Biological monitoring of metals in marine waters often employs tissue residue analyses from bivalves. Chemical residues in tissues do not elucidate biological injury, however, whereas biomarkers, do afford sensitive stress indices. Physiological and cytological responses were evaluated in laboratory experiments from the mussel Perna viridis in order to assess the species’ suitability for pollution monitoring in the tropics. Periodic copper exposure increased tissue residues but failed to perturb cardiac activity. Copper destabilized haemocyte lysosomal membranes in a exposure-dependent relationship showing a sensitive, rapid, indication of stress. Cytological biomarkers from P. viridis have potential as indices of pollutant exposure and could facilitate effective biomonitoring of water quality throughout South-east Asia.  相似文献

Due to their close proximity to the point sources of pollution, estuaries and harbours are exposed to metals. Mussels are used worldwide to monitor marine pollution due to their ability to take up contaminants and the ease of transplantation. Mussels were collected from two reference sites and transplanted in Richards Bay Harbour (2006 and 2010) and in three harbours. Transplanted and resident mussels were removed after 6 weeks exposure and metal concentrations were analysed using ICP-MS techniques. Transplanted mussels had higher metal concentrations than the resident mussels. This was attributed to regulation of metals by the resident mussels. Metal regulation was greater in mussels that are continuously submerged as opposed to those that undergo tidal influences. For monitoring it is essential that the reference population for transplantation is considered carefully as upwelling events in the pristine reference location results in significant increases in metal bioaccumulation.  相似文献

Mussels are used as sentinel organisms and bioindicators to evaluate the toxic effects of chemical pollutants in marine organisms, especially heavy metals, representing an important tool for biomonitoring environmental pollution in coastal areas. Antioxidant defence enzymes play an important role in cellular antioxidant defence systems and protect from oxidative damage by reactive oxygen species (ROS). Indigenous mussels Mytilus galloprovincialis of the Saronikos Gulf of Greece were used for monitoring heavy metal pollution in three polluted sites in the area and in one unpolluted site. Seasonal variations of the activity of antioxidant defence enzymes, superoxide dismutase (SOD) and catalase (CAT), as well as lipid peroxidation (LP) were measured as biomarkers in a period of three years in relation to concentrations of trace metals in their gills and mantle and compared to mussels from an unpolluted sampling site. SOD activity increased at least 2 fold at the polluted sites when compared to the control site (the high activity was recorded in the spring time). CAT activity was increased 2-3 times at the polluted sites, with high activity in the winter and spring time, compared to the control site. LP concentration was twice higher at the polluted sites, following the same seasonal pattern. Trace metals contents in mussels collected at polluted sites were 3-4 fold higher compared to the control site and showed moderate variations along the months, with a winter maximum followed by a summer pre-spawning minimum matching the seasonal trends of temperature and salinity. Our results showed that metal pollution in the Elefsis Bay (the most polluted coastal area) causes relatively medium levels of oxidative stress in tissues of mussels due to cellular oxy-radical generation. This study, which is the first in the area, showed that seasonal variations of the activity of antioxidant defence enzymes and LP concentrations in mussels can be used as potential biomarkers of toxicity for long-term monitoring in marine coastal ecosystems.  相似文献

The published literature on marine pollution monitoring research in South Africa from 1960 to present was evaluated. There has been a general decline in the number of papers from the 1980s and this can be linked to the absence of a marine pollution monitoring programme in South Africa. General trends observed were that contaminant exposure monitoring of metals predominates the research conducted to date. Monitoring results indicate that there has been a general decrease in metal concentrations in South African coastal waters and concentrations of metals and most organics in mussels are lower than in other industrialised nations. This is reflected in the general pristine nature and high biodiversity of the South African coastline. The establishment of a national marine pollution monitoring framework would stimulate marine pollution research.  相似文献

Biomarkers, which can detect changes at the biochemical level, have been used for many years as early warning tools in environmental monitoring. In order to distinguish between natural variability and the potential effects of anthropogenic pollution, it is essential to identify background levels and establish the potential influence of abiotic (season, temperature and salinity) and biotic (gametogenesis) factors. In this study, we examined various biomarkers of stress (glutathione S-transferase (GST)), reproduction (vitellin-like proteins) and damage (lipid peroxidation (LPO) and DNA damage (DNA)) in marine mussels (Mytilus spp.) from a known pristine hybrid zone. Levels of all biomarker expression varied between seasons and appeared to be linked to the reproductive cycle. Oxidative stress in winter, with low GST expression and a higher expression of LPO and DNA damage displayed could be explained by low food availability. These data provide vital baseline information for future ecotoxicological and environmental monitoring studies.  相似文献

Oxidative stress related investigations to monitor the impact of the pollutant discharges into the Venice lagoon (Italy) originating from anthropogenic activities (raw sewage water, agricultural and industrial effluents, oil tanker traffic), on marine organisms have classically been carried out by analyzing specific, single antioxidants (i.e. catalase, superoxide dismutase). In this paper, two studies are reported where the total oxyradical scavenging capacity assay (TOSC) was selected and measured toward peroxyl, and hydroxyl free radicals, and peroxynitrite in mussels (Mytilus galloprovincialis) transplanted into the Venice city and throughout the lagoon to measure the biological effects of anthropogenic activities. In the first experiment, mussels from a clean site (farm) were transplanted to the urban area of Venice for 0, 1, 2 and 4 weeks; cytosolic TOSC toward peroxyl and hydroxyl free radicals, and peroxynitrite revealed that the transplantation process caused a stress (handling stress, anoxia, oxidative burst) resulting in a reduction of TOSC in both control and urban sites, therefore, preventing clear interpretation of the data after one week. At week 2, a significant TOSC reduction (P < 0.05) toward peroxyl and hydroxyl radicals in the urban site revealed that mussels experienced oxidative pressure exerted by pollutants. Most TOSC values returned to initial levels at week 4; however, TOSC induction was noticed in the control group toward peroxyl and hydroxyl radicals while in the exposed group it was not indicating an inhibition of the oxidative metabolism. In the second experiment, mussels were deployed at seven different sites throughout the lagoon. After five weeks of exposure, significant TOSC reduction was measured (P < 0.05) toward peroxyl for Palude della Rosa, Chioggia and Valle Millecampi, toward hydroxyl radicals for Valle Millecampi and Campalto and toward peroxynitrite for Valle Millecampi. Although these data indicate a depletion of the low molecular weight scavengers, additional biomarkers are needed to draw a conclusion on the health of the mussels. TOSC was proved to be an interesting health index parameter to measure pollution impact in a transplantation study provided that the mussels are exposed for two weeks and a control is run in parallel.  相似文献

In this study, a suite of sublethal stress biomarkers were analysed in the sediment-dwelling bivalve, Austrovenus stutchburyi, with a view to using them as pollution monitoring tools. Shellfish were transplanted from a relatively uncontaminated site to four sites in a different harbour with different levels of contaminants in the sediment, where they were exposed in-situ for 2 and 8 weeks. A range of sublethal stress biomarkers were selected on the basis of their potential to provide ecologically relevant information (adenylate energy charge, total adenylate nucleotide pool, glycogen and RNA). Adenylate energy charge was lower at contaminated sites after 2 weeks, but recovered after 8 weeks, whereas total adenylate nucleotide pool was depressed after 8 weeks at these sites. Glycogen and RNA were found to be significantly higher at the uncontaminated site after both 2 and 8 weeks. These data suggest the potential application of biomarkers in coastal pollution monitoring.  相似文献

The physiological and biochemical characteristics of tissue samples of the marine mussel, Donax trunculus, from an oil-polluted site (Qiryat Yam) and from a site adjacent to an industrial factory (Frutarom) producing polyvinyl chloride (PVC) were compared with samples from a clean reference site (Akko). All sites were located along the sandy shores of the Israeli Mediterranean Sea. The mussels from the oil-polluted site showed increased activity of the system of active transport of organic anions (SATOA) in the gills and the renocardial organ and also of the multixenobiotic resistance transporter (MXR) in the gills. In contrast, samples collected near the PVC factory showed a decrease in SATOA activity and no increase in the activity of MXR in the gills. Specimens from the reference site demonstrated a redox state of equilibrium between energy production and utilization, while in Donax from both the oil polluted and the PVC-polluted sites, the mitochondrial redox state reflected intensive consumption of energy. No significant changes were found in the activity of reduced glutathione s-transferase (GST) in the cytosolic fraction of the digestive gland of Donax collected from any of the three sites. The data demonstrate a differential increase in the anti-chemical defense systems and an intensification of energy metabolism in the mussels exposed to pollution.  相似文献

Over the last three decades there has been a significant decline in the number of marine pollution monitoring-related studies in South Africa. Thus, the current study was conducted to assess the current state of metal contamination within the South African marine environment through the validation of the artificial mussel (AM). Indigenous reference mussels (Perna perna) were deployed alongside the passive device within the South African marine environment for a 6 week exposure period. Analysis of metal uptake (Cd, Cr, Cu, Pb and Zn) was determined by filtration and elution of the AM chelex resins, microwave digestion of the transplanted mussels, and determination of their metal concentrations by ICP-MS and ICP-OES analysis. Uptake patterns between the AM and transplanted mussels showed significant temporal and spatial correlation for the majority of the elements analysed. While the AM provided relevant and complementary information on the dissolved metal concentrations, limitations were also observed.  相似文献

In this study, the Active BioMonitoring (ABM) approach has been investigated using species of indigenous (from a pristine site) and transplanted (to a polluted area) hard clam Mercenaria mercenaria as an indicator organism of water/sediment pollution in Tampa Bay, Florida, a highly urbanized and industrialized subtropical estuary. A range of possible induced stress indices was selected and used on the basis of potential pollutant toxicity mechanisms including specific enzymatic responses (MFO as BPH and NADPH cytochrome c reductase, antioxidant enzymes as SOD and caltalase, PDH) and general cellular damage (lysosomal latency, histopathological lesions). The organic sediment contamination compared to the animal body burden revealed bioaccumulation activity in transplanted clams. The marked increases of BPH and catalase activities, together with the significant reduction of lysosomal membrane stability in transplanted animals, suggested their potential for application as biomarkers in subtropical coastal pollution monitoring.  相似文献

Bivalves are commonly used to detect metal pollution in the marine environment. Commercially cultured Milky oysters (Saccostrea commercialis) were transplanted in various sites along the North Queensland coast and analyzed for two metals of potentially anthropogenic origin (Cd, Zn). To provide additional information, naturally occurring Black Lip oysters (Saccostrea echinata) were also collected at the transplantation sites. The study demonstrated that the oysters species transplanted are good bioindicators of these metal concentrations in tropical waters, sensitive to variations in the environment at concentrations which are much smaller than pollution signals commonly reported for temperate waters. Three transplant experiments were carried out from May 1999 to February 2000. Milky oysters transplanted to offshore areas (Orpheus Is., Kelso Reef) accumulated Cd in the soft parts whereas oysters sampled from cages placed in Ross Creek and the Herbert River estuaries showed a decrease in Cd concentration, which resulted from an increase in dry weight. Dry weight appeared to be an important covariant affecting Cd concentration in the oysters whereas it does not unambiguously affect Zn concentrations. For the duration of the experiments, oysters sampled from the Magnetic Is. reference site showed effectively constant Cd concentrations and total Cd contents which indicates that any seasonal cycle affecting metal concentration is weak. It was found that Cd accumulation in oysters increased as ambient dissolved Cd concentration decreased, from which it was concluded that for these oysters, the predominant source of Cd was from the particulate phase rather than the dissolved phase.  相似文献

Within the framework of the biointegrator network (RINBIO), 92 man-made cages containing mussels (Mytilus galloprovincialis), distributed over 1800 km of the French Mediterranean coast, made it possible to assess chemical contamination by heavy metals (Cd, Hg, Zn, Pb, Cu, Ni, Cr, As) and organic compounds (DDT, PCBs, HAP). The caging technique was adopted to compensate for the scarcity of natural shellfish stocks in the Mediterranean and to enable comparison of the sites regardless of their physicochemical and trophic characteristics. Models linking a biometric parameter (the condition index) to pollutant levels make it possible to adjust raw data on contamination for a reference individual, by making a clear distinction between physiological factors (growth) and environmental ones. The results demonstrate significant levels of pollutants in sites heretofore unsampled; the pollution occurs at limited areas, and the sources have been identified. Moreover, average contamination levels are comparable to those measured in wild species and in other areas. Overall, the research demonstrates the reliability of this methodological approach for monitoring marine pollution, especially in the Mediterranean.  相似文献

Baseline information is presented on embryo malformation rate and biomarkers in fish as indicators of sub-lethal stress caused by pollution in coastal waters of Xiamen, PR China. Fish and eggs were sampled from several areas in Xiamen coastal waters (Xiamen Harbour, Maluan and Tongan Bays and East Channel), where varying levels of pollutant input have been documented. Comparative sampling was done at a "cleaner" reference site at Dongshan Island. Embryonic malformation rates, which indicate general water quality, varied with location and species of fish, and exceeded background levels for unpolluted waters (assumed approximately 5%) by up to eightfold at some sites. Generally, sites around Xiamen Harbour show signs of poor water quality having highest mean levels of embryo deformity (20-30%) and these decreased towards open waters (Tongan Bay, Eastern Channel) where abnormalities approached background levels. An indication that toxic contaminants may be having a localised effect in the region, particularly in the harbour was reinforced by the biomarker assays. However, activities of the biomarkers ethoxyresorufin O-deethylase (EROD) and glutathione S-transferase in fish livers indicate no clear pattern, and there is no evidence that fish from the four sampling areas have been more or less exposed to PAHs and other compounds that induce these biomarkers. Antioxidant biomarkers (glutathione peroxidase, catalase, superoxide dismutase, and reduced glutathione) suggest that exposure to xenobiotics appears to be lowest in Dongshan and Maluan and highest in the harbour and Tongan. Inhibition of acetylcholinesterase in fish muscle indicated possible effects by organophosphate and carbamate pesticides in Xiamen waters and these effects may be greatest in the area of the harbour.  相似文献

Impacts of anthropogenic pollution on marine ecosystems are being addressed by legislation to protect and restore coastal and transitional waters. A range of biological measures have been investigated for their ability to indicate anthropogenic disturbance in subtidal soft-sediment habitats, but little work to date has focussed in intertidal habitats. This study investigated the sensitivity of communities, individual taxa, diversity indices and biotic indices to nutrient and organic enrichment in intertidal soft-sediment habitats. Variation in macrofaunal communities was more strongly associated with anthropogenic stressors than with natural environmental variation. Two multimetric indices, M-AMBI and IQI, were more closely associated with nutrient and organic pollution than the AMBI and ITI indices. Intertidal monitoring based on existing monitoring tools offers a cost effective alternative to subtidal monitoring and has potential to form the basis for an ecosystem level approach.  相似文献

Samples were also collected at a “cleaner” reference site near Dongshan Island for comparison. Clearance rate, absorption efficiency and oxygen consumption of the mussels were measured under controlled laboratory condition; organic pollutants in their tissues were also analyzed. Scope for growth (SFG) was employed as a general biomarker to evaluate the stress of pollutants. Specimens from Tong’an site had the lowest SFG values (1.14 J/h/g); while specimens from the I’Maluan site and Huoshao site had SFG values of 5.01 and 6.72 J/h/g, respectively. Specimens from Dongshan (reference site) had a relatively high SFG value of 10.96 J/h/g. There was a significant negative correlation between the SFG of the different populations of green lipped mussels and the concentration of DDT in their tissues.  相似文献

Contamination of persistent organochlorines (OCs) such as PCBs (polychlorinated biphenyls), DDT and its metabolites (DDTs), HCH (hexachlorocyclohexane) isomers (HCHs), chlordane compounds (CHLs), and HCB (hexachlorobenzene) were examined in mussels collected from coastal waters of Asian countries such as Cambodia, China, Hong Kong, India, Indonesia, Japan, Korea, Malaysia, Philippines, Far East Russia, Singapore, and Vietnam in 1994, 1997, 1998, 1999, and 2001 to elucidate the contamination status, distribution and possible pollution sources and to assess the risks on aquatic organisms and human. OCs were detected in all mussels collected from all the sampling sites investigated. Considerable residue levels of p,p(')-DDT and alpha-HCH were found in mussels and the concentrations of DDTs and HCHs found in mussels from Asian developing countries were higher than those in developed nations suggesting present usage of DDTs and HCHs along the coastal waters of Asian developing countries. On the other hand, lower concentrations of PCBs detected in mussels from Asian developing countries than those in developed countries indicate that PCBs contamination in mussels is strongly related to industrial and activities. To our knowledge, this is a first comprehensive report on monitoring OCs pollution in the Asia-Pacific region.  相似文献

The artificial mussel (AM), a novel chemical sampling device, has been developed for monitoring dissolved trace metals in marine environments. The AM consists of Chelex-100 suspended in artificial seawater within Perspex tubing and enclosed with semi-permeable polyacrylamide gel at both ends. To validate the field performance of the AM in temperate waters, we deployed AMs alongside transplanted blue mussels Mytilus edulis in coastal environments in Scotland (Holy Loch, Loch Fyne, Loch Striven and Millport) and Iceland (Reykjavikurh?fn, Gufunes, South of thornerney, Hofsvik, Hvalfj?rethur and Sandgerethi) for monitoring trace metals. While uptake patterns of Cd between the AM and M. edulis were highly comparable, discrepancies were found in the accumulation profiles of the other metals (Cu, Cr, Pb and Zn), in particular Zn. Nonetheless, the AMs gave a better resolution to accurately reveal the spatial difference in dissolved metal contamination when compared with M. edulis. AMs complement the use of mussels since AMs indicate dissolved metals in seawater, whereas uptake by mussels indicates a mixture of dissolved and particulate metals. Our results also indicated that historical metal exposure of the transplanted M. edulis could significantly confound their metal concentrations especially when the deployment period was short (i.e. <34d). This study suggested that the AM can overcome problems associated with variable biological attributes and pre-exposure history in the mussel, and provides a standardized and representative time-integrated estimate of dissolved metal concentrations in different marine environments.  相似文献

Lysosomal membrane stability, the content of neutral lipids and lipofuscin as cellular biomarkers were measured in the digestive gland of mussels, Mytilus galloprovincialis Lam., during a one year survey at selected sites along the coast of the northern Adriatic. The ability of mussels to survive in air as a general physiological response was tested at the same sites. Annual changes of sea temperature, salinity, O(2) saturation, pH and chlorophyll a concentration were measured and the effects of these environmental factors on the biomarkers selected for this study were determined. Survival of mussels after aerial exposure was significantly (p < 0.05) reduced in mussels collected from the contaminated harbors of Pula and Rijeka compared to clean control sites. Lysosomal membrane stability of mussels from urban and industrial areas was also lower indicating exposure to different environmental contaminants. At the same sites, the content of neutral lipids and lipofuscin in digestive cells was significantly higher compared to the reference sites, suggesting alterations in lipid metabolism and contaminant-induced peroxidation of cell membranes. The greatest influence of natural environmental factors was observed for the aerial survival of mussels and the neutral lipids and lipofuscin content in the digestive gland, indicating that the annual cycles of biomarker responses should be well understood for a correct interpretation of biomonitoring results.  相似文献
