Abstract: | Yeast two-hybrid systems have contributed ver y significantly to the unraveling of molecular regulatory networks by facilitati ng the discovery of protein-protein, protein-DNA, protein-RNA and protein-sm all molecule interactions. In this article, we describe the two-hybrid technolo gy, mainly based on the Gal4-based system, and its applications and limitations . We also introduce several variations of the two-hybrid systems, i.e. one-hyb rid, three-hybrid and reverse two-hybrid systems for different applications. F inally, the Sos recruitment system (SRS) is presented. Judging from the robust s uccess in other fields, application of yeast two-hybrid systems and their varia tions to marine molecular biology will promise to have significant discoveries. |
