Trare amounts of benzene hydrocarbons obtained in Jiaozhou Bay (Qindao) were enriched bysorption on a GDX-102 column and eluted by carbon disulfide. The eluted was concenttaled and then de-temened by capillary column gas cbornatognphy.The contents of virious kinds of benzene hydrocarbons in Jiaozhou Bay coastal water were benzene(22.3-141.6)× 10~(-9)g/L, toluate (15.2-94.0) × 10~(-9) g/L, ethyl benzene(11.8-85.1)×10~(-9) g/L, p -xylene(15.2-78.5) ×10(-9) g/L, m-xylene (10.9-79.4) ×10(-9) g/L, o -xylene (12.4-80.1) x ×10(-9)g/L; iso-propyl(8.4- 73.1) x ×10(-9)g/L, n -propyl (6.9-76.4) ×10(-9) g/L, 1, 3, 5-trimethylbenzene (10.9- 35.9)×10(-9) g/L, 1,2, 4-trimethybenzene (10.0- 38.0)×10(-9) g/L, n - butydriare (8. 1 - 34.6) ×10(-9)g/L. Effects of water temperature (17, 21, 25, 30 and 35℃) and body size (14.75-281.41 g initial body weight) on food consumption, growth, feed conversion, and dry matter content in orange-spotted grouper fed to satiation were investigated. The combined effect of temperature (T, ℃) and body weight (W, g) on maximum food consumption (Cmax, g/day) was described as: InCmax=-7.411+0.828 InW+0.317T4).004 7T2, and the optimum feeding temperature was 33.9℃. The combined effect of temperature and body weight on growth (G) was described as: lnG=-4.461-0.2081nW+0.394T-0.006 3T^2. The optimum growth temperature was 31.4℃, whereas overall growth rates were high at 25, 30 and 35 ℃. Feed conversion efficiencies (FCE, %), increasing first and then decreasing with increasing temperature, averaged from 1.8 to 2.1 in terms of dry weight of food fish. The optimum temperature for FCE tended to be lower than that for growth or feeding. Dry matter content increased with both increasing water temperature (17, 25, 30 and 35℃) and body weight, and the combined effect of temperature and body weight on dry matter content (DM, %) was described as: lnDM =3.232+0.01 4 lnW-0.004 4T+0.001 2TInW.  相似文献

From Oct., 1999 to Oct., 2000, the heterotrophic bacterial flora in the aquaculture area around Xuejiadao was investigated. The result shows that the populations of the heterotrophic bacteria are heavier in summer and autumn than those in winter and spring. The average populations in seawater, sediment, the surface of seaweed and the surface of fish are 1.4×104cfu mL−1, 5.4×106cfu g−1, 1.5×106cfu g−1 and 1.8×103cfu cm−2, respectively. A total of 301 strains were isolated, among them 259 were Gram-negative. All the Gram-negative bacteria belong to 13 genera and some genera of Enterobacteriaceae. The communities of bacteria are slightly different among the samples. In the body surface of fish, Genus vibrio is dominant. In the remaining samples, dominant genus is Aeromonas.  相似文献

Biofouling is an important factor that affects the bivalve farming industry. Fouling organisms may reduce growth and survival rate of the cultured species. Fouler are often filter feeders, so they are potential competitors for food resource with the cultured species. The present study was conducted to measure the impact of fouling on food uptake and nutrient release in April and June, 2006 in Daya Bay near Guangzhou, China. Results showed that fouling organisms had significant effect on food uptake and nutrient release. The chlorophyll a uptake rate of fouled scallops was 7.53Lh-1±1.416Lh-1 and 11.94Lh-1±2.497Lh-1 in April and June, respectively, significantly higher than those of cleaned scallops, i.e., 4.23 Lh-1 ±2.744Lh-1 and 2.57Lh-1± 1.832 Lh-1 respectively.The consumption of total particulate matter by fouled scallops in April and June was 5.52Lh-1±0.818Lh-1 and 3.07Lh-1±0.971 Lh-1,respectively; the corresponding results for cleaned scallops are 2.49Lh-1 ±0.614Lh-1 and 2.37± 1.214Lh-1, respectively. Fouling increased ammonia release significantly. The ammonia release rate of fouled scallops was 33.81Lh-1±7.699Lh-1 and 76.39Lh-1 ±9.251Lh-1 in April and June, while cleaned scallops released 2.46Lh-1± 0.5 1 1Lh-1 and 7.23Lh-1± 1.026Lh-1 ammonia, respectively. Phosphate release of fouled scallops was 22.72Lh-1 ± 9.978 Lh-1 in June and cleaned scallops released phosphate 6.01Lh-1 ±0.876 Lh-1 in April. Therefore, fouling contributed much to food reduction and concentration increase of ammonia and phosphate in water.  相似文献

A new kinetic spectrophotometric method has been developed for the determination of iron (Ⅲ). The method is based on the catalytic effect of iron (Ⅲ) on the oxidation of weak acid brilliant blue dye (RAWL) by KIO4 in acid medium. The advantages of the proposed method are that it is sensitive, accurate, rapid, inexpensive, can be operated under room temperature and has a large determination concentration range compared to other techniques. The obtained optimum conditions are pH 3.15, RAWL (200 mgL-1) 5.00mL, Potassium periodate solution (0.01 molL-1) 0.30mL, phenanthroline (0.02molL-1) 1.00mL, reaction temperature 25℃ and reaction time 7 miu. With this method iron could quantitively be determined in the range 0.00-0.02 mgL-1, the detection limit being 4.10×10-10mL-1. The relative standard deviations (RSD) in five replicate determinations for 3 μgL-1 and 5μgL-1 iron (Ⅲ) are 3.1% and 1.9%, respectively. The method has been applied to the determination of iron (Ⅲ) in tap water samples and seawater samples (from the South China Sea), the recovery rates being 98.0% and 100.5%, respectively.  相似文献

Heavy paint coating loss and localized corrosion of the Al alloy hull occur red after less than a year's service of a ship at sea. Examination and analysis showed that the Mg-basic sacrificial anodes originally used for cathodic protection were not suitable as the resulting −1.3V-plus (vs Ag/AgCl) protection potential for the hull was too negative for the purpose. A new protection system was designed in which the protection current densities were 15mA/m2 for the hull and 350mA/m2 for the propeller. The hull protection potential under actual condition at sea was between −920mV and −1100mV (Ag/AgCl) and the protection efficiency was over 90%. The protection areas were calculated by using the formulas A=1.8×LBP×D+BC×LB×B for the hull and As=2Aas for the propeller. A new kinetic spectrophotometric method is developed for the measurement of manganese (II) in water. The method is based on the catalytic effect of manganese (II) with the oxidation of weak acid brilliant blue dye (RAWL) by KIO4 using the Nitrilo triacetic acid (NTA) as an activation reagent. The optimum conditions obtained are 40 mgL−1 RAWL, 1×10−4molL−1 KIO4, 2×10−4 molL−1 Nitrilo triacetic acid (NTA), pH = 5.8, the reaction time of 3.00 min and the temperature of 20.0 °C. Under the optimum conditions, the proposed method allows the measurement of manganese (II) in a range of 0–50.0 ng mL−1 and with a detection limit of down to 0.158 ng mL−1. The recovery efficiency in measuring the standard manganese (II) solution is in a range of 98.5%–102%, and the RSD is in a range of 0.76%–1.25%. The new method has been successfully applied to the measurement of manganese (II) in both fresh water and seawater samples with satisfying results. Moreover, few cations and anions interfere with the measurement of manganese (II). Compared with other kinetic catalytic methods and instrumental methods, the proposed method shows fairly good selectivity and sensitivity, low cost, cheapness, low detection limit and rapidity. The SCENTO-System was used to study the carbon dynamics between phytoplankton primary production and heterotrophic bacterial secondary production. Most of the methods used nowadays in situ for limnological synecology studies were applied. Primary production measurement showed an increasing tendency with increasing content of chlorophylla. It provided a true photosynthetic rate lying within the range of eutrophic lakes. Net EOC released from the algae ranged from 8.5 to 27.5 μg C l−1(6h)−1. Accompanying the algal products the number of bacteria increased from 1.475 ×109 to 8.074×109 cells l−1. The bacterial mean cell volume was small, between 0.0315 and 0.0548μm3. Bacterial carbon production from direct growth estimates was compared with independent calculations of bacterial growth from EOC uptake and3H-thymidine incorporation. Direct estimates were 2.97–10.0 μg Cl−1 (24h)−1 with the exception of a zero-growth on the third day. EOC uptake was 123.5–191.0 μg Cl−1 (6h)−1. That calculated from3H-thymidine incorporation was 0.2–0.5 μg Cl−1 (6h)−1.14C-glucose dark uptake ran parallel to the increasing bacterial biomass. The respiration of glucose was 6.5% (avg.) of the gross uptake. Since the system operated without grazing pressure, a real carbon flow from primary production to bacterial secondary production could be observed.  相似文献

The standing stock and primary production of benthic microalgae on tidal flats were measured seasonally at 3 transects (Puqing, Dahengchuang and Puqi) in Yueqing Bay during 2002 2003. The results showed that the integral chlorophyll a (Chl a) concentration in tidal flat mud exhibited a seasonal variation with the order of magnitude: winter (14.0 4.2 mg m-2) > spring (13.0 6.3 mg m-2) > autumn (7.7 5.9 mg m-2) > summer (4.6 3.2 mg m-2). The primary production showed an order of magnitude: spring (270.5 224.9 mgC m-2 d-1)>winter (238.7 225.5 mgC m-2 d-1)>autumn (214.1 56.2 mgC m-2 d-1)>summer (71.6 44.6 mgC m-2 d-1). Both chlorophyll a and primary production showed maximum values in the surface layer of sediment, and decreased rapidly with increasing depth due to sun light limitation. The results of variance analysis indicated that seasonal variation and tidal flat condition affected Chl a greatly, but had no significant effect on primary production. The annual primary production of benthic microalgae on tidal flats in Yueqing Bay was estimated at 16143 tons carbon, which is sufficient to support 1.02×105 tons shellfish production. The environmental factors affecting chlorophyll and primary production on the tidal flats in Yueqing Bay were discussed. By comparing with other bays on China’s coast, it was observed that Yueqing Bay is a region with high benthic microalgae standing crop and primary production, which may be related to the type of its sediment.  相似文献

Gametophytes of Laminaria japonica were cryopreserved in liquid nitrogen using encapsulation-dehydration with two-step cooling method. Gametophytes cultured at 10℃ and under continuous irradiance of 30 μmol m-2 s-1 for 3 weeks were encapsulated in calcium alginate beads. The beads were dehydrated in 0.4 molL-1 sucrose prepared with seawater for 6 h, desiccated in an incubator set at 10℃ and 70% relative humidity for 4 h, pre-frozen at either -40℃ or -60℃ for 30 min, and stored in liquid nitrogen for >24 h. As high as 43% of survival rate was observed when gametophytes were thawed by placing the beads in 40℃seawater and re-hydrated in 0.05 molL-1 citrate sodium prepared using 30‰ NaC1 7 d later. More cells of male gametophytes survived the whole procedure in comparison with female gametophytes. The cells of gametophytes surviving the preservation were able to grow asexually and produce morphologically normal sporophytes.  相似文献

Graded levels of manganese were supplemented to a semi-purified diet containing 45% crude protein, to provide six levels of manganese (i. e. containing 5, 25, 50, 70, 140 and 210×10−6, respectively) for two experiments with these experimental diets. The initial weight of shrimp used in the 35–day experiment I was 0.30±0.04 g, and that in the 70–day Experiment II was more than one gram. The results showed that optimum content in the semi-purified diet for the more than 1 gram shrimp ranged from 70 ×10−6, to 140×10−6, but supplementation of Mn was not necessary for the small shrimp. Cryopreservation of Porphyra yezoensis conchocelis was conducted with cryoprotectants and a proposed pretreatment procedure and thawing methods explored. Six cryoprotectants combined by DMSO with ethylene glycol (EG), propylene glycol (PEG), sorbitol and sucrose were developed. The effect of prefreezing at -40℃ or -20℃ for different time durations was compared and the thawing methods were screened. It was shown that the cryoprotectant including 10% DMSO with 0.5 molL^-1 sorbitol exhibited the optimal effect. The ideal pretreatment was that conchocelis segments were stayed for 20min at -40℃ before stored in liquid nitrogen, and 40℃ water bath was proper for quick thawing. The highest recovery rate of cryopreserved P. yezoensis conchocelis reached 89.41%.  相似文献

Discharge of heavy metals from metal processing industries is known to have adverse effects on the environment. Biosorption of heavy metals by metabolically inactive biomass of microbial organisms is an innovative and alternative technology for removal of these pollutants from aqueous solution. The search of marine actinobacteria with potential heavy metal biosorption ability resulted in the identification of a novel alkalophilic Streptomyces VITSVK5 species. The biosorption property of Streptomyces VITSVK5 spp. was investigated by absorbing heavy metals Cadmium (Cd) and Lead (Pb). Physiochemical characteristics and trace metal concentration analysis of the backwater showed the concentrations of different metals were lead 13±2.1 μg L−1, cadmium 3.1±0.3μg L−1, zinc 8.4±2.6μg L−1 and copper 0.3±0.1μg L−1, whereas mercury was well below the detection limit. The effect of pH and biomass dosage on removal efficiency of heavy metal ions was also investigated. The optimum pH for maximal biosorption was 4.0 for Cd (II) and 5.0 for Pb (II) with 41% and 84% biosorption respectively. The biosorbent dosage was optimized as 3 g L-1 for both the trace metals. Fourier transform infrared absorption spectrum results indicated the chemical interactions of hydrogen atoms in carboxyl (-COOH), hydroxyl (-CHOH) and amine (-NH2) groups of biomass with the metal ions. This could be mainly involved in the biosorption of Cd (II) and Pb (II) onto Streptomyces VITSVK5 spp. The results of our study revealed Streptomyces metabolites could be used to develop a biosorbent for adsorbing metal ions from aqueous environments.  相似文献

Chondrus is a type of commercially produced red seaweed that widely used for food and carrageen extraction. Although the natural life history of the alga had been well understood, the factors influencing development of the tetraspore and carpospore remain poorly understood. In the perspective of seedling resources, the regulation of early development is crucial for the seedling nursing; therefore, it is necessary to understand the physiological influences during its early development. In this study, we studied the effects of temperature and irradiance on the early development of Chondrus ocellatus Holm under laboratory conditions. The released tetraspores and carpospores were cultivated at different temperatures (10–28°C) and irradiances (10, 60 μmol photons m−2s−1) with a photoperiod of 12L:12D. The results indicate that both tetraspores and carpospores are tolerant to temperatures of 10–25°C, and have the highest relative growth rate at 20°C. Irradiance variances influenced the growth of the discoid crusts, and the influence was more significant with increasing temperature; 60 μmol photons m−2s−1 was more suitable than 10 μmol photons m−2s−1. The optimum temperature and irradiance for the development of seedlings was 20°C and 60 μmol photons m−2s−1, respectively.  相似文献

The effects of several nutritional factors on the growth and eicosapentaenoic acid (EPA) production of diatom Nitzschia laevis were studied. 4 LDM (quadrupled concentration of the nutrient salt) was the optimal concentration of nutrient salt for the growth and EPA production of N. laevis. The growth of N. laevis was inhibited when the glucose concentration was either lower than 10 gL−1 or higher than 15 gL−1. Both sodium nitrate and urea were good nitrogen sources for the growth and EPA production, while ammonium chloride seriously decreased the dry cell weight (DW) and the EPA content. Silicate seriously influenced the growth of N. laevis. The maximum DW of 2.34 gL−1 was obtained in the presence of 150 mgL−1 Na2SiO3·9H2O. The EPA content remained almost the same when the silicate concentration was lower than 150 mgL−1; however, higher silicate concentrations resulted in a steady decrease of EPA content. Low medium salinity (⩽29) did not seem to influence the DW of N. laevis, and high salinity resulted in a decrease of DW. The highest EPA content (4.08%) and yield (110 mgL−1) were observed at the salinity of 36 and 29, respectively.  相似文献

A cruise was conducted from late August to early September 2004 with the intention of obtaining an interdisciplinary understanding of the Yangtze River Estuary including the biological, chemical and physical subjects. Water sample analysis indicated that total phytoplankton species richness was 137. Of them 81 were found in Bacillariophyta and 48 in Pyrrophyta, accounting for 59.1% and 35.0% respectively. The average cell abundance of surface water samples was 8.8×104 cells L-1, with the maximum, 102.9×104 cells L-1, encountered in the area (31.75°N, 122.33°E) and the minimum, 0.2×104 cells L-1, in (30.75°N, 122.17°E). The dominant species at most stations were Skeletonema costatum and Proboscia alata f. gracillima with the dominance of 0.35 and 0.27. Vertical distribution analysis indicated that obvious stratification of cell abundance and dominant species was found in the representative stations of 5, 18 and 33. Shannon-Wiener index and evenness of phytoplankton assemblage presented negative correlation with the cell abundance, with the optimum appearing in (30.75°N, 122.67°E). According to the PCA analysis of the environmental variables, elevated nutrients of nitrate, silicate and phosphate through river discharge were mainly responsible for the phytoplankton bloom in this area.  相似文献
