A laboratory experiment was conducted to assess the bioaccumulation of Pb2+ and its effects on growth, morphology and pigment contents of Spirulina (Arthrospira) platensis. The specimen cultured in Zarrouk liquid medium was treated with various initial metal concentrations (0, 5, 10, 30, 50 and 100 μg mL−1). The growth of S. platensis was adversely affected by Pb2+ at high concentrations (30, 50 and 100 μg mL−1). However, at low concentrations (5 μg mL−1), Pb2+ could stimulate its growth slightly. The pigment contents (chlorophyll α and β carotene) were decreased in a dose-dependent manner. The highest reductions (67% and 53% respectively in chlorophyll α and β carotene) were observed in 100 μg mL−1 treatment group. The LC50 (96 h) of Pb2+ was measured as 75.34 μg mL−1. Apart from a few cases of filament breakages at elevated concentrations (50 and 100 μg mL−1), morphological abnormalities are not specific. Metal bioaccumulation increased with Pb2+ concentrations, but decreased with exposure time. The maximum accumulated amount was 188 mg g−1 dry weight. The bioconcentration factor (BCF) reached to a peak at day 2, followed by a gradual reduction for all the exposure concentrations. S. platensis is able to tolerate considerably high Pb2+ concentrations. Consequently it can be used as a potential species to remove heavy metal from contaminated waters.  相似文献

Preliminary studies on microzooplankton grazing were conducted with dilution method in Jiaozhou Bay from summer 1998 to spring 1999. Four experiments were carried out at St. 5 located at the center of Jiaozhou Bay. Chlorophyll a concentrations were consistently dominated by netphytoplankton (net-, >20μm), except during the autumn 1998 cruise, when they were dominated by nanophytoplankton(nano-, 2–20μm). The contribution of picophytoplankton (pico-, <2μm) to total chlorophyll a concentrations (<200μm) varied considerably between cruises. Instantaneous growth coefficients(u) of phytoplankton varied from 0.098 to 1.947d−1, with mean value of 0.902d−1. Instantaneous coefficients(g) of microzooplankton grazing on phytoplankton ranged from 0.066 to 0.567d−1, mean value of 0.265d−1, which was equivalent to daily lose of 21.9% of the initial standing stock and 58.1% of the daily potential production. The effect of Mg2+, NaCl and citric acid on the accumulation of β-carotene inDunaliella Salina was studied. The experimental results showed that 10.5 mmol/L Mg2+, 5 mol/L NaCl, 3 μmol/L citric acid, and CO2 are favorable forDunaliella Salina cell growth and β-carotene accumulation. After 144 h culture under the above conditions, theDunaliella Salina biomass increased by 7.18 times; β-carotene reached 9.61%. The standing stock and primary production of benthic microalgae on tidal flats were measured seasonally at 3 transects (Puqing, Dahengchuang and Puqi) in Yueqing Bay during 2002 2003. The results showed that the integral chlorophyll a (Chl a) concentration in tidal flat mud exhibited a seasonal variation with the order of magnitude: winter (14.0 4.2 mg m-2) > spring (13.0 6.3 mg m-2) > autumn (7.7 5.9 mg m-2) > summer (4.6 3.2 mg m-2). The primary production showed an order of magnitude: spring (270.5 224.9 mgC m-2 d-1)>winter (238.7 225.5 mgC m-2 d-1)>autumn (214.1 56.2 mgC m-2 d-1)>summer (71.6 44.6 mgC m-2 d-1). Both chlorophyll a and primary production showed maximum values in the surface layer of sediment, and decreased rapidly with increasing depth due to sun light limitation. The results of variance analysis indicated that seasonal variation and tidal flat condition affected Chl a greatly, but had no significant effect on primary production. The annual primary production of benthic microalgae on tidal flats in Yueqing Bay was estimated at 16143 tons carbon, which is sufficient to support 1.02×105 tons shellfish production. The environmental factors affecting chlorophyll and primary production on the tidal flats in Yueqing Bay were discussed. By comparing with other bays on China’s coast, it was observed that Yueqing Bay is a region with high benthic microalgae standing crop and primary production, which may be related to the type of its sediment.  相似文献

Activities of the respiratory electron transport system (ETS), C uptake, chlorophyll and temperature were determined at a station in the center of Donghu Lake. Seasonal, fluctuations in ETS-activities ranged from 100 to 460μg O2 h-1.L-1. Production: respiration ratio (P/R) calculated from C uptake and ETS-activity, ranged from 0.2 to 6.8. The test on cultured algae ETS-activity revealed marked differences in different growth phases. ETS activity is suggested as an indicator of lake nutrient status.  相似文献

Gametophytes of Laminaria japonica were cryopreserved in liquid nitrogen using encapsulation-dehydration with two-step cooling method. Gametophytes cultured at 10℃ and under continuous irradiance of 30 μmol m-2 s-1 for 3 weeks were encapsulated in calcium alginate beads. The beads were dehydrated in 0.4 molL-1 sucrose prepared with seawater for 6 h, desiccated in an incubator set at 10℃ and 70% relative humidity for 4 h, pre-frozen at either -40℃ or -60℃ for 30 min, and stored in liquid nitrogen for >24 h. As high as 43% of survival rate was observed when gametophytes were thawed by placing the beads in 40℃seawater and re-hydrated in 0.05 molL-1 citrate sodium prepared using 30‰ NaC1 7 d later. More cells of male gametophytes survived the whole procedure in comparison with female gametophytes. The cells of gametophytes surviving the preservation were able to grow asexually and produce morphologically normal sporophytes.  相似文献

The effects of several nutritional factors on the growth and eicosapentaenoic acid (EPA) production of diatom Nitzschia laevis were studied. 4 LDM (quadrupled concentration of the nutrient salt) was the optimal concentration of nutrient salt for the growth and EPA production of N. laevis. The growth of N. laevis was inhibited when the glucose concentration was either lower than 10 gL−1 or higher than 15 gL−1. Both sodium nitrate and urea were good nitrogen sources for the growth and EPA production, while ammonium chloride seriously decreased the dry cell weight (DW) and the EPA content. Silicate seriously influenced the growth of N. laevis. The maximum DW of 2.34 gL−1 was obtained in the presence of 150 mgL−1 Na2SiO3·9H2O. The EPA content remained almost the same when the silicate concentration was lower than 150 mgL−1; however, higher silicate concentrations resulted in a steady decrease of EPA content. Low medium salinity (⩽29) did not seem to influence the DW of N. laevis, and high salinity resulted in a decrease of DW. The highest EPA content (4.08%) and yield (110 mgL−1) were observed at the salinity of 36 and 29, respectively.  相似文献

Statistical analysis on data collected in the Jiaozhou Bay (Shandong, China) from May 1991 to February 1994 and those collected in Hawaii from March 1958 to December 2007 shows dynamic and cyclic changes in atmospheric carbon in the Northern Pacific Ocean (NPO), as well as the variation in space-time distribution of phytoplankton primary production and atmospheric carbon in the study regions. The study indicates that the human beings have imposed an important impact on the changing trends of the atmospheric carbon. Primary production in the Jiaozhou Bay presents a good example in this regard. In this paper, dynamic models of the atmospheric carbon in the NPO, the cyclic variations in the atmospheric carbon, and primary production in the Jiaozhou Bay are studied with simulation curves presented. A set of equations were established that able to calculate the rate and acceleration of increasing carbon discharged anthropologically into the atmosphere and the conversion rate of phytoplankton to atmospheric carbon. Our calculation shows that the amount of atmospheric carbon absorbed by one unit of primary production in the Jiaozhou Bay is (3.21−9.74)×10−9/(mgC·m−2d−1), and the amount of primary production consumed by a unit of atmospheric carbon is 102.66–311.52 (mgC·m−2d−1/10−6). Therefore, we consider that the variation of atmospheric carbon is a dynamic process controlled by the increase of carbon compound and its cyclic variation, and those from anthropologic discharge, and phytoplankton growth.  相似文献

The toxic effects of benzo[α]pyrene (B[α]P) at different concentrations (0.1, 0.5, 1, 2.5 and 7.5 μgL−1) on scallop (Chlamy farreri) immune system were studied. The results showed that B[α]P had significant toxic effects on the haemocyte counts, neutral red uptake, phagocytosis, bacteriolytic and antibacterial activity (P<0.05), while the seawater control and acetone control had no significant differences. The haemocyte counts, neutral red uptake, phagocytosis and bacteriolytic activity in all B[α]P treatment groups as well as antibacterial activity in groups of 0.5, 1, 2.5 and 7.5 μgL−1 B[α]P decreased significantly (P<0.05). Some of these indices tended to be stable on the sixth day and others on the ninth day, and the indices showed clear time- and concentration-response to B[α]P. Bacteriolytic activity in 0.1μgL−1 B[α]P treatment group and antibacterial activity in 0.1 μgL−1 and 0.5 μgL- B[α]P treatment groups increased at the beginning of exposure and reached their peaks on day 1 and day 6, respectively. Following that, both activities decreased gradually and became stable after day 9. When all the indices reached stability, they were significantly lower than those in control group (P<0.05), except for antibacterial activity in 0.1 μgL−1 B[α]P treatment group (P>0.05). Thus, B[α] has evident toxic effects on scallop immune system, which supports the view that a relationship exists between pollution and immunomodulation in aquatic organisms.  相似文献

Jiaozhou Bay data collected from May 1991 to February 1994, in 12 seasonal investigations, and provided the authors by the Ecological Station of Jiaozhou Bay, were analyzed to determine the spatiotemporal variations in temperature, light, nutrients (NO3^--N, NO2^--N, NH4^ -N, SIO3^2--Si, PO4^3--P), phytoplankton, and primary production in Jiaozhou Bay. The results indicated that only silicate correlated well in time and space with, and had important effects on, the characteristics, dynamic cycles and trends of, primary production in Jiaozhou Bay. The authors developed a corresponding dynamic model of primary production and silicate and water temperature. Eq. ( 1 ) of the model shows that the primary production variation is controlled by the nutrient Si and affected by water temperature; that the main factor controlling the primary production is Si; that water temperature affects the composition of the structure of phytoplankton assemblage; that the different populations of the phytoplankton assemblage occupy different ecological niches for C, the apparent ratio of conversion of silicate in seawater into phytoplankton biomas and D, the coefficient of water temperature‘s effect on phytoplankton biomass. The authors researched the silicon source of Jiaozhou Bay, the biogeochemical sediment process of the silicon, the phytoplankton predominant species and the phytoplankton structure. The authors considered silicate a limiting factor of primary production in Jiaozhou Bay, whose decreasing concentration of silicate from terrestrial source is supposedly due to dilution by current and uptake by phytoplankton; quantified the silicate assimilated by phytoplankton, the intrinsic ratio of conversion of silicon into phytoplankton biomass, the proportion of silicate uptaken by phytoplankton and diluted by current; and found that the primary production of the phytoplankton is determined by the quantity of the silicate assimilated by them. The phenomenon of apparently high plant-nutrient concentTations but low phytoplankton biomass in some waters is reasonably explained in this paper.  相似文献

Costaria costata (C. Agardh) Saunders is one of common kelps distributed in many coastal areas worldwide; however, in China, no reports have been made on cultivation of the genus. To investigate potential cultivation of the species in the northern part of China, trials on isolation and preservation of the gametophytes were conducted using C. costata from Korea; growth and development of the gametophytes were observed. We showed that at 10±1°C, 60 μmol m−2s−1 and 12:12 h (L:D), freshly released zoospores settled down within 1 hour, and then developed into the primary cell during the following 2 days. After a vegetative growth phase lasting 6–8 days, female gametophytes became 3–4 times larger in diameter than that of the primary cell, but still remained at a unicellular stage, while male gametophytes divided into 4–10 cells with only a slight change in size. Fertilization occurred within 10 days after the zoospores were released from the sporangia, and the apical and basal tissues of the juvenile sporophyte divided and differentiated into the blade and stipe. Temperature and irradiance influenced gametophytic vegetative growth and developmental patterns. Generally, low irradiance (15 μmol m−2s−1 and 30 μmol m−2s−1) was unfavorable to the induction of fertility, but it enhanced female gametophyte division. The optimal conditions for vegetative growth were 15°C and 30 μmol m−2s−1. After transplantation of the juvenile seedlings and after eight months cultivation, the harvested mature blade reached 194 cm in length and 32.7 cm in width. Our study proves that it is feasible to implement propagation and large scale cultivation of C. costata in northern China.  相似文献

When cultured under certain environmental conditions (25°C, light intensity 80 μmol/m2·s, LD 12/12, in enriched seawater medium with 7×10−4 mol/L NO3−N, 1.56×10−4 mol/L, PO4−P and supplements of other elements like Mn, Fe, I, etc.), male and female gametophytes ofU. pinnatifida kept growing vegetatively and propagated fast at average daily fresh weight increase rate of about 20%. The empirical formulaG m=G o·3m was established to estimate the output of vegetative gametophytes. Vigorous vegetative gametophyte cells began to form reproductive structures (oogonium and spermatangium, when the temperature was lower than 25°C and other environmental factors were kept optimal. The sufficient supply of gametophyte cells provided enough seeds for raisingUndaria sporelings on production scale. Controlled cross-breeding experiments using selected male and female gametophyte clones which increase their cell number by mitosis instead of meiosis were also carried out in vitro. Juvenile sporophytes from the cross-breeding had almost the same length and width increase rates as those of the control. The fact that vegetative gametophytes can be purposely selected, propagated quickly, cross-bred, with the resulting sporophytes having almost the same characteristics leads to a new way to select desiredUndaria strains for long time use without losing the desired economic characteristics. Chondrus is a type of commercially produced red seaweed that widely used for food and carrageen extraction. Although the natural life history of the alga had been well understood, the factors influencing development of the tetraspore and carpospore remain poorly understood. In the perspective of seedling resources, the regulation of early development is crucial for the seedling nursing; therefore, it is necessary to understand the physiological influences during its early development. In this study, we studied the effects of temperature and irradiance on the early development of Chondrus ocellatus Holm under laboratory conditions. The released tetraspores and carpospores were cultivated at different temperatures (10–28°C) and irradiances (10, 60 μmol photons m−2s−1) with a photoperiod of 12L:12D. The results indicate that both tetraspores and carpospores are tolerant to temperatures of 10–25°C, and have the highest relative growth rate at 20°C. Irradiance variances influenced the growth of the discoid crusts, and the influence was more significant with increasing temperature; 60 μmol photons m−2s−1 was more suitable than 10 μmol photons m−2s−1. The optimum temperature and irradiance for the development of seedlings was 20°C and 60 μmol photons m−2s−1, respectively.  相似文献

The distributions of SiO_3-Si, PO_4-P, NO_3-N and NO_2-N concentrations in the seawater, iceand snow from 82°45′21″W, 88°01′20″N to the North Pole are reported and the exchange of thenutrients among the seawater, ice and snow are discussed. The average concentrations of the nutrients inthe seawater near the North Pole were 4.9±2.4μmol/L for SiO_3-Si; 0.60±0.10μmol/L for PO_4-P,3.4±1.7μmol/L for NO_3-N and 0.13±0.04μmol/L for NO_2-N. The nutrient concentrations inthe ice and snow in the North Pole were 0.20 and 0.65μmol/L for SiO_3-Si; 0.22 and 0.25μmol/L forPO_4-P; 1.6 and 5.8μmol/L for NO_3-N, 0.19 and 0.15μmol/L for NO_2-N.  相似文献

Interactions between Skeletonema costatum (S. costatum) and Prorocentrum donghaiense (P. donghaiiense) were inves-tigated using bi-algal cultures at different concentrations of phosphate (PO4-P) and nitrate/phosphate (N/P) ratios. Experiments were conducted under P-limited conditions and the Lotka-Volterra mathematical model was used to simulate the growth of S. costatum and P. donghaiense in the bi-algal cultures. Both of these two species were inhibited significantly in bi-algal culture. The results of the simulation showed that the inhibitory degree of S. costaum by P. donghaiense was high when the concentration of PO4-P was low (0.1μmolL-1/2 d), but that of P. donghaiense by S. costaum was high with increased PO4-P supply (0.6μmolL-1/2 d). At low concen-tration of PO4-P (0.1μmolL-1/2 d), or high concentration of PO4-P (0.6μmolL-1/2 d) with high N/P ratio (160), the interactions be-tween S. costatum and P. donghaiense were dependent on the initial cell densities of both species. At high concentration of PO4-P (0.6μmolL-1/2 d) with low N/P ratio (25 or 80), S. costatum exhibited a survival strategy superior to that of P. donghaiense. The de-gree of inhibition of P. donghaiense by S. costaum increased with elevated N/P ratio when the medium was supplemented with con-centration 0.1μmolL-1/2 d of PO4-P. The degree of inhibition to P. donghaiense by S. costaum increased with elevated N/P ratio at low concentration of PO4-P (0.1 μmolL-1/2 d). This trend was conversed at high concentration of PO4-P (0.6μmolL-1/2 d). However, the degree of inhibition of S. costaum by P. donghaiense increased with the increased N/P ratio at different PO4-P concentrations (0.1μmolL-1/2 d and 0.6μmolL-1/2 d). These results suggested that both phosphate concentration and N/P ratio affected the competition between S. costaum and P. donghaiense: P. donghaiense is more competitive in environments with low phosphate or high N/P ratio and the influence of N/P ratio on the competition was more significant with lower phosphate concentration.  相似文献

The abundance and biomass of benthic heterotrophic bacteria were investigated for the 4 typical sampling stations in the northern muddy part of Jiaozhou Bay, estuary of the Dagu River, raft culturing and nearby areas of Huangdao in March, June, August and December, 2002. The abundance and biomass range from 0.98×107 to 16.87×107 cells g−1 sediment and 0.45 to 7.08 μg C g−1 sediment, respectively. Correlation analysis showed that heterotrophic bacterial abundance and biomass are significantly correlated to water temperature (R=0.79 and 0.83, respectively,P<0.01).  相似文献

The SCENTO-System was used to study the carbon dynamics between phytoplankton primary production and heterotrophic bacterial secondary production. Most of the methods used nowadays in situ for limnological synecology studies were applied. Primary production measurement showed an increasing tendency with increasing content of chlorophylla. It provided a true photosynthetic rate lying within the range of eutrophic lakes. Net EOC released from the algae ranged from 8.5 to 27.5 μg C l−1(6h)−1. Accompanying the algal products the number of bacteria increased from 1.475 ×109 to 8.074×109 cells l−1. The bacterial mean cell volume was small, between 0.0315 and 0.0548μm3. Bacterial carbon production from direct growth estimates was compared with independent calculations of bacterial growth from EOC uptake and3H-thymidine incorporation. Direct estimates were 2.97–10.0 μg Cl−1 (24h)−1 with the exception of a zero-growth on the third day. EOC uptake was 123.5–191.0 μg Cl−1 (6h)−1. That calculated from3H-thymidine incorporation was 0.2–0.5 μg Cl−1 (6h)−1.14C-glucose dark uptake ran parallel to the increasing bacterial biomass. The respiration of glucose was 6.5% (avg.) of the gross uptake. Since the system operated without grazing pressure, a real carbon flow from primary production to bacterial secondary production could be observed.  相似文献

Polysiphonia urceolata is one type of potential commercial red seaweeds used for breeding and cultivation, because of its significant biochemical and biomedical application. However, the information of breeding and seedling incubation for cultivation is limited, especially the early development. In this study, tetrasporohyte and gametophyte of P. urceolata were taken as the study materials in Huiquan Bay, Qingdao, China. The cleaned and sterilized tetrasporophytes and gametophytes were pre-cultured in sterilized seawater, then nurtured at 18°C, 25 μmol photons m−2 s−1 in 12:12 h (light:dark) photoperiod. Continuous observation under microscope showed that the early development consists of bipolar division stage and seedling stage. In the division stage, tetraspores germinate into bipolar sporelings that further differentiate into a colorless rhizoidal portion and a lightly pigmented upright shoot. The lightly pigmented rhizoidal cell develops to a rhizoid and the larger pigmented cell transforms to an erect axis. In the seedling stage, several quasi-protuberances appear on the erect axis and form juvenile seedlings. The results demonstrate the culture of P. urceolata from tetraspores under laboratory conditions.
