Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) was applied in the detection of the end point temperature (EPT) of thermal denatured protein in fish and meat in this study. It was also used in studying the thermal denatured temperature range of proteins in salmon and chicken meat. The results show that the temperature ranges of denatured proteins were from 65 ℃ to 75 ℃ , and these temperature ranges were influenced by the processing methods. 顶进力是顶管工程设计和施工中最重要的参数之一, 顶进力计算的精确性将直接影响顶管的工程的成败。随着顶管施工工艺越来越多的应用于岩石地层, 但鲜有关于岩石地层中顶管顶进力的计算模型。为了更加准确地计算岩石地层中的迎面阻力, 假设掌子面稳定, 通过分析岩石顶管机刀盘滚刀破岩机理、受力模型和影响规律, 在此基础上推导出岩石地层顶管迎面阻力计算公式。结果表明: 岩石顶管中, 岩石抗压强度和抗剪强度以及顶管机滚刀切入深度直接决定了迎面阻力的大小, 实测迎面阻力除个别点在计算值上下限以外, 其余与公式预测值相一致, 证明其具有适用性。   相似文献

The new polyenzyme method for making gravy from Harengula zunasi offal involves protein enzymolysis with flavorase after proper alkaline and neutral protease levels were established by orthogonal trials to select the best hydrolytic conditions for processing offal with alkaline and neutral protease. The conditions for the polyenzyme method were pH of 7.0, temperature of 50℃ , alkaline and neutral protease concentration of 1.5% respectively, hydrolysis time of 120 min, andflavorase concentration of 2.0% , for 60 min. The new gravy-making technology yields a nutritious and delicious gravy containing 40.3 % of total essential amino acids, with delicious amino acids Glu, Asp, Gly, Ala, Pro and Ser comprising 49.5% , total and amino nitrogen being respectively1.9 and 1.1 g/100 g (amino acid nitrogen being 61.0% of total nitrogen), The polyenzyme method was used to make 14.8% protein gravy from Harengula zunasi offal. As fish is one source of the ‘big eight’ food allergens,the prevalence of fish allergy has increased over the past few years.In order to better understand fish allergy,it is necessary to identify fish allergens.Based on the sera from fish-allergenic patients,a 28 kDa protein from local mackerel (Scomber japonicus),which has not been reported as a fish allergen,was found to be reactive with most of the patients’ sera.The 28 kDa protein was analyzed by MALDI-TOF-MS (Matrix-Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometry).Mascot search in NCBI database (Date: 08/07/2010) showed that the top protein matched,i.e.triosephosphate isomerase (TPI) from Xiphophorus maculatus and Poecilia reticulata,had a mowse (molecular weight search) score of 98.In addition,TPI from Epinephelus coioides also matched this mackerel protein with a mowse score of 96.Because TPI is con-sidered as an allergen in other non-fish organisms,such as lychee,wheat,latex,archaeopotamobius (Archaeopotamobius sibiriensis) and crangon (Crangon crangon),we consider that it may also be an allergen in mackerel.  相似文献

Status and historical changes in the fish community in Erhai Lake   总被引:1,自引:0,他引:1  
Erhai Lake is the second largest freshwater lake on the Yunnan Plateau, Southwest China. In recent decades, a number of exotic fish species have been introduced into the lake and the fish community has changed considerably. We evaluated the status of the fish community based on surveys with multimesh gillnet, trap net, and benthic fyke-net between May 2009 and April 2012. In addition, we evaluated the change in the community using historical data (1952–2010) describing the fish community and fishery harvest. The current fish community is dominated by small-sized fishes, including Pseudorasbora parva, Rhinogobius giurinus, Micropercops swinhonis, Hemiculter leucisculus, and Rhinogobius cliffordpopei. These accounted for 87.7% of the 22 546 total specimens collected. Omnivorous and carnivorous species dominated the community. A canonical correspondence analysis (CCA) plot revealed that the distribution of fishes in the lake is influenced by aquatic plants, water temperature, pH, and season. The abundance of indigenous species has declined sharply, and a majority of endemic species have been extirpated from the lake (a decrease from seven to two species). In contrast, the number of exotic species has increased since the 1960s to a total of 22 at present. The fishery harvest decreased initially following the 1960s, but has since increased due to the introduction of non-native fish and stocking of native fish. The fishery harvest was significantly correlated with total nitrogen, not total phosphorus, during the past 20 years. Based on our results, we discuss recommendations for the restoration and conservation of the fish resources in Erhai Lake.  相似文献

In order to determine the effect of twine thickness on the size-selectivity of the driftnet used for the yellow croaker, size-selectivity tests were conducted with three different twine thicknesses (monofilament diameters of 0.279 mm (number’s method; No. 3), 0.321 mm (No. 4), and 0.360 mm (No. 5)) of driftnets for the yellow croaker in the seas around Chooja-do, Jeju Islands. The selectivity curve was estimated by using Kitahara’s method. In order to determine the physical properties of the twine used in the experimental fishing nets, we measured the breaking load, elongation, and stiffness under both dry and wet conditions. In terms of physical properties, the thinnest twine (No. 3) had the strongest breaking strength per unit cross-sectional area, along with good elongation and excellent flexibility. The thickest twine (No. 5) had the lowest flexibility. In terms of selectivity, the net of No. 3 twine showed the broadest selection range and, thus, a relatively low selectivity compared with the other nets, while the less flexible net of No. 5 twine showed the narrowest selectivity range and high selectivity. In addition, it was found that a thicker twine resulted in a smaller haul of small fish. Therefore, it can be inferred that the thickness of the twine affects the size of the catch and selectivity, and thus the size composition of the catch as well.  相似文献

水泥粉体深层喷射搅拌桩与干振碎石桩〔1〕、干振灰土桩等一样,也是处理松软地基土的一种有效方法。但与干振碎石桩等其他挤密型复合地基处理方法不同,水泥粉体深层喷射搅拌桩属于摩擦桩型。由于天然地基土对桩体的侧摩擦力和端阻力的共同作用,使复合地基的承载力大幅度提高。然而人们普遍认为,水泥粉体深层喷射搅拌桩法复合地基处理不能解决场地土的液化问题。本文通过工程实例和瑞雷波检测结果,证实了该方法处理的复合地基,除了承载力有较大幅度的提高外,地基土的抗液化能力也可以得到相应的加强。  相似文献

The aim of the present study was to investigate the protein-sparing effect of carbohydrate in diets for juvenile turbot(Scophthalmus maximus) reared at five salinities(12,18,24,30,and 36).The fish were fed three isocaloric and isolipidic diets for 60 days.The results show that specific growth rate(SGR)and feed conversion efficiency(FCE) were higher in fish reared at salinities of 18 and 36,but lower at 12.Fish fed with diet C25P40(25%carbohydrate and 40%protein) had lower SGR and FCE values compared with those fed with the C5P52(5%carbohydrate and 52%protein) and C15P46(15%carbohydrate and 46%protein) diets;however,there was no statistical difference between diet C5P52 and C15P46.SGR and FCE values were unaffected by diet composition in fish reared at salinity 36.Hepatic lipogenic enzyme activities were higher in fish reared at 18 and 36,but lower at 12,while glucokinase(GK) activity was higher in fish reared at 12,and lower at 18 and 36.Dietary starch enhanced GK activity while depressing lipogenic enzyme activity.However,lipogenic enzyme activity increased with increasing dietary starch in fish reared at 36.It is recommended that salinity should be maintained 12 in the farming of juvenile turbot.In addition,an increase in gelatinized starch from 5%to 15%could spare 6%dietary protein in fish reared at salinities of 18-30,while higher salinity(36) could improve dietary carbohydrate use and enhance the protein-sparing effect,which is linked with the induction of lipogenic capacities.  相似文献

The protease inhibitor was purified from five different fish eggs. The molecular weights of Pacific herring, chum salmon, pond smelt, glassfish, and Alaska pollock egg protease inhibitors were 120, 89, 84.5, 17, and 16.8kDa, respectively. The specific inhibitory activity of glassfish egg protease inhibitor was the highest followed by those of Pacific herring and Alaska pollock in order. The specific inhibitory activity and purity of glassfish egg protease inhibitor were 19.70 U mg^- 1 protein and 164.70 folds of purification, respectively. Glassfish egg protease inhibitor was reasonably stable at 50 - 65℃ and pH 8, which was more stable at high temperature and pH than protease inhibitors from the other fish species. This study was carried out to investigate the biogenic amines(BAs), physicochemical property and microorganisms in dry salted fish, a traditional aquatic food consumed in China. Forty three samples of dry salted fish were gathered from retail and wholesale markets and manufacturers, which had been produced in various regions in China. Cadaverine(CAD) and putrescine(PUT) were quantitatively the most common biogenic amines. About 14% of the samples exceeded the histamine content standards established by the FDA and/or EU. The highest histamine content was found in Silver pomfret(Pampus argenteus)(347.79 mg kg-1). Five of forty three samples exceeded the acceptable content of TYR(100 mg kg-1), and 23.26% of dried-salted fish contained high contents of biogenic amines(above 600 mg kg-1). In addition, species, regions, pickling processes and drying methods made the physicochemical property, microorganisms and biogenic amines in dry salted fish to be different to some extents. The total plate count(TPC) was much higher than that of total halophilic bacteria in all samples. The biogenic amines, physicochemical property and microbiological counts exhibited large variations among samples. Furthermore, no significant correlation between biogenic amines and physicochemical property and TPC was observed. This study indicated that dry salted fish may still present healthy risk for BAs, depending on the processing methods, storage conditions among others.  相似文献

Smoking is widely used in fish processing for the color and flavor. Smoke flavorings have evolved as a successful alter- native to traditional smoking. The hazards of the fish products treated by liquid-smoking process are discussed in this review. The smoke flavoring is one important ingredient in the smoke-flavored fish. This paper gives the definition of smoke flavorings and the hazard of polycyclic aromatic hydrocarbons (PAHs) residue in the smoke flavorings on the market. It gives also an assessment of chemical hazards such as carcinogenic PAHs, especially Benzo-[a]pyrene, as well as biological hazards such as Listeria monocyto- genes, Clostridium botulinum, histamine and parasites in smoke-flavored fish. The limitations in regulations or standards are dis- cussed. Smoke flavored fish have lower content of PAHs as compared with the traditional smoking techniques if the PAHs residue in smoke flavorings is controlled by regulations or standards.  相似文献

The changes that have taken place in Nigerian fisheries are reviewed. Artisanal fishery has continued to dominate the fisheries,contributing over 85% of total fish production. The inland water and coastal seas are fully exploited and the increase in fishery production is not likely. Aquaculture potentials remain untapped as much as deep-sea fisheries. The combined potential of the fisheries resources-freshwater,marine and aquaculture can meet over 90% of the nation's demand for fish. An experiment on the optimum dietary protein content for the black sea bream using semi-purified dietsshowed the optimum content is 50.19% by weight as inferred from the growth rate and food consumptionof the fish. Another experiment on the proper ratio of animal to plant protein in its diet using sandeel as theanimal protein source and peanut cake meal as plant protein source showed that the ratio had great ef-fect on its growth and should be 1.00:0.89—0.96 as judged from the growth rate and food consumptionof the fish.  相似文献

Flavobacterium columnare causes columnaris disease in freshwater fish. In the present study, the antigenic regions of five outer membrane proteins (OMPs), including zinc metalloprotease, prolyl oligopeptidase, thermolysin, collagenase and chondroitin AC lyase, were bioinformatically analyzed, fused together, and then expressed as a recombinant fusion protein in Escherichia coli. The expressed protein of 95.6 kDa, as estimated by 10% sodium dodecyl sulfate-polyacrylamide gel electrophoresis, was consistent with the molecular weight deduced from the amino acid sequence. The purified recombinant protein was used to vaccinate the grass carp, Ctenopharyngodon idella. Following vaccination of the fish their IgM antibody levels were examined, as was the expression of IgM, IgD and IgZ immunoglobulin genes and other genes such as MHC Iα and MHC IIβ, which are also involved in adaptive immunity. Interleukin genes (IL), including IL-1β, IL-8 and IL-10, and type I and type II interferon (IFN) genes were also examined. At 3 and 4 weeks post-vaccination (wpv), significant increases in IgM antibody levels were observed in the fish vaccinated with the recombinant fusion protein, and an increase in the expression levels of IgM, IgD and IgZ genes was also detected following the vaccinations, thus indicating that an adaptive immune response was induced by the vaccinations. Early increases in the expression levels of IL and IFN genes were also observed in the vaccinated fish. At four wpv, the fish were challenged with F. columnare, and the vaccinated fish showed a good level of protection against this pathogen, with 39% relative percent survival (RPS) compared with the control group. It can be concluded, therefore, that the five OMPs, in the form of a recombinant fusion protein vaccine, induced an immune response in fish and protection against F. columnare.  相似文献

Hydrogel has high water content and structural similarity with natural extracellular matrix.So it has been widely studied and applied in the field of biomedicine.In order to further develop multifunctional hydrogels,we prepared mixed gels with antiultraviolet properties.This study found that the addition of polysaccharides and polyphenols was beneficial to the rheological,me-chanical properties,and biological activity of the protein.Chitosan(CS)could significantly improve the viscoelasticity,hardness,gel strength,thermal stability and crystallinity of gelatin.Interestingly,the addition of gallic acid(GA)could not only provide significant cross-linking effect,improve gel properties and microstructure,but also improve the UV resistance of the mixed gel.  相似文献

Preparation of Fe^2+ chelate of fish protein hydrolysate (Fe-FPH) obtained from low value fish proteins was introduced and its bioactivity was studied by compound enzymolysis. The optimum conditions for hydrolysate chelating Fe^2+ are DH (degree of hydrolysis) at 5%, pH 7.0, 20℃ and 15 min chelating time for FM (material not being defatted). Four types of Fe-FPH including CA (deposit after chelating), CB (deposit in 50% of absolute ethanol solution), CC (suspended deposit in 80% of absolute ethanol solution), and CD (bottom deposit in 80% of absolute ethanol solution) were fractionated with absolute ethanol from FM. Structural analysis through infra-red spectrum revealed that Fe^2+ was combined strongly with amino-group and carboxyl-group in each chelate and each Fe^2+ could form two five-member ring structures. All of the four chelates were shown more significant antioxidative activity and can be used as natural hydrophobic and hydrophilic antioxidant. Among all the chelates, the CB possesses the most effective antioxidative activity at 92% as high as that of a-tocopherol. Among all Fe-FPHs, only CD showed the most effective antibacterial activity against Escherichia coli, Staphylococcus aureus, Salmonella typhi, and Bacillus subtilis and can be used as natural antibacterial. It provides a more effective way for utilization of low value fish proteins and key information of Fe-FPH as additive in food industry.  相似文献

A feeding trial was conducted for 75 d to evaluate the nutritive value of a mixture of animal by-products(MAB) as a possible protein source in diets for juvenile mangrove red snapper,Lutjanus argentimaculatus(mean initial body weight,30 g). Fish were fed one of five isonitrogenous diets(40% crude protein) replacing 0,25%(MAB25),50%(MAB50),75%(MAB75) and 100%(MAB100) of fish meal protein with similar percentages of MAB. The MAB consisted of 25% cow liver meal,20% leather meal,20% meat and bone meal,15% blood meal,10% APC(poultry feather meal),8% poultry manure dried,1.5% choline and 0.5% chromic oxide. After 75 d of feeding,fish fed with diets MAB50,MAB75 and MAB100 exhibited significantly lower growth per-formance than that of fish fed with control and MAB25 diets. The optimum level of MAB was estimated to be 23%. Replacement of fish meal by MAB23% showed the following performance:maximum weight gain,510%;SGR,2.39% and FCE,2.83%. The MAB substitution up to 75% of fish meal protein in diets did not show differences in apparent protein digestibility(83.6% for MAB25,79.2% for MAB50,78.7% for MAB75) compared with control(83.4%),whereas in MAB100 group digestibility(65.3%) was sig-nificantly lower than in other groups. The apparent phosphorus absorption of test diet groups was significantly higher(37.1% for MAB25,28.5% for MAB50,55.6% for MAB75 and 54.5% for MAB100) than that of control(11.2%). The levels of protein and ash in the whole body,carcass and viscera increased as MAB substitution in diets increased,whereas lipids and moisture remained con-sistent among all treatment groups. These results showed that approximately 23% of fish meal protein could be replaced by a mixture of animal by-products for juvenile snapper growing from 30 g to 167 g in 75 d without compromising growth performance and feed efficiency.  相似文献
