biomarker indices: IBR (Integrated Biomarker Response) and HSI (Health Status Index). This multibiomarker approach enabled discrimination of a time and space trend between sites with different degrees of anthropogenic contamination, identifying one of them (site 2) as the most stressful and summer months as the most critical period for clams due to an increase of environmental stress (anthropogenic pressure along with extreme environmental conditions, e.g. temperature, dissolved oxygen, organic matter in the sediments, etc). The selected biomarkers provided an integrated response to assess the environmental quality of the system, proving to be a useful approach when complex mixtures of contaminants occur.  相似文献

Bioavailable contaminant concentrations are an important component in assessing environmental effects as they directly affect ecosystem health. Shellfish contaminant monitoring programmes have traditionally filled this requirement but are being phased out in some jurisdictions. Passive sampling devices (PSDs) have the potential to replace shellfish monitoring; however, there are still knowledge gaps to address before this can occur. This study assessed the suitability of three different PSDs in providing the required information to replace shellfish monitoring. PSDs were deployed at three historic mussel monitoring sites with different levels of urban influence in the Waitemata Harbour, Auckland, New Zealand. Contaminants of interest were urban heavy metals, plus current and emerging organic contaminants. PSDs provided extremely low detection limits and, for some contaminants, very strong correlations to shellfish. PSDs can currently complement shellfish in monitoring, but it is premature to make conclusions as to the suitability of PSDs in replacing shellfish monitoring until more information is available.  相似文献
