应用分子生态学方法, 研究在不同芳香族化合物胁迫下海洋沉积物中细菌群落结构的演替 规律。在实验室建立驯化系统, 在海洋沉积物中分别添加芘和苯甲酸钠, 采用Illumina MiSeq 高通量 测序技术分析实验组和对照组细菌群落16S rRNA 基因V3-V4 可变区数量的组成及变化, 研究沉积物 中添加芘、苯甲酸钠后, 微生物群落多样性和群落结构的变化。实验结果表明: 芘、苯甲酸钠极大地 降低了海洋沉积物中菌群丰度和多样性; 在添加苯甲酸钠的实验组, 变形杆菌门中海杆菌属 (Marinobacter)、假海源菌属(Pseudidiomarina)的细菌显著富集; 在添加芘实验组中拟杆菌门中的 Balneola 属、碳酸噬胞菌属(Aequorivita)极大富集。初步分析了在海洋沉积物环境中不同多环芳烃胁迫 下细菌群落结构的演替特征, 实验结果为海洋环境中针对不同芳香烃的微生物修复提供理论依据。  相似文献

在分析废旧金属拆解场地浅层地下水中芘的检出和分布特征的基础上,对地下水中芘的污染来源、污染途径和污染控制因素进行了探讨.研究结果表明,场地中心和地下水下游2km范围内芘污染明显,垂直于地下水流向芘迁移距离较小,一般小于0.5km.芘在地下水中的迁移转化受地下水中pH、Eh及DO等因素的控制,弱酸性或中性环境以及厌氧强还原条件下,芘易于检出和迁移,一般难以生物降解.  相似文献

用频率分析、逐步聚类和最小二乘拟合等方法研究了天津地区表土中蒽和芘含量的频率分布特征。结果表明,两者都呈双峰分布,其中蒽的双峰特征尤为典型。可以用双高斯密度函数很好地拟合经对数变换的实际观测数据,证明天津地区蒽和芘都包括两个相对独立的对数正态分布总体。根据拟合频率密度函数求得的蒽的两个子总体几何均值分别为2.73±2.17ng/g和26.52±1.55ng/g,芘则为11.05±2.11ng/g和82.52±3.24ng/g。芘的两个总体在空间上的分异比蒽更明显,其高值总体主要集中在城区、近郊区和开发区,而蒽的高值总体仅有局部聚集的一般趋势。芘与芘/苯并[a]芘之比的空间分布特征十分相似,表明两个芘含量总体并存的原因与排放源特征有关。  相似文献

In animals, biotransformation and the immune system interact with each other, however, knowledge of the toxic mechanism of benzo[a]pyrene (BaP) on these two systems is not well known. The present study investigated the toxic effects of BaP on the biotransformation system, cortisol level and DNA integrity of red sea bream (Pagrus major). The results showed that cortisol level was induced under the challenge of lipopolysaccharide (LPS). Short-term exposure (96 h) of BaP at environmental concentration significantly increased the cortisol level, hepatic EROD activity and CYP1A1 mRNA expression. When P. major was exposed to BaP for 14 d followed by LPS challenge this increased the cortisol level, EROD activity and hepatic DNA damage except CYP1A1 mRNA expression. Combined with our previous data, which showed that BaP exposure can modulate the immunologic response in P. major challenged with LPS, a hypothetical adverse outcome pathway of BaP on fish was suggested.  相似文献

Scallop Chlamys farreri was exposed to different concentrations of benzo(a)pyrene (BaP) (0.5 μg/L, 1.0 μg/L, 10.0 μg/L and 50.0 μg/L) for 30 days in seawater. The 7-ethoxyresorufin O-deethylase (EROD) activity was significantly induced, and increased with the increasing BaP concentration. The glutathione-S-transferase (GST), superoxide dismutase (SOD), catalase (CAT), Glutathione peroxidase (GPx) activities increased in short time at low concentration of BaP, and was significantly depressed at high concentrations. Scallop gill was more sensitive to BaP than the digestive gland, and the digestive gland was the main tissue to deal with oxyradicals. The contents of malondialdehyde (MDA) increased with the exposure time and there was a positive correlation (concentration-effect) between the MDA content and the concentration of BaP. The biomarkers determined in this experiment had important roles in detoxification, and showed great potential as biomarkers for oxidative stress. Controlled laboratory experiments designed to simulate field exposure scenarios are particularly useful in ascertaining biomarkers suitable for use with complex contaminant mixtures in the marine environment. The toxic effects of benzo[α]pyrene (B[α]P) at different concentrations (0.1, 0.5, 1, 2.5 and 7.5 μgL^-1) on scallop (Chlamys farreri) immune system were studied. The results showed that B[α]P had significant toxic effects on the haemocyte counts, neutral red uptake, phagocytosis, bacteriolytic and antibacterial activity (P〈0.05), while the seawater control and acetone control had no significant differences. The haemocyte counts, neutral red uptake, phagocytosis and bacteriolytic activity in all B[α]P treatment groups as well as antibacterial activity in groups of 0.5, 1, 2.5 and 7.5 μgL^-1·B[α]P decreased significantly (P〈0.05). Some of these indices tended to be stable on the sixth day and others on the ninth day, and the indices showed clear time- and concentration-response to B[α]E Bactenolytic activity in 0.1 μgL^-1 B[α]P treatment group and antibacterial activity in 0.1 ggLl and 0.5 μgL^-1 B[α]P treatment groups increased at the beginning of exposure and reached their peaks on day 1 and day 6, respectively. Following that, both activities decreased gradually and became stable after day 9. When all the indices reached stability, they were significantly lower than those in control group (P〈0.05), except for antibacterial activity in 0.1 μgLl B[α]P treatment group (P〉0.05). Thus, B[α]P has evident toxic effects on scallop immune system, which supports the view that a relationship exists between pollution and lmmunomodulation in aquatic organisms.  相似文献

To investigate the genotoxic effect of marine sediments on aquatic organism, sediment samples were collected from 13 sites along the coast of Gwangyang Bay (Korea). Concentrations of polycyclic aromatic hydrocarbons (PAHs) in sediments were determined and the relationship between exposure of flounder blood cells to sediment extracts and DNA single-strand breakage in the blood cells was examined using the comet assay. Levels of DNA damage were proportionally increased by exposure concentration and the highest sediment-associated DNA damage was observed at the station showing the highest PAHs contamination. DNA damage in blood cells exposed to five types of PAHs (benzo[a]pyrene, fluoranthene, anthracene, pyrene and phenanthrene) and in flounder (Paralichthys olivaceus) exposed to benzo[a]pyrene (BaP) for 0, 2 and 4 days were assessed by measuring comet tail length. The tail lengths of five PAHs-exposed groups at 50 and 100 ppb were significantly different from the non-exposed group, and the genotoxic effect of BaP correlated with both concentration and duration of exposure. Throughout the study, significant differences in DNA breakage were recorded between cells exposed to sediment extracts or PAHs and non-exposed control. This study demonstrated the comet assay as a successful tool in monitoring contamination of marine sediments and assessing genotoxicity of PAHs in marine organisms, either in vitro or in vivo.  相似文献

海洋环境中微塑料和多环芳烃(PAHs)污染日益严重,以滤食动物菲律宾蛤仔(Ruditapes philippinarum)为研究对象,探讨了聚苯乙烯微塑料和芘单一及联合暴露对菲律宾蛤仔的毒性效应.分别采用两个聚苯乙烯微塑料粒径(0.3μm和6μm,20 μg/L)和两个芘浓度水平(10 μg/L和100μg/L)单独和联合暴露21d,测定其对菲律宾蛤仔生理活动(肥满度和摄食率)、免疫防御、氧化应激等相关生物标志物的影响.研究结果表明,除芘单一暴露组外,其余暴露组菲律宾蛤仔的摄食率与对照组相比都显著降低,但这两种污染物对菲律宾蛤仔的肥满度没有显著影响.微塑料和芘暴露均导致菲律宾蛤仔免疫功能受损,表现为血细胞凋亡率增加和吞噬活性被抑制;其中,在小粒径微塑料和高浓度芘存在的暴露组血细胞凋亡率均显著高于对照组,并且二者联合暴露组血细胞凋亡率最高;除大粒径微塑料暴露组外,各暴露组血细胞吞噬活性均显著低于对照组.菲律宾蛤仔抗氧化酶系统不能及时清除体内产生的自由基,导致机体出现氧化损伤现象,表现为抗氧化酶活性变化和丙二醛(MDA)含量升高.微塑料和芘联合暴露导致鳃组织谷胱甘肽疏基转移酶(GST)活性显著升高,消化腺组织过氧化氡酶(CAT)和超氧化物歧化酶(SOD)活性显著降低.综合生物标志物指数(IBR)结果显示,联合暴露对菲律宾蛤仔造成的胁迫压力高于单一暴露组.  相似文献

传统的液液萃取技术主要采用与水不相溶的有机溶剂作萃取剂,这种异相萃取的传质速率一般较低,需反复振荡或多次萃取.本文建立了以乙腈-硫酸铵-水的双水相萃取体系高效液相色谱法测定地下水中苯并(a)芘和苯并(e)芘的方法,以密度比水小的乙腈为萃取剂,将其与水样混合(互溶)获得乙腈-水体系,然后加入硫酸铵,溶液澄清、分相后取上层有机相(乙腈)进样分析.考察了萃取剂的选择、双水相的形成条件、离子强度以及pH等对测定结果的影响.两种化合物线性范围为2.00~ 400.00 ng/mL,相关系数大于0.999.方法检出限为0.012~0.020 ng/mL,平均回收率为94.6%~97.3%,相对标准偏差为1.3% ~2.5%.与传统萃取方法相比,该方法具有操作流程简单、快速的特点,可实现对野外环境水样进行原地样品前处理.  相似文献

从红树植物红海榄叶片中分离一株对芘具有较好降解作用的海洋细菌,命名为B11,并对其菌体特征、生长条件及降解效能等进行了系统研究。结果发现该菌株为革兰氏阴性菌,结合其生理生化特征和16S rDNA序列比对,表明此菌株属于交替单胞菌属(Alteromonas sp.)。菌株在芘的起始浓度为100 mg/L,pH为中性或偏碱性条件下,盐度35时对芘的降解作用最强。外加碳源对B11对芘的降解效果都具有一定的抑制作用,其中水杨酸的抑制作用更为显著。综合菌株B11的生长特性和降解效能,初步认为B11菌株较适合用于降解多环芳烃芘,可用于红树植物多环芳烃生态污染的原位修复。  相似文献
