Alkaloids are a class of natural products with a wide range of biological activities. Due to the special living environment, the alkaloids from marine sponges have exhibited different biological activities and promising medical application potential. Neolamellarin A is a marine alkaloid possessing bisaryl-pyrrole structural features. Here, the synthesis of 12 different 3,4-bisaryl-N-alkylated permethylated analogues of neolamellarin A and their outstanding neuroprotective activity in PC12 cells are presented and discussed.  相似文献

The present study aimed at assessing the antifouling activity of bacteria associated with marine sponges. A total of eight bacterial strains were isolated from the surface of sponge Sigmadocia sp., of them, SS02, SS05 and SS06 showed inhibitory activity against biofilm-forming bacteria. The extracts of these 3 strains considerably affected the extracellular polymeric substance producing ability and adhesion of biofilm-forming bacterial strains. In addition to disc diffusion assay, microalgal settlement assay was carried out with the extracts mixed with polyurethane wood polish and coated onto stainless steel coupons. The extract of strain SS05 showed strong microalgal settlement inhibitory activity. Strain SS05 was identified as Bacillus cereus based on its 16S rRNA gene. Metabolites of the bacterial strains associated with marine invertebrates promise to be developed into environment-friendly antifouling agents.  相似文献

The PI3 K/Akt/m TOR signaling pathway is one of the most frequently dysregulated pathways in cancer. Targeting the PI3 K-mediated pathway has been an important strategy for developing novel anticancer agents. In the past decades, more than 40 inhibitors of the PI3 K/Akt/m TOR pathway have been developed at different clinical stages. Temsirolimus, everolimus, idelalisib, and copanlisib have been approved for clinical use by the Food and Drug Administration of the United States(FDA). However, the toxicity and drug resistance limit their efficiency in the treatment. Novel compounds with greater potency and selectivity, as well as improved therapeutic indices with reduced toxicity, are clearly required. Over the past three decades, a lot of bioactive ingredients with anticancer effects by affecting the PI3 K-mediated pathways have been found from marine organisms. In the present mini-review, anticancer compounds from marine source that target the PI3 K/Akt/m TOR signaling were reviewed. The molecular entities and their modes of action were presented. The marine compounds targeting special factors of the PI3 K/Akt/m TOR were highlighted.  相似文献

【目的】通过化学诱导方法从一株海洋真菌Hypocrea lixii中获得多样化的活性小分子化合物。【方法】对菌株在盐酸普鲁卡因的化学诱导作用下进行固体发酵,代谢产物采用硅胶减压柱层析、Sephadex LH-20以及pHPLC等手段分离纯化,对化合物运用NMR、MS等波谱学技术并比对文献进行结构鉴定,采用Ellman比色法及DPPH自由基清除法对化合物进行初步抗乙酰胆碱酯酶(AChE)和抗氧化活性测试。【结果】从海洋真菌Hypocrea lixii在盐酸普鲁卡因诱导下的发酵提取物中分离得到4个异黄酮类化合物,包括6'-O-crotonylgenistin(1)、Genistein(2)、Daidzein(3)和Genistin(4),以及对氨基苯甲酸甲酯(5)、对羟基苯丙酸(6)和胞嘧啶(7),其中化合物1和6为该种中首次报道,化合物5为生物转化来源首次报道,化合物2~4可能源自培养基。在100μmol/L剂量浓度下,化合物1~4对AChE的抑制以及DPPH自由基的清除活性均较弱,化合物5和6显示了相对强的AChE抑制活性,抑制率分别为40%和38%。【结论】盐酸普鲁卡因对该菌次级代谢产生了显著的影响,为深入研究该菌株化学诱导次级代谢产物提供基础。  相似文献

Marine sponge Tedania anhelans distributes throughout the intertidal zone of Fujian, southeastern China, and is a potential source of natural bioactive products. The sponge harbors a large number of bacterial groups that have been identified using various techniques, including fluorescent in situ hybridization (FISH). Fractionation of dissociated sponge allowed isolation of 25 bacterial species. Based on 16S rRNA gene sequencing, phylogenetic analysis attributed most of these eubacteria to α-Proteobacteria, γ-Proteobacteria, Cytophaga/Flavobacterium/Bacteroidetes (CFB group), and the family Bacillaceae of Gram-positive bacteria. In sequence similarity, five putatively novel species were identified with less than 98% similarity to other strains in the NCBI database. Tests for antimicrobial activities were performed against Gram-positive bacteria, Gram-negative bacteria, fungi, antitumor indicators Escherichia coli 343/591 (with DNA repair deficiency), regular E. coli 343/636 (with different DNA repair capacity), and 10 bacterial isolates exhibited inhibitory bioactivities. Among these strains, three isolates were detected involving function gene NRPS-A domains, which were most closely related to the amino acid sequences of linear gramicidin synthetase and pyoverdine synthetase. These results contribute to our knowledge of the microbes associated with marine sponges and further reveal novel bacterial resources for the screening of bioactive marine natural products.  相似文献

Three species of sponges and a tunicate were collected from Obhur creek of Jeddah coast for this bioactivity study. In order to assess the antifouling efficacy of selected marine organisms, methanolic extracts of these organisms were tested against different fouling bacterial forms and II-instar stage of the barnacle, Balanus amphitrite. Antibiosis, bioactivity and followed by multivariate analyses were carried out to check the efficacy of antifouling effect of the selected marine organisms. Principal component analysis revealed the exemplary antifouling efficacy of the sponge extracts of Stylissa sp. observed followed by Hyrtios sp. against bacterial forms in the laboratory study. De-trended correspondence analysis confirmed that the contribution of antifouling efficacy of the selected sponge extracts was observed to be more towards Bacillus sp., Vibrio sp. and Alteromonas sp. Moreover, the efficacy of Hyrtios sp. extract(20.430 μg m L~(-1)) followed by Stylissa sp.(30.945 μg m L~(-1)) showed higher against barnacle instar compared with other extracts in the bioactivity assay. Bray-Curtis cluster analysis under paired linkage categorized all the sponge extracts into one major cluster with 75% similarity, and one outlier tunicate. More than 80% similarity observed between Hyrtios sp. and Stylissa sp. Fourier transform infrared spectroscopy(FTIR) showed that the contribution of major peaks found in the marine organisms were towards sulfones, sulfoxides, cyanates and ketones.  相似文献

With the global ban on the application of organotin-based marine coatings by the International Maritime Organization, the development of environmentally friendly, low-toxic and nontoxic antifouling compounds for marine industries has become an urgent need. Marine microorganisms have been considered as a potential source of natural antifoulants. In this study, the antifouling potential of marine dinoflagellate Amphidinium carterae, the toxic and red-tide microalgae, was investigated. We performed a series of operations to extract the bioactive substances from Amphidinium carterae and tested their antialgal and antilarval activities. The crude extract of Amphidinium carterae showed significant antialgal activity and the EC50 value against Skeletonema costatum was 55.4 μg mL?1. After purification, the isolated bioactive substances (the organic extract C) exhibited much higher antialgal and antilarval activities with EC50 of 12.9 μg mL?1 against Skeletonema costatum and LC50 of 15.1 μg mL?1 against Amphibalanus amphitrite larvae. Subsequently, IR, Q-TOFMS, and GC-MS were utilized for the structural elucidation of the bioactive compounds, and a series of unsaturated and saturated 16- to 22-carbon fatty acids were detected. The data suggested the bioactive compounds isolated from Amphidinium carterae exhibited a significant inhibiting effect against the diatom Skeletonema costatum and Amphibalanus amphitrite larvae, and could be substitutes for persistent, toxic antifouling compounds.  相似文献

The sources of marine oil spills are mainly from accidents of marine oil tankers or freighters, marine oil-drilling platforms, marine oil pipelines, marine oilfields, terrestrial pollution, oil-bearing atmosphere, and offshore oil production equipment. It is concluded upon analysis that there are two main reasons for marine oil spills: (I) The motive for huge economic benefits of oil industry owners and oil shipping agents far surpasses their sense of ecological risks. (II) Marine ecological safety has not become the main concern of national security. Oil spills are disasters because humans spare no efforts to get economic benefits from oil. The present paper draws another conclusion that marine ecological damage caused by oil spills can be roughly divided into two categories: damage to marine resource value (direct value) and damage to marine ecosystem service value (indirect value). Marine oil spills cause damage to marine biological, fishery, seawater, tourism and mineral resources to various extents, which contributes to the lower quality and value of marine resources.  相似文献

Phenolic compounds have become one kind of the important pollutants of the marine environment. Single-walled Carbon nanotubes, as one-dimensional nano materials, have light weight and perfect hexagonal structure of connections, with many unusual mechanical, chemical and electrical properties. In recent years, with the research of carbon nanotubes and other nano materials, the application prospect is also constantly discussed. In this paper, homemade single-walled carbon nanotubes(SWCNTs) coating was used for establishing an analytical approach to the determination of five kinds of phenolic compounds in seawater using SPME-GC-MS. Optimal conditions: After saturation was conducted with Na Cl, and p H was adjusted to 2.0 with H_2SO_4, the extract was immersed in a water bath at 40 for GC℃-MS determination through 40-min agitating extraction at 500 rmin~(-1) and 3-min desorption at 280℃. The liniearities ranged between 0.01-100 μg L~(-1), and the determination limits ranged between 1.5-10 ngL~(-1). The relative standard deviation(RSD, n = 5) was less than 6.5%. For the phenolic compounds obtained from the spiked recovery test for actual seawater samples, the rates of recovery were 87.5%-101.7%, and the RSDs were less than 8.8%, which met the requirements of determination. Due to its simplicity, high efficiency and low consumption, this approach is suitable for the analysis of trace amounts of phenolic compounds in marine waters.  相似文献

The changes in heme (associated with hemoglobin), hemoglobin and hematin in the coelomic fluid of marine worm,Urechis unicinctus, exposed to different concentrations of sulfide, were investigated using biochemical techniques. When exposed to different sulfide concentrations for up to 96 h, the relative amounts of the three components changed in a regular pattern suggesting that the coelomocytes play an important role in the worm's tolerance to sulfide. The possible roles of heme compounds in sulfide tolerance of this species are discussed on the basis of our experimental data. Soft corals are an important group of organisms that live in reef ecosystems. In this study, the chemical defense of soft coral Sinularia polydactyla against biofilm-forming bacteria was assessed. The soft coral S. polydactyla was collected from the Obhur creek of the Red Sea and the bioactive compounds were extracted under different conditions using methanol and hexane as solvents. Results revealed that the bioactive compounds produced by the soft coral S. polydactyla were active against the growth, hydrophobicity and extracellular polymeric substances production of the biofilm-forming bacteria. However, the damage or injury in soft coral tissue caused a decrease in the activity against biofilm-forming bacteria. GC-MS analysis revealed that sesquiterpenes were abundant in normal coral sample extract while cembranoids were abundant in damaged coral sample extracts. In general, the results indicated that the soft corals either under stress or with damage may not have a strong chemical defense against the colonizing marine organisms and competitors.  相似文献

Environmental efficiency standards are often used to evaluate the costs of oceanic economic development. A variety of statistical analyses were applied in this study to quantify the marine environmental efficiency of 11 Chinese coastal provinces and municipalities between 2000 and 2014. Results initially reveal that environmental efficiency measures that incorporate undesirable outputs are more consistent with real production conditions and thus the use of marine economic and environmental efficiencies supplement and complement one another. Second, overall marine environmental efficiency across China tends to be low and can be spatially characterized by a transformation such that the inefficiencies noted in 2000 have subsequently been transformed to comprise a three-tiered structure that encompasses northern, central, and southern cores. Third, variation in absolute and relative marine environmental efficiency differences for the coastal regions of China have been consistent over time; values initially decreased before increasing again in a fluctuating manner over the time period of this analysis. Fourth, data show that the Pearl River Delta area has experienced the highest rate of change in marine environmental efficiency over time when economic zones are used as basic research units, although values have nevertheless fluctuated significantly. Fifth, values for total factor productivity as well as technical efficiency and change across the Chinese marine economy all fluctuated over time but increased. Data show that changes in marine environmental efficiency across China can primarily be attributed to progress in marine science and technology. Finally, levels of capital investment and marine industrial pollution intensity are not significantly correlated with marine environmental efficiency. Indeed, both marine industrial structural levels and environmental protection technologies have had a positive effect on environmental efficiency while levels of investment in marine scientific research as well as the scale of economic development, the marine economy, and the degree of external openness have all exerted negative effects on this key variable.  相似文献

In recent years, red tides occurred frequently in coastal areas worldwide. Various methods based on the use of clay, copper sulfate, and bacteria have been successful in controlling red tides to some extent. As a new defensive agent, marine microorganisms are important sources of compounds with potent inhibitory bioactivities against red-tide microalgae, such as Gymnodinium sp. (Pyrrophyta). In this study, we isolated a marine bacterium, HSB07, from seawater collected from Hongsha Bay, Sanya, South China Sea. Based on its 16S rRNA gene sequence and biochemical characteristics, the isolated strain HSB07 was identified as a member of the genus Halomonas. A crude ethyl acetate extract of strain HSB07 showed moderate inhibition activity against Gymnodinium sp. in a bioactive prescreening experiment. The extract was further separated into fractions A, B, and C by silica gel column chromatography. Fractions B and C showed strong inhibition activities against Gymnodinium. This is the first report of inhibitory activity of secondary metabolites of a Halomonas bacterium against a red-tide-causing microalga.  相似文献
