The aim of this study was to determine the species composition, biodiversity and, relative abundance of epiphytic algae and their relationship with environmental variables on three different macrophytes( Nymphaea alba, Ceratophyllum demersum, Typha latifolia) at Acarlar Floodplain Forest(AFF). Epiphytic algae were gathered monthly by collecting aquatic plants between November 2011 and October 2012, except in winter when there were no plants. In this study, 67 taxa on N. alba, 66 taxa on C. demersum and 66 taxa on T. latifolia were identified as epiphytic algae. The mean value of species richness was 17, that of diversity was 1.5 and that of evenness was 0.54 for epiphytic algae on N. alba, 17, 1.1, and 0.39 on C. demersum, and 18, 1.64, and 0.56 on T. latifolia, respectively. Oscillatoria sp. and Komvophoron crassum(Vozzen) Anagnostidis and Komárek were the most abundant and consistent epiphytic algal species, occurring in high abundance on all macrophytes. Results show that species composition of epiphytic algae was different, but diversity values were similar on all the macrophytes. The hydrological pulse is one of the most important factors determining the physical and chemical environment of the epiphytic algal community. However, substrate type also affected the colonization by F. capucina, O. sancta, P. catenata, and L. truncicola more than the epiphytic algal seasonality.  相似文献

Relationships between microbial communities and geochemical environments are important in marine microbial ecology and biogeochemistry. Although biogeochemical redox stratification has been well documented in marine sediments, its impact on microbial communities remains largely unknown. In this study, we applied denaturing gradient gel electrophoresis (DGGE) and clone library construction to investigate the diversity and stratification of bacterial communities in redox-stratified sandy reef sediments in a microcosm. A total of 88 Operational Taxonomic Units (OTU) were identified from 16S rRNA clone libraries constructed from sandy reef sediments in a laboratory microcosm. They were members of nine phyla and three candidate divisions, including Proteobacteria (Alpha-, Beta-, Gamma-, Delta-, and Epsilonproteobacteria), Actinobacteria, Acidobacteria, Bacteroidetes, Chloroflexi, Cyanobacteria, Firmicutes, Verrucomicrobia, Spirochaetes, and the candidate divisions WS3, SO31 and AO19. The vast majority of these phylotypes are related to clone sequences from other marine sediments, but OTUs of Epsilonproteobacteria and WS3 are reported for the first time from permeable marine sediments. Several other OTUs are potential new bacterial phylotypes because of their low similarity with reference sequences. Results from the 16S rRNA, gene clone sequence analyses suggested that bacterial communities exhibit clear stratification across large redox gradients in these sediments, with the highest diversity found in the anoxic layer (15–25 mm) and the least diversity in the suboxic layer (3–5 mm). Analysis of the nosZ, and amoA gene libraries also indicated the stratification of denitrifiers and nitrifiers, with their highest diversity being in the anoxic and oxic sediment layers, respectively. These results indicated that redox-stratification can affect the distribution of bacterial communities in sandy reef sediments.  相似文献

Freshwater shallow lakes typically exhibit two alternative stable states under certain nutrient loadings:macrophyte-dominated and phytoplankton-dominated water regimes.An ecosystem regime shift from macrophytes to phytoplankton blooming typically reduces the number of species of invertebrates and fishes and results in the homogenization of communities in freshwater lakes.We investigated how microbial biodiversity has responded to a shift of the ecosystem regime in Dianchi Lake,which was previously fully covered with submerged macrophytes but currently harbors both ecological states.We observed marked divergence in the diversity and community composition of bacterioplankton between the two regimes.Although species richness,estimated as the number of operational taxonomic units and phylogenetic diversity(PD),was higher in the phytoplankton dominated ecosystem after this shift,the dissimilarity of bacterioplankton community across space decreased.This decrease in beta diversity was accompanied by loss of planktonic bacteria unique to the macrophyte-dominated ecosystem.Mantel tests between bacterioplankton community distances and Euclidian distance of environmental parameters indicated that this reduced bacterial community differentiation primarily reflected the loss of environmental niches,particularly in the macrophyte regime.The loss of this small-scale heterogeneity in bacterial communities should be considered when assessing long-term biodiversity changes in response to ecosystem regime conversions in freshwater lakes.  相似文献

Sulfate-reducing bacteria (SRB), which obtain energy from dissimilatory sulfate reduction, play a vital role in the carbon and sulfur cycles. The dissimilatory sulfite reductase (Dsr), catalyzing the last step in the sulfate reduction pathway, has been found in all known SRB that have been tested so far. In this study, the diversity of SRB was investigated in the surface sediments from the adjacent area of Changjiang Estuary by PCR amplification, cloning and sequencing of the dissimilatory sulfite reductase beta subunit gene (dsrB). Based on dsrB clone libraries constructed in this study, diversified SRB were found, represented by 173 unique OTUs. Certain cloned sequences were associated with Desulfobacteraceae, Desulfobulbaceae, and a large fraction (60%) of novel sequences that have deeply branched groups in the dsrB tree, indicating that novel SRB inhabit the surface sediments. In addition, correlations of the SRB assemblages with environmental factors were analyzed by the linear model-based redundancy analysis (RDA). The result revealed that temperature, salinity and the content of TOC were most closely correlated with the SRB communities. More information on SRB community was obtained by applying the utility of UniFrac to published dsrB gene sequences from this study and other 9 different kinds of marine environments. The results demonstrated that there were highly similar SRB genotypes in the marine and estuarine sediments, and that geographic positions and environmental factors influenced the SRB community distribution.  相似文献

This study was conducted to evaluate the impact of hazy and foggy weather on the bacterial communities in bioaerosols, for which samples were collected from the Qingdao coastal region on sunny, foggy, and hazy days in January and March 2013. Bacterial community compositions were determined using polymerase chain reaction denaturing gradient gel electrophoresis (PCRDGGE). The bacterial community diversity was found to be high on foggy and hazy days, and the dominant species differed during hazy weather. The Shannon-Wiener index revealed that the bacterial community diversity of coarse particles was higher than that of fine particles in the bioaerosols. The bacterial community diversity of fine particles significantly correlated with relative humidity (RH; r2 = 0.986). The cluster analysis results indicated that the bacterial communities on sunny days differed from those on hazy and foggy days. Compared with sunny days, the bacterial communities in the fine particles during hazy weather exhibited greater changes than those in the coarse particles. Most of the sequenced bacteria were found to be closely affiliated with uncultured bacteria. During hazy weather, members of the classes Bacilli and Gammaproteobacteria (Pseudomonas and Acinetobacter) were dominant. The DGGE analysis revealed that Proteobacteria and Firmicutes were the predominant phyla, and their relative percentages to all the measured species changed significantly on hazy days, particularly in the fine particles. Haze and fog had a significant impact on the bacterial communities in bioaerosols, and the bacterial community diversity varied on different hazy days.  相似文献

The intestinal bacteria of vertebrates form a close relationship with their host.External and internal conditions of the host,including its habitat,affect the intestinal bacterial community.Similarly,the intestinal bacterial community can,in turn,influence the host,particularly with respect to disease resistance.We compared the intestinal bacterial communities of grass carp that were collected from farm-ponds or a lake.We conducted denaturing gradient gel electrophoresis of amplified 16S rRNA genes,from which 66 different operational taxonomic units were identified.Using both the unweighted pair-group method with arithmetic means clustering and principal component analysis ordination,we found that the intestinal bacterial communities from the two groups of pond fish were clustered together and inset into the clusters of wild fish,except for DF-7,and there was no significant correlation between genetic diversity of grass carp and their intestinal bacterial communities(Mantel one-tailed test,R=0.157,P=0.175).Cetobacterium appeared more frequently in the intestine of grass carp collected from pond.A more thorough understanding of the role played by intestinal microbiota on fish health would be of considerable benefit to the aquaculture industry.  相似文献

Macroalgal surfaces are prone to being attached by bacteria. Epibacterial community structures on marine macroalgae are host-specific but temporally and spatially variable. In this study, we investigated the structure of epibacterial communities on the surfaces of four red macroalgae, Gracilaria lemaneiformis, Gloiopeltis furcata, Mazzaella sp. and Porphyra yezoensis, by analyzing the sequences of 16 S r RNA gene libraries. Healthy individuals of all macroalgae species were collected in winter from a farm at Dalian, China. The results showed that the epibacterial communities were mainly dominated by α-Proteobacteria, γ-Proteobacteria and Bacteroidetes. Deinococcus-Thermus, Spirochaetes and ε-Proteobacteria were also found. The majority of cloned sequences shared the greatest similarity to those of culturable organisms. A large portion of sequences from the α-Proteobacteria homed in Roseobacter clade, i.e., genera Ahrensia, Roseovarius, Litoreibacter, Octadecabacter, Thaiassobacter and Sulfitobacter, while members of Bacteroidetes mainly belonged to family Flavobacteriaceae. The cloned sequences could be separated into 66 OTUs at 0.01 distance value, and rare common OTUs were found among libraries. At genus level, Pseudoalteromonas dominated Gr. lemaneiformis and Gl. furcata libraries, accounting for 72.2% and 47.3%, respectively. Sulfitobacter dominated P. yezoensis library, accounting for 35.4%. A previously undefined cluster within Deinococcus-Thermus dominated Mazzaella sp. library, accounting for 24.6% of the all. These results indicated that a broad range of bacteria inhabited the surfaces of these macroalgae.  相似文献

Pollution has a considerable effect on biological communities, in terms of size and diversity of the populations. Yet, the precise consequences of human activity on microbial communities in the marine environment are poorly understood. Therefore, in an ongoing collaborative research programme between Heriot-Watt University and the Ocean University of Qingdao, bacteria were isolated in 1999 and 2000 from marine sediment, seawater, seaweed, fish and shellfish, taken from locations in Shandong Province adjacent to Qingdao. Sampling locations were comprised of industrial and aquacultural sites and a clean, control site. In order to analyse microbial diversity, a polyphasic approach was adopted for characterisation of these isolates, specifically through examination of key phenotypic traits, i.e. using Biolog GN MicroPlate profiles, bacterial whole cell protein profiles and 16S and 23S rRNA gene sequences. These techniques yielded complex taxonomic data, which were subjected to statistical and cluster analyses. The application of these methods to studies of microbial communities is discussed.  相似文献

Bioremediation, is an effective and environment-friendly method of cleaning up crude oil pollution after an oil spill. However, the in situ bioremediation of crude oil is usually inhibited by deficiency of inorganic nutrients. To understand the effects of nutrient addition on the bioremediation of crude oil and the succession of bacterial communities during process of bioremediation, microcosms containing oilcontaminated sediments were constructed and biodegradation of crude oil was assessed based on the depletion of different ingredients. We used two culture-independent methods, denaturing gradient gel electrophoresis and a 16 S rRNA gene based clone library, to analyze the succession of bacterial communities. The results suggested n-alkanes were degraded after 30 days and that nutrient amendments significantly improved the efficiency of their biodegradation. Moreover, oil contamination and nutrient amendments could dramatically change bacterial community structures. Lower diversity was detected after being contaminated with oil. For instance, bacterial clones affiliated with the phylum Armatimonadetes, Firmicutes, Gemmatimonadetes, and Planctomycetes and the class Deltaproteobacteria and Epsilonproteobacteria could not be identified after 30 days of incubation with crude oil. However, "professional hydrocarbonocastic bacteria" became abundant in samples treated with oil during the bioremediation period, while these clones were almost completely absent from the control plots. Interestingly, bioinformatics analysis showed that even when dramatic differences in oil biodegradation efficiency were observed, bacterial communities in the plots with nutrient amendments were not significantly different from those in plots treated with oil alone. These findings indicated that nutrient amendments could stimulate the process of biodegradation but had less impact on bacterial communities. Overall, nutrient amendments might be able to stimulate the growth of n-alkane degrading bacteria.  相似文献

Illumina sequencing and quantitative PCR(qPCR) based on the 16 S ribosomal RNA(rRNA) gene were conducted to characterize the vertical distribution of bacterial and archaeal communities in the sediments of two sites from the South Yellow Sea. Both bacterial and archaeal communities showed a clear stratified distribution with sediment depth. The microbial communities in the upper layers were distinct from those in the deeper layers; the relative abundances of sequences of Thaumarchaeota, Gammaproteobacteria, and Actinobacteria were higher in the upper than in the deeper sediments, whereas the sequences of Bathyarchaeia, Lokiarchaeota, Euryarchaeota, Chloroflexi, and Deltaproteobacteria were relatively more abundant in the deeper sediments. Sediment depth and total organic carbon(TOC) can significantly influence both the bacterial and archaeal communities. Furthermore, bacterial and archaeal groups potentially involved in nitrogen, sulfur, and methane metabolism were detected in both sites. In our study, both ammonia-oxidizing bacteria( Nitrospira) and ammonia-oxidizing archaea( Candidatus Nitrosopumilus) were responsible for ammonia oxidization. Additionally, sulfur-reducing bacteria SEEP-SRB1 forming consortia with anaerobic methane-oxidizing archaea ANME-2 a-2 b were capable of anaerobic methane oxidation(AOM) in the 3400-02 sediment samples.  相似文献

Marine sponge Tedania anhelans distributes throughout the intertidal zone of Fujian, southeastern China, and is a potential source of natural bioactive products. The sponge harbors a large number of bacterial groups that have been identified using various techniques, including fluorescent in situ hybridization (FISH). Fractionation of dissociated sponge allowed isolation of 25 bacterial species. Based on 16S rRNA gene sequencing, phylogenetic analysis attributed most of these eubacteria to α-Proteobacteria, γ-Proteobacteria, Cytophaga/Flavobacterium/Bacteroidetes (CFB group), and the family Bacillaceae of Gram-positive bacteria. In sequence similarity, five putatively novel species were identified with less than 98% similarity to other strains in the NCBI database. Tests for antimicrobial activities were performed against Gram-positive bacteria, Gram-negative bacteria, fungi, antitumor indicators Escherichia coli 343/591 (with DNA repair deficiency), regular E. coli 343/636 (with different DNA repair capacity), and 10 bacterial isolates exhibited inhibitory bioactivities. Among these strains, three isolates were detected involving function gene NRPS-A domains, which were most closely related to the amino acid sequences of linear gramicidin synthetase and pyoverdine synthetase. These results contribute to our knowledge of the microbes associated with marine sponges and further reveal novel bacterial resources for the screening of bioactive marine natural products.  相似文献

The microbial communities colonized on microplastics (MPs) have attracted widespread attention. However, few studies focused on the MPs impacts on mangrove ecosystems, particularly on bacterial communities. We investigated the MPs pollution in mangrove of Zhujiang (Pearl) River estuary (ZRE). To study the potential risk posed by MPs to the mangrove ecosystems, the differences in bacterial communities, functions, and complexity between MPs and sediment samples were reported for the first time. Microplastics (2 991±1 586 items/kg dry weight (dw)) in sediment were mainly fibers and polyethylene, mostly transparent, and in size less than 0.5 mm. Bacterial communities and functions significantly differed from MPs in mangrove sediment. Compared with sediment, MPs significantly enriched members of Proteobacteria, Bacteroidetes, and Actinobacteria, as well as the bacteria associated with plastic-degrading and human diseases on their surface, suggesting that microbial communities on MPs may promote MPs degradation and the spread of diseases, posing potential risk to mangrove ecosystems and human health. Although bacteria on MPs exhibited a lower diversity, the co-occurrence network analysis indicated that network of bacteria colonized on MPs was bigger and more complex than those of mangrove sediment, illustrating that MPs can act as a distinct habitat in this special ecosystem. This study provides a new perspective for increasing our understanding of microplastic pollution in mangrove ecosystems.

Eighteen agar media were tested for the culture of gut-associated bacteria from farmed adult turbot(Scophthalmus maximus),including 16 agar media with or without 1% gastrointestinal(GI)supernatant,or with 2% or 4% GI supernatant.A total of 1 711 colonies were analyzed and 24 operational taxonomic units(OTUs)were identifi ed.The greatest bacterial diversity was isolated on Zobell 2216E/Zobell 2216E+ agar media,whereas MRS/MRS+ agar media produced a low diversity of colonies.Agar media with GI supernatant(1%,2%,or 4%)showed increased diversity and yielded different profi les of OTUs from the corresponding original media,suggesting that GI supernatant provides substances that enhance the culture effi ciency of bacteria from the turbot GI tract.The large majority of the colonies(82%)were γ-Proteobacteria,whereas 15.6% and 2.4% of colonies were Firmicutes and Actinobacteria,respectively.At the genus level,49.4% of all colonies were assigned to Vibrio.Other potential pathogens,including Pseudomonas,Photobacterium,and Enterobacter,and potential probiotics,including Bacillus,Paenibacillus,and Pseudomonas,were also isolated on agar media.Most cultured bacteria belonged to species that were fi rst described in the turbot GI tract.The impact of these species on turbot physiology and health should be investigated further.  相似文献

Denitrification and anammox processes are major nitrogen removal processes in coastal ecosystems.However,the spatiotemporal dynamics and driving factors of the diversity and community structure of involved functional bacteria have not been well illustrated in coastal environments,especially in human-dominated ecosystems.In this study,we investigated the distributions of denitrifiers and anammox bacteria in the eutrophic Bohai Sea and the northern Yellow Sea of China in May and November of2012 by constructing clone libraries employing nosZ and 16 S rRNA gene biomarkers.The diversity of nosZ-denitrifier was much higher at the coastal sites compared with the central sites,but not significant among basins or seasons.Alphaproteobacteria were predominant and prevalent in the sediments,whereas Betaproteobacteria primarily occurred at the site near the Huanghe(Yellow)River estuary.Anammox bacteria Candidatus Scalindua was predominant in the sediments,and besides,Candidatus Brocadia and Candidatus Kuenenia were also detected at the site near the Huanghe River estuary that received strong riverine and anthropogenic impacts.Salinity was the most important in structuring communities of nosZdenitrifier and anammox bacteria.Additionally,anthropogenic perturbations(e.g.nitrogen overloading and consequent high primary productivity,and heavy metal discharges)contributed significantly to shaping community structures of denitrifier and anammox bacteria,suggesting that anthropogenic activities would influence and even change the ecological function of coastal ecosystems.  相似文献

Using multi-mesh gillnets and trawls, the fish communities in Dianshan Lake at 6 stations from Oct. 2009 to Jul. 2010 were investigated seasonally to reveal the biodiversity and its spatial and temporal distribution patterns. The long-term changes in their structural characteristics were then analyzed to identify the main infl uencing factors and several measures for lake restoration were put forward. Thirty six species, belonging to 9 family and 30 genera, were collected, amongst which, the order Cypriniformes accounted for 61.1% of the total species number. In terms of importance value, Cypriniformes was the predominant group, Coilia nasus the dominant species, while Cyprinus carpio and Rhinogobius giurinus were the subdominant taxa. The community types did not differ among stations, but between seasons. There were no significant differences between seasons and among stations in species diversity, but richness differed both spatially and seasonally. Along with the process of eutrophication and the drastic reduction of the area colonized by macrophytes from 1959 to 2009–2010, the fish diversity declined markedly, and species numbers of herbivores and piscivores declined proportionately more than those of invertivores, omnivores, and planktivores. The decline of potamophilus and river-lake migratory fish was more marked than those of sedentary, river-sea migratory, and estuarine fishes. Eutrophication concomitant with sharp reduction of macrophyte area and overfishing may be the main reasons for the decline in fish diversity in Dianshan Lake.  相似文献

Ecological variables play a significant role in determining the diversity and distribution of any living organism on earth. Lichens are not exceptional and are quite sensitive in comparison to other organisms; hence the present study focuses on the impact of ecological variables on the diversity and distribution of epiphytic macrolichens colonizing Quercus leucotrichophora across eight different sites (50 m × 50 m) in Thal Ke Dhar forest, Kumaun Himalaya, Uttarakhand, India. For sampling of macrolichens, 200 trees (25 trees from each site) of Q. leucotrichophora were selected from each site and five quadrats of 5 cm × 10 cm (1000 quadrats in totality) were drawn at the tree trunk. From all the sampled trees, a total of 54 species of epiphytic macrolichens belonging to 18 genera and five families were recorded. Various ecological variables, namely altitude, aspect, slope, diameter at breast height (DBH), and lopping percent (partial cutting of the twigs as disturbance), were also analyzed to investigate their influence on macrolichen species composition and distribution pattern in the study area. For the determination of relationships between these variables, statistical analysis, namely Pearson’s Correlation Coefficient, Polynomial regression analysis and Principal Component Analysis (PCA) were performed. Out of all variables, lopping was significantly correlated to species richness of epiphytic macrolichens (0.712*, p<0.05) and it was confirmed by Pearson’s Correlation Coefficient. Despite of having high anthropogenic pressure or impact through lopping, the maximum number of macrolichen species was recorded at elevation 2267 meter above sea level (m asl). The present study revealed that besides other ecological variables, lopping practices can act as a key parameter in controlling the diversity and distribution not only of epiphytic macrolichens but also of other life forms such as bryophytes, pteridophytes, insects, birds etc. and can be either negatively or positively correlated.  相似文献

We studied distribution and site conditions of epiphytic orchids in a gradient of human interference in Kathmandu valley, central Nepal. The aim was to understand the recent distribution pattern of epiphytic orchids, with respect to (i) the micro-site conditions and (ii) the type and intensity of land use. The occurrence of epiphytic orchids was recorded for a grid with 1.5 km cell size. The cells represent different types and intensities of human impact. Site factors such as bark rugosity, bark pH, diameter at breast height (dbh; 1.3 m) of host trees, exposure to wind and sunlight intensity were recorded. With regard to the species richness and abundance of epiphytic orchids, we compared different human impact categories from very strong human impact (settlement area) to very low human impact (national park). Remote sensing was used for a supervised classification of land cover. Ficus religiosa turned out to be the most important host species for orchids in urban areas, while Schima wallichii and Alnus nepalensis significantly host orchids in the other categories. Both species richness and abundance of epiphytic orchids were significantly higher under very low human impact (forest in national park) and also some remaining patches of primary forest than the other regions. Micro-climate is crucial for orchid populations. Host bark pH, bark rugosity, sunlight intensity and host exposure were significantly different for all human impact categories in order to harbour epiphytic orchid species. Habitats with a mixture of mature trees are suitable and essential for the conservation of viable populations of epiphytic orchids in settled areas. The study reveals that to improve the population size of orchids it is essential for future urban forestry to: (i) Protect old trees as carriers of existing epiphytic orchid diversity, (ii) protect medium old trees to ensure that they may become old trees, (iii) plant new host trees for the future, (iv) plant in groups instead of single isolate trees. Trees should especially be planted in areas where orchids still exist to provide more trees for orchid population enlargement (e.g. along riparian system). Native species should be favoured; the pool of such native host species is wide.  相似文献

Cultured Apostichopus japonicus in China suffers from a kind of skin ulceration disease that has caused severe economic loss in recent years. The disease, pathogens of which are supposed to be bacteria by most researchers, is highly infectious and can often cause all individuals in the same culture pool to die in a very short time. The 16S rRNA gene phylogenesis of the culturable bacteria from the lesions of diseased individuals was conducted to study the biodiversity of the bacterial communities in the lesions and to identify probable pathogen(s) associated with this kind of disease. S. japonica samples were selected from a hatchery located in the eastern part of Qingdao, China. Bacterial universal primers GM5F and DS907R were used to amplify the 16S rRNA gene of bacteria colonies, and touchdown PCR was performed to amplify the target sequences. The results suggest that γ- proteobacteria (Alteromonadales and Vibrionales) of CFB group, many strains of which have been also determined as pathogens in other marine species, are the predominant bacterial genera of the diseased Apostichopus japonicus individuals.  相似文献

From May to August 2008, a large "green tide", consisting of the alga Ulva (Enteromorpha) prolifera, occurred in the Yellow Sea, China, affecting the local marine ecosystem and human activities. We investigated the influence of the green tide on the microbial community in the surface seawater, at four sites from July to August 2008, using bacterial 16S rRNA gene clone libraries. We sequenced 228 clones of unique patterns identified by restriction fragment length polymorphism (RFLP) techniques. The results show that 228 sequenced clones fell into six bacterial phyla:Proteobacteria, Bacteroidetes, Cyanobacteria, Verrucomicrobia, Actinobacteria, and Planctomycetes. Alphaproteobacteria (33%), Gammaproteobacteria (25%), Bacteroidetes (23%) and Cyanobacteria (9%) dominated the assemblage. Comparison between samples collected in July (during the tide) and those collected in August (after the tide) showed that, in the microbial community, diversities of Alphaproteobacteria and Cyanobacteria increased after the tide, while those of Gammaproteobacteria and Bacteroidetes decreased. These results indicate that the green tide influenced the growth of some bacteria, and provide information for further studies on the interactions and relationships between U. prolifera and the bacterial community. This study suggests that microbial community analysis is a good approach to monitoring green tides.  相似文献
