底栖蓝藻是一些底栖动物如介形虫的良好食物,在自然状态下呈丝状,但经室内培养后会形成很大的且难以分开的球形群体。本研究着眼于这种蓝藻在室内培养条件下的光合效率的测定,运用同位素^14C方法和黑白瓶测氧法对不同光照强度和温度下它的光合效率和呼吸率进行对比测定。 相似文献

本文以组织培养技术培养的菹草无菌苗为实验材料,研究了2 mmol/L外源鸟氨酸(L-Orn)对40μmol/L镉(Cd)胁迫下菹草体内Cd含量、光合色素水平、超氧阴离子(O·-2)产生速率、过氧化氢(H2O2)、丙二醛(MDA)与可溶性蛋白含量以及脯氨酸和多胺代谢的影响.结果显示:(1)在单一Cd胁迫下,菹草体内的Cd大量积累,达到335.00μg/g,并诱导了明显的氧化胁迫,具体表现为O·-2产生速率以及H2O2和MDA含量显著升高,可溶性蛋白以及光合色素含量明显下降.外源L-Orn的添加很大程度上抑制了菹草体内Cd的积累,Cd含量降至232.50μg/g,并降低了活性氧(ROS)水平,减轻了膜脂过氧化程度,减缓了光合色素和可溶性蛋白的分解.(2)在单一Cd胁迫下,伴随着吡咯啉-5-羧酸合成酶(P5CS)活性的增加和脯氨酸脱氢酶(ProDH)活性的下降,菹草体内的脯氨酸含量轻微增加,仅为对照组的1.91倍,而外源L-Orn显著增强了P5CS和δ-鸟氨酸转氨酶(OAT)的活性,使脯氨酸含量大幅度升高,为对照组的8.36倍.(3)单一Cd胁迫分别通过激活精氨酸脱羧酶(ADC)、鸟氨酸脱羧酶(ODC)和多胺氧化酶(PAO)的活性,使腐胺(Put)的含量增加,亚精胺(Spd)和精胺(Spm)含量减少.而施用外源L-Orn后,ADC和ODC的活性进一步增强,Put的含量大幅增加,并促进Put向Spd和Spm的转化,同时PAO的活性下降,使Spd和Spm含量增加,从而使总多胺含量增加.由此可见,外施L-Orn可通过抑制Cd的进入和参与调节脯氨酸以及多胺代谢,抑制ROS的积累、膜脂过氧化、光合色素以及蛋白质的分解,一定程度上增强菹草对Cd胁迫的抗性. 相似文献

在纯培养的铜绿微囊藻种群中添加有效牧食者棕鞭毛虫.进行为期9d的实验,用流式细胞仪检测棕鞭毛虫牧食作用对铜绿微囊藻的影响.结果表明棕鞭毛虫的牧食导致微囊藻种群迅速下降,同时微囊藻种群出现了群体形成现象.棕鞭毛虫直接牧食作用诱发微囊藻形成群体后,能在一定程度上能防御棕鞭毛虫的进一步牧食,使得种群得以延续.在棕鞭毛虫的牧食作用下.以酯酶活性和叶绿素荧光强度为代表的微囊藻细胞活性在实验后期显著增强.而牧食处理后细胞尺寸有所变小,这可能是伴随着微囊藻诱发性群体形成过程中一种生态对策的调整,保持较小的个体有利于种群的迅速增殖. 相似文献

The isolate, Pesudoalteromonas sp. TBT1, could grow to overcome the toxicity of tributyltin chloride (TBTCl) up to 30 μM in the absence of Cl− in the medium until the cells reached an exponential phase of growth. The viability, however, was reduced after the cells reached a stationary phase. The degradation products, such as dibutyltin (DBT) and monobutyltin (MBT), were not detected in the growth medium, indicating that the isolate has no ability to degrade TBT into less toxic DBT and MBT. Up to about 107.5 TBT molecules were adsorbed by a single cell. The observation of morphological changes with an electron microscope showed that the cell surface became wrinkled after exposure to the lethal concentration of 10 mM TBTCl. These results indicate that the resistance of the isolate toward the toxicity of TBTCl is not related to the unique cell surface, which seems to play an important role in preventing the diffusion of TBTCl into the cytoplasm. 相似文献

以巢湖优势种铜绿微囊藻(Microcystis aeruginosa)和鱼腥藻(Anabaena sp.)为研究对象,研究不同温度(35、25和10℃)对这两种藻生长特性和胞外有机物产生的影响.结果表明,温度对铜绿微囊藻和鱼腥藻的藻细胞密度、碱性磷酸酶活性和胞外有机物浓度影响显著.25℃是铜绿微囊藻和鱼腥藻最适宜的生长温度,最高细胞密度分别达到3.12×107cells/ml和2.03×107 cells/ml.不同温度下两种藻的碱性磷酸酶活性特征,证实了高温对鱼腥藻生长的抑制和低温对铜绿微囊藻生长的抑制.胞外有机物释放总量受蓝藻生物量和单位细胞有机物释放速率的影响.铜绿微囊藻的溶解性有机碳和胞外总多糖释放量在25℃最高,最大值分别为49.28和38.46 mg/L;而鱼腥藻在35℃时释放量最高,最大值分别为45.82和40.60 mg/L;10℃条件抑制了两种藻的生长及胞外有机物的释放.鱼腥藻胞外多糖含量在35℃培养条件下最高,而铜绿微囊藻在10℃条件下最高,说明不利的生长条件会促进蓝藻胞外多糖的分泌.三维荧光图谱分析结果表明,铜绿微囊藻和鱼腥藻胞外有机物以类蛋白质和类腐殖酸为主,温度主要影响藻细胞胞外有机物浓度,而对有机物种类组成没有影响. 相似文献

Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous pollutants in the environment and are derived from both man-made and natural resources. The present study is focused on the degradation of PAHs by a halotolerant bacterial strain under saline conditions. The bacterial strain VA1 was isolated from a PAH-degrading consortium that was enriched from marine water samples that were collected from different sites at Chennai, India. In the present study, a clearing zone formed on PAH-amended mineral salt agar media confirmed the utilization of PAH by the bacterial strain VA1. The results show that the strain VA1 was able to degrade anthracene (88%), phenanthrene (98%), naphthalene (90%), fluorene (97%), pyrene (84%), benzo(k)fluoranthene (57%) and benzo(e)pyrene (50%) at a 30 g/L NaCl concentration. The present study reveals that the VA1 strain was able to degrade PAHs in petroleum wastewater under saline conditions. The promising PAH-degrading halotolerant bacterial strain, VA1, was identified as Ochrobactrum sp. using biochemical and molecular techniques. 相似文献

Microorganisms resistant to 1–500 ppm cadmium were isolated over an 11 month period from sediments at the sewage sludge, dredge spoils and industrial acid waste disposal sites, as well as at an estuarine outflow to the New York Bight apex. Tests for antibiotic resistance in these isolates revealed that 94% of the isolates were resistant to one or more antibiotics and that 91% of the original isolates could be characterized by multiple drug resistance. Different selective pressures may account for the various genera and antimicrobial resistance patterns observed. The possibility of extrachromosomal linkage of cadmium and streptomycin resistance is discussed. 相似文献

Microorganisms play an important role in the biodegradation of petroleum contaminants, which have attracted great concern due to their persistent toxicity and difficult biodegradation. In this paper, a novel hydrocarbon-degrading bacterium HZ01 was isolated from the crude oil-contaminated seawater at the Daya Bay, South China Sea, and identified as Achromobacter sp. Under the conditions of pH 7.0, NaCl 3% (w/v), temperature 28 °C and rotary speed 150 rpm, its degradability of the total n-alkanes reached up to 96.6% after 10 days of incubation for the evaporated diesel oil. Furthermore, Achromobacter sp. HZ01 could effectively utilize polycyclic aromatic hydrocarbons (PAHs) as its sole carbon source, and could remove anthracene, phenanthrene and pyrence about 29.8%, 50.6% and 38.4% respectively after 30 days of incubation. Therefore, Achromobacter sp. HZ01 may employed as an excellent degrader to develop one cost-effective and eco-friendly method for the bioremediation of marine environments polluted by crude oil. 相似文献

The life history of the amphipod Orchestia sp. cf. cavimana (Heller, 1865) was studied throughout the course of a year with monthly samplings and the use of pitfall traps along the shores of Lake Albano in central Italy. The data thus obtained showed two peaks in abundance (the first in June and the second in October) and a minimum capture frequency in February. Egg-bearing females were recorded in spring and late summer, whilst recruitment occurred from spring to autumn, with maximum in June and October. The data collected also indicated the presence of a positive correlation between the abundance of talitrids and both temperature and sediment moisture. Morphological analysis enabled the identification of four different cohorts (with a lifespan of the species of approximately 12–15 months) all four of which remained distinguishable throughout the course of the year. It, moreover, enabled sex determination in individuals of 5 or more millimetres. The subsequent distribution indicated that, when significant, the sex ratio was female biased and that maximum size was greater in males. In particular, maximum sizes were recorded in spring for both males and females and were of 17.5 and 13.0 mm, respectively. Furthermore, the total body length recorded for egg-bearing females was positively correlated with the number of eggs in their brood pouch. 相似文献

Biodegradation of polycyclic aromatic hydrocarbons (PAHs) in contaminated sediment is an attractive remediation technique and its success depends on biodegradation kinetics, and the optimal condition for the PAH-degrading isolates; however, information on this aspect is still scarce. The effects of multi-factors on biodegradation of phenanthrene, a 3-ring model PAH, in contaminated sediment slurry by Sphingomonas sp. a bacterial strain isolated from surface mangrove sediment, were investigated using the orthogonal experimental design (form L16(45)). The most significant factors were salinity and inoculum size, while the effects of phenanthrene concentrations, nutrient addition and temperatures were insignificant. The optimal biodegradation condition in contaminated mangrove sediment slurry was 30 °C, 15 ppt salinity, a carbon/nitrogen ratio of 100:1 (the background ratio in sediment) and an inoculum size of 106 most probable number g−1 sediment. The phenanthrene biodegradation could be best described by the first order rate model, C = C0e−kt, where k (the rate constant) is equaled to 0.1185, under the optimal condition. The kinetic model was verified and its validity in predicting biodegradation by Sphingomonas sp. at various phenanthrene concentrations was proved by experimental data. 相似文献

为了加强长江中游地区大型无脊椎底栖动物的研究工作,2009年3月至2010年3月,对长江中游巴河支流的胜利河毛翅目优势种的生产力和营养基础进行了为期一年的调查研究.结果表明,胜利河中主要毛翅目优势种纹石蛾的生活史为一年3代.一至四级河流中,其生物量周年平均值分别为1.77、7.80、44.82、51.55 mg/m2.采用体长频率法测算的年均生产量为7.72 mg/m2,P/B系数为13.21.利用肠内含物分析法研究了胜利河中纹石蛾的营养结构.比较一至四级河流的营养基础发现,无形态碎屑为纹石蛾最常见的食物,占肠内含物的45.22%~62.92%;而对生产量贡献率最高的多是动物组织,达52.86%~68.58%.各季节的营养基础状况也有差异,春季无形态碎屑所占比例和对生产量的贡献率较高,而夏、秋和冬季动物组织则有更高的生产量贡献率.一至四级河流的年均物质通量分别为11.83、34.76、237.52和273.17g/(m2·a).由此可见,该优势种在河流生态系统中发挥着重要作用. 相似文献

Cometabolism has been suggested as an attractive approach to enhance the degradation rates of high-molecular-weight polycyclic aromatic hydrocarbons (PAHs). However, the effects of these recalcitrant PAHs on the degradation characteristics of low-molecular-weight PAHs are largely unknown. This study was conducted to investigate the effects of pyrene (PYR) and fluoranthene (FLA) on the degradation characteristics of phenanthrene (PHE) in the cometabolism process by Sphingomonas sp. strain PheB4 isolated from mangrove sediments. Based on the kinetics and characteristics of PHE metabolites, it was proposed that the transformation of hydroxylated PHE into 1-hydroxy-2-naphthoic acid was a rate-limiting step in the degradation of PHE by strain PheB4. Compared to a single addition of PYR or FLA, the presence of a mixture of PYR and FLA decreased the degradation rate of PHE to a larger extent, which was likely because it could inhibit the production of 1-hydroxy-2-naphthoic acid more effectively. 相似文献
