Biotransformation of polycyclic aromatic hydrocarbons in marine polychaetes   总被引:2,自引:0,他引:2  
Deposit-feeding polychaetes constitute the dominant macrofauna in marine environments that tend to be depositional centers for organic matter and contaminants. Polychaetes are known to accumulate polycyclic aromatic hydrocarbons (PAHs) from both particulate and dissolved phases but less is known about the mechanisms underlying elimination of accumulated PAHs. An important pathway of elimination is through biotransformation which results in increased aqueous solubility of the otherwise hydrophobic PAHs. Biotransformation in marine polychaetes proceeds in a two phased process similar to those well studied in vertebrates, phase I enzymes belonging to the Cytochrome P450 (CYP) enzyme family, along with a few phase II enzymes have been identified in marine polychaetes. In this review we aim at highlighting advances in the mechanistic understanding of PAH biotransformation in marine polychaetes by including data obtained using analytical chemistry and molecular techniques. In marine polychaetes induction of CYP enzyme activity after exposure to PAHs and the mechanism behind this is currently not well established. Conflicting results regarding the inducibility of CYP enzymes from polychaetes have led to the suggestion that induction in polychaetes is mediated through a different mechanistic pathway, which is corroborated by the apparent lack of an AhR homologous in marine polychaetes. Also, none of the currently identified CYP genes from marine polychaetes are isoforms of those regulated by the AhR in vertebrates. Relatively few studies of phase II enzymes in marine polychaetes are currently available and most of these studies have not measured the activity of specific phase II enzymes and identified phase II metabolites but used an extraction technique only allowing determination of the overall amount of phase II metabolites. Studies in insects and various marine invertebrates suggest that in invertebrates, enzymes in the important phase II enzyme family, UDP-glucuronosyl transferases primarily use glucoside as co-substrate as opposed to the vertebrate cosubstrate glucuronic acid. Recent studies in marine polychaetes have however identified glucuronidation of PAHs indicating no mechanistic difference in co-substrate preference among UDP-glucuronosyl transferases between vertebrates and marine polychaetes but it might suggest a mechanistic difference between marine polychaetes and insects.  相似文献

Previous studies have shown that aryl hydrocarbon receptor (AhR) agonists possess anti-estrogenic activities and several mechanisms have been proposed to explain the interactions between AhR and estrogen receptor (ER) signalling pathways. In the present study, we show that 3,3'4,4'5-pentachlorobiphenyl (PCB126 - a dioxin-like AhR agonist) produced estrogenic responses in the absence of ER agonist, in fish in vitro system. We exposed salmon primary hepatocytes to PCB126 (1, 10 and 50 pM) and the ER agonist nonylphenol (NP; 5 and 10 microM) singly and also in combination. Vitellogenin (Vtg) and zona radiata proteins (Zr-proteins) levels were analysed by semi-quantitative ELISA. We observed that the protein levels of Vtg and Zr-proteins were significantly induced in a concentration-specific manner in cells treated with PCB126 and NP, singly or in combination. In general, these results show a novel aspect of dioxin-like PCB effect not previously demonstrated in fish system.  相似文献

Barium/calcium profiles of bivalve shells are characterized by flat background signals periodically interrupted by sharp peaks, with the background signals correlated with water Ba/Ca. To test if the peaks are an environmental signal related to productivity, we analyzed high-resolution Ba/Ca profiles in bivalve shells that grew adjacent to one another. Two aragonitic Saxidomus giganteus show remarkable similarity over a decade of growth, clearly indicating an environmental forcing. Four calcitic Pecten maximus shells also record synchronous Ba/Ca peaks, again indicating an exogenous control. The Ba/Ca peaks, however, start ~40 days after the crash of a bloom, while sedimentation takes place immediately following the bloom. Barite formation in settling phytoplankton flocs, as has been previously proposed, is clearly not the cause of these peaks. Other possible causes, such as dissolved Ba in ambient water, spawning, shell organic matter content, and kinetic growth rate effects are also discussed, but none provide satisfactory explanations. Background shell Ba partition coefficients (Ba/Cacarbonate/Ba/Cawater) for both the calcitic shells (0.18) and aragonitic shells (0.16) are similar to that previously reported for the calcitic Mytilus edulis (~0.1). We suggest that Ba/Ca peaks in bivalve shells are caused by an as yet undetermined environmental forcing, while background Ba/Ca levels are a good indication of dissolved Ba/Ca in the water; both are independent of shell mineralogy.  相似文献

Tridacnidae shells, a valuable archive of past environments, are common in the Balobok Rockshelter archaeological site on Sanga‐Sanga Island in the south Philippines. This site was occupied during the mid‐Holocene (ca. 5000–8800 14C yr B.P.), a period of Neolithic cultural expansion in the Philippines. This paper focuses on the preservation of two shell specimens, Hippopus hippopus and Tridacna maxima, unearthed from two mid‐Holocene layers within the rockshelter. The shells' mineralogy and microstructure (prismatic and crossed‐lamellar) were studied using micro‐Fourier transform infrared spectroscopy and scanning electron microscopy to determine if the samples were suitable as paleoenvironmental records. Both shells are still aragonitic but aragonite crystals of both microstructure types are partly dissolved. This dissolution, characteristic of meteoric water alteration, precludes their utility in paleoenvironmental geochemical studies. Nevertheless, these shells are abundant in archaeological sites in the region and may be better preserved in other depositional contexts; more studies on Philippine Tridacnidae shell diagenesis are needed. © 2011 Wiley Periodicals, Inc.  相似文献
