摘    要:本实验研究了高温(28℃,33℃,35℃,37℃)胁迫下草鱼(Ctenopharyngodon idellus)体内热休克蛋白(Heat shock protein,HSP)的表达模式以分析高温影响草鱼存活的生理机制。在不同温度下,实时记录草鱼存活状态和呼吸频率,并利用蛋白杂交方法测定热休克蛋白(Hsp70,Hsp73)表达量的变化。结果表明,当水温超过28℃,草鱼存活率随温度升高而降低,在35℃和37℃时全部死亡;呼吸频率随温度升高而升高,各温度间差异显著(P<0.05);温度升高会诱导草鱼肝脏和肌肉中Hsp73表达量升高,表明热休克蛋白的上调表达是草鱼抵御温度应激的重要生理响应机制。

Abstract:Effect of thermal stress on survival of the grass carp(Ctenopharyngodon idellus) was studied.And through analyzing the mode of the expression of the heat-shock protein in the liver and muscle under different thermal regimes,the related physiological mechanism was preliminary elucidated.Fishes were reared under four different temperatures(28℃,33℃,35℃,and 37℃).During the experiment,the survival and respiratory frequency were observed and recorded timely.The expression of the 70-kD heat shock protein(Hsp70,Hsp73) was determined by using Western blotting.When the water temperature exceeded 28℃,the survival of grass carp decreased with temperature increasing.At 35℃ or 37℃,all fishes died.The respiratory frequency increased rapidly with the increasing temperature either,and significant differences existed among different temperatures(P<0.05).The high temperature can also induce upregulation of Hsp73.The pattern of Hsp73 expression had a close relationship with the thermal tolerance of the carp.
