冷胁迫下红树植物白骨壤和桐花树叶片热值的变化   总被引:10,自引:0,他引:10  
对九龙江口不同土壤盐度的白骨壤和桐花树离体叶片进行1-2℃冷胁迫处理,结果表明,随着冷胁迫时间的延长,白骨壤和桐花树叶片的热值逐渐下降,电解质渗出率呈现不同程度的上升,其中土壤盐度高的澳头白骨壤和桐花树叶片的变化幅度较土壤盐度低的白骨壤和桐花树小。白骨壤和桐花树叶片总含水量在冷处理过程中变化不大,但其自由水含量呈显著下降趋势,而束缚水含量及束缚水与自由水含量的比值则呈显著上升趋势。还原糖、可溶性糖和淀粉含量在冷胁迫过程中逐渐下降,蔗糖含量则逐渐上升。土壤盐度高的澳头白骨壤和桐花树叶片的自由水和束缚水含量的变化幅度大于土壤盐度低者,而还原糖、蔗糖、可溶性糖和淀粉含量的变幅则小于土壤盐度低者。  相似文献

2004年冬季在集美凤林类似底质类型、盐度和潮位的海湾地段4个断面,就不同红树植物林中的小型底栖动物数量和自由生活海洋线虫群落进行了研究。结果表明:海洋线虫是凤林红树林中小型底栖动物中的绝对优势类群,占到小型底栖动物的76.1%~96.3%;从丰度来看,旧区白骨壤(Avicennia marina)林中小型底栖动物的数量较少,有污水流过的光滩数量较大;4个断面共鉴定出海洋线虫37种(分类实体单元),其中新区秋茄(Kandelia candel)林中的生物多样性指数较低,旧区白骨壤林中群落具有较高的物种多样性;从出现的物种来看,新区秋茄林、旧区白骨壤林和光滩上出现的优势种和摄食功能群的类型各不相同。从4个断面13个站位进行的聚类分析和MDS标序分析结果推断自由生活海洋线虫的群落结构与不同的红树植物形成的沉积物有关。  相似文献

揭示极端天气影响的红树林潮滩潮流能量耗散过程是理解生物海岸演变及滨海湿地生态修复工程的核心内容。以北部湾防城港东湾白骨壤红树林潮滩为例,基于声学多普勒流速仪(ADV)获取区域2020年8月连续6天的水动力和白骨壤红树林下垫面植被实测数据,分析白骨壤潮滩近底层动力响应台风“森拉克”的耗散过程。结果表明:1)正常天气涨、落潮期间,自光滩到白骨壤红树林林内近底层湍流动能变化具有潮汐不对称性特征。涨潮期间光滩—白骨壤红树林边缘、白骨壤红树林边缘—林内潮流挟沙能量分别通过泥沙沉降以及搬运泥沙两种方式沿程消耗,落潮期间潮流挟沙能量主要以泥沙净沉降的方式消耗。2)白骨壤通过其潮滩表层向上发育长约10 cm的呼吸根以降低湍流垂向紊动;红树林边缘的枝、叶通过影响水平方向上的水流动力,致使潮流挟沙能耗降低。3)与正常天气比较,台风“森拉克”期间东湾自光滩到白骨壤红树林林内近底层水体流速无明显变化,但流向偏转幅度明显变大。同时湍流动能的耗散率和用以搬运泥沙为主的潮流挟沙能耗亦均增大。  相似文献

以红树植物白骨壤(Avicennia marina)基因组DNA为模板,根据超氧化物歧化酶基因保守序列设计特异引物进行PCR扩增,得到特异基因片段。回收该基因片段,与pMD18-T载体连接,并转化到感受态大肠杆菌ER2566细胞,获得铜锌超氧化物歧化酶基因片段的克隆。序列分析表明白骨壤铜锌超氧化物歧化酶基因片段含4个外显子和3个内含子,编码78个氨基酸,与水稻、玉米、红薯和白杨相应氨基酸序列的同源性分别为83.3%,84.6%,84.6%和87.2%。  相似文献

研究了环境盐度对低盐诱导长牡蛎(Crossotrea gigas)三倍体的影响。在海水温度为(25±2)℃条件下,长牡蛎亲贝,经过不同盐度(22、26、30、34和38)海水培育,采取人工受精,在受精卵中出现40%~50%第一极体时,进行15min的低盐(4、6、8、10、12和14)持续诱导,然后放入不同盐度海水中进行孵化。研究表明:同一环境盐度下,随着低盐逐渐升高至环境盐度,卵裂率和孵化率均逐渐升高;与其他环境盐度相比,环境盐度为38时,卵裂率和孵化率均明显较低。当环境盐度为26、30、34和38时,在低盐为时8三倍体率最高;当环境盐度为22时,在低盐为6时三倍体率最高。当环境盐度为22、26、30、34和38时,在低盐为8时综合评价指数最高。通过双因子方差分析发现,环境盐度和低盐均对卵裂率、孵化率及三倍体率有显著影响(P0.05)。  相似文献

以坛紫菜的4个品系(A、B、C、D)的丝状体为材料,探讨和比较了各品系在异常条件下,即高温(30℃、32℃)、高盐(盐度35、38)、低盐(盐度10、15)、低氮磷含量(N:0.0252mg/dm3;P:0.0041mg/dm3)的生长发育状况,筛选出抗逆的品系实验结果表明:(1)在高温32℃下,品系C生长较好,其瞬时生长率为0.40,表现出较强的耐高温特性;(2)在盐度38的海水中,品系A较能适应,藻色正常,瞬时生长率高达3.66,较耐高盐;(3)在盐度10的海水中,品系D的瞬时生长率为0.46,且形成了大量的、正常的孢子囊枝;(4)在低氮、磷的环境下,品系D适应性较强,其瞬时生长率为2.27,部分藻丝颜色恢复,并形成较多的、正常的孢子囊枝.  相似文献

在水温28.6—29.5℃、pH8.2条件下,研究了管角螺Hemifusus tuba稚贝[(壳高19.5±1.23)mm、体重(661.4±48.6)mg]在不同盐度下的生长和存活,并用二点法计算出最适宜存活和生长的盐度范围。结果表明,管角螺稚贝适宜存活盐度和最适存活盐度分别为17.7‰—40.8‰和25.1‰—35.9‰,适宜生长盐度和最适生长盐度分别为17.5‰—39.9‰和27.8‰—33.2‰。在最适生长盐度范围内,稚贝壳高日平均增长率为0.358—0.397mm.d 1,体重日平均增长率为44.64—49.09mg.d 1。超出上述范围,其存活率下降、生长率降低。稚贝在盐度17.0‰和41.3‰的海水中适应20d后再分别移至盐度14.0‰和45.0‰的海水中,72 h后存活率分别为84.0%和96.0%,而直接把生活在盐度为30.5‰中的稚贝放入到盐度为14.0‰和45.0‰的海水,72h后其存活率为8.0%和0,可见稚贝对盐度的适应能力与原生活海水盐度有关,经过驯化后的稚贝能够扩大对盐度的适应范围,提高对盐度的耐受力。  相似文献

1 材料和方法实验的大部分于1994年3~7月完成。虾夷马粪海胆(Strongylocentrotusintermedius)幼胆为大连水产学院1993年人工培育的F3代幼海胆(1.0±0.1cm)。1.1 低盐及正常海水中虾夷马粪海胆幼胆的成活率低盐海水为经暴气的自来水与自然海水(30.5)配制而成,自然海水为经砂滤的黑石礁沿岸水。盐度测定采用SYY-1型光学折射盐度计。实验容器采用2.0L烧杯代用,每个盐度组采用壳直径为1.0±0.1cm幼胆40个,分为2~4组,具体是将幼胆放入各盐度水中一定时间后取出放回到自然海水中并观察120~168h后计数成活率,盐度19.0以上放入时间为120h以…  相似文献

In coastal marshes, fish larvae may be exposed simultaneously to extreme salinities and to atrazine, a widely used herbicide. To assess the effects of salinity on the toxicity of atrazine, newly-hatched mummichog (Fundulus heteroclitus) were exposed to atrazine (0, 5, 50 and 500 microg/L) at three salinities (3, 15 and 35PSU). Whole body cortisol was measured after 24 h. Body length, condition factor, whole body proteins, lipids, residual masses and water contents were assessed after 96 h. Significants effects were found for both condition factor and water content. Condition factors were lower at salinity 3 and 35 compared to near isoosmotic salinity, 15PSU. In addition, atrazine decreased condition factor at 500 microg/L. Reduction in condition was likely due to retardation in axial growth since body length, percentages of proteins or lipids were not affected. In the absence of atrazine, salinity had no effect on the prevalence of dehydrated (81% water) or hyperhydrated (85% water) larvae. In larvae exposed to atrazine, the prevalence of hyperhydrated larvae increased at 3PSU and 5 microg/L atrazine and that of dehydrated larvae increased at 15 and 35PSU and 5 microg/L atrazine. Severity of dehydratation increased with atrazine concentration at salinity 35PSU. Thus, a short-term exposure to environmentally realistic concentrations of atrazine affects osmotic control in mummichog larvae with possible effects on buoyancy, survival and recruitment.  相似文献

采用双向电泳技术,对浙江乐清无花纹和有花纹文蛤外套膜的全蛋白进行了研究,并利用质谱和软件对差异蛋白进行了分析。经PDQuest软件分析,在pH4—7,分子量14.3—200kD之间,无花纹文蛤样品中检测到682±24个蛋白质点,有花纹样品检测到600±35个蛋白质点,平均匹配率为84.5%。研究结果显示,两种文蛤共有256个点存在差异表达,这些差异点可能与贝壳的颜色和花纹形成有关。其中98个点为无花纹文蛤特有,57个点为有花纹文蛤特有,另有101个点在表达量上存在两倍以上差异。在无花纹贝壳中发现分子量为42kD左右,等电点为6.4左右,与库蚊的肌动蛋白(分子量为41.7kD,等电点为5.3)匹配率较高;分子量为29kD,等电点为5.6,与飞鱿的肌动蛋白(分子量为29kD,等电点为5.25)匹配率较高的两个蛋白质。在有花纹文蛤中发现了分子量为27kD左右,等电点为4.8,与丽文蛤属的肌浆钙结合蛋白(分子量为20.8kD,等电点为4.98)匹配率较高。  相似文献

对长时间和短时间盐胁迫下红树植物秋茄(Kandelin candel)叶片膜脂过氧化作用、叶绿素含量、电解质渗透率、叶片肉质化程度等与盐胁迫强度之间的关系进行了对比研究.结果表明,短时间盐胁迫和长时间盐胁迫下秋茄叶片超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、叶片肉质化程度等随盐胁迫强度的提高而有不同的变化规律.在短时间盐胁迫下,随基质盐度的提高,SOD活性(fw)变化呈“U”型,在盐度15~20处叶片SOD活性最低,MDA含量变化不明显,电解质渗透率和叶片肉质化程度先升高而后下降,在盐度20处最高;在长时间盐胁迫下,SOD活性先随盐度的提高而提高,当盐度高于10时又随盐度的提高而下降,而MDA含量随盐胁迫程度的提高而下降,电解质渗透率和肉质化程度却上升,不同盐胁迫时间下红树植物秋茄具有不同的耐盐机制.膜脂过氧化不是盐胁迫下秋茄叶片膜损伤的主要原因.  相似文献

The extent to which marine organic matter is associated with surfaces and the consequences of such associations for organic matter remineralization are the focus of considerable attention. Since extracellular enzymes operating outside microbial cells are required to hydrolyze organic macromolecules to sizes sufficiently small for substrate uptake, the effects of surface interactions–on enzymes as well as on substrates–for hydrolytic activity also require investigation. We used a simplified laboratory system consisting of a free (dissolved) polysaccharide (pullulan) and the same polysaccharide tethered to agarose beads to restrict mobility, plus the corresponding free enzyme (pullulanase) and the same enzyme sorbed to montmorillonite (Mte), to investigate systematically the consequences of surface associations of enzymes and of substrates on hydrolytic activity. Changes in substrate molecular weight were monitored with time to measure the course of enzymatic hydrolysis. Although hydrolysis of free substrate was nearly complete after 2 min incubation with the free enzymes, the sorbed enzymes also effectively hydrolyzed free substrate, and the data suggest that they retained activity longer in solution compared to the free enzymes. Sorbed enzymes progressively hydrolyzed the free substrate from > 50 kD to lower molecular weights during a 24 h incubation, with a final product distribution on average showing only 1.4% and 10.3% of substrate still in the > 50 kD and 10 kD size classes, while 46.6%, 29.3%, and 12.5% of substrate was in the 4 kD, monomer, and free tag size classes, respectively. This product distribution was very similar to that of the free substrate/free enzyme experiment. Tethering the substrate to agarose beads led to lower substrate release (2–3% of total substrate after 98 h incubation) into solution compared to the free substrate case. For tethered substrates, the state of the enzyme (free or sorbed) measurably affected the molecular weight distribution of the hydrolysis products, with free enzymes producing a higher fraction of high molecular weight hydrolysis products (28.7 ± 5.4% of substrate > 50 kD at the end of the incubation) compared to sorbed enzymes (11.6 ± 2.8% of substrate > 50 kD at the end of the incubation.) Tethered substrates were also hydrolyzed in a sediment slurry from surface sediments from Cape Lookout Bight, North Carolina; 0.1% of total substrate was released by enzymes naturally present in 1 cm3 of sediment after 144 h incubation, demonstrating that the enzymes naturally present in marine sediments are also capable of accessing tethered substrates. These investigations suggest that surface associations of enzymes in marine systems may extend the active lifetime of such enzymes, providing an opportunity for hydrolysis over longer periods of time and producing a different size spectrum of hydrolysis products relative to free enzymes. Furthermore, in well-mixed systems, surface-associated enzymes can hydrolyze substrates whose mobility is restricted, highlighting the importance of processes such as resuspension and bioturbation on organic matter remineralization.  相似文献

根据大通水文站的流量和引水船站的盐度系列资料,应用谱分析方法研究长江口径流、盐度的变化规律及两者的相互关系.分析表明,长江口径流存在4~8a,2~3a,1.5a,1a,30~60d及20d的及化周期;盐度存在5~10a,2~3a,1.5a,1a,0.5a,40~50d,26~30d,14~15d7~8d及3.5d的变化周期.径流和盐度的谱结构在各水文年基本一致,均以1a周期变化占优势,但盐度谱结构在高频部分较径流的谱结构具有更大的振荡.盐度与流量的5~10a,2~3a,1.5a,1a的周期振动上存在高相关.径流对盐度变化的影响主要体现在低频部分.在低频变化中表现为盐度变化滞后于径流变化,在高频带呈同步变化.  相似文献

研究了天津厚蟹Helice tientsinensis对盐度和温度的耐受性。天津厚蟹对盐度的耐受能力极强,在温度9~34℃盐度为0的淡水介质中均能存活96h以上;在10℃、20℃及30℃的高渗介质中暴露96h后,天津厚蟹全部存活的盐度分别是65、56和50,全部死亡的盐度分别为83、74和68。天津厚蟹对低温的耐受力较强,在0、30和50盐度水平下,耐受低温的能力差别较大,能耐受96h的最低温度分别是9℃、2℃和6℃。天津厚蟹对高温也有较强的耐受能力,在0、30和50盐度水平下,分别在34℃、36℃和35℃下能耐受96h;盐度50温度38℃时,在开始的24h内无死亡,但48h后死亡率急剧上升,同样规律也出现在30和0溶液中,前者温度是39℃,后者为35℃。研究结果还表明盐度和温度的交互效应显著,二者的交互作用对天津厚蟹的存活亦有显著性影响(P0.05),温度升高(从10℃到20℃再到30℃)致使其耐盐能力显著下降,而盐度的剧烈变化(从30到50或从30到0)也致使其耐温能力显著下降。  相似文献

利用硫酸铵分级盐析、DEAE-纤维素阴离子交换层析、Sephadex G-75凝胶层析法对由海洋细菌FE92-8分泌的纤维蛋白溶酶进行了分离纯化,结果得到SDS-聚丙烯酰胺凝胶电泳单一区带。性质研究发现,该酶分子量为12.6kD,等电点为7.45,琼脂糖-纤维蛋白平板法测得该酶作用的最适温度为50℃,最适pH为8.0,在37℃,pH为8.0的情况下表现出很好的稳定性,温度起来50℃热稳定性较差,在碱性环境中稳定性较在酸性环境中稳定性强,阴性平板和阳性平板对比实验发现,该酶只有纤溶活性而无激酶活性,体外实验发现,该酶具有快速溶解血栓的能力。  相似文献

由于黄河口区域盐度偏低且不稳定,刺参(Apostichopus japonicus)的生长和存活受到严重威胁。为评估4个不同品系(日本刺参F2代RC-F2、抗病刺参F3代KC-F3、多刺刺参F2代DC-F2、青青刺参F1代QQ-F1)的低盐耐受力及其生理生化响应,本实验以青岛野生群体(QW)为对照,通过测定低盐胁迫下幼参的生长、存活以及非特异性免疫酶活力变化,评价了4个刺参不同选育品系对低盐的耐受状况。结果表明,在盐度17以下,除了QQ-F1表现为正生长,其他群体均为负生长。RC-F2、KC-F3、DC-F2、QQ-F1和QW的30天半致死盐度(LS50-30d)分别为15.02、15.19、16.48、14.26和15.13,QQ-F1显著低于其他4个群体;盐度14时,5个群体的半致死时间(ST50)分别为19.84 d、18.43 d、10.11 d、23.54 d和19.01 d,QQ-F1的ST50显著长于其他4个群体。在低盐胁迫时QQ-F1的ACP、AKP、SOD、LZM酶指标的下降幅度显著低于其他4个群体。不同选育群体的低盐耐受力排序为QQ-F1RC-F2QWKC-F3DC-F2,QQ-F1群体表现出良好的低盐耐受能力,在低盐度海域或盐度不稳定的海域具有良好的推广应用前景。本实验研究结果可为刺参健康养殖、良种性状评价、选育与推广等方面提供参考依据。  相似文献

Sediments from the lower Hudson River estuary and two other coastal environments were examined experimentally for their ability to mineralize (convert to CO2) the polycyclic aromatic hydrocarbons (PAHs) naphthalene and anthracene over a range of salinities. Routine assays employed 1:1 (vol fresh sed:vol water) sediment slurrys in order to overcome natural variability in mineralization rates among replicates. Mineralization rates were stimulated by about 2·5 fold, compared to unslurried controls, while the coefficient of variation fell from 13% to 3·5%.Rates of naphthalene mineralization in surface sediments from along the mainstem of the Hudson River (salinities from 2 to 27%) ranged from 0·011 to 1·5 nmol cm−3 day−1 (pool turnover [Tn] from 60 to 2040 days) with no discernible trends along the estuarine gradient. For two stations examined experimentally (mile point 5, salinity 23%; mile point 26, salinity 5%), microbial assemblages appeared acclimated to broad salinity variations as alter rates of mineralization compared to controls.Sediments from two upstream marshes of the Hudson (mile points 36 and 45) showed rates of naphthalene mineralization from 0·007 to 0·15 nmol cm−3 day−1 (Tn from 14 to 368 days), while sediments from a third marsh in freshwater (mile point 76) had high rates (66 nmol cm−3 day−1; Tn 40 days). For the two upstream marsh stations which rarely experienced salt intrusion, there was a substantial decrease in mineralization of naphthalene and anthracene with increasing salinity.Consistently high rates of naphthalene mineralization (780 to 1600 nmol cm−3 day−1; Tn 5 to 6 days) were observed in petroleum contaminated sediments from Port Jefferson Harbor (PJH) on the north shore of Long Island. PJH has a relatively constant salinity regime (about 27%) and imposed decreases in salinity effected decreases in rates of naphthalene and anthracene mineralization. Lowest rates of naphthalene mineralization (0·003 to 0·004 nmol cm−3 day−1; Tn from 714 days to 833 days) were found in sediments from two stations in the relatively pristine Carmans River estuary on the south shore of Long Island.The ability of increases or decreases in salinity to affect the rate of model PAH mineralization appeared to be dependent on the natural variation in the salinity regime from which a sample was obtained. Data from all the environments studied indicated a strong positive correlation between PAH concentration and the rates of mineralization of naphthalene. Rates of PAH mineralization in all environments examined appear to be primarily controlled by the extent of pollutant loading and not by natural variations in the salinity regime.  相似文献
