To better understand the development of the annually recurring late summer red water blooms of the phototrophic ciliate Myrionecta rubra in the Columbia River estuary we examined its standing stocks and measured its growth rates both in the estuary main channels and in Baker Bay, a peripheral embayment situated near the river mouth. Data collected during two summers show a biphasic development of M. rubra blooms, with an initial phase when the protist was only detected in Baker Bay, followed by an established phase when red waters were observed throughout the lower estuary. Ilwaco harbor (Baker Bay’s seaward-end) is at least one of the locations where the bloom starts since M. rubra was detected there at concentrations >100s cell L−1 before Chinook harbor (Baker Bay’s upriver-end) or the estuary main channels. In 2010, this initial phase lasted about 1.5 months, spanning the neap tide of early July to the beginning of the neap tide of mid-August. While high growth rates were measured in Ilwaco harbor during the initial phase (1.2–3.1 d−1) and in the estuary main channels in both surface red (0.7 d−1) and adjacent non-red (1.1 d−1) waters during the established period, growth of the ciliate was not detected in Ilwaco harbor during this second phase. Growth rate data obtained during the established bloom phase also suggest that M. rubra cells in the estuary mostly divide during the daytime and that red water patches might experience self-shading.  相似文献

An aggregate flux event was observed by ship and by four underwater gliders during the 2008 sub-polar North Atlantic spring bloom experiment (NAB08). At the height of the diatom bloom, aggregates were observed as spikes in measurements of both particulate backscattering coefficient (bbp) and chlorophyll a fluorescence. Optical sensors on the ship and gliders were cross-calibrated through a series of simultaneous profiles, and bbp was converted to particulate organic carbon. The aggregates sank as a discrete pulse, with an average sinking rate of ∼75 m d−1; 65% of aggregate backscattering and 90% of chlorophyll fluorescence content was lost between 100 m and 900 m. Mean aggregate organic carbon flux at 100 m in mid-May was estimated at 514 mg C m−2 d−1, consistent with independent flux estimates. The use of optical spikes observed from gliders provides unprecedented coupled vertical and temporal resolution measurements of an aggregate flux event.  相似文献

The role of copepod grazing on the ecosystem dynamics in the Oyashio region, western subarctic Pacific was investigated during six cruises from June 2001 to June 2002. In situ grazing rates of the copepod community (CGR) were measured by the gut fluorescence method in respect to developmental stages of dominant species. In terms of biomass, more than 80% of the copepod community was dominated by six large calanoid species (Neocalanus cristatus, Neocalanus flemingeri, Neocalanus plumchrus, Eucalanus bungii, Metridia pacifica and Metridia okhotensis) throughout the year. Resulting from the observed pattern of the interzonal migrating copepods, the CGR in the Oyashio region was divided into three phases, i.e. spring (bloom), summer (post-bloom) and autumn-winter phase. During the spring bloom, late copepodites of the interzonal migrating species, N. cristatus, N. flemingeri and E. bungii appeared in the surface layer (0-50 m) to consume the production of the bloom, resulting in a high grazing rate of the copepod community (7.9 mg Chl m−2 d−1), though its impact on phytoplankton community was low due to the high primary productivity. During the post-bloom period, although the copepod community which was dominated by N. cristatus, N. plumchrus, M. pacifica and newly recruited E. bungii still maintained a high biomass, the CGR was generally lower (1.8-2.6 mg Chl m−2 d−1 for June and August 2001), probably due to the lower availability of phytoplankton. Nevertheless, the highest CGR was also observed during this period (10.5 mg Chl m−2 d−1 in June 2002). The high CGR on autotrophic carbon accounted for 69% of the primary production, suggesting that the copepod community in the Oyashio region potentially terminates the phytoplankton bloom. Abundant occurrence of young E. bungii, which is a characteristic phenomenon in the Oyashio region, was largely responsible for the high grazing pressure in June 2002 suggesting that success of reproduction, growth, and survival in E. bungii during the spring bloom is an important factor in controlling phytoplankton abundance during the post-bloom season. During autumn and winter, CGR was the lowest in the year (0.29-0.38 mg Chl. m−2 d−1) due to the disappearance of the interzonal migrating copepods from the surface layer. Diel migrant M. pacifica was the most important grazer during this period. The annual ingestion of the copepod community is estimated as 37.7 gC m−2 on autotrophic carbon (converted using C:Chl ratio of 30) or 137.9 gC m−2 on suspended particles (using C:Chl ratio of in situ value, 58-191), accounting for 13% and 46% of annual primary production, respectively. This study confirms that copepod grazing is an important pathway in carbon flow in the Oyashio region and in particular their role in the phytoplankton dynamics is significant for the termination of the spring bloom.  相似文献

We studied the microbial food web in the upper 100 m of the water column in iron-limited sub-Antarctic HNLC waters south-east of New Zealand in the SAGE experiment in 2004, with focus on bacterioplankton. Samples were collected daily from inside and outside the iron enriched patch. Short term enrichment experiments were conducted on board in 4 L polycarbonate bottles with water outside the iron enriched patch to study single and combined effects of micronutrient additions on microbial food web. Low bacterial growth was recorded in the study area with community turnover times of 50 h or more during the study period. Measurements of bacterial standing stocks and production rates in the study show minor responses to the large scale iron enrichment, with increase in rates and stocks after the first enrichment and at the end of the study period after the third iron enrichment when solar radiation increased and wind mixing decreased. The average daily bacterial production rates were 31.5 and 33.7 mgCm−2 d−1 for the OUT and IN stations, respectively; thus overall there was not a significant difference between the control and the iron-enriched patch. In the bottle experiments bacterial thymidine incorporation showed responses to single iron and silicic acid enrichments and a major growth response to the combined iron and sucrose enrichments. Phytoplankton chlorophyll-a showed clear stimulation by single additions of iron and silicic acid and silicic acid enhanced the iron impact. Cobalt additions had no effect on bacteria growth and a negative effect on phytoplankton growth. Low bacterial in situ growth rates and the enrichment experiments suggest that bacteria are co-limited by iron and carbon, and that bacterial iron uptake is dependent on carbon supply by the food web. With the high iron quota (??mol Fe mol C−1) bacteria may scavenge considerable amounts of the excess iron, and thus influence the relative importance of the microbial food web as a carbon sink.  相似文献

We conducted studies of phytoplankton and hydrological variables in a semi-enclosed bay in northern China to understand the spatial–temporal variability and relationship between these variables. Samples were collected during seven cruises in Jiaozhou Bay from November 2003 to October 2004, and were analyzed for temperature, nutrients and phytoplankton pigments. Pigments from eight possible phytoplankton classes (Diatoms, Dinoflagellates, Chlorophyceae, Prasinophyceae, Chrysophyceae, Haptophyceae, Cryptophyceae and Caynophyceae) were detected in surface water by high performance liquid chromatography (HPLC). Phytoplankton pigment and nutrient concentrations in Jiaozhou Bay were spatially and temporally variable, and most of them were highest in the northern and eastern parts of the sampling regions in spring (May) and summer (August), close to areas of shellfish culturing, river estuaries, dense population and high industrialization, reflecting human activities. Chlorophyll a was recorded in all samples, with an annual mean concentration of 1.892 μg L−1, and fucoxanthin was the most abundant accessory pigment, with a mean concentration of 0.791 μg L−1. The highest concentrations of chlorophyll a (15.299 μg L−1) and fucoxanthin (9.417 μg L−1) were observed in May 2004 at the station close to the Qingdao Xiaogang Ferry, indicating a spring bloom of Diatoms in this area. Although chlorophyll a and other biomarker pigments showed significant correlations, none of them showed strong correlations with temperature and nutrients, suggesting an apparent de-coupling between the pigments and these hydrological variables. The nutrient composition and phytoplankton community composition of Jiaozhou Bay have changed significantly in the past several decades, reflecting the increasing nutrient concentrations and decline of phytoplankton cell abundance. The unchanged total chlorophyll a levels indicated that smaller species have filled the niche vacated by the larger species in Jiaozhou Bay, as revealed by our biomarker pigment analysis.  相似文献

We characterized the community composition of phytoplankton in the western subarctic Pacific from the pre-bloom to the decline phase of the spring bloom with special reference to decreases in the silicic acid concentration in surface waters as an index for diatom bloom development. Furthermore, responses of heterotrophic bacteria and viruses to the spring bloom were also concomitantly investigated. Under pre-bloom conditions when nutrients were abundant but the surface mixed layer depth was relatively deep, chlorophyll (Chl) a concentrations were consistently low and green algae (chlorophytes and prasinophytes), cryptophytes, and diatoms were predominant in the phytoplankton assemblages as estimated by algal pigment signatures. Together with the shallowing of the mixed layer depth and the decrease in silicic acid concentration, diatoms bloomed remarkably in the Oyashio region, though the magnitude of the bloom in the Kuroshio-Oyashio transition (hereafter Transition) region was relatively small. A total of 77 diatom species were identified, with the bloom-forming diatoms mainly consisting of Thalassiosira, Chaetoceros, and Fragilariopsis species. It has become evident that the carotenoid fucoxanthin can serve as a strong indicator of the diatom carbon biomass during the spring diatom bloom. Differences in the species richness of diatoms among stations generally enabled us to separate the Oyashio bloom stations from the Transition and the Oyashio pre-bloom stations. Relatively high values of the Shannon-Wiener index for the diatom species were also maintained during the Oyashio bloom, indicating that a wide variety of species then shared dominance. In the decline phase of the Oyashio bloom when surface nutrient concentrations decreased, senescent diatom cells increased, as inferred from the levels of chlorophyllide a. Although the cell density of heterotrophic bacteria changed little with the development of the diatom bloom, viral abundance increased toward the end of the bloom, suggesting an increased likelihood of mortality among organisms including diatoms resulting from viral infection. This is the first report on the microbial trophodynamics, including viruses, during the spring diatom bloom in the western subarctic Pacific.  相似文献

The variability of bottom dissolved oxygen (DO) in Long Island Sound, New York, is examined using water quality monitoring data collected by the Connecticut Department of Environmental Protection from 1995 to 2004. Self-organizing map analysis indicates that hypoxia always occurs in the Narrows during summer and less frequently in the Western and the Central Basins. The primary factor controlling the bottom DO, changes spatially and temporally. For non-summer seasons, the levels of bottom DO are strongly associated with water temperature, which means DO availability is primarily driven by solubility. During summer, stratification intensifies under weak wind conditions and bottom DO starts to decrease and deviate from the saturation level except for stations in the Eastern Basin. For the westernmost and shallow (<15 m) stations, bottom DO is correlated with the density stratification (represented by difference between surface and bottom density). In contrast, at deep stations (>20 m), the relationship between oxygen depletion and stratification is not significant. For stations located west of the Central Basin, bottom DO continues to decrease during summer until it reaches its minimum when bottom temperature is around 19–20 °C. In most cases the recovery to saturation levels at the beginning of fall is fast, but not necessarily associated with increased wind mixing. Therefore, we propose that the DO recovery may be a manifestation of either the reduced microbial activity combined with the depletion of organic matter or horizontal exchange. Hypoxic volume is weakly correlated to the summer wind speed, spring total nitrogen, spring chlorophyll a, and maximum river discharge. When all variables are combined in a multiple regression, the coefficient of determination (r2) is 0.92. Surprisingly, the weakest variable is the total nitrogen, because when it is excluded the coefficient r2 only drops to 0.84. Spring bloom seems to be an important source of organic carbon pool and biological uptake of oxygen plays a more crucial role in the seasonal evolution of bottom DO than previously thought. Our results indicate that the reassessment phase of the Long Island Sound Total Maximum Daily Load policy on nitrogen loading will most likely fail, because it ignores the contributions of the spring organic carbon pool and river discharge. Also, it is questionable whether the goal of 58.5% anthropogenic nitrogen load reduction is enough.  相似文献

The effects of the oxygen minimum zone on the metabolism of the heterotrophic microplankton community (0.22-100 μm) in the Humboldt Current System, as well as the factors controlling its biomass production, remain unknown. Here we compare the effect of four sources of dissolved organic carbon (glucose, oxaloacetate, glycine, leucine) on microbial biomass production (such as ATP-P) and the potential enzymatic activities involved in catabolic pathways under oxic and suboxic conditions. Our results show significant differences (p < 0.05) in the ATP-P production when induced by the different substrates that are used as dissolved organic carbon herein. The induction of ATP-P production is enhanced from glucose < oxaloacetate < glycine < leucine. Nevertheless, for individual substrates, no significant differences were found between incubation under oxic and suboxic conditions except in the case of leucine. For this amino acid, the induction of ATP-P synthesis was higher under suboxic than oxic conditions. The data sets of all the substrates used showed greater potential ATP-P production under suboxic than oxic conditions. The results of the potential enzymatic activities suggest that malate dehydrogenase has the highest signal of NADH oxidization activity in the microbial assemblage. Furthermore, for all experiments, the malate dehydrogenase activity data set had a significant relationship with ATP-P production. These findings suggest that the microbial community inhabiting the oxygen minimum zone has the same or greater potential growth than the community inhabiting more oxygenated strata of the water column and that malate dehydrogenase is the activity that best represents the metabolic potential of the community.  相似文献

For studies in bio-optical oceanography, visible light properties by classes of dissolved organic matter should be characterized. The regional adjustment of model parameters is one approach which is being widely used to refine bio-optical models. In the present study, buoy and laboratory data were acquired, and during the 15-day observation period an algal bloom event occurred. The absorption coefficient of CDOM at 443 nm, ag(443), changed in the range of 0.09–0.35 m−1 and 0.1–0.34 m−1 for two depths, 0.32 m and 2.3 m., respectively, throughout the entire period. CDOM absorption was larger for bloom conditions than for non-bloom conditions. In addition, the fraction of CDOM in total absorption was higher during the bloom event than that of non-bloom conditions. The spectral slope of CDOM absorption, Sg, regressed over 400–500 nm, ranged from 0.015 to 0.0185 nm−1 with an average of 0.0166 nm−1. CDOM fluorescence intensity (Fcdom) was obtained using an internal Raman standard and varied over the range of 467.44–1538.23 in relative units. Variations in Fcdom showed a similar pattern to that of CDOM absorption. A robust non-linear relationship between Fcdom and CDOM absorption was found, with a correlation coefficient of 0.893, throughout the whole observation. An attempt to describe Sg with absorption showed a promising consequence that can be described with a negative correlation during the bloom, however, without any distinguishable tendency for non-bloom time span. A two-band ratio algorithm was also carried out for retrieving CDOM absorption. The variations in optical properties of CDOM may be related to the complicated environment in the estuarine waters, which may result from different sources of CDOM.  相似文献

Size-fractionated bacterial production, abundance and α- and β- glucosidase enzyme activities were studied with respect to changes in hydrography, total suspended matter (TSM), chlorophyll a, particulate organic carbon and nitrogen ratio (POC:PON), 1.5 M NaCl-soluble and 10 mM EDTA-soluble carbohydrates (Sal-PCHO and CPCHO) and transparent exopolymeric particles (TEP) in the surface waters from July 1999–2000 at a shallow coastal station in Dona Paula Bay, west coast of India. The bulk of the total bacterial production and glucosidase activity were associated with particles (75% and >80%, respectively). Total bacterial production was linearly correlated to chlorophyll a (r = 0.513; p < 0.05) whereas enzyme activity was significantly correlated to TSM (α-glucosidase: r = 0.721 (p < 0.001); β-glucosidase: r = 0.596 (p < 0.01)). Both α-glucosidase (r = 0.514; p < 0.05) and β-glucosidase enzymes (r = 0.598; p < 0.01) appeared to be involved in the degradation of CPCHO and Sal-PCHO, respectively. Changes in α-glucosidase/β-glucosidase ratios highlighted the varying composition of particulate organic matter. The bacterial uptake of 14C-labeled bacterial extracellular carbohydrate measured over 11 days showed a strong linear correlation between 14C-uptake and bacterial production using tritiated thymidine. The turnover rate of 14C-labeled carbohydrate-C was 0.52 d−1, higher than the estimated annual mean potential carbohydrate carbon turnover rate of 0.33 ± 0.2 d−1. Our study suggests that carbohydrates derived from sediments may serve as an important alternative carbon source sustaining the bacterial carbon demand in the surface waters of Dona Paula Bay.  相似文献

The trophic efficiency of the planktonic food web in the Phaeocystis-dominated ecosystem of the Belgian coastal waters was inferred from the analysis of the carbon flow network of the planktonic system subdivided into its different trophodynamic groups. A carbon budget was constructed on the basis of process-level field experiments conducted during the spring bloom period of 1998. Biomass and major metabolic activities of auto- and heterotrophic planktonic communities (primary production, bacterial production, nanoproto-, micro- and mesozooplankton feeding activities) were determined in nine field assemblages collected during spring at reference station 330. In 1998, the phytoplankton spring flowering was characterised by a moderate diatom bloom followed by a massive Phaeocystis colony bloom. Phaeocystis colonies, contributing 70% to the net primary production, escaped the linear food chain while the early spring diatom production supplied 74% of the mesozooplankton carbon uptake. The rest of mesozooplankton food requirement was, at the time of the Phaeocystis colony bloom, partially fulfilled by microzooplankton. Only one-third of the microzooplankton production, however, was controlled by mesozooplankton grazing pressure. Ungrazed Phaeocystis colonies were stimulating the establishment of a very active microbial network. On the one hand, the release of free-living cells from ungrazed colonies has been shown to stimulate the growth of microzooplankton, which was controlling 97% of the nanophytoplankton production. On the other hand, the disruption of ungrazed Phaeocystis colonies supplied the water column with large amounts of dissolved organic matter available for planktonic bacteria. The budget calculation suggests that ungrazed colonies contributed up to 60% to the bacterial carbon demand, while alternative sources (exudation, zooplankton egestion and lysis of other organisms) provided some 30% of bacterial carbon requirements. This suggests that the spring carbon demand of planktonic bacteria was satisfied largely by autogenic production. The trophic efficiency was defined as the ratio between mesozooplankton grazing on a given source and food production. In spite of its major contribution to mesozooplankton feeding, the trophic efficiency of the linear food chain, restricted to the grazing on diatoms, represented only 5.6% of the available net primary production. The trophic efficiency of the microbial food chain, the ratio between mesozooplankton grazing on microzooplankton and the resource inflow (the bacterial carbon demand plus the nanophytoplankton production) amounted to only 1.6%. These low trophic efficiencies together with the potential contribution of ungrazed Phaeocystis-derived production to the bacterial carbon demand suggest that during spring 1998 most of the Phaeocystis-derived production in the Belgian coastal area was remineralised in the water column.  相似文献

Spring diatom blooms are important for sequestering atmospheric CO2 below the permanent thermocline in the form of particulate organic carbon (POC). We measured downward POC flux during a sub-polar North Atlantic spring bloom at 100 m using thorium-234 (234Th) disequilibria, and below 100 m using neutrally buoyant drifting sediment traps. The cruise followed a Lagrangian float, and a pronounced diatom bloom occurred in a 600 km2 area around the float. Particle flux was low during the first three weeks of the bloom, between 10 and 30 mg POC m−2 d−1. Then, nearly 20 days after the bloom had started, export as diagnosed from 234Th rose to 360-620 mg POC m−2 d−1, co-incident with silicate depletion in the surface mixed layer. Sediment traps at 600 and 750 m depth collected 160 and 150 mg POC m−2 d−1, with a settled volume of particles of 1000-1500 mL m−2 d−1. This implies that 25-43% of the 100 m POC export sank below 750 m. The sinking particles were ungrazed diatom aggregates that contained transparent exopolymer particles (TEP). We conclude that diatom blooms can lead to substantial particle export that is transferred efficiently through the mesopelagic. We also present an improved method of calibrating the Alcian Blue solution against Gum Xanthan for TEP measurements.  相似文献

Acute and severe hypoxia associated with harmful algal bloom has become one of the major causes for the environmental deterioration of coastal areas. Although it is generally thought that a large part of the dissolved oxygen consumption at a bloom site is initiated by heterotrophic bacteria that attack organic matter derived from dead or dying algal cells, precise microbial processes leading to the hypoxia are yet to be examined. Here we show temporal dynamics of extracellular dissolved organic carbon (DOC) of the red tide forming raphidophyte Chattonella marina and bacterial populations associating with the algae under laboratory conditions. During the growth of non-axenic strains of C. marina, we monitored abundance of algae, associated bacteria, and DOC in the culture media. Bacterial cell abundance increased in response to the increase in DOC both at the beginning and the late log phase of the algal growth. Flow cytometric analysis revealed that transient increase in the percentage of respiratory-active bacterial cells also coincided with the timing of the increase in bacterial abundance and DOC. These results strongly suggest that DOC released from growing C. marina fuels respiration and growth of planktonic bacteria surrounding the algae. This has implications for the role of DOC released from C. marina bloom before the collapse in mediating interactions between neighboring algae and bacterial assemblage which may eventually lead to algal bloom-associated hypoxia.  相似文献

The inner zone of the Bahía Blanca Estuary is shallow, nutrient-rich and turbid. Tidal energy and water turbulence strongly affect the water column resulting in a well-mixed structure and high concentrations of suspended sediment. The phytoplankton community is mostly dominated by diatoms and the annual pattern has been characterized by a recurrent winter-early spring bloom. Here, we investigated to what extent the temporal variations of suspended particulate matter (SPM) regulate the phytoplankton blooms in the head of the estuary by light-limitation. Sampling was done on a fortnightly basis (weekly during the blooming season) at a fixed station in the inner zone of the estuary from January 2007 to February 2008. SPM concentrations and light extinction coefficients (k) in the water column were significantly correlated and showed relatively lower values during the phytoplankton maximal biomass levels. During winter, SPM and k reached values of 23.6 mg l−1 and 0.17 m−1 which were significantly lower than the annual means of 77.6 mg l−1 and 2.94 m−1, respectively. The particulate organic matter (POM) concentration was significantly correlated with the calculated phytoplankton biomass although the contribution of the latter to the total POM was rather low. Both, POM and biomass, had maximal values during winter (21.8 mg l−1 and 393.5 μg C l−1) and mid summer (24.3 mg l−1 and 407.0 μg C l−1), with cell densities up to 8 × 106 cells l−1 and chlorophyll a up to 24.6 μg l−1. Our results suggest that the decrease of SPM concentrations in the water column with a concomitant increase in the penetration of solar radiation seems to be one of the main causes for the development of the phytoplankton winter bloom in the Bahía Blanca Estuary.  相似文献

High concentrations of the phytoplankton metabolite dimethylsulfoniopropionate (DMSP) and its degradation product dimethylsulfide (DMS) are associated with blooms of Phaeocystis antarctica in the Ross Sea, Antarctica. Episodic and rapid vertical export of Phaeocystis biomass to deep water has been reported for the Ross Sea, therefore we examined the distribution and microbial consumption rates of DMSP and DMS throughout the sub-euphotic water column. Total DMSP (dissolved+particulate; DMSPt) was present at 0.5–22 nM at depths between 70 and 690 m during both the early bloom (November) and the late bloom (January). Sub-euphotic peaks of DMSP were sometimes associated with mid-water temperature maxima, and elevated DMSP below 70 m was found mainly in water masses characterized as Modified Circumpolar Deep Water or Antarctic Shelf Water. Overall, 50–94% of the integrated water-column DMSPt was found below the euphotic zone. At one station during the early bloom, local maxima of DMSPt (14 nM) and DMS (20 nM) were observed between 113 and 240 m and these maxima corresponded with high chlorophyll a concentrations, P. antarctica cell numbers, and Fv/Fm (the quantum yield of photosystem II). During the late bloom, a sub-euphotic maximum of DMSPt (15.8 nM) at 250 m cooccurred with peaks of chlorophyll a concentration, DMSP lyase activity, bacterial production and dissolved DMSP consumption rates. DMSP turnover contributed ~12% of the bacterial carbon demand between 200 and 400 m. DMS concentrations peaked at 286 m but the maximum concentration (0.42 nM) was far lower than observed during the early bloom, probably because of relatively rapid biological consumption of DMS (1–3 turnovers per day) which, in turn, contributed to elevated dissolved dimethylsulfoxide (DMSO) concentrations. Relatively stable DMSPt distributions at some sites suggest that rapid sinking of Phaeocystis biomass is probably not the major mechanism responsible for mesopelagic DMSP accumulations. Rather, subduction of near-surface water masses, lateral advective transport or trapping of slowly sinking P. antarctica biomass in intermediate water masses are more likely mechanisms. We found that a culture of P. antarctica maintained cellular integrity during 34 days of darkness, therefore the presence of intact cells (and DMSP) at depth can be explained even under a slow sinking/advection scenario. Dinoflagellates are unicellular eukaryotic protists that dominate in all coastal waters, and are also present in oceanic waters. Despite the central importance of dinoflagellates in global primary production, the relationship between dinoflagellates and bacteria are still poorly understood. In order to understand the ecological interaction between bacterial and dinoflageUates communities, denaturing gradient gel electrophoresis (DGGE) and SSU rRNA sequencing were applied to monitoring the population dynamics of bacteria and dinoflagellates from the onset to disappearance of a dinoflagellates bloom occurred in Baltimore Inner Harbor, from April 15 to 24, 2002. Although Prorocentrum minimum was the major bloom forming species under the light microscopy, DGGE method with dinoflagellate specific primers demonstrated that Prorocentrum micans, Gymnodinium galatheanum and Gyrodinium uncatenum were also present during the bloom. Population shifts among the minor dinoflagellate groups were observed. DGGE of PCR-amplified 16S rRNA gene fragments indicated that cyanobacteria, α, β, γ-proteobacteria, FlavobacteriumBacteroides-Cytophaga (FBC), and Planctomcetes were the major components of bacterial assemblages during the bloom. DGGE analysis showed that Cytophagales and α-proteobacteria played important roles at different stages of dinoflagellates bloom. DGGE can be used as a rapid tool to simultaneously monitor population dynamics of both bacterial and dinoflagellates communities in aquatic environments, which is demonstrated here.  相似文献

Biogenic bottom features, animal burrows and biological activities interact with the hydrodynamics of the sediment–water interface to produce altered patterns of sediment erosion, transport and deposition which have consequences for large-scale geomorphologic features. It has been suggested that depending on the hydrodynamic status of the habitat, the biological activity on the bottom may have a variety of effects. In some cases, different bioturbation activities by the same organism can result in different consequences. The burrowing crab Neohelice granulata is the most important bioturbator at SW Atlantic saltmarshes and tidal plains. Because of the great variety of habitats that this species may inhabit, it is possible to compare its bioturbation effects between zones dominated by different hydrodynamic conditions. Internal marsh microhabitats, tidal creeks bottoms and basins, and open mudflats were selected as contrasting zones for the comparison on a large saltmarsh at Bahía Blanca Estuary (Argentina). Crab burrows act as passive traps of sediment in all zones, because their entrances remain open during inundation periods at high tide. Mounds are generated when crabs remove sediments from the burrows to the surface and become distinctive features in all the zones. Two different mechanisms of sediment transport utilizing mounds as sediment sources were registered. In the first one, parts of fresh mound sediments were transported when exposed to water flow during flooding and ebbing tide, with higher mound erosion where currents were higher as compared to internal marsh habitats and open mudflats. In the second mechanism, mounds exposed to atmospheric influence during low tide became desiccated and cracked forming ellipsoidal blocks, which were then transported by currents in zones of intense water flow in the saltmarsh edge. Sedimentary dynamics varied between zones; crabs were promoting trapping of sediments in the internal saltmarsh (380 g m−2 day−1) and open mudflats (1.2 kg m−2 day−1), but were enhancing sediment removal in the saltmarsh edge (between 10 and 500 g m−2 day−1 in summer). The implication is that biologically mediated sedimentological changes could be different among microhabitats, potentially leading to contrasting geomorphologic effects within a particular ecosystem.  相似文献

The study of lysogeny in aquatic systems is an often overlooked aspect of microbial ecology, especially in tropical environments. Herein, the fraction of lysogenized cells (FLC) was detected in the surface waters of 20 coastal stations distributed from the eutrophicated shoreline to seaward waters of Hann Bay (Senegal). Concurrently, viral lytic infection rates were extrapolated from the frequency of visibly infected bacterial cells (FVIC), as determined from transmission electron microscopy observations. The experimental induction of prophage was observed in less than 3% of indigenous marine bacteria, suggesting that lysogenic stages of infection are rare in Hann Bay. Similarly, only 0.5–4.7% of bacteria showed visible signs of lytic infection. However, the positive correlation between the fraction of lysogenic and lytic cells (r = 0.67, p < 0.05, n = 20) may actually indicate that the coexistence of both lifestyles may be due to the massive and rapid induction of lysogens, potentially from the high levels of local UV radiation. Overall, we suggest that the determination of FVIC and FLC to examine the predominance of one type of cycle versus the other may be a source of misinterpretation in some particular aquatic environments.  相似文献

We report results from two surveys of pCO2, biological O2 saturation (??O2/Ar) and dimethylsulfide (DMS) in surface waters of the Ross Sea polynya. Measurements were made during early spring (November 2006-December 2006) and mid-summer (December 2005-January 2006) using ship-board membrane inlet mass spectrometry (MIMS) for high spatial resolution (i.e. sub-km) analysis. During the early spring survey, the polynya was in the initial stages of development and exhibited a rapid increase in open water area and phytoplankton biomass over the course of our ∼3 week occupation. We observed a rapid transition from a net heterotrophic ice-covered system (supersaturated pCO2 and undersaturated O2) to a high productivity regime associated with a Phaeocystis-dominated phytoplankton bloom. The timing of the early spring phytoplankton bloom was closely tied to increasing sea surface temperature across the polynya, as well as reduced wind speeds and ice cover, leading to enhanced vertical stratification. There was a strong correlation between pCO2, ??O2/Ar, DMS and chlorophyll a (Chl a) during the spring phytoplankton bloom, indicating a strong biological imprint on gas distributions. Box model calculations suggest that pCO2 drawdown was largely attributable to net community production, while gas exchange and shoaling mixed layers also exerted a strong control on the re-equilibration of mixed layer ??2 with the overlying atmosphere. DMS concentrations were closely coupled to Phaeocystis biomass across the early spring polynya, with maximum concentrations exceeding 100 nM.During the summer cruise, we sampled a large net autotrophic polynya, shortly after the seasonal peak in phytoplankton productivity. Both diatoms and Phaeocystis were abundant in the phytoplankton assemblages during this time. Minimum pCO2 was less than 100 ppm, while ??O2/Ar exceeded 30% in some regions. Mean DMS concentrations were ∼2-fold lower than during the spring, although the range of concentrations was similar between the two surveys. There was a significant correlation between pCO2, ??O2/Ar and Chl a across the summer polynya, but the strength of these correlations and the slope of O2 vs. CO2 relationship were significantly lower than during the early spring. Summertime DMS concentrations were not significantly correlated to phytoplankton biomass (Chl a), pCO2 or ??O2/Ar. In contrast to the early spring time, there were no clear temporal trends in summertime gas concentrations. Rather, small-scale spatial variability, likely resulting from mixing and localized sea-ice melt, was clearly evident in surface gas distributions across the polynya. Analysis of length-scale dependent variability demonstrated that much of the spatial variance in surface water gases occurred at scales of <20 km, suggesting that high resolution analysis is needed to fully capture biogeochemical heterogeneity in this system.  相似文献

Eucampia zodiacus Ehrenberg is harmful, as it causes reduction in the quality of the aquacultured Porphyra thalli owing to nutrient depletions during dense blooms in the late winter in the macrotidal Ariake Sea, Japan. To understand the mechanism of bloom development, changes in the abundance of E. zodiacus during a bloom were investigated along vessel transects from February to April 2012. In addition, marine environmental variables were continuously monitored by the Ariake Sea Monitoring Tower, which revealed that turbidity periodically decreased during neap tides. During the 16 February neap tide, a high Secchi depth (4.3 m) was recorded at offshore stations and the Z 1% depth, at which the light intensity attenuates to 1 % of that at the sea surface, exceeded the water depth. On 16 February, the abundance of E. zodiacus was 52–732 cells mL?1, peaking at 7.0 m depth offshore. Subsequently, abundance increased at all stations. During the 22 February spring tide, abundance became vertically uniform. On 19 March, abundance at the tower reached 3758 cells mL?1 at the surface. We conclude that an improvement in light conditions in the deeper layer triggered the bloom, although the size and the duration of the bloom were determined by nutrient availability. Thus, decreases in turbidity during neap tides and subsequent strong vertical mixing during spring tides may be instrumental in the population dynamics of the large diatom E. zodiacus in macrotidal environments.  相似文献
