Studies on the diversity and distribution of bacterial populations will improve the overall understanding of the global patterns of marine bacteria and help to comprehend local biochemical processes and environments. We evaluated the composition and the dynamics of bacterial communities in the sediment of Jiaozhou Bay (China) using PCR-denaturing gradient gel electrophoresis (DGGE). Sediment samples were collected from 10 different sites in May, August, and November 2008 and in February 2009. There was significant temporal variation in bacterial community composition at all sites. However, the spatial variation was very small. The DGGE analyses of bacterial communities were used to divide the 10 stations into three types. Canonical correspondence analysis (CCA) revealed that the changes in bacterial communities were driven by sediment properties. Sequence analysis of DGGE band-derived 16S rRNA gene fragments revealed that the dominant bacterial groups in the sediment were of the classes γ-proteobacteria and δ-proteobacteria and phyla Bacteroidetes and Nitrospirae. Our results provide considerable insight into the bacterial community structure in Jiaozhou Bay, China.  相似文献

The seasonal dynamics of a crustacean zooplankton community in Erhai Lake was investigated from May 2010 to April 2011. In total, 11 species were recorded, including six (6 genera) cladoceran and five (5 genera) copepod species. The crustacean zooplankton densities ranged from 24.3 to 155.4 ind./L. In winter and spring, the large-bodied cladoceran Daphnia galeata dominated the crustacean plankton community. In summer and autumn, when the colonial or filamentous algae dominated the phytoplankton communities, the small-bodied species (e.g. Bosminafatalis, Ceriodaphnia quadrangular, and Mesocyclops leuckarti) replaced the large-bodied ones. One-way ANOVA and redundancy analysis revealed that community structure was dependent upon total nitrogen, total phosphorus, water temperature, transparency, and the biomass of small algae. The variation in both phytoplankton structure and environmental variables were important factors in the seasonal succession of crustacean zooplankton structure in Erhai Lake.  相似文献

The intestinal bacteria of vertebrates form a close relationship with their host.External and internal conditions of the host,including its habitat,affect the intestinal bacterial community.Similarly,the intestinal bacterial community can,in turn,influence the host,particularly with respect to disease resistance.We compared the intestinal bacterial communities of grass carp that were collected from farm-ponds or a lake.We conducted denaturing gradient gel electrophoresis of amplified 16S rRNA genes,from which 66 different operational taxonomic units were identified.Using both the unweighted pair-group method with arithmetic means clustering and principal component analysis ordination,we found that the intestinal bacterial communities from the two groups of pond fish were clustered together and inset into the clusters of wild fish,except for DF-7,and there was no significant correlation between genetic diversity of grass carp and their intestinal bacterial communities(Mantel one-tailed test,R=0.157,P=0.175).Cetobacterium appeared more frequently in the intestine of grass carp collected from pond.A more thorough understanding of the role played by intestinal microbiota on fish health would be of considerable benefit to the aquaculture industry.  相似文献

Four soil types(peat, marsh, meadow, and sandy) in the Zoige Plateau of China are associated with the severity of wetland degradation. The effects of wetland degradation on the structure and abundance of fungal communities and cellulase activity were assessed in these 4 soil types at 3 depths using DGGE(Denatured Gradient Gel Electrophoresis), q PCR(Quantitative Real-time PCR),and 3,5-dinitrosalicylic acid assays. Cellulase activity and abundance of the fungal community declined in parallel to the level of wetland degradation(from least to most disturbed). DGGE analysis indicated a major shift in composition of fungal communities among the4 soil types consistent with the level of degradation.Water content(WC), organic carbon(OC), total nitrogen(TN), total phosphorus(TP), available nitrogen(AN), and available phosphorus(AP) were strongly correlated with cellulase activity and the structure and abundance of the fungal community.The results indicate that soil physicochemical properties(WC, OC, TN, TP, AN, and AP), cellulase activity, and diversity and abundance of fungal communities are sensitive indicators of the relative level of wetland degradation. WC was the major factorinvolved in Zoige wetland degradation and lower WC levels contributed to declines in the abundance and diversity of the fungal community and reduction in cellulase activity.  相似文献

This study assessed the effects of changes in organic carbon content on soil bacterial community composition and diversity in the Antarctic Fildes Peninsula. 16 S rRNA gene sequencing was performed to investigate bacterial community composition. Firstly, we found that organic carbon(OrC) and nutrients showed an increasing trend in the lake area. Secondly, soil geochemistry changes affected microbial composition in the soil. Specifically, we found 3416 operational taxonomical units(OTUs) in 300 genera in five main phyla: Proteobacteria, Actinobacteria, Acidobacteria, Chloroflexi, and Bacteroidetes. Although the diversity was similar among the four sites, the composition was different. Among them, Hungateii content changed very significantly, from 16.67% to 33.33%. Canonical correspondence analysis showed that most measured geochemical factors were relevant in structuring microbiomes, and organic carbon concentration showed the highest correlation, followed by NO_(3-)~-N. Hungateii was significantly correlated with the content of organic carbon. Our finding suggested organic carbon played an important role in soil bacterial communities of the Antarctic coastal lake region.  相似文献

Characteristics of a microbial community are important as they indicate the status of aquatic ecosystems. In the present study, the metabolic and phylogenetic profile of the bacterioplankton community in Guishan coastal water(Pearl River Estuary), South China Sea, at 12 sites(S1–S12) were explored by community-level physiological profiling(CLPP) with BIOLOG Eco-plate and denaturing gradient gel electrophoresis(DGGE). Our results showed that the core mariculture area(S6, S7 and S8) and the sites associating with human activity and sewage discharge(S11 and S12) had higher microbial metabolic capability and bacterial community diversity than others(S1–5, S9–10). Especially, the diversity index of S11 and S12 calculated from both CLPP and DGGE data(H 3.2) was higher than that of others as sewage discharge may increase water nitrogen and phosphorus nutrient. The bacterial community structure of S6, S8, S11 and S12 was greatly influenced by total phosphorous, salinity and total nitrogen. Based on DGGE fingerprinting, proteobacteria, especially γ- and α-proteobacteria, were found dominant at all sites. In conclusion, the aquaculture area and wharf had high microbial metabolic capability. The structure and composition of bacterial community were closely related to the level of phosphorus, salinity and nitrogen.  相似文献

An artificial oyster shell reef was deployed in Rongcheng Bay, East China. However, the effects of this reef on the surrounding macrobenthic communities were unknown. We compared sedimentary factors, macrobenthic biomass, abundance, and community composition and ecological indicators between the reef and non-reef areas over a one year period. The mean values for chlorophyll a (Chl a), total organic matter (TOM), total organic carbon (TOC), and total nitrogen (TN) content in surface sediments in the reef area were slightly higher than those in the non-reef area. The Chl a levels differed significantly between the two areas, but the TOM, TOC, and TN were not significantly different. The abundance of crustaceans was significantly different between the two areas, but the abundance and biomass ofpolychaetes, echinoderms, mollusk did not differ significantly. The permutational multivariate analysis of variance (PERMANOVA) revealed that the macrobenthic community differed significantly through time and analysis of similarity multivariate analyses (ANOSIM) revealed that the macrobenthic community differed significantly in some months. The ecological indicators revealed that the environmental quality of the reef area was slightly better than that of the non-reef area. Overall, our results suggest that the artificial oyster shell reef may change the macrobenthic community and the quality of the environment. Despite the lack of an effect in the short term, long-term monitoring is still needed to evaluate the effects of artificial oyster shell reefs on macrobenthic communities.  相似文献

Among many reports investigating microbial diversity from environmental samples with denaturing gradient gel electrophoresis (DGGE), limited attention has been given to the effects of universal primers and DNA extraction on the outcome of DGGE analysis. In this study, these effects were tested with 16S rRNA gene-based DGGE on a bacterial community from farming water samples. The results indicate that the number of discernable bands in the DGGE fingerprint differed with the primer pairs used; the bands produced by 63f/518r, 341f/926r and 933f/1387r primer pairs were obviously fewer than those by 968f/1401r. Also, we found that each DNA extraction method resulted in different community profiles, reflected by the number and intensity of bands in the DGGE fingerprint. Furthermore, the main bands (theoretically representing dominant bacteria) differed with the extraction methods applied. It is therefore believed that the effects of universal primers and DNA extraction should be given more attention and carefully chosen before performing an investigation into a new environment with DGGE.  相似文献

The diversity, community structure and seasonal variation in demersal nekton off the Changjiang (Yangtze) River estuary was evaluated using monthly trawl survey data, collected between December 2008 and November 2009. A total of 95 species (56 teleosts, 11 cephalopods, and 28 decapod crustaceans) from 69 genera, 49 families and 15 orders were collected. These species could be classified into six groups on the basis of temporal distribution patterns. The resident crab Ovalipes punctatus dominated the community, both in number and biomass. A clear seasonal succession was observed in the species composition. Cluster analysis revealed three primary seasonal groups corresponding to the samples collected in winter-spring, late spring-summer and late summer-autumn. The highest biomass and lowest diversity were observed in summer, while the lowest biomass and highest diversity in winter. The abundance-biomass comparison curves and community composition suggested that the investigated community was moderately disturbed. The results suggest that reduction in fishing pressure and in the degree of seasonal hypoxia are essential for sustainable resource management off the Changjiang River estuary.  相似文献

To identify impact factors on the distribution and characters of natural plants community in reclamation area, with survey data from 67 plant quadrats in July 2009, soil properties data from 216 sampling points in April 2009, and TM (30 m) data in 2006, the composition and characteristics of natural plants community in different time of the Fengxian area in the Changjiang (Yangtze) River estuary were analyzed with two-way indicator species analysis (TWINSPAN), multivariate analysis of variance (MANOVA), detrended canonical correspondence analysis (DCCA) and canonical correspondence analysis (CCA). The results show that: 1) The plant communities in the reclaimed area are mainly mesophytes and helophytic-mesophytic transitional communities, showing a gradient distribution trend with the change in reclamation years. Species richness (MA), species diversity (H) and above-ground biomass also increase with the increase of reclamation years. Nevertheless, they appear to decline slightly in the middle and late reclamation period (> 30 years). 2) With the rise in land use levels, the changes in species richness and species diversity tend to increase at first and then decrease; species dominance (D), however, tends to decline; and above-ground biomass increases slightly. 3) The distribution of the plant community is mainly influenced by the following factors: land use levels (R = 0.55, p < 0.05), soil moisture (R = 0.53, p < 0.05), soil salinity (R = 0.43, p < 0.05) and reclamation time (R = 0.40, p < 0.05).  相似文献

We characterized variations in bacterioplankton community composition(BCC) in mesocosms subject to three different treatments. Two groups contained fish(group one: Cyprinus carpio; group two: Hypophthalmichthys molitrix); and group three, the untreated mesocosm, was the control. Samples were taken seven times over a 49-d period, and BCC was analyzed by PCR-denaturing gradient gel electrophoresis(DGGE) and real-time quantitative PCR(q PCR). Results revealed that introduction of C. carpio and H. molitrix had a remarkable impact on the composition of bacterioplankton communities, and the BCC was significantly diff erent between each treatment. Sequencing of DGGE bands revealed that the bacterioplankton community in the different treatment groups was consistent at a taxonomic level, but differed in its abundance. H. molitrix promoted the richness of Alphaproteobacteria and Actinobacteria, while more bands affiliated to Cyanobacteria were detected in C. carpio mesocosms. The redundancy analysis(RDA) result demonstrated that the BCC was closely related to the bottom-up(total phosphorus, chlorophyll a, phytoplankton biomass) and top-down forces(biomass of copepods and cladocera) in C. carpio and control mesocosms, respectively. We found no evidence for top-down regulation of BCC by zooplankton in H. molitrix mesocosms, while grazing by protozoa(heterotrophic nanoflagellates, ciliates) became the major way to regulate BCC. Total bacterioplankton abundances were significantly higher in C. carpio mesocosms because of high nutrient concentration and suspended solids. Our study provided insights into the relationship between fish and bacterioplankton at species level, leading to a deep understanding of the function of the microbial loop and the aquatic ecosystem.  相似文献

Soil microbial communities and enzyme activities play key roles in soil ecosystems. Both are sensitive to changes in environmental factors,including seasonal temperature, precipitation variations and soil properties. To understand the interactive mechanisms of seasonal changes that affect soil microbial communities and enzyme activities in a subtropical masson pine(Pinus massoniana) forest, we investigated the soil microbial community structure and enzyme activities to identify the effect of seasonal changes on the soil microbial community for two years in Jinyun Mountain National Nature Reserve, Chongqing, China. The soil microbial community structure was investigated using phospholipid fatty acids(PLFAs). The results indicated that a total of 36 different PLFAs were identified, and 16:0 was found in the highest proportions in the four seasons, moreover, the total PLFAs abundance were highest in spring and lowest in winter. Bacteria and actinomycetes were the dominant types in the study area. Seasonal changes also had a significant(P 0.05) influence on the soil enzyme activity. The maximum and minimum values of the invertase and catalase activities were observed in autumn and winter, respectively. However, the maximum and minimum values of the urease and phosphatase acid enzymatic activities were found in spring and winter, respectively. Canonical correspondence analysis(CCA) analysis revealed that the seasonal shifts in soil community composition and enzyme activities were relatively more sensitive to soil moisture and temperature, but the microbial community structure and enzyme activity were not correlated with soil pH in the study region. This study highlights how the seasonal variations affect the microbial community and function(enzyme activity)to better understand and predict microbial responses to future climate regimes in subtropical area.  相似文献

This study aims to investigate the seasonal variations in copepod community structure and prosome length of dominant species from March 2009 to January 2010 around artificial reefs in Xiaoshi Island,Yellow Sea,Weihai,China.Samples were collected using two types of plankton net(Model I and Model II) for different-sized copepods.The number of taxon was calculated from the data of both the net types,while the copepod abundance was done using the samples from Model II only.Sixteen species of planktonic copepods,including 5 dominant species,were recorded.Results reveal that Oithona similis was the first dominant species from March to June,and was replaced by Paracalanus parvus in September;both dominated the copepod community in January.Acartia hongi was the second dominant species from March to September.Centropages abdominalis was the third dominant species from March to June,and was replaced by O.similis in September and Corycaeus affinis in January.C.affinis was the fourth dominant species in September.Population density of the dominant copepods was compared with that of other similar regions.We found that the dominant species were mostly small copepods(<1 mm) except for adult Centrapages abdominalis.Seasonal variation in prosome length of O.similis,C.abdominalis,and C.affinis,and their copepodites were studied for the first time in China.For P.parvus and A.hongi,seasonal trends in prosome length variation were similar with those in Jiaozhou Bay,Yellow Sea,Qingdao,China,in a similar temperate domain.The results are helpful for future calculation of copepod biomass and production,and for investigation of the relationship between copepods and fish resources.  相似文献

Relationships between microbial communities and geochemical environments are important in marine microbial ecology and biogeochemistry. Although biogeochemical redox stratification has been well documented in marine sediments, its impact on microbial communities remains largely unknown. In this study, we applied denaturing gradient gel electrophoresis (DGGE) and clone library construction to investigate the diversity and stratification of bacterial communities in redox-stratified sandy reef sediments in a microcosm. A total of 88 Operational Taxonomic Units (OTU) were identified from 16S rRNA clone libraries constructed from sandy reef sediments in a laboratory microcosm. They were members of nine phyla and three candidate divisions, including Proteobacteria (Alpha-, Beta-, Gamma-, Delta-, and Epsilonproteobacteria), Actinobacteria, Acidobacteria, Bacteroidetes, Chloroflexi, Cyanobacteria, Firmicutes, Verrucomicrobia, Spirochaetes, and the candidate divisions WS3, SO31 and AO19. The vast majority of these phylotypes are related to clone sequences from other marine sediments, but OTUs of Epsilonproteobacteria and WS3 are reported for the first time from permeable marine sediments. Several other OTUs are potential new bacterial phylotypes because of their low similarity with reference sequences. Results from the 16S rRNA, gene clone sequence analyses suggested that bacterial communities exhibit clear stratification across large redox gradients in these sediments, with the highest diversity found in the anoxic layer (15–25 mm) and the least diversity in the suboxic layer (3–5 mm). Analysis of the nosZ, and amoA gene libraries also indicated the stratification of denitrifiers and nitrifiers, with their highest diversity being in the anoxic and oxic sediment layers, respectively. These results indicated that redox-stratification can affect the distribution of bacterial communities in sandy reef sediments.  相似文献

Macrobenthos samples were collected from the Yellow and East China Seas in four seasons during 2011 to 2012. The seasonal distribution of macrobenthos and its relationship with environmental factors were analyzed. A total of 562 macrobenthic species were identified, with polychaetes and mollusks accounting for 67% of the total number of species. A similarity percentage(SIMPER) analysis showed that the dominant species were bivalve mollusks in the Yellow Sea and small-sized polychaetes in the East China Sea. A two-factor analysis of variance showed significant seasonal variations in species number, density and diversity index, and significant regional differences of biomass and density. Two-factor community similarity analysis also showed significant seasonal and regional differences in macrobenthic communities. Canonical correspondence analysis indicated that the main environmental factors af fecting the macrobenthic communities were water depth, temperature, dissolved oxygen, and inorganic nitrogen. The results demonstrate significant regional differences and seasonal variations in macrobenthos in the two seas. Sediment properties and water mass characteristics are speculated to be the causes of regional differences.  相似文献

研究2005-2008年广东省湛江市英罗湾高桥3个红树林区(红坎、两头流、西村)虾类群落的变化特征,并分析虾类群落组成与水质指标(盐度,pH)和沉积物指标(pH、可溶性盐、有机质、黏土、粉砂、砂砾)的相关性。结果表明:3个区域捕获虾类13596尾,属3科5属9种,刀额新对虾是高桥虾类的优势种;虾类种类年际变化较大,年间相似度平均为70.4,区域间相似度平均为60.6;红坎虾类多样性较丰富,两头流虾类的优势种明显,各年优势度指数均达0.95以上;3个区域的主要生态因子年间差异不显著(P>0.05);红坎3个采样点的底质性状较复杂,其沉积物特性差异显著(P<0.05),两头流和西村的底质性状较为单一,其沉积物特性均差异不显著(P>0.05)。群落变化特征和沉积物性状有显著相关关系,表明高桥虾类的渔获量受人为因素影响,多样性和分布受环境影响。  相似文献

Soil fauna have been receiving more and more attention because they play an important role in nutrient cycling. However, there is a lack of information on soil arthropods in the forest-steppe ecotone in the mountainous region of northern Hebei, which makes it difficult to meet the need of protecting biodiversity in this area. Soil arthropod communities were investigated in the forest-steppe ecotone in northern Hebei province to provide basic information on changes in mountain soil fertility, which could promote the development of soil arthropod communities in mountain ecotones. From the preliminary identification, a total of 7994 individual soil arthropods were collected, which belonged to 25 groups, 6 classes and 24 orders. Acarina, Hymenoptera and Collembola were the dominant groups in the ecotone. The number of Acarina was higher than Collembola, and this phenomenon was obviously different from other areas in the same climate zone. The increased abundance of rare groups in the Forest zone with the richer vegetation, higher arthropod abundance and more substantial litter depth, could be interpreted as a reaction to the suitable soil environment and food supply. And these rare groups were sensitive to environmental changes, which could be regarded as biotic indicators for evaluating soil quality. The analysis of community diversity showed that the abundance index (d), the Shannon-Wiener index (H′), the evenness index (J) and the density-group index (DG) were significantly higher in the forest zone, lower in the forest-steppe zone, and lowest in the meadow-steppe zone. Seasonal variations in community composition correlated with changes in average air temperature and precipitation in this ecotone. Groups and individuals of soil arthropod communities in the three zones were present in greater numbers in the middle of the rainy season than in the early or late periods of the rainy season as a whole. At the same time, seasonal changes in soil arthropod communities from different plots were also influenced by habitat condition.  相似文献

To understand the relationship between the plankton community and environmental factors and water quality in the Bayuquan Port of Liaodong Bay, China, and investigations were carried out during six different periods (April 2009, April 2010, October 2011, April 2012, October 2012, and April 2013). This area was characterized by high levels of nutrient and suspended solids (SPS) during survey periods, and eutrophication led to the occurrence of red tides in April and October 2012 and April 2013. Our analyses revealed that the plankton communities of Bayuquan Port lacked stability and were affected seriously by external disturbance (e.g., oceanographic engineering and river runoff). Our data indicate that oil, dissolved inorganic nitrogen (DIN), SPS, and chlorophyll a (Chl-a) were key factors that regulated the phytoplankton and zooplankton communities. The partial redundancy analysis (partial RDA) suggested that oil and SPS were the most important environmental variables affecting the phytoplankton community in April 2010 and 2012, whereas DIN concentration played a governing role in zooplankton dynamics. Oil and Chl- a concentrations affected significantly the zooplankton community in October 2012. Therefore, the plankton communities could reflect both dynamic changes in coastal environmental factors and the ongoing eutrophication process caused by anthropogenic activities in this area.  相似文献

The knowledge of prey small fish stock, distribution and abundance is necessary to guide stocking of piscivorous fish for the biomanipulation in domestic tap water lakes. This study describes the current status of small fish community in Lake Kuilei (China), and examines the spatial and seasonal variations of the community in relation to key environmental factors. Based on submerged macrophyte cover and water depth, the lake was divided into five major habitats: (1) macrophyte covered shallow habitat of water depth < 2.00 m, (2) uncovered or less-covered shallow habitat (2.00 m–3.50 m), (3) uncovered medium shallow habitat (3.50 m–5.00 m), (4) uncovered medium deep habitat (5.00 m–6.50 m) and (5) uncovered deep habitat (6.50 m–8.50 m). The abundance and composition of small fish were monitored by benthic fykenet sampling from April 2013 to January 2014. A total of 2881 individuals belonging to 5 families and 21 species were collected. Based on their abundance (accounted for 88.96% of the total) and occurrence (more than 33.33%), Acheilognathus chankaensis, Acheilognathus macropterus, Microphysogobio microstomus, Pseudorasbora parva and Rhinogobius giurinus were recognized as dominant small fish species. The results of correlation analysis identified that species richness ( Sr ), Shannon-Wiener diversity index ( H′ ) and Margalef′s richness index ( D ) were significantly negatively correlated with water depth, but positively correlated with biomass of submerged macrophytes.Redundancy analysis (RDA) revealed that the spatial distributions of most small fishes were negatively associated with water depth. The details of these findings are beneficial to understanding the adaptation of the small fishes in degraded environments, and to developing suitable biomanipulation strategies for the management of fish resources and water quality in the lakes along the lower reach of the Changjiang (Yangtze) River basin.
