Iron-reducing activity of autochthonous bacteria from two temporary hydromorphic soils is evaluated by the study of iron reductive dissolution, as a function of water content. The release of ferrous iron in solution is coupled to the mineralization of soil organic carbon. Water soil saturation is not necessary for iron reductive dissolution, since the highest dissolution is obtained for a wet, but not water-saturated soil (100% of water holding capacity WHC), and dissolution is also very high in a soil at 75% WHC. 半乳糖凝集素-8基因(galectin-8,Gal-8)属于凝集素家族一员。已知Gal-8在哺乳动物的先天免疫中发挥重要作用,然而在鱼类中却鲜有报道。本研究通过转录组测序获得大弹涂鱼(Boleophthalmus pectinirostris)半乳糖凝集素-8样基因(galectin-8 like,Bp Gal-8L)c DNA序列。首先确定其序列特征,其次采用实时定量PCR(real-time quantitative PCR,q PCR)的方法鉴定Bp Gal-8L组织表达分布及其与迟缓爱德华氏菌(Edwardsiella tarda)感染相关性;最后分别通过原核表达获得N端和C端CRD区重组蛋白Bp Gal-8L-N和Bp Gal-8L-C,并采用玻片法观察这两个CRD区的细菌凝集活性和糖类结合特性。多重序列比对显示,Bp Gal-8L蛋白含有两个糖识别结构域(carbohydrate recognition domain,CRD),但只有C端的CRD区存在Gal家族典型的β-半乳糖苷糖类结合基序。系统进化树表明Bp Gal-8L属于鱼类Gal-8的一种亚型。q PCR结果显示,Bp Gal-8L基因m RNA在各组织中普遍表达,并且在迟缓爱德华氏菌感染后大弹涂鱼肝和脾中表达量呈上调趋势。细菌凝集实验结果显示,只有Bp Gal-8L-C重组蛋白对检测的5种革兰氏阴性菌及2种革兰氏阳性菌有凝集作用,并与乳糖和脂多糖特异性结合。综上所述,Bp Gal-8L与大弹涂鱼抗菌免疫密切相关,且其N端和C端CRD区发挥不同作用。  相似文献

环境样品的低生物量是微生物宏基因组学研究面临的首要挑战,通过基因组扩增技术来满足高通量测序对DNA样品量的需求是最常用的解决策略。MALBAC(Multiple Annealing and Looping Based Amplification Cycles)基因组扩增试剂盒最初为扩增和研究哺乳动物的单细胞基因组而研发。本文中,我们通过人工构建的微生物群落来检测该试剂盒在微生物宏基因组扩增方面的效率和应用可行性。结果表明,每个标准反应中,10 pg的DNA模板量足以满足MALBAC试剂盒对样品扩增的需要。每个标准反应DNA模板用量为10和100 pg时,所扩增DNA样品的基因组覆盖度与原始未扩增样品表现出高度的一致性,证明MALBAC试剂盒扩增效果的高度稳定性和一致性。常用的GenomePlex全基因组扩增试剂盒使我们可以在每个标准反应DNA模板量为100 pg的条件下扩增获得足够的DNA样品,但是结果表明该参照试剂盒无法有效的实现对群落中低丰度细菌菌株基因组的线性扩增。对于MALBAC试剂盒和参照试剂盒而言,在扩增高GC含量的微生物物种基因组DNA方面效率低下。我们的实验结果表明MALBAC试剂盒在高效扩增环境样品宏基因组DNA方面的可行性,但对该试剂盒在扩增环境样品中高GC含量微生物物种方面的适用性存在疑虑。  相似文献

The KwaZulu-Natal Bight is a shallow indentation of the eastern seaboard of South Africa, characterised by a narrow (45 km wide) extension of the continental shelf, with a shelf break at about 100 m. It has a complex hydrography: the waters of the bight are derived from the fast-flowing, southward-trending Agulhas Current, which is fed mostly by the tropical and subtropical surface waters of the South-West Indian Ocean subgyre, which are generally oligotrophic in nature, notably depleted in reduced nitrogen and phosphate except at river mouths and during periodic upwelling of deeper nutrient-rich water. Despite this, the bight is believed to be relatively productive, and it is suggested that efficient nutrient recycling by prokaryotes may sustain primary productivity efficiently, even in the absence of new nutrient inputs. Here we have measured bacterial numbers, biomass and heterotrophic productivity during summer and winter in conjunction with phytoplankton standing stock and factors that influence it. Bacterial distribution closely matched phytoplankton distribution in surface waters, and was highest close to the coast. Bacterial standing stocks were similar to those of oligotrophic systems elsewhere (0.5–5.0 × 105 cells ml–1; 1 × 10–8 to 1.25 × 10–7 g C ml–1) and increased in association with the development of phytoplankton blooms offshore and with inputs of allochthonous material by rivers at the coast. Heterotrophic productivity in summer was lowest in the far south and north of the bight (0.5 × 10–10 g C ml–1 h–1) but higher close to the shore, over shallow banks, and in association with increased phytoplankton abundance over the midshelf (1.0–3.5 × 10–9 g C ml–1 h–1). There were marked seasonal differences with lower bacterial standing stocks (5 × 104 to 2 × 105 cells ml–1; 4–5 × 10–9 to 1–2 × 10–8 g C ml–1) and very low bacterial productivity (4 × 10–11 to 1 × 10–10 g C ml–1 h–1) in winter, probably resulting from lowered rates of primary productivity and dissolved organic matter release as well as reduced riverine allochthonous inputs during the winter drought.  相似文献

针对我国东北某石油烃污染场地地下水中的萘污染,筛选出了AcinetobacterPseudomonas菌属的高效萘降解菌群。该菌群对萘的耐受性较高,且具有良好的乳化性,能够自动调节细胞表面疏水性和自聚集性。不同pH值、萘初始浓度、温度和菌接种量对萘降解效率的影响研究表明:最适菌群生长的pH范围为7.08.0;萘降解效率和速率在1.005.00 mg/L范围内与其初始浓度成正比;在1030 ℃温度范围内均表现出较高的萘降解效率。在此基础上,利用Guass、GuassAmp、LogNomal、Poisson和Pulse数学模型对萘的降解过程进行拟合,其降解速率更符合GuassAmp模型。通过将降解动力学模型中的常数与影响因素相关联,推导出了拟合度较高的多因素影响下的萘降解动力学方程。  相似文献

A systematic study of the lipid composition of thirteen bacterial species and three Recent sediments (methanogenic sediment, cyanobacterial mat and evaporative gypsum crust) was undertaken in an attempt to recognize bacterial organic matter in sediments. A sequential method, which distinguishes between three different modes of occurrence of lipid moieties (free, OH- and H+-labile), was applied. The acid-labile fractions are discussed.The three main groups of bacteria, archaebacteria, gram-positive eubacteria and gram-negative eubacteria, are easily distinguished. Methanogenic and extremely halophilic archaebacteria are characterized by the presence of diphytanyl glyceryl ether and the absence of fatty acids. The gram-positive eubacteria contain primarily iso- and anteiso-branched fatty acids whereas the gram-negative bacteria and sediments are dominated by β- and α-hydroxy fatty acids. A wide variety of H+-labile hydroxy fatty acids was observed which included several, as yet unknown, structures.β-Hydroxy fatty acids in this H+-labile mode of occurrence are exclusively present in bacteria. Their distribution patterns in sediments are considered “fingerprints” of past and present bacterial populations. The specific differences in β -hydroxy fatty acid compositions observed in the different bacteria and the three sediments investigated, suggest that amide-linked β-hydroxy fatty acid patterns are useful as markers of bacterial populations and therefore of environmental conditions.  相似文献

采用生物氧化技术和化学方法,利用堆浸工艺分步提取,对含Au 140~150 g/t、Ag 900~1 000 g/t、B i 9.0%~10.0%、Cu14%~17%的多金属浮选金精矿的综合回收工艺进行了研究。经小型搅拌试验,确定了生物氧化浸铜—氰化浸金、NaC l-FeC l3-HC l体系(氯盐法)浸出铋和银的技术方法。通过柱浸试验,研究了利用该方法分步浸取Au、Ag、Cu和B i在堆浸工艺中的可行性。在10 t精矿的扩大试验验中,验证了柱浸试验所取得的工艺指标和参数。试验中掺入骨架材料,解决了精矿在堆浸中的渗透问题;生物氧化过程中,菌种在含高浓度铜离子浸出液中反复驯化,对铜的耐受力提高到30 g/L;在NaC l-FeC l3-HC l体系浸出B i和Ag时,用H2O2将浸出液中的Fe2+氧化成Fe3+,使浸出液能循环使用,且B i和Ag同时浸出。Au、Ag、Cu和B i的最终浸出率分别为92.98%、65.09%、45.33%和53.49%。  相似文献

Bacterial concentration (Escherichia coli) is used as the key indicator for marine beach water quality in Hong Kong. For beaches receiving streamflow from unsewered catchments, water quality is mainly affected by local nonpoint source pollution and is highly dependent on the bacterial load contributed from the catchment. As most of these catchments are ungauged, the bacterial load is generally unknown. In this study, streamflow and the associated bacterial load contributed from an unsewered catchment to a marine beach, Big Wave Bay, are simulated using a modelling approach. The physically based distributed hydrological model, MIKE‐SHE, and the empirical watershed water quality model (Hydrological Simulation Program – Fortran) are used to simulate streamflow and daily‐averaged E. coli concentration/load, respectively. The total daily derived loads predicted by the model during calibration (June–July 2007) and validation (July–October 2008) periods agree well with empirical validation data, with a percentage difference of 3 and 2%, respectively. The simulation results show a nonlinear relationship between E. coli load and rainfall/streamflow and reveal a source limiting nature of nonpoint source pollution. The derived load is further used as an independent variable in a multiple linear regression (MLR) model to predict daily beach water quality. When compared with the MLR models based solely on hydrometeorological input variables (e.g. rainfall and salinity), the new model based on bacterial load predicts much more realistic E. coli concentrations during rainstorms.
