Design and application of the method for isolating magnetotactic bacteria   总被引:1,自引:0,他引:1  
A simple apparatus was designed to effectively isolate magnetotactic bacteria from soils or sediments based on their magnetotaxis. Through a series of processes including sample incubation, MTB harvesting, isolation, purification and identification, several strains of bacteria were isolated from the samples successfully. By Transmission Electron Microscopy (TEM) and Energy-Dispersive X-ray Analysis (EDXA), these bacteria were certificated to be magnetotactic bacteria. The phylogenetic relationship between the isolated magnetic strains and some known magnetotactic bacteria was inferred by the construction of phylogenetic tree based on 16SrDNA sequences. This apparatus has been proven to have the advantages of being inexpensive, simple to assemble, easy to perform and highly efficient to isolate novel magnetotactic bacteria. The research indicated that the combined approach of harvesting MTB by home-made apparatus and the method of plate colony isolation could purify and isolate magnetotactic bacteria effectively.  相似文献

Although magnetotelluric sounding method applied to the land is advanced, there are many difficulties when it is applied to marine environment, one of which is how to lay magnetic field sensors down to the seafloor to complete measurements. To protect the magnetic field sensors from intense erosion and high pressure, suitable high-pressure sealed cabins must be designed to load them. For the consideration of magnetic measurement and marine operation, the sealed pressure cabin should be nonmagnetic and transportable. Among all optional materials, LC4 super-hard aluminum alloy has the highest performance of price/quality ratio to make the sealed pressure cabin. However, it does not mean that the high-pressure sealed cabin made using LC4 will be perfect in performance. In fact, because of its weak magnetism, the pressure cabin made using LC4 has distorting effect on frequency responses of the magnetic field sensors sealed in it. This distorting effect does not affect the use of the magnetic field sensor, but if we want to eliminate its effect, we should study it by experimental measurements. In our experiment tests, frequency sweep magnetic field as excitation signal was used, and then responses of the magnetic field sensor before and after being loaded into the high-pressure sealed cabin were measured. Finally, normalized abnormal curves for the frequency responses were obtained, through which we could show how the high-pressure sealed cabin produces effects on the responses of the magnetic field sensor. Experimental results suggest that the response distortion induced by the sealed pressure cabin appears on mid- andhigh-frequency areas. Using experimental results as standardization data, the frequency responses collected from seafloor magnetotelluric measurements can be corrected to restore real information about the seafloor field source.  相似文献

Geotopes or geological sites provide information on the evolution, structure and properties of the Earth‘s crust and on the geological past. They also represent the great variety of features that characterize a landscape, For this reason, their preservation is of special concern to geosciences. As early as in the 19th century, outstanding rock formations became important tourist sites although the modern terms ““geosites““ and ““geotourism““ were unknown. Research on international student’s post migration plans treats migration as a binary stay-return category and focuses on push-pull factors as the cause of this migration. In this paper we expand the definition of migration and consider the role of life experiences and aspirations, particularly the concept of home. We ask, what are the different conceptualizations of home and how are these tied to differential migratory plans? We analyze data from 232 interviews with international students from more than 50 countries who attended a flagship public university in Canada from 2006–2013.We find that students have four ways of thinking about home: as host, as ancestral, as cosmopolitan, and as nebulous. These understandings of home correspond to particular post-migration plans. While students who view home as a host plan to stay, and those who view home as ancestral plan to return, those with cosmopolitan and nebulous conceptions of home have more open migration plans.  相似文献

A Dirichlet–Neumann representation method was recently proposed for upscaling and simulating flow in reservoirs. The DNR method expresses coarse fluxes as linear functions of multiple pressure values along the boundary and at the center of each coarse block. The number of flux and pressure values at the boundary can be adjusted to improve the accuracy of simulation results and, in particular, to resolve important fine-scale details. Improvement over existing approaches is substantial especially for reservoirs that contain high-permeability streaks or channels. As an alternative, the multiscale mixed finite-element (MsMFE) method was designed to obtain fine-scale fluxes at the cost of solving a coarsened problem, but can also be used as upscaling methods that are flexible with respect to geometry and topology of the coarsened grid. Both methods can be expressed in mixed-hybrid form, with local stiffness matrices obtained as “inner products” of numerically computed basis functions with fine-scale sub-resolution. These basis functions are determined by solving local flow problems with piecewise linear Dirichlet boundary conditions for the DNR method and piecewise constant Neumann conditions for MsMFE. Adding discrete pressure points in the DNR method corresponds to subdividing faces in the coarse grid and hence increasing the number of basis functions in the MsMFE method. The methods show similar accuracy for 2D Cartesian cases, but the MsMFE method is more straightforward to formulate in 3D and implement for general grids.  相似文献

The Zermatt–Saas-Fee ophiolites (ZSFO) are one of the best preserved slices of eclogitic oceanic crust in the Alpine chain. They formed during the opening of the Mesozoic Tethys and underwent subduction to HP/UHP conditions during Alpine compression. A cathodoluminescence-based ion microprobe (SHRIMP) dating of different zircon domains from metagabbros and oceanic metasediments was carried out to constrain the timing of formation and subduction of this ophiolite, two fundamental questions in Alpine geodynamics. The formation of the ophiolitic sequence is constrained by the intrusion ages of the Mellichen and the Allalin metagabbros (164.0 ± 2.7 Ma and 163.5 ± 1.8 Ma) obtained on magmatic zircon domains. These data are in line with the maximum deposition age for Mn-rich metasediments which overlie the mafic rocks at Lago di Cignana (161 ± 11 Ma) and at Sparrenflue (ca. 153–154 Ma). An Eocene age of 44.1 ± 0.7 Ma was obtained for whole zircons and zircon rims from an UHP eclogite and two metasediments at Lago di Cignana. One of the Eocene zircons contains a rutile inclusion indicating formation at HP conditions. As the temperature and pressure peak of these rocks nearly coincide, the Eocene zircons probably constrain the age for the deepest subduction of the ZSFO. This Eocene age for the UHP metamorphism implies that the ZSFO were subducted later than the Adriatic margin (Sesia-Lanzo Zone) and before the Late Eocene subduction of the European continental crust below Apulia. A scenario with three subduction episodes propagating in time from SE to NW is proposed for the geological evolution of the Central and Western Alps. In order to constrain the crustal wave velocity structure in the southern Tibetan crust and provide insight into the contribution of crustal composition, geothermal gradient and partial melting to the velocity structure, which is characterized by low average crustal velocities and widespread presence of low-velocity zone(s), the authors model the crustal velocity and density as functions of depth corresponding to various heat flow values in light of velocity measurements at high temperature and high pressure. The modeled velocity and density are regarded as comparison standards. The comparison of the standards with seismic observations in southern Tibet implies that the predominantly felsic composition at high heat flow cannot explain the observed velocity structure there. Hence, the authors are in favor of attributing low average crustal velocities and low-velocity zone(s) observed in southern Tibet mainly to partial melting. In this paper, we systematically analyze the pressure projection stabilization method for the Darcy and coupled Darcy–Stokes flow problems in multiple dimensions. Stability results for this stabilization method are established. For the Darcy flow, optimal error estimates in the divergence norm for velocity and suboptimal error estimates in the $L^{2}$ -norm for pressure are obtained, and a superconvergence result for the pressure is derived; a local postprocessing scheme is constructed to generate optimal error estimates in the L 2-norm for pressure. For the coupled Darcy–Stokes flow, error estimates of optimal order are obtained in terms of the energy norm of velocity and pressure. The mid-Pleistocene transition (MPT, c. 1.2 to 0.5 Ma) records fundamental changes in Earth's climate state, where low-amplitude 41-kyr obliquity-dominated cycles gave way progressively to the high-amplitude, quasiperiodic (c. 100-kyr) fluctuations that characterize the later Pleistocene and Holocene. We use wavelet analysis on the LR04 δ^l8O benthic foraminiferal stack to confirm low-frequency power as early as 1.25-1.20 Ma, determine the persistence of obliquity-dominated cyclicity through and beyond the MPT, and reveal new levels of complexity in climate evolution.  相似文献

The eastern Himalayan syntaxis in Namjagbarwa is a high-grade metamorphic terrain formed by the India-Eurasia collision and northward indentation of the Indian continent into Asia. Right- and left-lateral slip zones were formed by the indentation on the eastern and western boundaries of the syntaxis respectively. The Dongjug-Mainling fault zone is the main shear zone on the western boundary. This fault zone is a left-lateral slip belt with a large component of thrusting. The kinematics of the fault is consistent with the shortening within the syntaxis, and the slipping history along it represents the indenting process of the syntaxis. The Ar-Ar chronological study shows that the age of the early deformation in the Dongjug-Mainling fault zone ranges from 62 to 59 Ma. This evidences that the India-Eurasia collision occurred in the early Paleocene in the eastern Himalayan syntaxis.  相似文献

Many Archean terranes are interpreted to have a tectonic and metamorphic evolution that indicates intra-crustal reorganization driven by lithospheric-scale gravitational instabilities. These processes are associated with the production of a significant amount of felsic and mafic crust, and are widely regarded to be a consequence of plume-lithosphere interactions. The juvenile Archean felsic crust is made predominantly of rocks of the tonalite–trondhjemite–granodiorite (TTG) suite, which are the result of partial melting of hydrous metabasalts. The geodynamic processes that have assisted the production of juvenile felsic crust, are still not well understood. Here, we perform 2D and 3D numerical simulations coupled with the state-of-the-art of petrological thermodynamical modelling to study the tectonic evolution of a primitive Archean oceanic plateau with particular regard on the condition of extraction of felsic melts. In our numerical simulations, the continuous emplacement of new, dry mafic intrusions and the extraction of the felsic melts, generate an unstable lower crust which drips into the mantle soon after the plume arrival. The subsequent tectonic evolution depends on the asthenosphere TP. If the TP is high enough (≥ 1500 C) the entire oceanic crust is recycled within 2 Myrs. By contrast at low TP, the thin oceanic plateau slowly propagates generating plate-boundary like features.  相似文献

As a kind of abnormal natural gas formed with special mechanism, the deep-basin gas, accumulated in the lower parts of a basin or syncline and trapped by a tight reservoir, has such characteristics as gas-water inversion, abnormal pressure, continuous distribution and tremendous reserves. Being a geological product of the evolution of petroliferous basins by the end of the middle-late stages, the formation of a deep-basin gas accumulation must meet four conditions, i.e., continuous and sufficient gas supply, tight reservoirs in continuous distribution, good sealing caps and stable structures. The areas, where the expansion force of natural gas is smaller than the sum of the capillary force and the hydrostatic pressure within tight reservoirs, are favorable for forming deep-basin gas pools. The range delineated by the above two forces corresponds to that of the deep-basin gas trap.
