为从一种组成未知的商用复合菌中分离并筛选出优势发酵菌株,本实验采用稀释平板培养法,对菌种进行分离纯化,同时参照《真菌鉴定手册》和《常见细菌鉴定手册》,根据其形态学特征、生理生化特性,并分别结合16S r DNA和18S r DNA序列的比对分析对菌种进行鉴定。另外,本研究通过生长曲线测定,单菌种发酵试验以及多菌种混合发酵发酵试验,以游离氨基酸态氮为指标,以期筛选出发酵低值水产品制备菌肥的最佳菌株组合。结果显示,共分离得到4株菌,分别为a热带假丝酵母、b罗伦隐球菌、c枯草芽孢杆菌、d蜡样芽胞杆菌。且菌种组合acd发酵后游离氨基酸态氮的含量较对照组上升最显著,为8.276g/L。故可确定最佳发酵菌株配方为acd(热带假丝酵母+枯草芽孢杆菌+蜡样芽胞杆菌)。  相似文献

Gut content examination and trophic markers (fatty acids, stable isotopes of C and N) were combined to delineate the diet of the dominant species of amphipods from Mediterranean Posidonia oceanica seagrass meadows and to highlight trophic diversity among this community. Our results indicate that, although all dominant species heavily relied on macroalgal epiphytes, considerable interspecific dietary differences existed. Carbon stable isotope ratios notably showed that some of the amphipod species favored grazing on epiphytes from leaves or litter fragments (Apherusa chiereghinii, Aora spinicornis, Gammarus aequicauda), while others such as Dexamine spiniventris preferred epiphytes from rhizomes. The remaining amphipods (Caprella acanthifera, Ampithoe helleri and Gammarella fucicola) readily consumed both groups. In addition, SIAR modeling suggested that most species had a mixed diet, and relied on several food items. Fatty acid analysis and gut contents revealed that contributions of microepiphytic diatoms and of benthic and suspended particulate organic matter to the diet of amphipods were anecdotal. None of the examined species seemed to graze on their seagrass host [low 18:2(n‐6) and 18:3(n‐3) fatty acids contents], but Gammarus aequicauda partly relied on seagrass leaf detritus, as demonstrated by the lesser 13C‐depletion of their tissues. Overall, our findings suggest that amphipods, because of their importance in the transfer of organic matter from primary producers and detritus to higher rank consumers, are key items in P. oceanica‐associated food webs.  相似文献

以中国近海典型赤潮藻中肋骨条藻(Skeletonema costatum)为研究对象,考察不同氧饱和度环境下底栖甲壳动物对有机物在沉积物中降解的影响。通过对比不同氧饱和度培养模拟体系中中肋骨条藻在有/无刀额新对虾(Metapenaeus ensis)幼体活动背景下的降解过程,追踪中肋骨条藻中4种主要脂肪酸(C14:0,C16:0,C16:1(7)和C20:5)在100%,50%,25%和0%的氧饱和度体系中的降解行为。结果表明,100%氧饱和度体系中刀额新对虾幼体的活动能明显加快脂肪酸在沉积物中的降解(约为对照组的1.2~1.7倍),特别是对饱和脂肪酸C14:0和C16:0的作用最为明显;而在50%氧饱和度的缺氧体系中,在前三天的培养中,刀额新对虾幼体的活动逐渐减弱直至最终死亡,死亡之前的扰动增加了有机物在次表层沉积物中的埋藏并降低了其降解速率。研究还发现,在100%氧饱和度体系中,刀额新对虾幼体的扰动作用促进大量脂肪酸转移到次表层沉积物和悬浮颗粒中;而在50%氧饱和度体系中,这种转移作用明显变小。对于氧饱和度25%和0%的环境,刀额新对虾幼体在模拟培养开始后迅速死亡,对有机物质降解几乎没有影响。  相似文献

This study examined the distributions and stable carbon isotopic compositions of saturated fatty acids (SaFAs) in one 300 cm long sedimentary profile, which was named as Site4B in Shenhu, northern South China Sea. The concentrations of total SaFAs in sediments ranged from 1.80 to 10.16 μg/g (μg FA/g dry sediment) and showed an even-over-odd predominance in the carbon chain of C12 to C32, mostly with n-C16 and n-C18 being the two major components. The short-chain fatty acids (ScFAs; n-C12 to n-C18) mainly from marine microorganisms had average δ13C values of −26.7‰ to −28.2‰, whereas some terrigenous-sourced long-chain fatty acids (LcFAs; n-C21 to n-C32) had average δ13C values of −29.6‰ to −34.1‰. The other LcFAs (n-C24 & n-C26  n-C28; average δ13C values are −26.1‰ to −28.0‰) as well as n-C19 and n-C20 SaFAs (average δ13C values are −29.1‰ and −29.3‰, respectively) showed a mixed signal of carbon isotope compositions.The relative bioproductivity calculation (marine vs. terrigenous) demonstrated that most of organic carbon accumulation throughout the sedimentary profile was contributed by marine organism. The high marine productivity in Shenhu, South China Sea may be related to the hydrocarbon seepage which evidenced by diapiric structures. Interestingly, there is a sever fluctuation of terrigenous inputs around the depth of 97 cm below the seafloor (bsf), probably resulting from the influence of the Dansgaard–Oeschger events and the Younger Dryas event as revealed by 14C age measurements.  相似文献

The ‘new glacial stratigraphy’ (NGS) of Britain postulates that deposits hitherto assigned to the Anglian glaciation in Marine Isotope Stage (MIS) 12 represent MIS 16, 12, 10 and 6. This controversial idea can be tested at Sidestrand in north Norfolk, where fossiliferous temperate‐stage deposits underlie the oldest and overlie the youngest of the glacial deposits. Previous work has considered biostratigraphy and amino acid dating of these temperate‐stage deposits, but did not achieve tight age constraint using the amino acid evidence alone. The total hydrolysable amino acid fraction in intra‐crystalline protein from Bithynia tentaculata opercula has previously been analysed for concentrations of serine and alanine. Statistical screening of these data gives alanine/serine ratios of 4.572 ± 0.114 for the Sidestrand Hall Member (beneath the glacial deposits) and 3.564 ± 0.091 for the Sidestrand Cliff Formation (overlying the glacial deposits). These ratios imply ages of MIS 13a and 11c, respectively, indicating the latest Cromerian and early Hoxnian interglacials and invalidating the NGS. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

Requirement for dietary n−3 HUFA (n−3 highly unsaturated fatty acid) for growth and survival of black sea bream (Sparus macrocephalus) larvae was studied using rotifers andArtemia at various levels of n−3 HUFA. Five treatments with rotifers andArtemia differing in n−3 HUFA were prepared by enriching them with various oil emulsions. Results indicated that dietary n−3 HUFA significantly influence fish n−3 HUFA levels and are essential for growth and survival of black sea bream larvae. The results also indicated that the incorporation of n−3HUFA TG (triacylglycerols) into tissues of larval black sea bream was more effective from natural fish oil in comparison with n−3 HUFA fatty acid ethyl esters from ethyl-esterified oil. 对大獭蛤(Lutraria maxima)软体部进行营养成分分析,并对其营养价值进行综合评定。结果表明,大獭蛤软体部粗蛋白含量为81.20%,干物质中水解氨基酸总量为83.50%,其中必需氨基酸为30.22%,占氨基酸总量的36.19%;氨基酸中6种呈味氨基酸含量丰富,占氨基酸总量的53.81%;大獭蛤软体部第一限制氨基酸为亮氨酸;矿物质含量丰富。可见,大獭蛤软体部营养价值高,呈味特性良好。  相似文献
