An optimized method for automated analysis of algal pigments by HPLC   总被引:3,自引:0,他引:3  
A recent development in algal pigment analysis by high-performance liquid chromatography (HPLC) is the application of automation. An optimization of a complete sampling and analysis protocol applied specifically in automation has not yet been performed. In this paper we show that automation can only be successful if the various methodological aspects of the sampling and analysis protocol are considered in coherence. We introduce an optimized protocol that involves freeze-drying of the sample, subsequent extraction in 90% acetone and the application of water-packing during analysis. The method was evaluated on both natural plankton populations and a broad spectrum of microalgal cultures: Thalassiosira weisflogii (Bacillariophyceae), Emiliania huxleyi (Prymnesiophyceae), Phaeocystis globosa and Phaeocystis antarctica (Prymnesiophyceae) and Pyramimonas sp. (Prasinophyceae). Whereas pigment extracts were unstable in methanol, with recorded chlorophyll a losses from 10% to 60% per day, pigment degradation rates in acetone were generally less than 1% over 18 h storage in the autosampler (4 °C). In addition, it was found that the extraction efficiency of acetone significantly increased upon freeze-drying prior to extraction. Increases as high as 50–60% were measured in P. antarctica. The application of water-packing of the sample during injection resulted in improved peak shape and peak separation, without diluting the pigment concentrations. Automation is especially beneficial for application in the field, when mixed algal assemblages and low biomass put a high demand on the sensitivity as well as reproducibility of the method.  相似文献

The detailed distribution of algal chlorophyll and carotenoid pigments was determined around the halocline (freshwater-seawater interface) in the Krka Estuary on the east coast of the Adriatic Sea; in May 1988. After collection of water along the estuary, particulate matter was extracted and analyzed for pigments by high-performance liquid chromatography coupled with absorbance and fluorescence detection. Bottom marine waters were characterized by lower chlorophyll a (chl a) concentrations than encountered in surface waters, decreasing downstream from 0.50 μg l−1 to 0.16 μg l−1 at the marine end-member. The highest concentrations of chl α (up to 26.34 μg l−1) were found in the interfacial layer, an particularly at one station located off the city of ibenik, where high inputs of nutrients supported the accumulation of living algae at the halocline. Fucoxanthin was the most abundant carotenoid, which indicates a euryhaline dominance of diatoms in the estuary, whereas the dinoflagellate-derived carotenoid peridinin was confined to the interfacial and bottom saline waters of the inner estuary. High concentrations of alloxanthin and chl b were found in the interfacial layer, which also suggests an accumulation of Cryptophyceae and green algae in the inner estuary. Phaeophorbides showed higher concentrations in bottom waters than in surface waters, whereas the highest concentrations occurred in the interfacial layer. These high levels could reflect a density trapping of dead cells in an early degradation state, as suggested by the importance of allomerized chl a and chlorophyllide a vs. total chl a, or of faecal pellets originating from zooplankton grazing in the interfacial layer.  相似文献

A pile-supported OWC breakwater is a novel marine structure in which an oscillating water column (OWC) is integrated into a pile-supported breakwater, with a dual function: generating carbon-free energy and providing shelter for port activities by limiting wave transmission. In this work we investigate the hydrodynamics of this novel structure by means of an analytical model based on linear wave theory and matched eigenfunction expansion method. A local increase in the back-wall draft is adopted as an effective strategy to enhance wave power extraction and reduce wave transmission. The effects of chamber breadth, wall draft and air chamber volume on the hydrodynamic performance are examined in detail. We find that optimizing power take-off (PTO) damping for maximum power leads to both satisfactory power extraction and wave transmission, whereas optimizing for minimum wave transmission penalizes power extraction excessively; the former is, therefore, preferable. An appropriate large enough air chamber volume can enhance the bandwidth of high extraction efficiency through the air compressibility effect, with minimum repercussions for wave transmission. Meanwhile, the air chamber volume is found to be not large enough for the air compressibility effect to be relevant at engineering scales. Finally, a two-level practical optimization strategy on PTO damping is adopted. We prove that this strategy yields similar wave power extraction and wave transmission as the ideal optimization approach.  相似文献

宏条形码分析(metabarcoding analysis)是通过对野外样本的分子标记(即条形码)进行PCR扩增、测序,并利用测序结果分析野外样本物种组成、物种相对丰度,及其时空动态变化。这项在近20年里逐步发展起来的分子技术,得益于DNA测序技术的快速发展,包括测序通量的提升和测序成本的降低以及生物信息学分析方法的快速开发。基于可操作分类单元(operational taxonomic units,OTUs)分析方法,特别是最近的扩增子序列变异(amplicon sequence variants,ASVs)方法的开发,极大促进了宏条形码分析的应用。在世界及我国的海洋生态领域,宏条形码分析的应用还处于起步阶段。随着分析方法的日趋成熟,可适用于不同应用领域的分子标记不断增加,以及参考数据库的不断完善,宏条形码分析将释放出广阔的应用潜力。可以预期,宏条形码分析将在系统分析我国近岸海域有害藻华物种(及其他浮游生物物种)的组成和相对丰度中起关键作用,通过发现新物种,辨别隐存种,跟踪物种的生物地理学,进而帮助解析有害藻华暴发机理。  相似文献

Transverse vibrations of a rectangular orthotropic plate with a free edge while the others are supported according to four different combinations of boundary conditions are considered in the present paper. The presence of a concentrated mass is also taken into account. In view of the difficulty of satisfying exactly the natural boundary conditions at the free edge, the fundamental frequency of vibration is determined by means of the optimized Rayleigh-Ritz method and a pseudo-Fourier expansion which contains, in its argument, the optimization parameters γi. The results are in excellent agreement with those obtained by means of a finite element algorithmic procedure.  相似文献

Photoacclimatization of zooxanthellae extracted from the coral Pocillopora verrucosa was studied through the determination of pigments, light absorption and photosynthetic parameters, for samples collected in summer and winter between 1 and 40 m on a northwestern reef of Tahiti (French Polynesia). The same measurements were also performed on phytoplanktonic samples collected at a stable oceanic site north of the island. For the zooxanthellae, the variations with depth of all the parameters were generally of small amplitude. Seasonal differences were also observed. The photosynthetic to non-photosynthetic pigments ratio was higher at depth in both seasons and was higher in winter. The intracellular concentration of chlorophyll a and photosynthetic pigments was higher in winter, as was the photosynthetic pigments/chlorophyll a ratio, whereas the non-photosynthetic pigments/chlorophyll a ratio was higher in summer. Variations in the light absorption properties were also small. The photosynthetic parameters showed limited changes with depth with the largest variations (a factor of ∼2) observed for PBmax. The trends observed for the phytoplankton assemblage were generally of much higher amplitudes than for the zooxanthellae (e.g. for photosynthetic to non-photosynthetic pigments ratio or the saturation parameter, Ek). These results suggest that, in the very clear Polynesian waters, the amount of energy that reaches the zooxanthellae of Pverrucosa is not variable enough in the 1–40 m depth range to result in a drastic modification of the photosynthetic apparatus of the algae.  相似文献

Over the past five decades, several approaches for estimating probabilities of extreme still water levels have been developed. Currently, different methods are applied not only on transnational, but also on national scales, resulting in a heterogeneous level of protection. Applying different statistical methods can yield significantly different estimates of return water levels, but even the use of the same technique can produce large discrepancies, because there is subjective parameter choice at several steps in the model setup. In this paper, we compare probabilities of extreme still water levels estimated using the main direct methods (i.e. the block maxima method and the peaks over threshold method) considering a wide range of strategies to create extreme value dataset and a range of different model setups. We primarily use tide gauge records from the German Bight but also consider data from sites around the UK and Australia for comparison. The focus is on testing the influence of the following three main factors, which can affect the estimates of extreme value statistics: (1) detrending the original data sets; (2) building samples of extreme values from the original data sets; and (3) the record lengths of the original data sets. We find that using different detrending techniques biases the results from extreme value statistics. Hence, we recommend using a 1-year moving average of high waters (or hourly records if these are available) to correct the original data sets for seasonal and long-term sea level changes. Our results highlight that the peaks over threshold method yields more reliable and more stable (i.e. using short records leads to the same results as when using long records) estimates of probabilities of extreme still water levels than the block maxima method. In analysing a variety of threshold selection methods we find that using the 99.7th percentile water level leads to the most stable return water level estimates along the German Bight. This is also valid for the international stations considered. Finally, to provide guidance for coastal engineers and operators, we recommend the peaks over threshold method and define an objective approach for setting up the model. If this is applied routinely around a country, it will help overcome the problem of heterogeneous levels of protection resulting from different methods and varying model setups.  相似文献

A broad spectrum of events come under the category of harmful algal blooms (HABs), the common denominator being a negative impact on human activities. Harmful algal blooms involve a wide diversity of organisms, bloom dynamics, and mechanisms of impact. Here we review the effects of natural and man-induced environmental fluctuations on the frequency and apparent spreading of these phenomena. This article highlights the need for interdisciplinary research aimed at shedding light on basic mechanisms governing the occurrence and succession of microalgae in coastal seas. Information integrated from various discipline coupled with improved, sustained monitoring systems, will help predict and manage problems caused by HABs over a wide range of space and time scales.  相似文献

Proper sample storage and analytical techniques are required when analyzing anoxic sediment for acid volatile sulfide (AVS), because a change in redox state may alter sediment parameters. The goal of this study was to determine the optimum sample handling, storage, and analytical techniques to adopt when determining AVS concentrations in sediment. To determine the correct handling and storage protocols for use with anoxic sediment, samples of three different AVS levels were manipulated under a combination of the following handling and storage plans: handled on benchtop or under nitrogen atmosphere; preserved with zinc acetate (ZnAc) or not preserved, and stored at 4 °C or frozen (< − 20 °C). The data indicate that sulfide levels are best maintained when samples are handled under a nitrogen atmosphere, stored at 4 °C or frozen, and analyzed within 2 weeks of collection. To determine the best analytical procedure for the determination of AVS in sediment, the following methods were compared: colorimetric, gravimetric, ion-selective electrode, and photoionization detection. The dynamics of phytoplankton diversity constitute crucial data in environmental monitoring, food-web studies and ecosystem modeling. Proper assessment of phytoplankton community composition requires large investments in offshore sampling and taxonomic competence because these communities can change fundamentally on a weekly scale. Cheap and high-frequent offshore phytoplankton sampling can be achieved by using ships-of-opportunity, and in the present study we used the Alg@line facilities onboard a passenger ferry during its regular route between Finland and Sweden in the northern Baltic Sea. The first aim was to test if pigment analysis, as a cheaper alternative to cell counts, can be used to detect environment-correlated variation in phytoplankton communities. It has been shown that spatial variation (salinity differences) and variation throughout one annual cycle (changes in temperature and nutrients) were strongly reflected by pigment composition. The second aim was to test if pigment analyses and cell counts detect environment-correlated variation in phytoplankton communities equally well. It has been shown that spatial and seasonal variations were reflected by pigment composition while cell counts reflected seasonal variation only. This suggests that the pigment analyses detected aspects of the phytoplankton that were neglected or misinterpreted by cell counts. It is advocated that available resources of long-term studies, such as environmental monitoring, can be used more efficiently by utilizing the high ecological resolution of pigment composition in combination with high-frequent ships-of-opportunity sampling. To assess heterotrophic and/or toxic species, such programs could also include sampling for cell counts in a complementary manner, with actually performing counts only for crucial samples (e.g. between large community changes) as indicated by multivariate statistical analysis of pigment composition. Some aspects of analyzing phytoplankton data with multivariate statistics are also discussed.  相似文献

Estimation and analysis of the uncertainty introduced by using a numerical model for the investigation and study of any type of flow problem have become common industry practice. Through understanding and evaluation of the uncertainty introduced by a numerical model, the accuracy and applicability of the model itself are evaluated. In this paper, the numerical uncertainty of a CFD-methodology developed to analyse the hydrodynamic performance of a collective and cyclic pitch propeller (CCPP) is estimated and analysed. The CCPP is a novel propulsion and manoeuvring concept for autonomous underwater vehicles, aimed to generate both propulsion and manoeuvring forces through advanced control of the propeller's blade pitch. The numerical uncertainty is established for three performance parameters, the generated propulsive force, the side-force magnitude, and the side-force orientation, by conducting a grid and time-step refinement study over three operational conditions. Additionally, the influence of the oscillatory uncertainty, introduced by the periodic nature of the problem, is investigated although shown to have a minimal effect when properly monitored. Based on a least-squares regression analysis of the refined simulation results, the numerical uncertainty is proven to be dominated by the introduced discretisation errors. In the case of the propulsive and side-force magnitude, the total uncertainty is dictated by the time discretisation uncertainty under bollard pull conditions, while the total uncertainty of the captive cases is mainly a result of the spatial discretisation uncertainty. The total uncertainty in the side-force orientation is observed to be primarily a consequence of the time discretisation uncertainty for all simulated cases. Overall, the total uncertainty for captive cases can be considered satisfactory for all three performance parameters, while further work is needed to reduce the observed uncertainty of the simulations under bollard pull conditions.  相似文献

随着海上风能的开发向深水发展,支撑风机的载体平台越来越受到关注。在经济性与安全性、稳定性的多重要求下,张力腿平台(TLP)在海洋风能资源的开发中体现出了重要地位。采用基于开源平台OpenFOAM开发的计算流体动力学(CFD)水动力学求解器naoe-FOAM-SJTU对一座处于中等水深下的风机基础水下TLP(STLP)的运动响应进行了数值模拟与研究。文中使用弹簧锚链模型模拟STLP的垂向系泊锁链系统,模拟该平台在不同波浪环境下的运动响应情况。首先将STLP单自由度自由衰减CFD模拟结果与已有全耦合时域分析结果进行对比,验证了naoe-FOAM-SJTU求解器及使用弹簧模型模拟STLP系泊系统的准确性与可靠性。随后在考虑非线性波浪载荷的情况下研究极端海况下与一般作业海况下STLP的运动响应情况,计算工况中的风机基础所受弯矩及锚链受力情况,并详细展示流场、速度场信息,分析高阶波浪成分、不同海况等条件对于STLP运动性能的影响。研究结果表明,TLP在中等水深中具有良好的运动性能,naoe-FOAM-SJTU求解器可以有效模拟水中生产平台在波浪环境下的水动力问题,并可以对整个流场进行可视化展示与分析。  相似文献

The shell traits and weight traits are measured in cultured populations of bay scallop, Argopecten irradians. The results of regression analysis show that the regression relationships for all the traits are significant (P<0.01). The correlative coefficients between body weight, as well as tissue weight with shell length, shell height and shell width are significant (P<0.05). But the correlative coefficients between the anterior and posterior auricle length with body weight as well as tissue weight are not significant (P>0.05). The multiple regression equation is obtained to estimate live body weight and tissue weight. The above traits except anterior and posterior auricle length are used for the growth and production comparison among three cultured populations, Duncan's new multiple range procedure analysis shows that all the traits in the Lingshuiqiao (LSQ) population are much more significant than those of the other two populations (P<0.01), and there is no significant difference between the Qipanmo (  相似文献

The purpose of the present study is to investigate the performance and three-dimensional flow fields in a water-jet pump. TASCflow is employed to simulate the rotator-stator coupling flow field. A standard k-ε turbulence model combined with standard wall functions is used. In order to investigate the effect of a rear stator on flow fields, the flows in two water-jet pumps with and without a rear stator are studied. Computational fluid dynamics (CFD)-predicted overall performances are in good agreement with the experimental results. Then the flow fields, such as the pressure distribution on the blade surfaces, and the axial and tangential velocity distribution, especially the radial loading distribution, are investigated at different flow rates. In addition, the effects of a rear stator and different spacings between the rotor and the stator on the overall performance and the flow fields of the water-jet pump are also investigated.  相似文献

Within larger ice-free regions of the western Arctic Seas, subject to ongoing trophic cascades induced by past overfishing, as well as to possible future eutrophication of the drainage basins of the Yukon and Mackenzie Rivers, prior very toxic harmful algal blooms (HABs) – first associated with ∼100 human deaths near Sitka, Alaska in 1799 – may soon expand. Blooms of calcareous coccolithophores in the Bering Sea during 1997–1998 were non-toxic harbingers of the subsequent increments of other non-siliceous phytoplankton. But, now saxitoxic dinoflagellates, e.g. Alexandrium tamarense, were instead found by us within the adjacent downstream Chukchi Sea during SBI cruises of 2002 and 2003. A previous complex, coupled biophysical model had been validated earlier by ship-board observations from the Chukchi/Beaufort Seas during the summer of 2002. With inclusion of phosphorus as another chemical state variable to modulate additional competition by recently observed nitrogen-fixers, we now explore here the possible consequences of altered composition of dominant phytoplankton functional groups [diatoms, microflagellates, prymnesiophyte Phaeocystis colonies, coccolithophores, diazotrophs, and dinoflagellates] in relation to increases of the toxic A. tamarense, responding to relaxation of grazing pressure by herbivores north of Bering Strait as part of a continuing trophic cascade. Model formulation was guided by validation observations obtained during 2002–2004 from: cruises of the SBI, CHINARE, and CASES programs; moored arrays in Bering Strait; other RUSALCA cruises around Wrangel Island; and SBI helicopter surveys of the shelf-break regions of the Arctic basin. Our year-long model scenarios during 2002–2003 indicate that post bloom silica-limitation of diatoms, after smaller simulated spring grazing losses, led to subsequent competitive advantages in summer for the coccolithophores, dinoflagellates, and diazotrophs. Immediate top-down control is exerted by imposed grazing pressures of the model’s herbivores and bottom-up control is also effected by light-, nitrate-, ammonium-, silicate-, and phosphate-modulated competition among the six functional groups of the simulated phytoplankton community. Similar to the history of the southern North Sea adjacent to the Rhine River, possible farming of northwestern Alaska and Canada, in conjunction with other human activities of ice retreat and overfishing, may lead to future exacerbations of poisonous phytoplankton. These potential killers include both toxic dinoflagellate and diazotroph HABs, deadly to terrestrial and marine mammals, as well as those of prymnesiophytes, some of which have already foamed beaches, while others have killed fishes of European waters.  相似文献

Abstract

In this paper, two analytical models used worldwide to assess salinity variation in alluvial estuaries are applied to the Ashtamudi estuary, a Ramsar site, southwest coast of India; and Bouregreg estuary, in northwest part of Morocco. The estuaries’ bathymetry is described by an exponential function. Both models are quite similar and use a predictive equation for the dispersion in the estuary mouth (D0). The major difference between the two models is the use of the constant value of K?=?0.5 for the Van der Burgh coefficient (K) and the introduction of the correction factor ζ, which is a function of damping (δ) and shape (γ). The performance of these two models was evaluated by comparing their results with field measurements; this revealed that both analytical models apply well to both the estimation of salinity distribution and the prediction of salt intrusion in the Ashtamudi and Bouregreg estuaries (Ashtamudi: RMSE = 0.60–1.22 ppt; Bouregreg: RMSE = 0.92–2.71 ppt). One model agrees more with the field measurements of salinity distribution along the estuaries axis; the second underestimate and overestimate some values of salinity distribution along the estuaries. Possibly, the constant value of K?=?0.5 for the Van der Burgh coefficient (K) has applicability limits for the estuaries under tidal conditions. The specifying of the parameterization may be a field of research.  相似文献
