Causes of deep-water variations: Reply to comment by E. Fahrbach,M. Hoppema,G. Rohardt,M. Schröder and A. Wisotzki

Vertically integrated heat content from the Weddell Sea along 69 °S in vicinity of the Greenwich meridian from the 1977–2001 period is presented. This demonstrates that the east-west variability in the area is lower than the natural variability. The sensitivity on the choice of lower boundary for the heat content integral is discussed in relation to upwelling from the deeper water masses. The cooling around Maud Rise during the 1990's on the order of 20 W/m² remains significant, and may be explained by increased surface heat fluxes during winter months with lower than normal sea ice concentration.     

氯化钠和碱度对罗氏沼虾仔虾的毒性研究

实验研究氯化钠、碱度对罗氏沼虾仔虾的急性毒性作用。用概率单位法求出了其氯化钠2 4、4 8、72、96 h的半致死浓度分别为 8.58、 5.32、 3.4 7、 2 .19g/ L;碱度的 2 4、4 8、72、96 h半致死浓度分别为 51.0 2、 37.0 7、 2 7.19、 2 1.54mmol/ L。对氯化钠和碱度混合作用的结果进行的方差分析表明 ,氯化钠和碱度对仔虾的死亡率影响显著 ,而两者的交互作用不显著。碱度 (AL K)与罗氏沼虾仔虾氯化钠 2 4、4 8h L C50 (S)关系式分别为 S=- 0 .12 78AL K+ 9.2 346 ,S=- 0 .0 715AL K +5.530 1。     

S. E. M. analysis of quartz sand grains from selected Hong Kong and Chinese littoral environments

This pilot study attempts to demonstrate some underlying scanning electron microscopy themes of quartz grain surface textures. A variety of textural patterns and individual features are described for grains selected from various littoral environments. An attempt was made to differentiate samples on surface textures alone, but limitations of using this technique in sedimentological isolation were apparent. Statistical analysis of checklist data and photographic evidence revealed some of the more important feature combinations used in environmental diagnosis. The use of discriminant analysis provided quantitative sample separation.     

GT-1M 海洋重力仪与KSS31M 海洋重力仪的对比

为了检验俄罗斯、加拿大合作生产的GT-1M海洋重力仪与德国KSS31M海洋重力仪的工作性能,验证GT-1M重力数据的可靠性,将GT-1M采集的3条重力剖面与KSS31M重力仪采集的重力测网进行了对比分析。结果显示:GT01剖面与26条KSS31M重力测线的交点差均值为0.31mGal,交点差均方根为2.51mGal;GT02剖面与11条KSS31M重力测线的,交点差均值为–0.78mGal,交点差均方根为1.97mGal;全部交点差均值为–0.01mGal,均方根(RMS)为2.32mGal。GT01剖面与KSS31M测网网格化切割的剖面的变化趋势一致,重力变化幅值基本吻合,相关系数为0.98;而GT02剖面位于KSS31测网边缘,网格化切割的KSS重力剖面不能反应真实的重力场,虽然GT02剖面变化趋势一致,但相关系数仅为0.94,在相位上也明显有误差。考虑到测网位于海底地形崎岖的深水区,而且两次测量时间间隔达3a,因此GT-1M重力仪测试的结果还是基本与KSS31M重力仪吻合的,数据是可靠的。该分析结果对今后的重力测量工作有参考价值。     

海洋链霉菌分离株M095遗传转化体系的建立

研究了海洋链霉菌分离株M095的基因转移系统。利用属间接合转移将具有oriT的大肠杆菌-链霉菌穿梭质粒pIJ8600转入EscherichiacoliET12567(pUZ8002)中,获得供体菌。将供体菌与预萌发的菌株M095的孢子进行接合转移,将质粒pIJ8600转入菌株M095中,其转化率为1.99×10-4个接合转化子/受体。Southern杂交证明质粒pIJ8600已经整合到菌株M095的染色体上。同时,将来自Spirulinamaxima(Cyanophyta)的别藻蓝蛋白基因(apc)克隆在质粒pIJ8600的XbaI和BglII位点,产生质粒pAPIJ。用接合转移法将质粒pAPIJ转入菌株M095。通过SDS-PAGE分析,得到2个大小为22ku和17ku的蛋白,分别相对应于别藻蓝蛋白的α和β亚基。这些结果进一步证明菌株M095的遗传转化体系已经成功建立起来,这将为其他海洋放线菌遗传转化工作的研究奠定基础。
