摘    要:取喂食东海海参60d的ICR小鼠肝脏,制备微阵列基因表达芯片,并与模式小鼠芯片进行杂交,筛选功能基因和差异表达蛋白.从食用海参的鼠肝中共筛选出了4180个基因,其中差异表达基因184个,上调基因107个,下调基因77个.利用MAS.2.0系统对其功能进行分析,结果显示,分子功能上有催化活性的基因占39.86%,结合功能...

Abstract:In order to make full use of sea cucumber Acaudina leucoprocta nutritional and health care values, this paper reported on screening differentially expressed genes of mice fed on sea cucumber using microarray hybridization. The results showed that a total of 4180 genes were screened from livers of mice fed with sea cucumber, of which 184 differentially expressed genes were found, 107 genes were found up-regulated and 77 genes were found down-regulated. Using MAS.2.0 to analyze molecular function of cluster, results showed that 39.86% of genes were with catalytic activities, genes with combinational function accounted for 30.1%, signal transduction, molecular transduction, electron transfer, molecular motors, and other functional genes accounted for 22.1%. Bio-chemical process analysis showed 67.4% of genes were with biochemical metabolic function, 16.2% were regulatory genes, and genes related to development were accounted for 9.1%. Analysis based on cell components, 51% of genes were responsible for making different components of cell, 21.6% were for organelles, proteins and other macromolecular complexes were 12.4%, 4.6% was for extracellular matrix, and 5.2% was for other small particulate matter. The differentially expressed proteins from the consumption of sea cucumber are involved in cell signaling factors, synthesis, catabolism enzymes and for the energetic materials. Therefore, long-term consumption of sea cucumbers can improve immunity, anti-fatigue, and anti-oxidation.
