The aim of the present study was to propose the immunofluorescence technique in cultured fibroblasts from Mediterranean cetaceans as a new "in vitro" tool to explore the susceptibility of these marine mammals to different xenobiotic compounds. The cell lines were cultured from integument biopsies of free-ranging and stranded cetaceans (dead within 12h). Using the indirect immunofluorescence assay, we detected endogenous proteins induced by different contaminants. Here we present the method used for qualitative and quantitative evaluation of cytochromes P450 (CYP1A1 and CYP2B) induced by some POPs (DDTs and PCBs) and emerging contaminants (PBDEs) in fibroblast cell cultures of striped dolphin (Stenella coeruleoalba) and bottlenose dolphin (Tursiops truncatus). Immunofluorescence was quantified with a specially designed Olympus macro, DetectIntZ. A major result was the possibility of using this "in vitro" assay to quantify induction of endogenous proteins.  相似文献

The identification of bacterial community structure has led, since the beginning of the 1990s, to the idea that bacterioplankton populations are stratified in the water column and that diverse lineages with mostly unknown phenotypes dominate marine microbial communities. The diversity of depth-related assemblages is also reflected in their patterns of activities, as bacteria affiliated to different groups can express different activities in a given ecosystem. We analysed bacterial assemblages (DGGE fingerprinting) and their activities (prokaryotic carbon production, protease, phosphatase, chitinase, beta-glucosidase and lipase activities) in two areas in the Ross Sea, differing mainly in their productivity regime: two stations are located in the Terra Nova Bay polynya area (highly productive during summer) and two close to Cape Adare (low phytoplankton biomass and activity). At every station a pronounced stratification of bacterial assemblages was identified, highlighting epipelagic communities differing substantially from the mesopelagic and the bathypelagic communities. Multivariate analysis suggested that pressure and indirectly light-affected variables (i.e. oxygen and fluorescence) had a great effect on the bacterial communities outcompeting the possible influences of temperature and dissolved organic carbon concentration. Generally activities decreased with depth even though a signal of the Circumpolar Deep Water (CDW) at one of the northern stations corresponded to an increase in some of the degradative activities, generating some ‘hot spots’ in the profile. We also found that similar assemblages express similar metabolic requirements reflected in analogous patterns of activity (similar degradative potential and leucine uptake rate). Furthermore, the presence of eukaryotic chloroplasts’ 16S rDNA in deep samples highlighted how in some cases the dense surface-water formation (in this case High Salinity Shelf Water—HSSW) and downwelling can affect, at least for some phylotypes, the bacterial (16S rDNA based) community structure of the dark ocean.  相似文献

Uncertainty assessment in quantitative rockfall risk assessment   总被引:4,自引:1,他引:3  
This study shows a quantitative rockfall risk assessment (QRA) for a slope of the Feifeng Mountain (China), including an explicit assessment of the uncertainties. For rockfall risk analysis, the annual probability of occurrence, reach probability, temporal–spatial probability and vulnerability of tourists were calculated for both dry and rainy day conditions. The resulting individual risk for exposed people visiting the historical site can be considered as acceptable for all scenarios, whereas the overall societal risk lies within the as low as reasonably practicable (ALARP) zone and therefore requires some mitigation actions. For the explicit assessment of uncertainty, an error propagation technique (first-order second moment (FOSM)) was adopted, starting from expert knowledge heuristic estimations of the coefficient of variation for each component of the risk analysis procedure. As a result, coefficients of variation of the calculated risk were obtained, ranging from 48 to 132 %, thus demonstrating the importance of accounting for uncertainty in rockfall risk modelling. A multi-criteria methodology is also proposed for the assessment of the standard deviation of the parameters adopted for the stochastic rockfall run-out model.  相似文献

The distribution of the dissolved labile and of the particulate Fe and Cu together with dissolved oxygen, nutrients, chlorophyll a and total particulate matter was investigated in the surface waters of Terra Nova Bay polynya in mid-January 2003. The measurements were conducted within the framework of the Italian Climatic Long-term Interactions of the Mass balance in Antarctica (CLIMA) Project activities. The labile dissolved fraction was operationally defined by employing the chelating resin Chelex-100, which retains free and loosely bound trace metal species. The dissolved labile Fe ranges from below the detection limit (0.15 nM) to 3.71 nM, while the dissolved labile Cu from below the detection limit (0.10 nM) to 0.90 nM. The lowest concentrations for both metals were observed at 20 m depth (the shallowest depth for which metals were measured). The concentration of the particulate Fe was about 5 times higher than the dissolved Fe concentration, ranging from 0.56 to 24.83 nM with an average of 6.45 nM. The concentration of the particulate Cu ranged from 0.01 to 0.71 nM with an average of 0.17 nM. The values are in agreement with the previous data collected in the same area. We evaluated the role of the Fe and Cu as biolimiting metals. The N:dissolved labile Fe ratios (18,900–130,666) would or would not allow a complete nitrate removal, on the basis of the N:Fe requirement ratios that we calculated considering the N:P and the C:P ratios estimated for diatoms. This finding partially agrees with the Si:N ratio that we found (2.29). Moreover we considered a possible influence of the dissolved labile Cu on the Fe uptake process.  相似文献

Abstract— The petrographic relationships in diogenites between orthopyroxene and minor phases such as chromite, troilite, diopside, plagioclase, and silica are often obscured by the intense brecciation that characterizes these meteorites. Although brecciated, Bilanga preserves numerous clasts displaying primary textural relations between orthopyroxene and these minor phases that are large enough to analyze by electron microprobe. In this study, we focus on the distribution, composition, and origin of the minor phases in Bilanga to provide new insights into the crystallization and metamorphic history of these rocks. The samples examined consist mainly of orthopyroxene grains plus five types of assemblages containing diopside + a Fe‐rich phase (chromite, troilite, and/or Fe‐Ni metal) ± plagioclase ± silica. We interpret type 1 assemblages as being remnants of intercumulus melt trapped in the interstices between orthopyroxene grains after crystal settling in a magma chamber. Type 2 assemblages appear to have formed by heterogeneous exsolution during thermal metamorphism. Type 3 assemblages are believed to be remnants of other assemblages that have been shocked, melted, and rapidly recrystallized by impact events. Type 4 assemblages consist of veins that also appear to have formed from trapped intercumulus melt. Regions of silica‐rich mesostasis (type 5) appear to be larger patches of more evolved intercumulus melt that have been significantly affected by late‐stage impact melting. Finally, large clasts containing plagioclase ± diopside are interpreted to be exotic fragments of a different but possibly related rock type incorporated in the Bilanga breccia.  相似文献

Abstract— We present numerical calculations of the peak temperatures experienced by micrometeorites during atmospheric entry. Results are given for particle diameters between 2 and 50 μm and for entry velocities between 10 and 25 km/s. A material density of 2 g/cm3 and an entry angle of 45° are used for the calculation, but the results presented here can be easily reinterpreted for other densities and for vertical entry.  相似文献

We conducted experiments in space to investigate the aggregation of millimeter‐ and submillimeter‐sized particles in microgravity, an important early step in planet formation. Particulate materials included salt (NaCl), sugar (sucrose), coffee, mica, ice, Bjurböle chondrules, ordinary and carbonaceous chondrite meteorite fragments, and acrylic and glass beads, all triply confined in clear plastic containers. Angular submillimeter particles rapidly and spontaneously formed clusters strong enough to survive turbulence in a protoplanetary nebula. Smaller particles generally aggregated more strongly and quickly than larger ones. We observed only a weak dependence of aggregation time on particle number density. We observed no strong dependence on composition. Round, smooth particles aggregated weakly or not at all. In a mixture of particle types, some phases aggregated more readily than others, creating selection effects that controlled the composition of the growing clumps. The physical process of aggregation appears to be electrostatic in nature.  相似文献

We present a semi-analytic treatment of galactic winds within high-resolution, large-scale cosmological N -body simulations of a Λ cold dark matter (ΛCDM) universe. The evolution of winds is investigated by following the expansion of supernova-driven superbubbles around the several hundred thousand galaxies that form in an approximately spherical region of space with diameter 52  h −1 Mpc and mean density close to the mean density of the universe. We focus our attention on the impact of winds on the diffuse intergalactic medium. Initial conditions for mass loss at the base of winds are taken from Shu, Mo & Mao. Results are presented for the volume filling factor and the mass fraction of the intergalactic medium (IGM) affected by winds, and their dependence on the model parameters is carefully investigated. The mass-loading efficiency of bubbles is a key factor to determine the evolution of winds and their global impact on the IGM: the higher the mass loading, the later the IGM is enriched with metals. Galaxies with 109 < M < 1010 M are responsible for most of the metals ejected into the IGM at   z = 3  , while galaxies with   M < 109 M   give a non-negligible contribution only at higher redshifts, when larger galaxies have not yet assembled. We find a higher mean IGM metallicity than Lyα forest observations suggest, and we argue that the discrepancy may be explained by the high temperatures of a large fraction of the metals in winds, which may not leave detectable imprints in absorption in the Lyα forest.  相似文献

Illness and mortality related to heat and worsening air quality are core public health concerns associated with climate change projections. We examined the historical relationship between age- and cause-specific mortality rates from 1980 through 2006 and heat events at the 99th percentile of humidex values in the historic period from January 1, 1970 to December 31, 2006 in the greater Seattle area (King, Pierce and Snohomish counties), Spokane County, the Tri-Cities (Benton and Franklin counties) and Yakima County; the relative risks of mortality during heat events were applied to population and climate projections for Washington State to calculate number of deaths above the baseline (1980–2006) expected during projected heat events in 2025, 2045 and 2085. Three different warming scenarios were used in the analysis. Relative risks for the greater Seattle area showed a significant dose-response relationship between heat event duration and daily mortality rates for non-traumatic deaths for persons ages 45 and above, typically peaking at four days of exposure to humidex values above the 99th percentile. The largest number of projected excess deaths in all years and scenarios for the Seattle region was found for age 65 and above. Under the middle warming scenario, this age group is expected to have 96, 148 and 266 excess deaths from all non-traumatic causes in 2025, 2045 and 2085, respectively. We also examined projected excess deaths due to ground-level ozone concentrations at mid century (2045–2054) in King and Spokane counties. Current (1997–2006) ozone measurements and mid-twenty-first century ozone projections were coupled with dose-response data from the scientific literature to produce estimates overall and cardiopulmonary mortality. Daily maximum 8-h ozone concentrations are forecasted to be 16–28% higher in the mid twenty-first century compared to the recent decade of 1997–2006. By mid-century in King County the non-traumatic mortality rate related to ozone was projected to increase from baseline (0.026 per 100,000; 95% confidence interval 0.013–0.038) to 0.033 (95% CI 0.017–0.049). For the same health outcome in Spokane County, the baseline period rate of 0.058 (95% CI 0.030–0.085) was estimated increase to 0.068 (95% CI 0.035–0.100) by mid-century. The cardiopulmonary death rate per 100,000 due to ozone was estimated to increase from 0.011 (95% CI 0.005–0.017) to 0.015 (0.007–0.022) in King County, and from 0.027 (95% CI 0.013–0.042) to 0.032 (95% CI 0.015–0.049) in Spokane County. Public health interventions aimed at protecting Washington’s population from excessive heat and increased ozone concentrations will become increasingly important for preventing deaths, especially among older adults. Furthermore, heat and air quality related illnesses that do not result in death, but are serious nevertheless, may be reduced by the same measures.  相似文献
