Actinomycetes in five marine sediments collected from the Arctic Ocean at depths of 43 to 3 050 m were cultivated using a variety of media. A total of 61 actinomycete colonies with substrate mycelia only were observed, and no colonies with aerial mycelia were observed under aerobic conditions at 15 ℃. From these colonies, 28 were selected to represent different morphological types.Denaturing gradient gel electrophoresis (DGGE) was used to check the purity of isolates and select representatives for subsequent sequencing. Phylogentic analyses based on nearly full-length 16S ribosomal RNA gene (rDNA) sequences indicated that the actinomycetes isolated were accommodated within genus Rhodococcus of family Nocardiaceae, genus Dietzia of family Dietziaceae,genera Janibacter and Terrabacter of family Instrasporangiaceae and genera Kocuria and Arthrobacter of family Micrococcaceae. One of the strains (P27-24) from the deep-sea sediment at depth of 3 050 m was found to be identical in 16S rDNA sequence(1474/1474)with the radiation-resistant Kocuria rosea ATCC 187T isolated from air. More than halfofthe isolates showed the similarities ranging from 99.5% to 99.9% in 16S rDNA sequence to dibenzofran-degrading, butyl 2-ethylhexanoate-hydrolysising and nitrile-metabolizing actinomycetes. All the strains isolated were psychrotolerant bacteria and grew better on the media prepared with natural seawater than on the media prepared with deionized water. Three of them (Dietzia sp. P27-10, Rhodococcus sp. S11-3 and Rhodococcus sp.P11-5)had an obligate growth requirement for salt, confirming that these strains are indigenous marine actinomycetes.  相似文献

PCR-DGGE approach was used to analyze bacterial diversity in the bottom section of seven arctic sea ice samples colleted from the Canada Basin. Thirty-two 16S rDNA sequences were obtained from prominent DGGE bands. The closest relatives of these sequences are found to be those of cultivated or uncultured bacteria from antarctic or arctic sea ice. Phylogenetic analysis clustered these sequences or phylotypes within α- proteobacteria, γ-proteobacteria and CFB (cytophaga-flexibacter-bacteroides) group. Sequences belonging to γ-proteobacteria were dominant and members of the CFB group were highly abundant. It was suggested that the CFB group was the representative of the bottom section of sea ice samples.  相似文献

To assess the significance of naturally occurring dissolved organic matter (DOM) on complexation of transuranic elements in seawater, a series of bioassay experiments was conducted in which the effect of DOM on the accumulation of 241 Am, 237Pu (III–IV), and 237Pu (V–VI) by the marine diatom Thalassiosira pseudonana was measured. EDTA at 0.3μM complexed both metals substantially, resulting in reduced radio-isotope uptake by the diatom; the greatest effect was on Pu (III–IV). In contrast, there was no apparent complexation of either element by equimolar concentrations of marine fulvic (MFA) or humic acids (MHA), naturally occurring photooxidizable DOM (uncharacterized), or diatom exudates, as none of these materials reduced isotope uptake; on the contrary, there were indications that some of this DOM enhanced transuranic bioaccumulation in the diatom slightly. Subsequent experiments showed this enhancement was probably due to complexation of transition metals by the DOM, leading to fewer ambient ions ‘competing’ for binding sites on the cells; 241 Am uptake rates were negatively correlated (r =? 0.846, P < .01) with Σ ASV-labile Cu + Zn + Cd + Pb. These experiments suggest that naturally occurring DOM may not appreciably complex Am or Pu or greatly affect their bioavailability in the sea.  相似文献

西北冰洋表层沉积物黏土矿物分布特征及物质来源   总被引:2,自引:3,他引:2  
西北冰洋表层沉积物黏土矿物分析结果显示其黏土矿物组成的区域分布和变化具有明显的规律性:从楚科奇海到北冰洋深水区,随着水深的增加,蒙皂石和高岭石含量增高,绿泥石和伊利石含量降低。自西往东,伊利石结晶度值降低,化学指数升高。根据Q型聚类分析获得的黏土矿物组合特征,结合周缘陆地的地质背景、河流及洋流情况,对研究区进行了黏土矿物组合分区,讨论了其黏土矿物来源。楚科奇海表层沉积物黏土矿物组合在靠近阿拉斯加一侧海域以Ⅰ类为主,靠东西伯利亚海一侧主要有Ⅱ类、Ⅲ类和Ⅳ类,中部主要为Ⅵ类,主要是西伯利亚和阿拉斯加的火山岩、变质岩以及一些含高岭石的沉积物以及古土壤等,经风化、河流搬运入海,在北太平洋的3股洋流及西伯利亚沿岸流的作用下沉积形成的。西北冰洋深水区表层沉积物的黏土矿物组合以Ⅰ类和Ⅴ类为主,表明其沉积物来源为欧亚陆架和加拿大北极群岛周缘海域的海冰沉积和大西洋水体的搬运以及加拿大马更些河的河流物质输入。  相似文献

Over the next several decades, arctic marine mammals will face threats from six areas of human influence: climate change, environmental contaminants, offshore oil and gas activities, shipping, hunting, and commercial fisheries. This paper reviews these factors, the nature and magnitude of the threats they pose, current scientific understanding and management of those threats, and the potential for effective conservation action. Climate change, offshore oil and gas activities, and commercial fisheries likely pose the greatest threats. Addressing the combined effects of all six factors, however, will be particularly difficult but essential to prevent declines beyond those that have already occurred.  相似文献

白令海、西北冰洋等高生产力海域在北冰洋"生物泵"中起到重要作用;海水升温、海冰消退等北极快速变化,将强烈影响该海域"生物泵"的结构与规模,并在沉积物中有机质的来源与新鲜程度上有所体现,可用脂肪酸加以指征。对第五次、第六次中国北极科学考察在以上海域采集的表层沉积物进行脂肪酸含量(以沉积物干重计)及组成分析,结果显示楚科奇海陆架总脂肪酸含量非常高((97.15±55.31)μg/g),白令海盆最低((15.00±1.30)μg/g),加拿大海盆、楚科奇海陆坡、白令海陆架居中(分别为(88.65±3.52)μg/g,(70.35±11.32)μg/g与(38.28±14.89)μg/g)。海源脂肪酸占总脂肪酸比例最高(86.82%±7.08%),陆源次之(8.45%±6.62%),细菌最低(4.63%±2.24%);硅藻指数(16:1ω9/16:0)在楚科奇海陆架(> 0.82)、白令海陆架边缘(> 0.65)较高,其他区域均较低。脂肪酸结果表明:(1)该海域沉积有机质主要来自海源,陆源贡献小;在北部、南部楚科奇海陆架、白令海陆架边缘,硅藻生物量占主要优势;细菌脂肪酸比例显著低于...  相似文献

通过对北冰洋西部(楚科奇海及北部边缘地带、加拿大海盆)34个表层沉积物样品中黏土组分的Sm-Nd同位素分析,结果表明:(1)黏土组分的Sm-Nd同位素分异明显,可将沉积物划分为北部、西部、南部和东部等多个同位素物源区;(2)在楚科奇海,太平洋入流的向北输运自西向东形成了3个不同的同位素物源区,沉积物的c(147Sm)/c(144Nd)比值、εNd(0)值和TDM年龄自西向东呈递减趋势;(3)在研究区北部,顺时针方向流动的波弗特涡流将马更些河物质向西搬运至加拿大海盆和楚科奇高地,使该区沉积物εNd(0)值明显偏低,TDM偏大;(4)在楚科奇海北部外陆架、海台和陆坡区,太平洋水、大西洋水和波弗特涡流的相互作用使该区沉积物TDM年龄的标准偏差大,同时出现了南-北源和西-南源等同位素混合类型。  相似文献

通过对中国第1~4次北极考察在西北冰洋采集的表层沉积物中底栖有孔虫丰度及其优势种分布特征与环境因素关系的综合研究发现,楚科奇海区低的底栖有孔虫丰度主要受较高的陆源物质输入的稀释作用影响;楚科奇海台和阿尔法脊较高的底栖有孔虫丰度主要受到暖而咸的大西洋中层水的影响;受碳酸钙溶解作用影响的门捷列夫深海平原和加拿大海盆底栖有孔虫丰度较低,并且水深3 597 m的站位出现了似瓷质壳的Pyrgo williamsoniQuinqueloculina orientalis,说明该区的CCD深度大于3 600 m。根据底栖有孔虫7个优势属种的百分含量分布特征可以划分出5个区域组合:南楚科奇海陆架-白令海峡组合以优势种Elphidium excavatumBuccella frigida为特征,可能反映受白令海陆架水影响的浅水环境;阿拉斯加沿岸-波弗特海组合以优势种Florilus scaphusElphidium albiumbilicatum为特征,可能反映受季节性海冰融化,低盐的阿拉斯加沿岸流以及河流淡水输入的低盐环境;大西洋中层水组合以优势种Cassidulina laevigata为特征,可能反映高温高盐的大西洋中层水影响的环境;北极深层水组合以优势种Cibicides wuellerstorfi为特征,可能反映水深大于1 500 m低温高盐的北极深层水环境;门捷列夫深海平原组合以优势种Oridorsalis umbonatus为特征,可能反映低营养的底层水环境。  相似文献

The chemical forms of transition metals in marine sediments are discussed on the basis of differential chemical leaching.A large part of hydrogenous transition metals in the near-shore sediments is leached with dilute acetic acid, whereas hydrogenous transition metals in the deep-sea sediments are slightly leached with the acid solution and mostly leached with the reducing agent solution. This difference is attributed to the different forms of hydrogenous iron and manganese between the near-shore and deep-sea sediments.The chemical behavior of hydrogenous transition metals in the deep-sea sediments for the acid and reducing agent leaching solutions is similar to that in ferromanganese nodules. This similarity indicates that the major parts of hydrogenous transition metals in deep-sea sediments are associated with iron-manganese oxide minerals dispersed in or coated on the sediments.  相似文献

通过对楚科奇海及邻近的北冰洋深水区表层沉积物中有机碳同位素含量(δ13C)、氮同位素含量(δ15N)及生物成因SiO2(BSiO2)含量分析,结果表明海源和陆源有机质的分布受海区环流结构和营养盐结构所制约.楚科奇海中西部和楚科奇海台受太平洋富营养盐海水的影响,海洋生产力高,沉积物中海源有机质和BSiO2含量高;靠阿拉斯加一侧海域海水的营养盐含量和生产力都偏低,沉积物中陆源有机质比重增加;在研究区北部和东北部的楚科奇高地和加拿大海盆,冰封时间较长,营养盐供应少,海洋生产力低,但来自马更些河和阿拉斯加北部的陆源有机质增多,沉积物中BSiO2含量小于5%,海源有机质百分含量小于40%.由于亚北极太平洋水通过楚科奇海向北冰洋海盆输送,研究区营养盐池表现为开放系统,营养盐的利用率与它的供应成反比,与海洋生产力成反比.  相似文献

利用Zobell 2216E培养基和涂布平板法对北极海洋沉积物中可培养细菌进行分离纯化,并利用16SrRNA基因进行分子鉴定与系统发育分析。根据菌落形态学特征,从59个站点的沉积物样品中共分离纯化获得570株细菌;基于16SrRNA基因的分子鉴定与系统发育分析表明,分离到的可培养细菌分别属于细菌域的4个门,5个纲,12个目,23个科,47个属,102个种,其中γ-Proteobactria占绝大多数;有14株菌株与模式菌株的16SrRNA基因序列相似性小于97%,为6个潜在的新种。北极海域的海洋沉积物中存在着丰富的微生物种质资源,为开发新型生物活性物质和特殊功能基因打下了基础。  相似文献

Deep-sea benthic communities and their structural and functional characteristics are regulated by surface water processes. Our study focused on the impact of changes in water depth and food supplies on small-sized metazoan bottom-fauna (meiobenthos) along a bathymetric transect (1200–5500 m) in the western Fram Strait. The samples were collected every summer season from 2005 to 2009 within the scope of the HAUSGARTEN monitoring program. In comparison to other polar regions, the large inflow of organic matter to the sea floor translates into relatively high meiofaunal densities in this region. Densities along the bathymetric gradient range from approximately 2400 ind. 10 cm-2 at 1200 m to approximately 300 ind. 10 cm-2 at 4000 m. Differences in meiofaunal distribution among sediment layers (i.e., vertical profile) were stronger than among stations (i.e., bathymetric gradient). At all the stations meiofaunal densities and number of taxa were the highest in the surface sediment layer (0–1 cm), and these decreased with increasing sediment depth (down to 4–5 cm). However, the shape of the decreasing pattern differed significantly among stations. Meiofaunal densities and taxonomic richness decreased gradually with increasing sediment depth at the shallower stations with higher food availability. At deeper stations, where the availability of organic matter is generally lower, meiofaunal densities decreased sharply to minor proportions at sediment depths already at 2–3 cm. Nematodes were the most abundant organisms (60–98%) in all the sediment layers. The environmental factors best correlated to the vertical patterns of the meiofaunal community were sediment-bound chloroplastic pigments that indicate phytodetrital matter.  相似文献

本文基于塞舌尔群岛东北部海域34个表层沉积物样品的黏土矿物测试结果分析其组成特征、分布规律和物质来源。研究区表层沉积物中黏土矿物平均相对含量从高到低依次为伊利石(35%)、坡缕石(20%)、蒙皂石(20%)、高岭石(16%)、绿泥石(10%)。根据沉积物黏土矿物相对含量和空间分布规律,可将研究区分为2个亚区(I区、II区)。I区覆盖研究区北部大部区域,物质来源主要为阿拉伯半岛、印度半岛西南部以及索马里海岸沙漠的风尘物质;II区位于研究区南部的塞舌尔群岛以东,主要接收塞舌尔高原风化物质。综合分析研究区细颗粒物质来源、空间分布规律及区域动力环境特征,南亚夏季风搬运阿拉伯半岛、印度西部及索马里海岸沙漠风尘物质向南输运是影响I区黏土矿物组成的主要因素;南赤道逆流驱动塞舌尔高原富高岭石、绿泥石的细粒风化物质向东扩散,控制了II区黏土矿物组成特征和空间分布规律。  相似文献

用连续提取法对选自西北冰洋的32个表层沉积物进行分析,测定了Cr、Cu、Pb在全样中及其在P1-2、P3、P4、P5中的含量.结果表明随着离岸距离增加,水深加深,Cr、Cu、Pb含量呈增高的趋势.研究区陆架沉积物中Cr、Cu、Pb含量分别为60.66、14.77、16.65μg/g;深海沉积物中Cr、Cu、Pb的含量分别为80.40、41.70、25.62μg/g.元素赋存形态分析表明,Cr、Cu、Pb主要赋存在残渣态,残渣态中三元素的含量平均值分别为64.97、19.67、17.56μg/g,占元素总量的比例分别为93.58%、75.02%和83.76%.三元素在各赋存形态中的含量分布是:Cu为残渣态>有机质和硫化物态>铁锰氧化态>可交换及碳酸盐态;Pb和Cr则为残渣态>铁锰氧化物态>有机质硫化物态>可交换态及碳酸盐态.研究区重金属元素在不同赋存形态中的含量分布与北太平洋深海沉积物中类似.  相似文献

为了准确解释环境磁学参数记录的极地古气候环境变化信息,本研究对白令海和西北冰洋61个站位的表层沉积物进行了高、低频质量磁化率(χ)、非磁滞磁化率(χARM)和磁化率-温度(k-T)分析,以探明该区沉积物中磁性矿物的种类、来源与搬运路径。结果显示,样品的χ具有明显的地域分布特征。白令海的χ值整体高于楚科奇海,并在育空河口外侧和圣劳伦斯岛南侧较高,向北和向西南方向逐渐减小。楚科奇海中东部陆架上表层沉积χ值高于阿拉斯加沿岸,而西北冰洋深海平原和洋脊区的χ值最低。χARM的变化趋势与质量磁化率相似,但频率磁化率的变化趋势与质量磁化率正好相反。k-T分析结果显示阿留申海盆沉积物中的铁磁性矿物以磁赤铁矿占主导,白令海陆架育空河口外侧和圣劳伦斯岛南北两侧为磁铁矿,白令海陆架西部和楚科奇海陆架中东部为磁赤铁矿和磁铁矿,楚科奇海阿拉斯加沿岸为黄铁矿,而西北冰洋陆坡、深海平原和洋脊区为胶黄铁矿和黄铁矿,但高纬度区沉积物中的胶黄铁矿含量更高。沉积物中磁性矿物的区域性分布受沉积物来源、洋流和底质环境等因素的控制。白令海和楚科奇海陆架磁赤铁矿来源于亚洲大陆,白令海陆架东部的磁铁矿来自育空河流域,阿拉斯加沿岸沉积物中的黄铁矿,应为阿拉斯加西北部陆源侵蚀来源的或早期成岩作用形成的,西北冰洋深海盆区的胶黄铁矿,为自生成因的。  相似文献
