Algae(and their extracts)are increasingly important for pharmaceutical applications due to the diversity of useful compounds they contain.The genus Fucus contains one of the most studied species,Fucus vesiculosus.The species F.ceranoides differs from the others of the genus by presenting longitudinal air-vesicles and a capacity to survive at low salinities.It is an alga that inhabits the Mondego River estuary(Portugal),at the southern limit of its distribution,and can serve as a role model to understand the effect of a salt gradient on the production of bioactive compounds.We assessed the phenolic content and antioxidant activity of different F.ceranoides extracts(e.g.methanolic,aqueous and polysaccharide)prepared from samples harvested from two different zones to evaluate if the adaptation of F.ceranoides to different salinity levels influenced its chemical composition.The antioxidant activity of the extracts was determined using 1,2-diphenyl-picrylhydrazyl(DPPH)and 2.2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)radicals.These assays demonstrated that the methanolic extract of lyophilized F.ceranoides that grew at low salinities was the most bioactive,i.e.DPPH(IC 50=50.39μg/mL)and ABTS(TEAC=2.42).The total phenolic content(Folin-Ciocalteu method)and the methanolic extract of the lyophilized F.ceranoides collected from a low salinity habitat exhibited the highest phenolic content(PGE=49.48μg/mg of lyophilized extract)amongst those sampled.Thin layer chromatography(TLC)and Fourier transform infrared spectroscopy(FTIR)were used for the identification of compounds in the extracts.This characterization allowed confirmation that the various extracts contained almost the same compounds but with notable quantitative differences.Based on these results,we conclude that there were differences in the quantity of the compounds due to the effect of salinity.The drying methods used were also found to have influenced the quality of the extracted compounds.  相似文献

Screening bioactive natural products from bacteria is a determinative step in the drug discovery programs. The present study aim to isolate actinobacteria from the Oman Sea sediments for determining the effects of different culture media and treatments on the yield of the isolation process, and measure the DPPH radical scavenging and Artemia cytotoxic activity of culture extracts of the actinobacterial isolates. A total of 290 actinobacterial isolates were collected from 14 sediment samples. Heat treatment(40.68%) and M_4 medium(29.31%) exhibited the maximum isolation rates of actinobacteria. Streptomyces isolates were dominantly distributed in all of the investigated stations according to 16 S rRNA gene sequencing. The distribution pattern of Streptomyces followed a depth-dependent frequency trend, whereas the members of rare genera including Micromonospora, Nocardia Actinoplanes, Nocardiopsis, Saccharopolyspora and Crossiella were distributed in deeper stations. Approximately,25% of the examined isolates could scavenge 90% of 10~(–4) mol/L DPPH solutions at 1 250 μg/mL final concentration of their ethyl acetate culture extracts. Furthermore, the most potent extracts could scavenge DPPH radicals with IC50 ranges from 356.8 to 566.4 μg/mL. Brine shrimp cytotoxicity tests showed that 38.88% of the examined culture extracts exhibited LC_(50) lower than 1 000 μg/mL against the Artemia cells. Moreover, the most potent culture extracts exhibited LC_(50) range from 335.4 to 534.4 μg/mL. Phylogenetic analysis by 16 S rRNA gene sequence revealed that the OS 005, OS 263 and OS 157 closely related to Streptomyces djakartensis, Streptomyces olivaceus and Nocardiopsis dassonvillei respectively. These results suggested the widespread distribution of the antioxidant and cytotoxic producing actinobacteria in the Oman Sea sediments, which could be considered as promising candidates for the discovery of microbial bioactive compounds.  相似文献

大气CO2浓度升高引起的海洋酸化可能对浮游植物造成不同程度的影响。而近海浮游植物不仅面临着海水酸化问题,还会受到海水溶解性CO2降低及pH升高(海水碱化)的影响。本实验以斑点海链藻(Thalassiosira punctigera)为研究对象,测定7个不同pCO2水平(25 μatm、50 μatm、100 μatm、200 μatm、400 μatm、800 μatm、1 600 μatm)下的生长、光合作用和呼吸作用速率、细胞粒径、叶绿素a和生物硅含量以及叶绿素荧光等参数。结果表明,与400 μatm相比,在海水酸化(pCO2 > 400 μatm)和海水碱化(pCO2 < 400 μatm) 条件下,斑点海链藻的生长速率和叶绿素a含量都显著降低,但是碱化条件下降低的程度更大。此外,碱化处理的藻细胞光合作用速率、最大量子产量(Fv/Fm)和最大相对电子传递速率(rETRmax)都显著低于400 μatm培养下的细胞,而呼吸作用速率显著升高,但是生物硅含量和细胞大小无明显变化。研究表明海水碱化和海水酸化均会抑制其生理活动,而且海水碱化对其影响更显著。这表明正常pCO2生长下的藻细胞具有最适的生理状态。本研究可为探究海水碳酸盐系统变化对海洋初级生产力的影响提供一定的数据支持。  相似文献

Paralytic shellfish toxins(PSTs) are notorious neurotoxins that threaten public health and food safety worldwide.Although PST monitoring programs have recently been established throughout China, the profiles and variation of PSTs in important commercial clams(e.g., Mactra veneriformis, Ruditapes philippinarum, and Meretrix meretrix) along the Jiangsu Province coastline remain largely unexplored. In this study, a validated hydrophilic interaction liquid chromatography–tandem mass spectrometry(HILIC-MS/MS) method was used to examine PST profiles and levels in 540 clam samples from natural production areas along Jiangsu Province coastline during2014–2016. Although the PST levels(≤6.38 μg saxitotoxin equivalents(eq)/kg) were consistently below European Union regulatory limits(≤800 μg saxitotoxin eq/kg) during this time period, saxitotoxin, decarbamoylsaxitotoxin,and gonyautoxins 1 and 4 were detected, and nearly 40% of the samples were saxitotoxin-positive. The PST levels also varied significantly by seasons, with peak values observed in May during 2014–2016. This is the first systematic report of PSTs in clams from Jiangsu Province, and additional research and protective measures are needed to ensure the safety of clams harvested in this area.  相似文献

Seasonal variation of marine plankton spatial distribution is important in understanding the biological processes in the ocean.In this study,we studied spatial distribution of planktonic ciliate abundance and biomass in the central deep area(station depth greater than 60 m) and the coastal shallow area(station depth less than 60 m) of the southern Yellow Sea(32°–36.5°N,121°–125°E) in spring(April) and autumn(October–November) of 2006.Our results showed that both ciliate abundance and biomass in the surface waters were higher in spring((1 490±2 336)ind./L;(4.11±7.81) μg/L) than in autumn((972±823) ind./L;(1.11±1.18) μg/L,calculated by carbon).Ciliate abundance and biomass in the surface waters of the coastal shallow area were similar in spring and autumn.However,in the central deep area,those values were much higher in spring((1 878±2 893) ind./L;(5.99±10.10)μg/L) than in autumn((738±373) ind./L;(0.74±0.76) μg/L).High values of ciliate abundance and biomass occurred in the central deep area in spring and in the coastal shallow area in autumn.Mixotrophic ciliate Laboea strobila was abundant in the central deep area in spring,when a phytoplankton bloom occurred.However,in autumn,L.strobila was abundant in the coastal shallow area.Boreal tintinnid Ptychocyli obtusa was found in spring.Both L.strobila and P.obtusa were concentrated in the surface waters when their abundance was more than 1 000 ind./L.Peaks of these species were in the subsurface waters when their abundance was less than 400 ind./L.This study showed that both high abundance and biomass of ciliates occurred in different areas in southern Yellow Sea seasonally.  相似文献

The study was to find out the effect of cadmium and mercury on levels of hemolymph glucose, crustacean hyperglycemic hormone (CHH) and hepatopancreas glycogen in the blue swimmer crab Portunus pelagicus. The experiments were performed in both intact and eyestalk ablated crabs. Quantification of CHH was done by the indirect ELISA with the aid of primary anti-Carcinus maenas-CHH antibody. Higher glucose concentration was observed on exposure to 8×10-6 of cadmium ((825.6±5.42) μg/mL) and 6×10-6 of mercury ((90.5±6.25) μg/mL) after 48 h and 24 h respectively. Higher level of hemolymph glucose was observed in eyestalk intact crabs on exposure to cadmium and mercury than eyestalk ablated crabs. Decrease in the levels of CHH was observed in both eyestalk intact and ablated crabs on heavy metal exposure. Decline of the hepatopancreas glycogen level was also witnessed with the exposure to heavy metal, which validated its utilization in the production of glucose. Thus this study brings to light, the variations in hemolymph glucose, CHH and hepatopancreas glycogen on heavy metal stress. These carbohydrate metabolites can be used as biomarkers in assessing heavy metal contamination in water bodies.  相似文献

白令海、西北冰洋等高生产力海域在北冰洋“生物泵”中起到重要作用;海水升温、海冰消退等北极快速变化,将强烈影响该海域“生物泵”的结构与规模,并在沉积物中有机质的来源与新鲜程度上有所体现,可用脂肪酸加以指征。对第五次、第六次中国北极科学考察在以上海域采集的表层沉积物进行脂肪酸含量(以沉积物干重计)及组成分析,结果显示楚科奇海陆架总脂肪酸含量非常高((97.15± 55.31) μg/g),白令海盆最低((15.00±1.30) μg/g),加拿大海盆、楚科奇海陆坡、白令海陆架居中(分别为(88.65 ± 3.52) μg/g,(70.35±11.32) μg/g与(38.28±14.89) μg/g)。海源脂肪酸占总脂肪酸比例最高(86.82%±7.08%),陆源次之(8.45%±6.62%),细菌最低(4.63%±2.24%);硅藻指数(16:1ω9/16:0)在楚科奇海陆架(> 0.82)、白令海陆架边缘(> 0.65)较高,其他区域均较低。脂肪酸结果表明:(1) 该海域沉积有机质主要来自海源,陆源贡献小;在北部、南部楚科奇海陆架、白令海陆架边缘,硅藻生物量占主要优势;细菌脂肪酸比例显著低于温暖海域,指示低温抑制细菌活动。(2) 楚科奇海陆架区硅藻生产力高、细菌活动弱,新鲜有机质沉降效率高,但对未来海水升温、浮游植物群落变化也较为敏感。(3) 加拿大海盆、楚科奇海陆坡的浮游植物群落由绿藻与金藻主导。以上结论说明脂肪酸可指示表层沉积物中有机质的来源与新鲜程度;未来,脂肪酸有望进一步揭示北冰洋“生物泵”对北极快速变化的响应。  相似文献

在每100g等氮、等能纯化饲料中分别添加0,100,200,400,800,1 000mg维生素C(Vc)多聚磷酸酯(LAPP),用这样配制成的试验饲料饲喂已淡化5d的河蟹大眼幼体及后续幼蟹25d,探讨Vc对大眼幼体和幼蟹生长、存活、蜕壳频率及抗逆性的影响,同时探明幼蟹对Vc的适宜需求量,结果表明,饲料Vc添加量为100~400mg/100g时,可以保证幼蟹正常生长、存活和蜕皮,尤其以投喂添加Vc200mg/100g(C2组)和400mg/100g(C3组)饲料的Ⅲ期幼蟹增重率、存活率和平均蜕皮频率较高,其中C2组(上述各指标值)分别为516.18%,62.83%,2.58,C3组分别为458.89%,66.17%,2.61;饲料中缺少Vc时(如C0组),Ⅲ期幼蟹的增重率、存活率和平均蜕皮频率最低,分别为265.31%,26.67%和2.26.饲料中Vc的添加量达到800mg/100g(C4)和1 000mg/100g(C5)时,幼蟹的蜕皮反而受到抑制,平均蜕皮频率降低,蜕皮周期延长,且存活率降低,到Ⅲ期幼蟹的增重率、存活率和平均蜕皮频率在C4组分别为355.7%,31.33%,2.39,在C5组分别为307.53%,28.56%,2.19.此外,各处理组幼蟹对抗逆性影响的试验表明,在一定的范围内,饲料中添加Vc能提高幼蟹的耐低氧能力和耐pH突变能力,而对耐盐度突变能力没有明显的影响.综合上述结果可以得出,为保持幼蟹良好生产性能,饲料中添加的Vc?  相似文献

为研究带鱼(Trichiuruslepturus)蛋白复合酶解液中抗菌肽(ABP)的抑菌性及表征活性,本文以舟山小带鱼为研究对象,制备蛋白抗菌肽。研究得到抗菌肽分子量在区间(3000Da, 3000—800Da,800Da)内均有分布,在3000—800Da区间内含量达到49.61%,酶解程度最深,氨基酸总量为881.83mg/g,必需氨基酸与氨基酸总量比为44.75%,必需氨基酸与非必需氨基酸比为81.33%,营养价值高。六类提升抗菌效力的氨基酸占氨基酸总量42.71%,抗菌效力较高,具有广谱抗菌活性,为阳离子型抗菌肽,对G–具有较强的抑菌效能。抗菌效力与稀释128倍(10.15U/mL)土霉素抗菌效力相当,与稀释4倍的硫酸链霉素(325U/mL)抗菌效力相当。对抗菌肽的表征活性进行研究表明:该抗菌肽在pH 2—12下溶解度良好,均大于88%,有强适应性;乳化性随pH增大而增强, pH 4时乳化性最低,为53.68%,在pH 12时达到最高值78.34%;乳化稳定性在pH 6时达到最高,为85.83%, pH环境过酸或者过碱的情况下乳化稳定性均较低;发泡性随pH上升呈现增长趋势,在pH 12时达到最大值57.33%, pH 4时抗菌肽泡沫稳定性最低,为22.86%, pH升高,泡沫稳定性也随之增强,在pH 12时达到最大值59.58%。该研究结果对探究带鱼抗菌肽的各项性质提供了理论依据,对带鱼抗菌肽的应用研究具有一定的理论指导意义。  相似文献

通过对采集自南海北部的D06和S0612两个短柱状沉积物样品中的不同赋存形态钡、有机碳和生物硅的含量以及有机质碳氮同位素组成的分析,结果表明沉积物中的钡主要以碎屑钡和自生的生物钡形式存在。沉积物中生物钡的含量在10.3~385.2 μg/g之间,平均值为177.0 μg/g,据此计算的新生产力在12.3~146.7 mg/(d·m2)(以碳计)之间,平均为78.9 mg/(d·m2);D06站计算的结果和附近站位的实测值相当。沉积物中有机碳的含量在0.64%~1.34%之间,平均值为0.89%,C/N比值为4.96~5.93,平均值为5.54。有机碳的δ13C值在-22.98‰~-20.73‰之间,平均值为-21.46‰,依据端元组份同位素组成计算的陆源有机质比率显示,D06站位的有机质主要来自海洋生物,而S0612站位则受陆源有机质的影响较大。沉积物中有机质的δ15N值在3.96‰~6.29‰之间,平均值为5.26‰,反映的是该海区氮源的同位素组成,而不受硝酸盐利用率的影响。  相似文献

The stalked barnacle Pollicipes pollicipes is a cirriped crustacean that lives on very exposed rocky shores. This barnacle is the most important economical resource on intertidal rocky shores of continental Portugal. It is highly prized as food and heavily exploited (professional and recreational fishery), but fishery data are scarce and do not estimate the real pressure upon this resource.Despite its socio-economic interest, specific regulations on this fishery are recent and different along the Portuguese coast. Four regions with different regulation can be identified: the marine reserve “Reserva Natural das Berlengas” (RNB) and the marine park “Parque Marinho Prof. Luiz Saldanha” (PMLS) (both in central Portugal); the natural park located in SW Portugal (“Parque Natural do Sudoeste Alentejano e Costa Vicentina”, PNSACV); and the rest of the coast.The main objective of the present study was to study the spatial patterns of percentage cover, biomass, density and size structure of P. pollicipes in areas with different exploitation regimes, including harvested areas and no-take areas. Additionally, variability between mid shore and low shore barnacles was also analysed.Seven areas were sampled with a variable number of sites (a total of 24) randomly sampled in each area during 2011. Photographs and image analysis (percentage cover) and destructive sampling (density, biomass and size) were used.In general, percentage cover, biomass and density were higher in mid shore when compared to low shore, namely in harvested areas. Low shore barnacles had a higher proportion of adults with moderate and high commercial value, while juveniles were relatively more abundant at mid shore.There were no consistent differences in the patterns of distribution and abundance of P. pollicipes among areas subject to different exploitation regimes. The most different area was the harvested area by professional fishers in RNB, where the highest biomass within the study was registered (mid shore, 7.7 kg·m 2). Barnacles within this area presented a higher proportion of adults with commercial value, while recruits and juveniles were relatively more abundant in other areas.The hypothesis of a highest percentage cover, density and biomass in the low shore of no-take sites was not supported.  相似文献

The biomass and size fraction of phytoplankton in terms of chlorophyll a(Chl a) was measured during four cruises conducted in April, July, October 2013 and January 2014 in mariculture area, the Sanggou Bay, China.Results show that total Chl a levels in the surface seawater of the Sanggou Bay generally range from 0.10 to 20.46μg/L, with an average value of 2.13 μg/L. Nano-phytoplankton was the most important size-fraction and accounted for about 65.1% of total Chl a. In order to evaluate the importance of the "protozoan trophic link" for energy transfer from the microbial loop to filter-feeding feeders, Zhikong scallop Chlamys farreri was then offered a natural planktonic community as potential prey. Results show that scallops obtained carbon source from natural plankton with the rate of 11 033.05 μg/(g·d). Protists(nanoflagellates and ciliates) were the dominant source of carbon retained by scallop(48.78%). The microbial loop provided 58.45% of the carbon source for farmed scallops. These results indicate that the microbial loop represent a valuable trophic resource in mariculture system of the Sanggou Bay.  相似文献

以三角褐指藻(Phaeodactylum tricornutum)为研究材料,比较了Na NO3、NH4HCO3和CO(NH2)2为氮源的两种培养基(m L1和ASW培养基)对其生长和生物活性成分(岩藻黄素、金藻昆布糖和二十碳五烯酸(C20:5,EPA))时相积累的影响,同时分析了脂肪酸组成和总脂含量的变化。结果表明:以m L1培养基培养时,三角褐指藻的生物质质量浓度明显高于ASW培养基培养时的生物质质量质量浓度,尿素优于其他两种氮源,最大生物质质量质量浓度为3.7 g/L。不同培养条件下岩藻黄素含量的时相变化规律一致,均随着培养时间的延长呈现先增加后减少的趋势,其最高积累量分别为:13.27 mg/g(Na NO3)、13.23 mg/g(CO(NH2)2)和13.89 mg/g(NH4HCO3)(m L1);13.2 mg/g(Na NO3)、14.92 mg/g(CO(NH2)2)和13.6 mg/g(NH4HCO3)(ASW),由此可知氮源对岩藻黄素积累量影响不大。金藻昆布糖含量随着培养时间延长逐渐增加,其最大积累量分别为9.82 mg/g(NH4HCO3)(m L1)和8.59 mg/g(Na NO3)(ASW)。不同培养条件下其总脂含量变化不显著,均在培养末期达到最大值,分别为24.18%(NH4HCO3)(m L1)和23.79%(Na NO3)(ASW);其主要脂肪酸组成为:豆蔻酸(C14:0)、棕榈酸(C16:0)、棕榈油酸(C16:1)、硬脂酸(C18:0)、油酸(C18:1)、亚油酸(C18:2)、花生一烯酸(C20:1)、木焦油酸(C24:0)和EPA,其中,EPA含量随着培养时间延长逐渐下降,尿素最有利于EPA的积累。  相似文献

于2005年5月至2006年3月对浙江沿岸重要的3种经济贝类泥蚶、缢蛏和僧帽牡蛎的呼吸率与排泄率进行了周年研究。结果表明:泥蚶、缢蛏和僧帽牡蛎耗氧率的变化范围分别为0.11~5.05 mg/(g·h)、0.77~5.97 mg/(g·h)和0.33~5.47 mg/(g·h),排氨率的变化范围分别为21.51~1 078.63 μg/(g·h),26.97~990.73 μg/(g·h)和32.12~1 378.67 μg/(g·h);耗氧率与排氨率9月份最高,1月份最低。经单因素方差分析发现,3种实验贝类月际间的耗氧率与排氨率均存在极显著差异(p<0.01),经配对T检验发现,缢蛏(SC)和僧帽牡蛎(SCA)之间的耗氧率存在着显著差异TSC-SCA=3.184,p=0.024),泥蚶(TG)与缢蛏、僧帽牡蛎的耗氧率之间都不存在差异(TTG-SC=1.887,p=0.118;TTG-SCA=0.246,p=0.815),泥蚶、缢蛏和僧帽牡蛎之间的排氨率的差异均不显著(TTG-SC=0.977,p=0.373;TTG-SCA=2.369,p=0.064;TSC-SCA=1.002,p=0.362)。3种贝类的耗氧率与排氨率均随温度的升高而升高,经回归分析发现,耗氧率和排氨率与温度的变化均呈较显著的幂指数性相关lnY=lna+blnTY=aebT)。泥蚶呼吸排泄O:N变化范围为4.69~28.55,缢蛏和僧帽牡蛎的O:N变化范围分别为:4.68~37.89和3.04~10.27。  相似文献

利用分级浸取分离法将黄海、东海陆架区沉积物氮分为转化态氮(TF-N)和非转化态氮,并将可转化态氮区分为4种形态:离子交换态氮IEF-N 、碳酸盐结合态氮CF-N 、铁锰氧化态氮IMOF-N及有机态和硫化物结合态氮OSF-N。 对各形态氮的平面、垂直和沉积剖面年际分布进行了分析,并进一步探讨了该区域氮形态与古生产力的替代指标——生物硅(BSi)的相关性,揭示了氮形态的地球化学特征及与浮游植物总量的关系。结果表明,黄海、东海陆架区表层沉积物中可转化态氮占总氮的百分比为16.81%,可转化态氮中4种形态氮的平均含量为:IMOF-N(66.65 μg/g)>IEF-N(22.96 μg/g)>OSF-N(17.40 μg/g)>CF-N(11.26 μg/g),IMOF-N是可转化态氮的优势形态;柱状沉积物中各形态氮垂直分布不同,长江口沉积物中各形态氮垂向变化幅度大于南黄海中部;离子交换态氮IEF-N和铁锰氧化态氮IMOF-N对浮游植物总量贡献较大。  相似文献

The distribution of inorganic nitrogen compounds and the metabolic rates of these compounds by microorganisms as a whole were investigated in the Seas of Hiuchi and Bingo. The results obtained are as follows:
  1. Of inorganic nitrogen compounds, the contents in sea water, those in bottom muds, the uptake or liberation rates of microorganisms as a whole in sea water, and the liberation rates from bottom muds to sea water are 0.2~4.0 μg at. N/l, 3~60 μg at.N/100 g, 0.01~0.5 μg at.N//lhr, and 0.3~1.9 μg at.N/100 cm2/hr, respectively, and these contents or rates of ammonia usually are the largest of these inorganic nitrogen compounds.
  2. From the above-mentioned results and the others, it is suggested that the nitrogen in the seas circulates mainly in sea water itself and the course of nitrogen cycle, which passes through bottom muds, is not so important, and further that, of the cycle of inorganic nitrogen compounds, the main course is the course which ammonia is liberated from organic nitrogen compounds and it is immediately uptaked by microorganisms, and the course which it is oxidized to nitrate and the others are not so important.
