The occurrence percentage and abundance of General Aerobic Hetero- trophic Bacteria (GAB) were determined by using the method of MPN for 182 sub- samples from 10 sediment cores taken from the Canadian basin and the Chukchi Sea at two different culturing temperatures. The results showed that the general occurrence percentage of GAB was quite high, average abundanees of GAB at cultured temperatares of 4℃ and 25℃ were 4.46 ×10^7 and 5.47×10^7 cells·g^-1(wt), respectively. The highest abundance of GAB occurred at 20 -22 cm section in the sediment. GAB abundances changed among the section of sediments, but there is a trend : the a-bundances at the middle or lower sections were lower than those at upper section. Cultivation at 25℃ could improve the occurrence percentage and abundances of GAB, which suggests that the increasing of temperature may change the living circum-stances of GAB. The differences of GAB among the latitudes areas indicated that occurrence percentage and abundances of GAB in middle latitude areas were higher than those in the higher or lower latitude areas, and were more obvious at 4℃ than those at 25℃. The GAB abundances in sediment under the shallower water seemed to be low- er than those in sediments under the deeper water and this status was more obvious at 25℃ than that at 4℃.  相似文献

The abundance and biomass of benthic heterotrophic bacteria were investigated for the 4 typical sampling stations in the northern muddy part of Jiaozhou Bay, estuary of the Dagu River, raft culturing and nearby areas of Huangdao in March, June, August and December, 2002. The abundance and biomass range from 0.98×107 to 16.87×107 cells g−1 sediment and 0.45 to 7.08 μg C g−1 sediment, respectively. Correlation analysis showed that heterotrophic bacterial abundance and biomass are significantly correlated to water temperature (R=0.79 and 0.83, respectively,P<0.01).  相似文献

We evaluated the effects of red and blue light on the repair of UV-B radiation-induced damage in tetraspores of Chondrus ocellatus Holm. Tetraspores of C. ocellatus were treated with different UV-B radiation levels(0,36,72,108,144 and 180 J/m2),and thereafter subjected to PAR,darkness,or red or blue light during a 2-h repair stage,each day for 48 days. The diameters and cellular contents of cyclobutane pyrimidine dimmers(CPDs),chlorophyll a(Chl a),phycoerythrin,and UV-B-absorbing mycosporinelike amino acids(MAAs) contents of the tetraspores were determined. Our results show that low doses of UV-B radiation(36 and 72 J/m 2) promoted the growth of C. ocellatus; however,increased UV-B radiation gradually reduced the C. ocellatus growth(greater than 72 J/m2). The MAAs(palythine and asterina-330) in C. ocellatus were detected and analyzed by LC/MS. Our results suggest that moderate red light could induce the growth of this alga in aquaculture. In addition,photorepair was inhibited by red light,so there may be some other DNA repair mechanism activated by red light. Blue light promoted the activity of DNA photolyase,greatly improving remediation efficiency. Red and blue lights were found to reduce the capacity of C. ocellatus to form MAAs. Therefore,PAR,red light,and blue light play different roles during the repair processes for damage induced by UV-B radiation.  相似文献

Inorganic carbon utilization in the non-calcifying marine microalgae,Nannochloropsis oculata, Phaeodactylum tricornutum andPorphyridium purpureum was compared with high- and low-calcifying strains ofEmiliania huxleyi grown in artificial seawater medium aerated with either air (0.03% V/V CO2) or CO2-free air. For high-calcifying strain ofE. oculata andP. tricornutem, similar growth patterns were observed in air-and CO2-free air-grown cultures.P. purpureum showed a less final cell density in CO2-free air than in air-grown culture. However, low-calcifying strain ofE. huxleyi was able to grow only in air-grown culture, but not in CO2-free air-grown culture. Measurements of alkalinity, pH, concentration of dissolved inorganic carbon (DIC) and free CO2 showed different patterns of DIC utilization. WithN. oculata, P. tricornutum andP. purpureum the pattern of DIC utilization was characterized by an increase of pH and a decrease of DIC but a constant alkalinity in the cultures aerated with air or CO2-free air, suggesting that bicarbonate utilization was concomitant with an efflux of OH. Both alkalinity and pH were maintained rather constant in air-grown culture of low-calcifying strain ofE. huxleyi, suggesting that diffusive entry of CO2 could meet the requirement of DIC for its photosynthesis and growth. High-calcifying strain ofE. huxleyi, however, showed a pattern of decrease of alkalinity and DIC but an almost constant pH, indicating that bicarbonate was the major form of inorganic carbon utilised by this organism and bicarbonate uptake is unlikely to be accompanied by an efflux of OH. The final pH values reached byN. oculata, P. tricornutum andP. purpureum in a closed system were 10.75, 10.60 and 9.85 respectively, showing that bicarbonate utilisation is concomitant with an efflux of OH. While the final pH of 8.4 in high-calcifyingE. huxleyi suggests that bicarbonate utilization was not accompanied by an efflux of OH. The dynamic microcosms were used to evaluate the effect of oil spills on microbial ecological system in marine sedi- ment and the enhancement of nutrient on the oil removal. The function and structure of microbial community caused by the oil pollu-tion and phosphate dosage were simultaneously monitored by dehydrogenase activity assay and PCR-denaturing gradient gel elec- trophoresis (DGGE) techniques. The results indicated that the amount of total bacteria in all dynamic microcosms declined rapidly with incubation time. The number of petroleum-degrading bacteria and the activity of sediment dehydrogenase were gradually en-hanced by petroleum in the oil-treated microcosms, while they both showed no obvious response to phosphate dosage. In comparison, phosphate spiked heterotrophic bacteria and they showed a significant increase in amount. DGGE profiles indicated that petroleum dosage greatly changed community structure, and the bacteria belonged to class Deltaproteobacteria, and phyla Bacteroidetes and Chlorobi were enriched. This study demonstrated that petroleum input greatly impacted the microbial community structure and con- sequently the marine sediment petroleum-degrading activity was enhanced. Phosphate dosage would multiply heterotrophic bacteria but not significantly enhance the petroleum degradation.  相似文献

Macroalgae in a littoral zone are inevitably exposed to UV-B irradiance.We analyzed the effects of UV-B on isoenzyme patterns and activities of superoxide dismutase(SOD),peroxidase(POX),catalase(CAT),and ascorbate peroxidase(APX) of red algae Grateloupia filicina(Lamour.) C.Agardh.The activities of SOD,CAT,and APX changed in response to UV-B in a time- and dose-dependent manner.POX activity increased significantly under all three UV-B treatments.The enzymatic assay showed three distinct bands of SODⅠ(Mn-SOD),SODⅡ(Fe-SOD),and SODⅢ(CuZn-SOD) under a low(Luv) and medium(Muv) dose of UV-B irradiation,while SODⅠ and SODⅢ activities decreased significantly when exposed to a high dose of UV-B irradiation(Huv).The activity of POX isoenzymes increased significantly after exposure to UV-B,which is consistent with the total activity.In addition,a clear decrease in activity of CATIV was detected in response to all the three doses of UV treatments.Some bands of APX isoenzyme were also clearly influenced by UV-B irradiation.Correspondingly,the daily growth rate declined under all the three exposure doses,and was especially significant under Muv and Huv treatments.These data suggest that,although the protection mechanisms of antioxidant defense system are partly inducible by UV-B to prevent the damage,G.filicina has incomplete tolerance to higher UV-B irradiation stress.  相似文献

Nitrogen deficiency is an effective strategy for enhancing lipid production in microalgae. Close relationships exist among lipid production, microalgal species, and nitrogen sources. We report growth, lipid accumulation, and fatty acid composition in four microalgae (Chloroeoccum ellipsoideum UTEX972, Chlorococcum nivale LB2225, Chlorococcum tatrense UTEX2227, and Scenedesmus deserticola JNU19) under nitrate- and urea-nitrogen deficiencies. We found three patterns of response to nitrogen deficiency: Type-A (decrease in biomass and increase in lipid content), Type-B (reduction in both biomass and lipid content), and Type-C (enhancement of both biomass and lipid content). Type-C microalgae are potential candidates for large-scale oil production. Chlorococcum ellipsoideum, for example, exhibited a neutral lipid production of up to 239.6 mg/(L'd) under urea-nitrogen deficiency. In addition, nitrogen deficiency showed only a slight influence on lipid fractions and fatty acid composition. Our study provides useful information for further screening hyper-lipid microalgal strains for biofuel production.  相似文献

In order to explore the train of thought for China‘s urbanizing development and coordinated rural eco-nomic development, and to find good ways of solving rural problems through urbanization, this paper absorbs the push-and-pull forces theory and the systematic dynamic theory in the traditional population migration theories, views urbanization as a dynamic system, makes research on the push-and-pull mechanism of urbanization. The pulling power of urbanization is analyzed according to two aspects, the agglomeration effect and the radiation effect of cities. The agglomeration effect provides continuous propelling force for urbanization, and the radiation effect further accelerates the urbanization process by pushing forward the development of rural economy. Of course, the slow de-velopment of urbanization can result in the hindrance to rural economic development.  相似文献

Seven surveys were carried out in April, September, October, December 2006 and March, May, August 2007 in the Yellow Sea, China. Variations in the spatial and temporal distribution of Synechococcus, picoeukaryotes and heterotrophic bacteria are quantified using flow cytometry. Synechococcus and heterotrophic bacteria are most abundant from late spring to autumn, while picoeukaryotes concentration is high in spring. Synechococcus and heterotrophic bacteria concentrated high in the northwest part of the Yellow Sea in spring and autumn, while picoeukaryotes distributed evenly over the whole study area except for a small frontal zone in the coastal area on the west (in spring) and central Yellow Sea (in autumn). Under mixing conditions, the vertical distribution of the three picoplankton groups showed a well-mixed pattern. Upon a well-established stratification, the maximum abundance of picoplankton occurred above the mixed layer depth (~30 m). Cell sizes of Synechococcus and picoeukaryotes were estimated by converting forward scatter signals (FSC) from cytometry analysis to cell diameter, showing the results of 0.65–0.82 μm for Synechococcus and 0.85–1.08 μm for picoeukaryotes. The average integrated carbon biomasses ranged 15.26–312.62 mgC/m2 for Synechococcus, 18.54–61.57 mgC/m2 for picoeukaryotes, and 402.63–818.46 mgC/m2 for heterotrophic bacteria. The distribution of Synechococcus and heterotrophic bacteria was temperature dependent, and picoplankton presence was poor in the Yellow Sea Cold Water Mass.  相似文献

Microalgal lipids are regarded as main future feedstock of biofuels for its higher efficiency of accumulation and sustainable production.In order to investigate the effect of various nitrogen to phosphorus ratios on cells growth,chlorophyll content and accumulation of lipids in Dunaliella tertiolecta,experiments were carried out in modified microalgal medium with inorganic nitrogen(nitrate-nitrogen) or organic nitrogen(urea-nitrogen) as the sole nitrogen source at initial N:P ratios ranging from 1:1 to 32:1.The favorable N:P of 16:1 in the nitrate-N or urea-N medium yielded the maximum cell density and specific growth rate.Decrease in chlorophyll content were observed at the N:P of 4:1 in both nitrate-N and urea-N cultures.It was also observed that the maximum lipids concentration was obtained at the N:P of 4:1 in both nitrate and urea nutrient medium.The lipid productivity and lipid content of cultures in the urea-N medium at the N:P of 4:1were markedly higher than those from cultures with other N:P ratios(p 0.05).The results of this work illustrate the possibility that higher ratios of nitrogen to phosphorus have enhancing effect on cells growth of D.tertiolecta.Conversely,higher lipid accumulation is associated with a decrease in chlorophyll content under lower ratios of nitrogen to phosphorus.The results confirm the hypothesis of this study that a larger metabolic flux has been channeled to lipid accumulation in D.tertiolecta cells when the ratios of nitrogen to phosphorus drop below a critical level.  相似文献

We evaluated the relationships between water masses and pico- and nano-phytoplankton and bacterial abundance in the Chukchi Sea. The abundance of picoplankton ranged from 0.01 ~ 103 cells.mL1 (100 m, station R05) to 2.21 x 103 cells.mL-1 (10 m, station R05) and that of nanoplankton ranged from 0.03 x 103 cells.mL-I (100 m, station R07) to 2.21 ~ 104 cells.mLq (10 m, station R05). The lowest abundance of bacteria in the whole water column (0.21 x 106 cells.mLq) was at 100 m at station R17, and the highest (9.61 x 106 cells.mLL) was at 10 m at station R09. Melting sea ice affected the physical characteristics of the Chukchi Sea by reducing salinity of the surface mixed layer, resulting in greater hydrodynamic stability of the water column. These changes were accompanied by increased bacterial abundance. The warm Pacific water brought nutrients into the Chukchi Sea, resulting in greater abundance of bacteria and nano-phytoplankton in the Chukchi Sea than in other regions of the Arctic Ocean. However, the abundance of pico-phytoplankton, which was related to chlorophyll a concentration, was higher in Anadyr water than in the other two water masses. The structures ofpico- and nanoplankton communities coupled with the water masses in the Chuk- chi Sea can serve as indicators of the inflow of warm Pacific water into the Chukchi Sea.  相似文献

Forest canopy reduces shortwave radiation and increases the incoming longwave radiation to snowpacks beneath forest canopies. Furthermore, the effect of forest canopy may be changed by complex topography. In this paper, we measured and simulated the incoming longwave radiation to snow beneath forest at different canopy openness in the west Tianshan Mountains, China(43°16'N, 84°24'E) during spring 2013. A sensitivity study was conducted to explore the way that terrain influenced the incoming longwave radiation to snow beneath forest canopies. In the simulation model, measurement datasets, including air temperature, incoming shortwave radiation above canopy, and longwave radiation enhanced by adjacent terrain, were applied to calculate the incoming longwave radiation to snow beneath forest canopy. The simulation results were consistent with the measurements on hourly scale and daily scale. The effect of longwave radiation enhanced by terrain was important than that of shortwave radiation above forest canopy with different openness except the 20% canopy openness. The longwave radiation enhanced due to adjacent terrain increases with the slope increase and temperature rise. When air temperature(or slope) is relatively low, thelongwave radiation enhanced by adjacent terrain is not sensitive to slope(or air temperature), but the sensitivity increases with the decrease of snow cover area on sunny slope. The effect of longwave radiation is especially sensitive when the snow cover on sunny slope melts completely. The effect of incoming shortwave radiation reflected by adjacent terrain on incoming longwave radiation to snow beneath forest canopies is more slight than that of the enhanced longwave radiation.  相似文献

Two cruises were conducted in January and July 1986 in the Changjiang (Yangtse River) Estuary and its adjacent East China Sea (30°45′ -32°00′N,121°00′-124°00′E). Direct epifluorescence counts of planktonic bacteria and determinations of ATP concentrations were made. Subsamples were taken for measurement of oxygen consumption rates and chlorophyll concentrations.Bacteria and ATP concentrations were higher in summer than in winter, highest in the river and the river mouth, and gradually lower offshore. The bacteria number was correlated positively with suspended matter, nitrates and oxygen consumption rates, and negatively with salinity.In winter bacteria were the main contributors of ATP and the main consumers of dissolved oxygen in the whole studied area. In summer two maxima of ATP were found along the salinity gradient. In order to understand the large-scale spatial distribution characteristics of picoplankton,nanophytoplankton and virio-plankton and their relationship with environmental variables in coastal and offshore waters,flow cytometry(FCM) was used to ana-lyze microbial abundance of samples collected in summer from four depths at 36 stations in the North Yellow Sea(NYS).The data revealed spatial heterogeneity in microbial populations in the offshore and near-shore waters of the NYS during the summer.For the surface layer,picoeukaryotes were abundant in the near-shore waters,Synechococcus was abundant in the offshore areas,and bacte-rial and viral abundances were high in the near-shore waters around the Liaodong peninsula.In the near-shore waters,no significant vertical variation of picophytoplankton(0.2-2μm) abundance was found.However,the nanophytoplankton abundance was higher in the upper layers(from the surface to 10 m depth) than in the bottom layer.For the offshore waters,both pico-and nanophytoplankton(2-20μm) abundance decreased sharply with depth in the North Yellow Sea Cold Water Mass(NYSCWM).But,for the vertical dis-tribution of virus and bacteria abundance,no significant variation was observed in both near-shore and offshore waters.Autotrophic microbes were more sensitive to environmental change than heterotrophic microbes and viruses.Viruses showed a positive correla-tion with bacterial abundance,suggesting that the bacteriophage might be prominent for virioplankton(about 0.45μm) in summer in the NYS and that viral abundance might play an important role in microbial loop functions.  相似文献

By using monthly historical sea surface temperature (SST) data for the years from 1950 to 2000, the Western Pacific Warm Pool (WPWP) climatology and anomalies are studied in this paper. The analysis of WPWP centroid (WPWPC) movement anomalies and the Niño-3 region SST anomalies(SSTA) seems to reveal a close, linear relation between the zonal WPWPC and Niño-3 region SSTA, which suggests that a 9° anomaly of the zonal displacement from the climatological position of the WPWPC corresponds to about a l°C anomaly in the Niño-3 region area-mean SST. This study connects the WPWPC zonal displacement with the Niño-3 SSTA, and it may be helpful in better understanding the fact that the WPWP eastward extension is conducive to the Niño-3 region SST increase during an El Niño-Southern Oscillation (ENSO) event.  相似文献

Acute toxicities of potassium permanganate, formalin, and Lugol’s iodine solution to a commonly occurred marine ciliate Pleuronema coronatum (Ciliophora, Scuticociliatida) were measured. Linear regression analysis of the results highlighted the close relationships between doses of the medicines and mortalities of the organisms, thus providing a capability to predict toxicity effects from the dose. Toxic effects of the medicines on the ciliates were described in the present paper, and the median lethal concentrations (LC50 values) were given. Results of measurements indicated that 2 h-LC50 and 12 h-LC50 values of formalin on P. coronatum were 59.00×10−6 and 43.57×10−6, while those of Lugol’s solutions were 90.13 and 67.84×10−6 respectively. The tolerance of P. coronatum to formalin is apparently lower than that to Lugol’s iodine solution and potassium permanganate is a suitable medicine to kill ciliates in short time.  相似文献

Time series of sea surface temperature (SST),wind speed and significant wave height (SWH) from meteorologicalbuoys of the National Data Buoy Center (NDBC) are useful for studying the interannual variability and trend of these quantities at the buoy areas.The measurements from 4 buoys (B51001,B51002,B51003 and B51004) in the Hawaii area are used to study theresponses of the quantities to EI Nino and Southern Oscillation (ENSO).Long-term averages of these data reflect precise seasonaland climatological characteristics of SST,wind speed and SWH around the Hawaii area.Buoy observations from B51001 suggest asignificant warming trend which is,however,not very clear from the other three buoys.Compared with the variability of SST andSWH,the wind speeds from the buoy observations show an increasing trend.The impacts of El Nifio on SST and wind waves arealso shown.Sea level data observed by altimeter during October 1992 to September 2006 are analyzed to investigate the variabilityof sea level in the Hawaii area.The results also show an increasing trend in sea level anomaly (SLA).The low-passed SLA in theHawaii area is consistent with the inverse phase of the low-passed Sol (Southern Oscillation Index).Compared with the low-passedSOl and PDO (Pacific Decadal Oscillation),the low-passed PNA (Pacific-North America Index) has a better correlation with thelow-passed SLA in the Hawaii area.  相似文献
