A rapid and sensitive fluorescence labeling method was developed and validated for the microanalysis of a sulfated polysaccharide drug,namely propylene glycol alginate sodium sulfate(PSS), in rat plasma. Fluorescein isothiocyanate(FITC) was selected to label PSS, and 1, 6-diaminohexane was used to link PSS and FITC in order to prepare FITC-labeled PSS(F-PSS) through a reductive amination reaction. F-PSS was identified by UV-Vis, FT-IR and 1H-NMR spectrum. The cell stability and cytotoxicity of F-PSS were tested in Madin-Darby canine kidney(MDCK) cells. The results indicated that the labeling efficiency of F-PSS was 0.522% ± 0.0248% and the absolute bioavailability was 8.39%. F-PSS was stable in MDCK cells without obvious cytotoxicity. The method was sensitive and reliable; it showed a good linearity, precision, recovery and stability. The FITC labeling method can be applied to investigating the absorption and metabolism of PSS and other polysaccharides in biological samples.  相似文献

The water vapor permeability (WVP) of films is important when developing pharmaceutical applications. Films are frequently used as coatings, and as such directly influence the quality of the medicine. The optimization of processing conditions for sodium alginate films was investigated using response surface methodology. Single-factor tests and Box-Behnken experimental design were employed. WVP was selected as the response variable, and the operating parameters for the single-factor tests were sodium alginate concentration, carboxymethyl cellulose (CMC) concentration and CaClz solution immersion time. The coefficient of determination (R2) was 0.97, indicating statistical significance. A minimal WVP of 0.389 8 g-mm/(m^2.h.kPa) was achieved under the optimum conditions. These were found to be a sodium alginate concentration, CMC concentration and CaCl2 solution immersion time at 8.04%, 0.13%, and 12 min, respectively. This provides a reference for potential applications in manufacturing film-coated hard capsule shells.  相似文献

Results of a series of experiments (on the adsorption of divalent metal ions by dried alginic acid, Na and Ca alginates of different composition and block structure) conducted in this systematic study of the effects of the composition and structure of alginates on the static adsorption equilibrium of divalent metal ions indicate that the properties of alginate adsorption to divalent metal ions are highly different, depending not only on the cations used, but also on the form and structure of the alginates. There is close correlation between the adsorption properties and the structure of the alginates. The selectivity coefficient of Na alginate for Cd-Sr ion exchange tends to increase with the increase of theM/G ratio in alginate, whereas the adsorption capacity of Ca alginate for Cu2+ ion decrease with the increase of the G-block or the average length of the G-block and the total adsorption capacity of alginic acid is found to vary in the same order as theF MM(diad frequency) in alginate in the mixed solution of Sr2+, Ba2+ and Cd2+. Carbohydrases secreted by marine Vibrio sp. YKW-34 with strong Laminaria cell wall degrading ability were screened, and among them alginate lyase was found to be dominant. The effects of medium composition and culturing condition on the production of alginate lyase by marine Vibrio sp. YKW-34 in flask were investigated in this study. In the culture medium of marine broth, no alginate lyase was produced. The activity of the alginate lyase, after being induced, reached 5 UmL^-1. The best inoculum volume and inoculum age were 10% and 12h, respectively. The optimal temperature for alginate lyase production was 25℃. The fermentation medium was composed of 0.5% of Laminaria powder and 0.2% of KNO3 with an initial acidity of pH 8.0. Alginate could induce alginate lyase production but not as efficiently as Laminaria powder did. The addition of fucoidan, cellulose and glucose had negative effect on the alginate lyase production. Other kinds of nitrogen sources, such as yeast extract, beef extract and peptone, had positive effect on the growth of the microorganism and negative effect on alginate lyase production. In addition, the time course of alginate lyase production under the optimized condition was described. The optimal harvest time was 48 h.  相似文献

Using the NCEP/NCAR reanalysis dataset from 1959-2004, the location and strength of the ITCZ (Inter-Tropical Con-vergence Zone), as well as their relations with typhoons in the northwestern Pacific were studied. It was found that the pentad loca-tion and strength of the ITCZ had close relations with the typhoon frequency. Higher latitude location or strengthened ITCZ were found to be favorable for the occurrence of typhoons over the Northwestern Pacific. An index was defined for ascertaining the loca-tion of the ITCZ. It was found that the index defined with the maximum value ofpentad and monthly meridional shear of zonal wind speed could better describe the location of ITCZ than another index defined with the maximum value of convergence. Correlation analysis between the index of ITCZ and the maximum cloud cover in the tropics showed that there were close relations between the ITCZ determined by the index and the maximum tropical cloud belt. The strength index of an ITCZ was defined as the zonal wind speed difference at latitudes south and north of the ITCZ. It was found that there are close relations between the ITCZ intensity and typhoon occurrence in the South China Sea [10°N-20°N, 100°E-120°E] and regions east of the Philippines and near the Mariana Islands [5°N-20°N, 127.5°E-150°E].  相似文献

Zygosaccharomyces rouxii is a salt-tolerant yeast species capable of removing cadmium(Cd) pollutant from aqueous solution. Presently, the physiological characteristics of Z. rouxii under the stress of sodium chloride(NaCl) and Cd are poorly understood. This study investigated the effects of NaCl and Cd on the growth, oxidative stress and antioxidant enzyme activities of Z. rouxii after stress treatment for 24 h. Results showed that NaCl or Cd alone negatively affected the growth of Z. rouxii, but the growth-inhibiting effect of Cd on Z. rouxii was reduced in the presence of NaCl. Flow cytometry assay showed that under Cd stress, NaCl significantly reduced the production of reactive oxygen species(ROS) and cell death of Z. rouxii compared with those in the absence of NaCl. The activities of superoxide dismutase(SOD), catalase(CAT), and peroxidase(POD) of Z. rouxii were significantly enhanced by 2%–6% NaCl, which likely contributed to the high salt tolerance of Z. rouxii. The POD activity was inhibited by 20 mg L-1Cd while the SOD and CAT activities were enhanced by 8 mg L-1 Cd and inhibited by 20 mg L-1 or 50 mg L-1 Cd. The inhibitory effect of high-level Cd on the antioxidant enzyme activities of Z. rouxii was counteracted by the combined use of NaCl, especially at 6%. This probably accounted for the decrease in Cd-induced ROS production and cell death of Z. rouxii after incubation with NaCl and Cd. Our work provided physiological clues as to the use of Z. rouxii as a biosorbent for Cd removal from seawater and liquid highly salty food.  相似文献

We used generalized additive models (GAM) to analyze the relationship between spatiotemporal factors and catch, and to estimate the monthly marine fishery yield of single otter trawls in Putuo district of Zhoushan, China. We used logbooks from five commercial fishing boats and data in government’s monthly statistical reports. We developed two GAM models: one included temporal variables (month and hauling time) and spatial variables (longitude and latitude), and another included just two variables, month and the number of fishing boats. Our results suggest that temporal factors explained more of the variability in catch than spatial factors. Furthermore, month explained the majority of variation in catch. Change in spatial distribution of fleet had a temporal component as the boats fished within a relatively small area within the same month, but the area varied among months. The number of boats fishing in each month also explained a large proportion of the variation in catch. Engine power had no effect on catch. The pseudo-coefficients (PCf) of the two GAMs were 0.13 and 0.29 respectively, indicating the both had good fits. The model yielded estimates that were very similar to those in the governmental reports between January to September, with relative estimate errors (REE) of <18%. However, the yields in October and November were significantly underestimated, with REEs of 36% and 27%, respectively.  相似文献
