本文研究一株地衣芽孢杆菌(Bacillus licheniformis)对提高泥蚶耐受镉(Cd2+)胁迫的作用,实验分析了不同浓度地衣芽孢杆菌对Cd2+胁迫下泥蚶(Tegillarca granosa)的存活率及组织中Cd残留量的影响;同时比较了地衣芽孢杆菌作用前后,泥蚶体内两类抗氧化酶,超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)的活性,进一步通过Real-time PCR分析了三类免疫相关基因金属硫蛋白(MT)、小热休克蛋白分子(sHSP)和铁蛋白(ferritin)的表达情况。研究结果显示,地衣芽孢杆菌能降低泥蚶组织中Cd残留量,提高其存活率。并能有效促进泥蚶体内SOD和GSH-Px酶活性的增加,同时降低MT、sHSP和ferritin在mRNA水平上的表达。数据表明该地衣芽胞杆菌可通过调节泥蚶的免疫系统,起到降低Cd2+对泥蚶毒害的作用。  相似文献

为构建泥蚶感染弧菌标准技术体系,本研究采用哈维氏弧菌浸泡感染和注射胁迫两种攻毒方式对泥蚶进行了急性毒性实验,观察泥蚶在菌液胁迫下的存活情况,分析哈维氏弧菌对泥蚶的半致死浓度。浸泡感染实验结果表明,随着感染水体中弧菌升高,泥蚶死亡起始时间和最大日死亡率时间均有所提前,泥蚶死亡情况呈现上升趋势,当浸泡浓度超过2×107CFU/mL,泥蚶15 d累计死亡率接近100%。注射胁迫实验结果表明,泥蚶死亡率与弧菌注射浓度呈显著正相关(r=0.964,P<0.01)。Probit回归分析结果显示,哈维氏弧菌浸泡感染泥蚶7 d后停止实验,15 d的半致死浓度(LC50)为1.35×107CFU/mL;注射胁迫下的96 h半致死剂量(96 h LD50)为2.12×107CFU/g,说明哈维氏弧菌对泥蚶有明显的毒害作用。研究结果为泥蚶对哈维氏弧菌耐受性提供数据参考,并且为泥蚶弧菌病防治提供了理论基础。  相似文献

为了解重金属Cd2+和Cu2+对泥蚶的毒害程度和泥蚶对重金属Cd2+和Cu2+的解毒能力,用毒理学实验方法研究了Cd2+和Cu2+对泥蚶(Tegillarca granosa)的急性毒性和联合毒性效应。结果表明:Cd2+、Cu2+对泥蚶的96 h半致死质量浓度分别为6.189、0.460 mg/L;安全质量浓度分别为0.062、0.005 mg/L;相对毒性Cu2+Cd2+。在1︰1毒性单位的联合作用下,Cd2+和Cu2+对泥蚶96 h的半致死质量浓度分别为1.984、0.147 mg/L;安全质量浓度分别为0.020、0.001 mg/L。2种重金属离子的联合毒性大于任一重金属离子的毒性,联合毒性表现为协同作用。  相似文献

用原子吸收光谱法(AAS)和原子荧光光度法(AFS)测定了采集于北海银滩镇、厦门海门岛、宁德漳湾镇、乐清西门岛、温岭坞根镇、三门花桥镇等地区自然滩涂以及温岭坞根镇养殖区的可口革囊星虫(Phascolosoma esculentas)及其栖息地的底泥样品中Zn、Cu、Mn、Fe、Cd、Pb、Hg 和As 等8 种重金属含量, 并对检测结果进行了含量特征、生物体与栖息地底泥中重金属含量相关性分析、生物体重金属富集系数分析。结果表明: 不同栖息地可口革囊星虫间的重金属含量差异性显著(P<0.05), 各群体中Pb 含量均超过标准, 厦门海门岛群体中Cd 含量超标; 厦门海门岛、乐清西门岛自然滩涂栖息地和温岭坞根镇养殖区等底泥中重金属含量相对较高, 所有采样点底泥中Cu 含量以及乐清西门岛自然滩涂底泥中Cd 含量达到沉积物质量标准Ⅱ类; 可口革囊星虫对Zn、Cu、Fe 的富集系数均小于1, 对Mn、Cd、Pb、Hg 和As 的富集系数均大于1。Zn、Cu、Fe、Pb 和As 的富集系数在大部分群体间差异性较小; 可口革囊星虫中的Fe 和As 含量与底泥中的Fe 和As 含量高度正相关, Zn 和Hg 含量的相关性次之, 其余重金属元素含量的相关性较差。  相似文献

The adsorption of trace metals (Pb, Zn, Cu, Cd and Hg) on calcite, kaolinite and bentonite has been studied in Krka river water of various salinities (S = 3, 20 and 38‰).Model experiments were performed at pH 8 in natural water and in UV-irradiated water samples to determine the importance of natural soluble organic matter. Cold vapour atomic absorption spectrometry was used for the Hg determination, and a differential pulse anodic stripping voltammetry (DPASV) method for determination of other trace metals, using either a hanging mercury drop electrode or a mercury-coated glassy carbon working electrode. The following results have been obtained on calcite:
1. (1) a powerful absorption of Pb and Zn was observed for both natural and UV-irradiated waters at S=20‰;
2. (2) a strong adsorption of Hg was observed in natural water, and significantly lower adsorption in UV-irradiated water at all three salinities, particularly at low salinity;
3. (3) Cd adsorption was of medium intensity in both waters at S=20‰;
4. (4) Cu was practically not adsorbable in both waters at S=20‰ if the total Cu ion concentration was lower than the copper complexing capacity value;
5. (5) Cu adsorption was of a medium intensity at S=38‰. It has not influenced by the EDTA model ligand.
Trace metals Pb, Zn and Cu are adsorbable on kaolinite and bentonite at S=38‰, whereas Cd is practically not adsorbed. The Krka River is a calcareous river, which supplies the sea with calcite and aluminosilicates. In the light of our model experiments, we suggest that the self-purification of the Krka River is remarkable in respect to Pb, Zn and Hg; this occurs to a lesser extent for Cu, and is negligible in the case of Cd.  相似文献

为对大天鹅(Cygnus cygnus)保护及湖区管理提供基础数据,作者于2011年11月~2012年3月研究了山东荣成天鹅湖越冬大天鹅食物来源及其粪便的重金属水平。结果表明:所选择的食物来源中,Zn、As和Cr在硬毛藻(Chaetomorpha linum)中含量最高,Cd在大叶藻(Zostera marina L.)叶片中含量最高,Pb在大叶藻根茎中含量最高,Cu在石莼(Ulva pertusa)中含量最高,Hg在小麦(Triticum aestivum)中含量最高;水生植物来源的粪便中Cd含量极显著高于陆地植物来源的粪便,陆地植物来源的粪便中Cr,Pb和Hg含量显著高于水生植物来源的粪便;摄食小麦苗可能造成大天鹅对Cd和Hg的积累。  相似文献

根据2012—2017年每年8月廉州湾贝类养殖区现场监测资料,探讨了重金属(Hg、Cu、Pb、Cd)在其海水和表层沉积物中的含量变化,评价了2种介质中重金属的污染状况和沉积物重金属的潜在危害状况.结果表明:海水中重金属平均含量为Cu Pb Cd Hg,除Cu外,Hg、Pb、Cd的年际变化均呈明显上升趋势,其污染程度顺序为Cu Hg Pb Cd,整体水质状况基本属于本底水平;表层沉积物中重金属平均含量为Pb Cu Cd Hg,除Hg外,Cu、Pb、Cd的年际变化趋势和水相中的变化基本一致,其污染程度排序为Cu Pb Hg Cd,整体处于中等污染水平;表层沉积物中重金属的潜在生态风险程度顺序依次为Hg Cd Cu Pb,6次调查结果的综合潜在生态风险指数RI值均低于140,平均值为76. 98,表明该海域属于低潜在生态风险水平.  相似文献

Special primers were designed based on the sequence information of superoxide dismutase genes reported in the Iiterature and were used for RT-PCR and rapid amplification of cDNA ends(RACE) to clone Tegillarca granosaMn-superoxide dismutase gene successfully.The length cDNA of Tegillarca granosaMn-superoxide dismutase gene was of 635bp by including a 3'untranslated region(UTR)of 230bp and an open reading frame(ORF) of 4.5bp encoding a polypeptide of 135 amino acids. TheTegillarca granosaMn-superoxide dismutase gene shares 66%~73% homology with Mizuhopecteb yessoensis's,Danio rerio's,Xenopus Laevis's,Human's,Callinectes sapidus'sSOD CDS, while the corresponding amino acids shares 68%-78% homology and 80%~89% comparability. These indicate the SOD gene is high evolutional conservation in animals.  相似文献

Here it is demonstrated that both Porphyra spp. and Enteromorpha spp. of macro-algae display similar and very marked seasonal variations in their concentration factor (CF) of Cu, Pb, Cd and Hg in field conditions. The CF variations are specific for each metal and reproducible over several years. The way variations in the biological activity affect the equilibrium and kinetics of the interaction between trace metals and live algae was studied in vitro. Natural seawater was used as the culture medium. Voltammetry was used for the determination of natural organic ligands and trace metals except Hg, which was determined by mercury cold vapour after on-line pre-concentration. Titrations with the relevant metal demonstrated that the maximum binding capacity of the algae was not significantly dependent on the season for Pb (ca. 100 μmol gdry algae−1), Cd (ca. 50 μmol g−1) and Hg (80–100 μmol g−1). Marked seasonal variations were observed for Cu (ca. 40 μmol g−1 in January; 70 μmol g−1 in May; and 100 μmol g−1 in August). The conditional stability constants of metal–algae complexation sites were seasonally independent and similar for both algae: logKMS′=8.5±0.3 (Cu), 5.6±0.2 (Pb), 5.3±0.2 (Cd) and 18.0±0.3 (Hg). Exudates with a strong Cu complexing capacity (logKCuL=12.47±0.06) were determined in cultures with added Cu, Pb or Cd concentrations, and identified by cathodic stripping voltammetry (CSV) as cysteine or glutathione. All the tested metals promoted the liberation of exudates, both cysteine- and glutathione-like ligands were exuded in the presence of Cu, only cysteine-like ligands in the presence of Pb, and only glutathione-like ligands in the presence of Cd, the rise depending of the season of the year, particularly for Cu. Highest levels were produced in the presence of added Pb. When exposed to either 1- or 100-μM total dissolved metal concentrations, the metal uptake, and its rate, varied with the season and the algae.  相似文献

泥蚶(Tegillarcagranosa)是一种比较特别的具有较强耐低氧能力的贝类,但目前其耐低氧分子调控机理尚不知。为探究泥蚶耐受低氧的分子调控机理,本研究对低氧胁迫下的泥蚶(Tegillarca granosa)血细胞转录组中富集的信号通路及生物学过程进行分析,并初步筛选分析耐低氧基因。对低氧(DO=0.5 mg/L)胁迫6、24、72、120 h的泥蚶血细胞进行转录组测序(RNA-seq)并开展生物信息学分析,筛选出差异基因集,并对胁迫72、120 h的DEGs进行GO富集分析、KEGG通路分析,采用qPCR方法检测7个DEGs的表达量并与转录组比较。结果显示从6 h开始到120 h的4个时间点的DEGs数量呈增多的趋势, GO功能分析主要富集在JUN激酶活性的负调控、蛋白质水解的负调控及免疫系统进程等主动抗凋亡、抗逆进程; KEGG通路分析主要富集在胰岛素及胰腺分泌的信号通路、HIF-1通路、钙信号相关通路和细胞凋亡相关通路。qPCR检测结果显示, 7个基因的表达上调/下调趋势与转录组测序一致,证实了转录组测序结果的可靠性。本研究推测胰腺分泌信号通路、钙信号通路及凋亡通路在泥蚶...  相似文献

The pathway by which "Co and 137C3 got into Area, and the type of uptake in animal were studied. The results showed that 60Co was taken up by gill and was transferred quickly to other organs through blood. There are more uptake pathways for 137Cs. The uptake of 60Co by Arca was mainly active transfer and the energy comes mainly from the process of biotic oxidation. On the contrary, the uptake of 137C3 by Arca was mainly a passive transfer, and for most organs the exchange through body-wall was a significant factor.  相似文献

对海南省花场湾内表层沉积物中重金属的含量进行测定,探讨其分布特征、相关性及潜在生态危害程度.含量测定结果表明,花场湾表层沉积物中的Cu、Pb、Zn、Cr、Cd、Hg、As的含量范围分别为8.41~51.45、5.96~30.73、16.87~99.15、18.60~57.80、0.12~0.76、0.020~0.073、6.02~17.56 mg/kg,均值分别为28.19、18.35、50.48、35.75、0.32、0.05、12.71 mg/kg.相关性分析结果表明,Cu、Pb、Zn、Hg、Cr及Cd具有相同污染源,主要受养殖影响;此外,Cu、As、Hg受潮汐影响较大,Pb与Zn受渔船及潮汐影响较大,Cd与Cr受沿岸虾塘影响较大.重金属潜在生态危害评价结果表明,研究区以中等生态危害为主,其次为强生态危害与轻微生态危害,多种重金属潜在生态危害指数(RI)高值基本位于花场湾潟湖中心.单金属潜在危害系数程度较高的是Cd与Hg,潜在生态风险大小依次为:CdHgAsCuPbCrZn.  相似文献

Some fish and crustacea caught near the northeastern Mediterranean coast of Turkey between 1977 and 1980 have been analysed by the AAS technique for their H Hg, Cd, Pb, Zn, Cu, Fe, Ni, Cr and Mn contents. In the crustacea Portunus pelagicus (crab) and Penaeus keratherus (shrimp), the two essential elements Zn and Cu, showed a linear correlation. Among the bony fish, Upeneus moluccensis (gold band goat fish), a member of the Mullidae family, and Portunus pelagicus showed a much greater tendency to accumulate Hg than other species. Although Cr concentrations in the sediments of the area studied were high, Cr levels in the organisms studied were found to be relatively low. The highly toxic metals Hg, Cd and Pb were found to be present in concentrations lower than those reported from other areas of the Mediterranean.  相似文献

镉(Cd)是水生生态环境中常见的重金属污染物,通常会抑制植物的生长,减少生物量,对植物产生毒害作用。为了研究羊栖菜幼苗对海水中Cd胁迫的抗氧化响应特性,我们在天然海水和Cd加富(100 μmol·L-1)海水两种不同的Cd浓度下培养羊栖菜幼体。实验结果表明:Cd加富胁迫显著抑制了羊栖菜幼苗生长,使藻体颜色变暗。此外,高Cd处理显著降低了羊栖菜中的色素含量,生长速率,过氧化物酶(POD)活性,脱氢抗坏血酸(DHA)含量和谷胱甘肽还原酶(GR)活性。相反,高Cd处理显著促进了藻体内的Cd元素积累、对Cd2+吸收速率,并增加了其暗呼吸/净光合速率比值(Rd/Pn),抗坏血酸(Vc)含量,可溶性蛋白质(SP)含量,且还原型谷胱甘肽(GSH)以及羊栖菜的超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性均显著增加,但对丙二醛(MDA)的影响不显著。虽然羊栖菜幼苗增加了其抗氧化活性并促进AsA-GSH循环,以产生H2O2并维持健康的新陈代谢,但高Cd胁迫显著限制了幼苗的光合作用,降低了其光合色素,抑制了其生长,并使其藻体颜色发生变化。海水中高浓度的Cd对羊栖菜幼苗具有毒性作用,并且极大地增加了这种海藻的食用风险。  相似文献

为了解九龙江口表层沉积物重金属的污染特征,作者于2013年9月在九龙江口进行了表层沉积物采样调查分析,结果表明:沉积物中Cu、Pb、Zn和Cd的空间分布较为相似,高值区均分布在南溪入海口和海门岛下游海域,Cr含量的高值区也分布在南溪入海口附近。聚类分析将采样点位分为4类:第一类主要分布在河口区外侧,沉积物中油类含量较高;第二类主要分布在南溪出海口,硫化物、Zn和Cr的含量较高;第三类分布在河口区中北部,污染物含量处于中等水平;第四类主要分布在海门岛下游海域,硫化物、TOC、Cu、Pb和Zn含量较高。地质累积系数评价显示:Hg、Cu、Pb、Zn、Cr、As处于未污染到轻度污染水平,Cd为轻度污染到重度污染水平;Zn、Cr、Cd的Igeo指数相对较高,第二类和第四类站位的Igeo指数相对较高。相关分析表明,硫化物与Cu、Zn、Cd正相关性较高,Cu与Zn、Cd、Cr正相关性较高,Pb与Zn、Cd正相关性较高,Zn与Cd、Cr正相关性较高。主成分分析表明,河口沉积物重金属主要有以下几个可能来源:流域和河口的工农业活动和采矿活动、流域水土流失、流域自然风化。针对河口沉积物重金属污染,流域-河口污染控制与生态修复需采取以下措施:工业污染的控制、农业污染控制、养殖污染的控制、植树造林与河岸带绿化和矿山生态恢复。  相似文献

Ag, Al, Ca, Cd, Cu, Fe, Mg, Mn, Pb and Zn concentrations and 210Po and 210Pb activities were measured in 26 specimens of the squid Nototodarus gouldi taken from the waters of Bass Strait in one jigging operation. All the elements show wide ranges in concentrations in specimens apparently subject to the same environmental conditions. Copper concentration was 27-1 200 μg/g, and 210Po activity 4·8–24·2 Bq/g. The animal wet weights, the elements Ag, Al, Cd, Fe and Zn, and the radionuclide 210Po have coefficients of variation in the range 40–60%; Ca, Mg and Mn show the smallest variability (CV = < 30%), and Cu the greatest (CV = 12%). Significant correlations (p < 0·001) were found between the following pairs of elements: Cd-Zn, Cd-Cu, Zn-Cu, Mg-Mn, Fe-Mn, Ca-Mg and Fe-210Po.  相似文献

为降低瘤背石磺(Onchidium struma)体内超标重金属铜和镉的含量,保证其食品安全,作者通过底泥添加木鱼石土(富含锌、硒等)、饲料添加蛋白核小球藻(Chlorella pyrenoidosa)粉以及维生素C对瘤背石磺体内Cd和Cu进行净化,探究不同实验组对瘤背石磺体内Cd和Cu的净化效果。结果表明,对照组单纯用玉米粉暂养对瘤背石磺重金属的排出有一定的效果,但远远低于试验组在底泥中添加木鱼石土、在饲料中添加蛋白核小球藻粉及维生素C的效果。在瘤背石磺产品生产过程中,通过在底泥中添加木鱼石土、在饲料中添加蛋白核小球藻粉及维生素C能高效地降低其体内重金属铜和镉的含量,达到可食用的目的,保证食品安全。  相似文献

This paper reports on a study of the toxicity of copper to three species of bivalve of commercial food value, Anadara granosa (Linnaeus), Meretrix casta, Deshayes and Crassostrea madrasensis (Preston) inhibiting the Vellar estuary at Porto Novo (southern India). The LC50 values were estimated as 60 μg Cu/litre for A. granosa, 72 μg Cu/litre for M. casta and 88 μg Cu/litre for C. madrasensis subjected to 96-h static bioassay tests after acclimation for a period of four days in the laboratory. The animals selected for study ranged in length from 29 to 44 mm in A. granosa, from 25 to 42 mm in M. casta and from 31 to 115 mm in C. madrasensis. The experiments were conducted at 25% salinity with pH 8·0 ± 0·1 at a temperature of 27°C ± 0·5°C. The revival rates of bivalves which survived exposure to the LC50 concentrations for the 96-h period (when marked and released in the natural environment) were 67% for A. granosa, and C. madrasensis and 83% for M. casta. The LT50 values for the three bivalves at different concentrations of the metal were also calculated.  相似文献

Synthesis of metallothionein (MT) as a possible mechanism by which Fundulus heteroclitus adapts to heavy metal pollution has been investigated in relation to responses to methylmercuric chloride (meHg) and HgCl2(Hg2+) at different life stages.Clutches of embryos from unpolluted sites exhibit teratogenic responses to a standard dose of meHg or of Hg2+ which range from little or no effect (tolerant) to severely affected or almost complete inhibition of development (susceptible). At the time of hatching, untreated embryos from tolerant clutches had twice as much of a protein which co-migrates in SDS-PAGE gels with Fundulus MT as do those of susceptible clutches. This difference could be a mechanism for conveying tolerance to Hg2+ and also to meHg if demethylation occurs. However, eggs at the time of deposition had virtually no MT, suggesting that the protein is synthesized too late to provide tolerance in the early stages of development which are critical in the etiology of malformations. Treatment of embryos with non-teratogenic doses of either meHg or Hg2+ did not increase the amount of MT-like protein. This may indicate that the thionein gene is already fully active, and would also explain observations that Hg2+ pretreatment of embryos does not enhance tolerance to Hg2+ in the larval stage.MT, isolated from livers of Hg2+- or Cu2+-dosed or control adults, is associated with Cu and Zn but not with Hg, although it can bind Hg2+in vitro. Fundulus, like other fish, methylates much (ca. 75 %) of its Hg burden, and meHg is considered by some not to react with MT. Fundulus liver has its Hg burden distributed between homogenate sediment (25 %), the cytosol (60 %) and the remainder in lipid. The liver responds to Hg2+ treatment by increasing lipid content. This may allow sequestration of some lipid-soluble meHg. Of the cytosolic Hg, most is associated with larger proteins and some (ca. 25 %) is dialyzable.These data suggest that metallothionein is not a significant factor in Hg tolerance in Fundulus adults, and has a questionable role in embryonic and larval tolerance to Hg2+ and none to the more toxic meHg.  相似文献

Concentrations of trace metals (As, Cd, Cr, Cu, Pb, Hg, Se, and Zn) were determined in the livers, kidneys, muscles, intestines, and hearts of twelve long-beaked common dolphins (Delphinus capensis) from the East Sea, Korea, in 2006. All specimens were entangled in various commercial fishing nets or traps and as such are recorded as by-catch. The concentrations of Cu, Hg, Se, and Zn were much higher in the liver than in the kidney, muscle, intestine, or heart. Trace metals that accumulated in the liver were, in descending order: Zn > Hg > Cd > Se > Cu > As > Cr > Pb. In contrast, the concentration of Cd was higher in the kidney than in any other organs. The trace metals accumulated in the kidney were, in descending order: Cd > Zn > Cu > Hg > Se > As > Pb > Cr. No significant differences were found in the concentrations of As, Cr, or Pb in all the tissues examined.  相似文献
