The early effects of chromosomal manipulation of eggs and sperm on the yields of trip-loid and gynogenefic diploid larvae of Paralichthys olivaceus were investigated. Triploidy was achieved by cold shocking fen.ilized eggs at 0 - 2℃ for 45 minutes duration 5 minutes after fen.ilization, and the in-duced triploidy rates were 31.2% - 50% and the relative hatching rates were 53.3% - 99%. Gynnge-aetic diploids were obtained when eggs were inseminated with irradiated sperm and cold shocked at 0 -2℃ for 45 minutes duration 5 minutes after fertihzation. The induced gynogenetic diploid rates and the relative hatching rates were 94 % - 96 % and 48.5 % - 68.5 % respectively. The embryonic development of the triploid experimental group and of the gynogenetic diploid experimental group was delayed at first compared with the control group. But from the gastrula stage, it was not delayed anymore. There were no significant differences in the growth of the triploid experimental group larvae and the control group larvae, and in the growth of the gynogenetic diploid experimental group larvae and the control group larvae ac-cording to Student‘s t-test (α = 0.05). The relationship between the early growth of the triploid experi-mental group larvae and that of gynogenetic diploid experimental group larvae was also studied.  相似文献

The living morphology, nuclear apparatus, infraciliature and silverline system of the poorly-known ciliatePseudocohnilembus hargisi Evans & Thompson, 1964, collected from a shrimp culture pond in Qingdao, China, were examined by observation of the live specimen; and by the pyridinated carbonate method and Chatton-Lwoff technique. The main characteristics: 13–14 (usually 13) longitudinal or slightly spirally arranged somatic kineties; single contractile vacuole with usually 2 (1–2) pores at posterior ends of the fourth and fifth somatic kineties to the right of the buccal cavity. Trumpet shell, Charonia sauliae, is an endangered and valuable species, but its artificial propagation protocol has not been successfully established. To estimate the possibility of cryopreservation for larvae of C. sauliae, which is a potential preparation for its artificial reproduction and further research, in this study a protocol for the cryopreservation of veliger larvae of trumpet shell was optimized. Through a two-step cryopreservation procedure, four kinds of cryoprotectants (ethylene glycol, 1, 2-propanediol, dimethyl sulfoxide and glycerol) were employed at three concentrations (1.0, 1.5 and 2.0 molL−1) respectively and survival rates of larvae were determined after a storage of 1h. The larvae frozen with these four cryoprotectants after 1 h storage were cultured, and then survival rates were determined at 24, 72 and 120 h after thawing. Dimethyl sulfoxide at a concentration of 1.5 molL−1 showed the best protective effect in all experiments (p<0.05). And survival rates of larvae frozen with dimethyl sulfoxide were determined after 1, 7 and 15 d of storage. The survival rates of larvae frozen with 1.5 molL−1 dimethyl sulfoxide after 1 h, 1 d, 7 d and 15 d of storage were 80.77% ±7.51%, 80.34% ±11.28%, 83.10% ±9.14% and 77.23% ±6.22% respectively. No significant differences in survival rates of larvae frozen with dimethyl sulfoxide were observed after various storage periods (p>0.05).  相似文献

A laboratory experiment was conducted to assess the bioaccumulation of Pb2+ and its effects on growth, morphology and pigment contents of Spirulina (Arthrospira) platensis. The specimen cultured in Zarrouk liquid medium was treated with various initial metal concentrations (0, 5, 10, 30, 50 and 100 μg mL−1). The growth of S. platensis was adversely affected by Pb2+ at high concentrations (30, 50 and 100 μg mL−1). However, at low concentrations (5 μg mL−1), Pb2+ could stimulate its growth slightly. The pigment contents (chlorophyll α and β carotene) were decreased in a dose-dependent manner. The highest reductions (67% and 53% respectively in chlorophyll α and β carotene) were observed in 100 μg mL−1 treatment group. The LC50 (96 h) of Pb2+ was measured as 75.34 μg mL−1. Apart from a few cases of filament breakages at elevated concentrations (50 and 100 μg mL−1), morphological abnormalities are not specific. Metal bioaccumulation increased with Pb2+ concentrations, but decreased with exposure time. The maximum accumulated amount was 188 mg g−1 dry weight. The bioconcentration factor (BCF) reached to a peak at day 2, followed by a gradual reduction for all the exposure concentrations. S. platensis is able to tolerate considerably high Pb2+ concentrations. Consequently it can be used as a potential species to remove heavy metal from contaminated waters.  相似文献

Analysis of cold tolerance on mangroveKandelia candel leaf growing in different soil salinity along Jiulong River Estuary in South China showed that the cold tolerance decreased as the increase of soil salinity. The lethal temperatures ofK. candel leaf were −10.4, −9.9 and −8.6 °C in Liaodong, Baijiao and Aotou, respectively. Under 1–2°C cold stress treatment on detached leaves ofK. candel, their caloric value gradually decreased, while electrolyte leakage gradually increased. The leaf's caloric value and electrolyte leakage in Aotou with higher soil salinity varied more largely than those in Liaodong with lower soil salinity. InK. candel leaf, total water content lowered a little, bound water content rose significantly and free water content dropped significantly with duration of cold stress. At the same time, reduction sugar, soluble sugar and starch content gradually decreased and sucrose content gradually increased. Bound water, free water and sucrose content inK. candel leaf from Aotou with higher soil salinity changed more slowly than those from Liaodong with lower soil salinity, but reduction sugar, soluble sugar and starch content inK. candel leaf from Aotou had faster variations than those from Liaodong. These data indicated that soil salinity can reduce cold tolerance ofK. candel leaf by increasing negative effect of salt ions in cell membrane, inhibiting variations of water content, and aggravating consumption of material and energy. The meiotic process in Noctiluca scintillans were observed under light microscope.Some abnormal cell divisions, incompletely separated “zoospores” and the changes of the zoospores are described in this paper. Together with the fmdings of field samplings and the previous results by other researchers, the process of meiosis in N. scintillans was supposed to be a pathway to reduce the extra high density of NH3-N within the cell in order to ensure normal population growth.  相似文献

In order to reveal transpiration rates of wetland plants and its relationships to micro- meterological factors in a mountain valley wetland, relative humidity, air temperature, leaf temperature, soil temperature, photo flux density and transpiration rates were measured once two hours in a Carex meyeriana wetland of the Changbai Mountain valley in dry (July) and wet (September) of 2003, respectively. Results showed that the tendency of "decreasing after increasing" was obvious in wet season. However, a relatively stable trend was observed for the transpiration in dry season.. Generally, the photon flux density of Carex meyeriana was higher in wet season than that in dry season. However, the variabilities of leaf temperature, air temperature and relative humidity were similar in both seasons. Higher transpiration rates of Carex meyeriana leaves were observed in July (varied from 40 to 150 mol.m^-2.s^-1) compared to those (varied from 7 to 14 mol.m^-2.s^-1) in September. Transpiration rates were significantly correlated with air temperature (P〈0.01), leaf temperature (P〈0.01), and wind speed (P〈0.05), but correlationship between relative humidity and photo flux density was not significant (P〈0.05).  相似文献

Isolated protoplasts from thalli ofPorphyra haitanensis andPorphyra yezoensis were treated with colchicine or irradiated by ultraviolet (UV). Several types of color variants were observed among the protoplast offspring. After treatment with colchicine: (1) 0.04–0.09% of red type variants inP. haitanensis were obtained; (2) The rate of red type variants and the variegated chimeral thalli composed of red type and wild type of sectors were 0.31–1.11% inP. yezoensis. After irradiation with UV: (1) 3.5–10.5% of red type variants inP. yezoensis were obtained: (2) 0.5–2.0% of red type variants and the variegated chimeral thalli composed of red type and wild type of sectors were obtained inP. haitanensis. Colchicine and UV’s mutangenic effects onP. yezoensis protoplasts were stronger than those onP. haitanensis protoplasts. The most efficient concentration of colchicine was 0.05%. The optimal length of UV-radiation was 1/2 min (radiation distance 5 cm). The red type variants induced by colchicine treatment grew faster than the wild type thalli. The clones of vegetative propagation from protoplasts of red type variants were still red type thalli. The red type variants will be good materials for genetic studies and improvement ofPorphyra strains.  相似文献

Twenty-four compounds including eight steroids (1–8), nine triterpenoids (9–16, 24), three flavonoids (20–22), and four benzenecarboxylic derivatives (17–19, 23) were isolated and identified from stems and twigs of medicinal mangrove plant Sonneratia caseolaris. The structures of the isolated compounds were determined by extensive analysis of their spectroscopic data. Among these metabolites, compounds 1, 4–20 and 22–24 were isolated and identified for the first time from S. caseolaris. In the in vitro cytotoxic assay against SMMC-7721 human hepatoma cells, compound 21 (3′,4′,5,7-tetrahydroxyflavone) exhibited significant activity with IC50 2.8 μg/mL, while oleanolic acid (14), 3,3′-di-O-methyl ether ellagic acid (18), and 3,3′,4-O-tri-O-methyl ether ellagic acid (19) showed weak activity. None of these compounds displayed significant antibacterial activites. The interspecific interactions between the rotifer Brachionus plicatilis and two harmful algal blooms (HAB) species were investigated experimentally by single culture method. B. plicatilis population and the growth of the two algae were compared at different algal cell densities. The results demonstrated that the B. plicatilis obtained sufficient nutrition from Prorocentrum donghaiense to support net population increase. With exposure to 2.5×104 cells mL−1 of P. donghaiense, the number of B. plicatilis increased faster than it did when exposed to other four algal densities (5, 10, 15 and 20 ×104 cells mL−1), and the increase rate of B. plicatilis population (r) at this algal density was 0.104 ± 0.015 rd−1. Cell densities of P. donghaiense decreased due to the grazing of B. plicatilis. In contrast, Heterosigma akashiwo had an adverse effect on B. plicatilis population and its growth was largely unaffected by rotifer grazing. In this case, B. plicatilis population decreased and H. akashiwo grew at a rate similar to that of the control.  相似文献

The effects of several nutritional factors on the growth and eicosapentaenoic acid (EPA) production of diatom Nitzschia laevis were studied. 4 LDM (quadrupled concentration of the nutrient salt) was the optimal concentration of nutrient salt for the growth and EPA production of N. laevis. The growth of N. laevis was inhibited when the glucose concentration was either lower than 10 gL−1 or higher than 15 gL−1. Both sodium nitrate and urea were good nitrogen sources for the growth and EPA production, while ammonium chloride seriously decreased the dry cell weight (DW) and the EPA content. Silicate seriously influenced the growth of N. laevis. The maximum DW of 2.34 gL−1 was obtained in the presence of 150 mgL−1 Na2SiO3·9H2O. The EPA content remained almost the same when the silicate concentration was lower than 150 mgL−1; however, higher silicate concentrations resulted in a steady decrease of EPA content. Low medium salinity (⩽29) did not seem to influence the DW of N. laevis, and high salinity resulted in a decrease of DW. The highest EPA content (4.08%) and yield (110 mgL−1) were observed at the salinity of 36 and 29, respectively.  相似文献

Morphology of the spermatozoon of Octopus ocellatus was studied by light, scanning electron, and transmission electron microscopes. Sperm are 600–700 μm long, with a large number of granules in diameter about 130 nm. Each spermatozoon is composed of a head, neck, and tail. The head is made up of an acrosomal complex anterior to the nucleus. The spiral acrosomal complex consists of an electron-lucent vesicle, lacuna, and an electron-dense acrosomal vesicle. Additionally, the spiral acrosomal vesicle has numerous equidistant striations, and is surrounded by many small granules (20 nm diameter). A long straight nucleus, which is electron-densed, has a deep posterior concavity, the nuclear vacuole. At the terminal end of the nucleus is a sleeve-like structure with a concave posterior nuclear fossa (PNF). The neck is short connecting the PNF. The basal body is located in the PNF and gives rise to the axoneme. This structure connects the head, neck, and tail. The tail is divided into a middle piece and a principal piece. The middle piece, having a 9+9+2 arrangement, is surrounded by a mitochondrial sheath and terminates by an electron-dense fibrous sheath. The principal piece is the longest part of the sperm with coarse fibers tapering posteriorly. The results of this study shall provide some useful information for artificial breeding of this species.  相似文献

The living morphology and infraciliature of two rare marine ciliates, Placus salinus Dietz, 1964 and Strombidium apolatum Wilbert and Song, 2005, collected from the coastal waters near Qingdao, China, were investigated by in vivo observation and protargol impregnation technique. The improved diagnosis for Placus salinus is as follows: medium-sized marine Placus, in vivo (50–60)μm×(30–40)μm; cell elliptical to barrel-shaped; 28–31 somatic kineties; single macronucleus usually ellipsoid and one micronucleus located in the indention of the macronucleus; one contractile vacuole posteriorly positioned. Strombidium apolatum is characterized by: marine strombidium (40–60)μm×(30–45)μmm in vivo, cordiform in shape with somewhat pointed posterior end and conspicuous apical protrusion; extrusomes prominent, about 15μm in length and evenly arranged along the circle kinety; about 16 collar and 5–6 buccal membranelles; one elongate macronucleus and one micronucleus; circle and ventral kineties consisting of about 53 and 45 dikinetids respectively.  相似文献

Heat shocks effectively produced triploids inPenaeus chinensis. Fertilized eggs heat shocked (28–32°C) for 8 to 16 minutes, starting from 8 to 20 minutes after spawning, resulted in triploidy induction rates of 39%–75%. Several triploid populations were cultured to 10 cm. In a triploid population, two kinds of ovaries were observed. Histological examination showed apparent differences between these two kinds of ovaries, whereas among male shrimp, there were no such differences. Vibrio harveyi cells (dose—<103 cells mL−1) and extracellular products (ECP; >25 μmg mL−1 of total protein concentration) destroyed haematopoietic cultures of Nephrops norvegicus within 24 h of exposure. Cytopathic effects (CPE) started after 4h of exposure to the bacterial cells, with some granularity in the cytoplasm, mostly in cells in the outer periphery of the explant growth. At the end of the infection, a considerable number of nuclei remained attached to the substrate, apparently unaffected. Following exposure to ECP, initial deterioration was observed at 2 h with the presence of granularity in the cytoplasm of<20% cells, and few cells displayed small vacuoles around the nuclei. Parallel results were obtained using whole animal experiments, with V. harveyi cells being lethal to nephrops within 24 h.  相似文献

The toxic effects of benzo[α]pyrene (B[α]P) at different concentrations (0.1, 0.5, 1, 2.5 and 7.5 μgL−1) on scallop (Chlamy farreri) immune system were studied. The results showed that B[α]P had significant toxic effects on the haemocyte counts, neutral red uptake, phagocytosis, bacteriolytic and antibacterial activity (P<0.05), while the seawater control and acetone control had no significant differences. The haemocyte counts, neutral red uptake, phagocytosis and bacteriolytic activity in all B[α]P treatment groups as well as antibacterial activity in groups of 0.5, 1, 2.5 and 7.5 μgL−1 B[α]P decreased significantly (P<0.05). Some of these indices tended to be stable on the sixth day and others on the ninth day, and the indices showed clear time- and concentration-response to B[α]P. Bacteriolytic activity in 0.1μgL−1 B[α]P treatment group and antibacterial activity in 0.1 μgL−1 and 0.5 μgL- B[α]P treatment groups increased at the beginning of exposure and reached their peaks on day 1 and day 6, respectively. Following that, both activities decreased gradually and became stable after day 9. When all the indices reached stability, they were significantly lower than those in control group (P<0.05), except for antibacterial activity in 0.1 μgL−1 B[α]P treatment group (P>0.05). Thus, B[α] has evident toxic effects on scallop immune system, which supports the view that a relationship exists between pollution and immunomodulation in aquatic organisms.  相似文献
