以单针藻Monoraphidium sp.FXY-10为研究对象,研究了黄腐酸对单针藻FXY-10生长、油脂及相关理化指标的影响,同时分析了黄腐酸对油脂生物合成相关的关键酶活性的影响。结果表明,添加25mg/L黄腐酸时,藻细胞总油脂含量比对照组提高了1.12倍,且最高油脂含量可达54.3%。藻细胞生物量、叶绿素含量无显著变化,蛋白质含量有所提升。此外,油脂生物合成相关的酶活性与油脂积累存在相关性,添加25mg/L黄腐酸诱导微藻细胞,乙酰辅酶A羧化酶(ACCase)和苹果酸酶(ME)的活性上调,而磷酸烯醇式丙酮酸羧化酶(PEPC)的活性下调。研究表明,黄腐酸作为一种外源诱导剂可促进微藻中油脂的合成,为提高微藻油脂积累提供了一种新的方法。  相似文献

许多微藻被认为是制备生物油燃料的理想原料,培养液中高浓度的Fe^3+对海水小球藻(Chlorella vulgaris)的油脂积累有明显的促进作用。采用荧光分光光度法对尼罗红染色后的海水小球藻的油脂积累过程进行了观测,同时测定了铁对小球藻生长和叶绿素含量的影响。结果表明,高铁处理的小球藻出现了两次油脂积累高峰,且第2次脂积累的量显著高于第1次。  相似文献

对新月菱形藻(Nitzschia closterium)藻细胞进行超声波破碎并离心,用分光光度计在672nm处测定其吸光度。这种方法与细胞计数法、叶绿素a含量测定法相比较,既快速又准确,且获得十分理想的线性相关性,其测定的吸光度A值可以作为衡量藻类生物量的指标。  相似文献

为拓展糖蜜酒精废醪液(molasseswastewater,MW)综合利用途径,本实验研究了在MW中添加Mg2+对单针藻Monoraphidium sp. FXY-10生长和油脂含量影响,并考察了Mg2+对MW中总氮、总磷、化学需氧量(chemicaloxygendemand,COD)的移除率。结果显示:当Mg2+浓度为800μmol/L时,微藻的生物量和油脂含量达到最高,为0.91g/L和59.58%,分别是对照组的1.62和1.26倍;CO2固定效率比对照增加了95.15%;总氮、总磷、COD的移除率分别达到88.37%、93.69%和87.98%。研究表明,适宜浓度的Mg2+可以促进MW中微藻生长和油脂含量的积累,将废水的处理和微藻油脂制备相结合,是微藻资源化利用的另一策略。  相似文献

设计4因子5水平的正交实验方案[采用正交表L25(5^6)],通过确定加速溶剂萃取法(ASE)处理的最优化条件,包括萃取溶剂、萃取温度、萃取时间和萃取循环次数,达到快速萃取微藻油脂并且萃取量高的效果.结果表明,除循环次数外,其他萃取条件因子均能显著影响油脂萃取量(P〈0.035).在萃取溶剂为无水乙醇或丙酮、温度为175℃、萃取时间为16min、循环次数为3次的最优处理条件下,微藻油脂萃取量最高(19.20%~21.36%).与常规的索氏萃取法相比,萃取量可提高39.08%~47.09%.采用ASE法在最优化条件下萃取微藻油脂可以达到良好效果.  相似文献

将1株产油淡水微藻——微拟球藻(Nannochloropsis sp.MASCC 11)分别接种到青岛市2家市政污水处理厂(STP)的尾水中,根据其生长、油脂产率和营养盐去除情况,评价了利用STP尾水培养微藻以生产富油的藻生物质同时深度净化尾水的可行性。结果表明,团岛污水处理厂(TD-STP)和李村河污水处理厂(LC-STP)尾水中无机氮和磷酸盐的浓度虽然远低于BG11培养基,但仍能支持微藻生长,而且以未经稀释的尾水更具优势,培养8 d后,藻生物量分别达到BG11培养基中生长微藻的65.23%和44.77%。STP尾水经稀释后处于营养盐缺乏状态,有利于藻细胞内脂质积累,但是油脂产率最大值(10.5 mg·L-1·d-1)仍出现在未经稀释的TD-STP尾水中,为未稀释LC-STP尾水的1.37倍。LC-STP尾水中微藻的油脂产率较低,可能与该处理厂接纳工业废水而在尾水中残留较多有害物质有关。在微藻的直接和间接作用下,TD-STP和LC-STP尾水中磷酸盐的去除率分别达到94.5%和100%;无机氮的去除率较低(分别为59.2%和45.4%),与尾水中初始N/P较高有关。上述结果表明,与接纳工业废水的污水厂相比,处理生活污水的污水厂所排尾水较适于培养产油微藻,能够实现产油微藻低耗培养与尾水深度净化相耦合。  相似文献

为了研究大黄鱼(Pseudosciana crocea)peroxiredoxin Ⅳ(Lyc-Prx Ⅳ)在细胞内的抗氧化功能,构建了表达大黄鱼Prx Ⅳ的重组质粒p CMV-Lyc-Prx Ⅳ,瞬时转染人胚肾细胞(HEK-293T),利用Western blotting方法检测转染后的细胞样品中Lyc-Prx Ⅳ的表达情况,并通过测定细胞内过氧化氢浓度来评价Lyc-Prx IV的体内抗氧化作用.结果显示,Lyc-Prx Ⅳ可在转染重组质粒p CMV-Lyc-Prx Ⅳ的HEK-293T细胞中表达,且细胞中A560处的吸光值在转染后6、12、24 h后分别为0.154、0.116以及0.162,而瞬时转染空载体p CMV后细胞样品在A560处的吸光值在转染后一直稳定在0.260左右,说明细胞中过氧化氢的浓度在转染后6、12、24 h时明显低于瞬时转染空载体p CMV细胞中的过氧化氢浓度(p〈0.01).表明Lyc-Prx Ⅳ在生物体内可以分解过氧化氢,参与生物体内氧化还原状况的调控.  相似文献

石油消耗,气候变化,能源安全等问题的日益突出,使得可再生能源研究逐渐受到世界各地的关注。微藻富含油脂,具有很多独特的优势,在生物柴油领域被寄予厚望,但利用微藻生产生物柴油也有许多问题亟待解决。通过基因工程的手段对微藻进行改造有可能是解决这一问题的突破点。国内外通过基因工程手段在高等植物、微藻、微生物中均实现过提高油脂含量或改变油脂组成的目的,而这些经验能够为基因工程在微藻中的应用起到很好的指导作用。对此,本文作了比较详细的介绍以期为利用微藻生产生物柴油提供些有价值的信息。  相似文献

优质产油藻种是实现微藻油脂产业化生产的基础,蹄形藻可以积累高含量的储藏性三酰甘油,但其是否具有微藻油脂开发潜力,目前仍然缺少系统的评价。利用形态学和分子技术对分离自暨南大学南湖的一株微藻进行鉴定,通过设置4种硝酸钠浓度(3.6、9.0、18.0和36.0 mmol/L),测定生物质浓度、总脂含量、脂肪酸组成、光合效率等指标,评价该藻株的产油能力,并利用现有模型,计算生物柴油的质量参数。经鉴定该藻株为蹄形藻JNU-3201 (Kirchneriella sp. JNU-3201),在整个培养周期内,其碳水化合物含量(干重)均低于20%,蛋白质含量呈降低趋势,总脂含量逐渐增加,说明脂类是该藻的主要储能物质;主要脂肪酸包括油酸(C18︰1)、棕榈酸(C16︰0)和亚油酸(C18︰2);该藻株的生长和油脂含量明显受氮素水平影响,在最低氮浓度条件下(3.6mmol/L),获得最高总脂含量(46.92%±1.52%,干重),在最高氮浓度条件下(36.0mmol/L),获得最高生物质浓度[(6.53±0.11)g/L],在18.0 mmol/L条件下,获得最高总脂产量[(2.43±0.06) g/L...  相似文献

通过气泡柱光反应器评价了两株硅藻筒柱藻(Cylindrotheca fusiformis)和纤细角毛藻(Chaetoceros gracilis)的生物量、油脂及甘油三脂的积累.结果发现,其生物量与油脂产率相当,脂肪酸组成方面,也均以C16:0和C16:1为主,但是筒柱藻的油脂组成以甘油三酯为主,纤细角毛藻的油脂以单酰甘油酯、磷脂和糖脂为主.通过培养条件的优化,发现相比于降低初始氮元素与硅元素,降低培养液盐度,能够获得更高的生物量、油脂以及甘油三脂产率.通过筒柱藻批次培养最高获得了0.36 g/(L·d)的甘油三脂产率,且甘三酯占收获干物质的50%以上,具有相当好的应用潜力.  相似文献

微藻固碳是一种新型节能减排技术,具有长期可持续发展的潜力。本文对两株富油微藻(球等鞭金藻和微拟球藻)进行了富碳培养下生长特性及中性脂积累特性的研究。两株富油微藻的最佳培养条件为10%CO2浓度和f培养基。本研究对两株富油微藻的最大生物量产率、总脂含量、最大油脂产率、微藻的C含量和CO2固定率进行了测定。球等鞭金藻的各参数指标分别为:142.42±4.58g/(m2·d),39.95%±0.77%,84.47±1.56g/(m2·d),45.98%±1.75%和33.74±1.65g/(m2·d)。微拟球藻的各参数指标分别为:149.92±1.80g/(m2·d),37.91%±0.58%,89.90±1.98g/(m2·d),46.88%±2.01%和34.08±1.32g/(m2·d)。实验结果显示,两株海洋微藻均属于高固碳优良藻株,适合应用于微藻烟气减排技术开发,具备用于海洋生物质能耦合CO2减排开发的潜力。  相似文献

Heavy nutrient loads in coastal waters often lead to excessive growth of microalgal and macroalgal epiphytes on seagrass leaves, with varying effects on the underlying seagrasses. This study evaluates the photosynthetic performance, epiphytic biomass and tissue nutrient content of two tropical seagrasses, Cymodocea serrulata and Thalassia hemprichii, in two intertidal areas along the Dar es Salaam coast in the Indian Ocean, a nutrient-rich region at Ocean Road (near the city centre), and a nutrient-poor region at Mjimwema (south of the city centre). Epiphyte biomass was significantly higher at the nutrient-rich site, and epiphytes were associated with reduced photosynthetic performance in both seagrass species at both sites. Likewise, nitrogen and phosphorus tissue content was higher in both species at the nutrient-rich site than at the nutrient-poor site. Epiphytic species composition on the seagrass leaves varied between seagrass species and between sites. Cymodocea serrulata had a higher number of epiphytic species at Mjimwema than at Ocean Road, whereas Thalassia hemprichii had more epiphytic species at Ocean Road than at Mjimwema. Seagrass photosynthetic performance, epiphytic biomass and nutrient content of the seagrasses were shown to be affected by nutrient concentration in the water column. Thus, for the future monitoring of the seagrass meadow, we recommend the use of combined measures such as seagrass performance, epiphytic biomass, nutrient contents and nutrient concentration levels in the water column.  相似文献

总结了目前脂类研究在海洋微藻化学分类学上的研究进展。所有的海洋微藻脂类都含一定的脂肪酸和甾醇,而不同种类的海洋微藻都有其特殊的脂肪酸和甾醇组成特征,我们可以运用这些特性来判别微藻可能所处的化学分类学地位:如果运用从某些藻类中发现的部分罕见脂类成分,还有可能对微藻的个别种类进行确定。随着对微藻中脂类物质分离,提取、纯化技术研究的不断深入,加上质谱、核磁共振等结构分析手段和脂类衍生化技术的日益完善,可以构建海洋微藻脂类物质组成结构快速查询信息库,并使其成为微藻化学分类的一个重要辅助手段。  相似文献

Mouth breaching is a recurrent event in temporarily open/closed estuaries (TOCEs). Such disturbances result in flushing and sediment scouring, reducing the microalgal biomass stock. The depletion of these microalgae may have negative repercussions in the form of depleted stocks of commercial fish, game fish, crustaceans and mollusks. The aim of this investigation was therefore: (1) to monitor the recovery of microalgal biomass and production following a breaching event; and (2) to determine the key environmental parameters influencing primary production during the open and recovery phases. Phytoplankton and benthic microalgal production was measured (14C-uptake method) successively during the closed, open and recovery phases of the Mdloti TOCE (South Africa). Upon breaching, 94–99% of microalgal biomass was washed out to sea through flushing and sediment scouring. A temporary recovery of phytoplankton and benthic microalgal biomass was observed during the open phase, but this was not sustained because of continual flushing and scouring of the sediment. During the re-closure (recovery phase), microalgal biomass immediately increased, reaching pre-breaching levels 35–40 days following the breaching event. In contrast to biomass, autochthonous pelagic primary production reached a maximum level (341 mg C m−2 h−1) during the open phase. Pelagic primary production normalized to biomass (PB) significantly increased during the open phase. This is attributed to a favorable combination of optimum light conditions, high influx of macronutrients and high water temperatures (33 °C). Similarly, benthic primary production normalized to biomass (PB) peaked during the open phase (35 mg C mg chl-a−1 h−1). Multivariate analysis showed that major variations in primary production were mainly controlled by temperature, dissolved inorganic nitrogen (DIN) to phosphorus (DIP) molar ratios (water-column and pore-water) and light extinction (Kd), all of which were regulated by the state of the mouth.  相似文献

微藻培养过程中氮缺失有利于油脂和生物量的积累,然而不同氮源条件下微藻生长与生物量的研究有限,限制了生物油脂的相关研究。本文研究通过研究南极冰藻Chlamydomonas sp.ICE-L在不同氮源条件下的细胞生长与油脂积累,进一步探究其作为富集油脂微藻的潜力。研究发现:在含有NH4CL的培养基中,Chlamydomonas sp.ICE-L生长速率最大;在含有NH4NO3的条件下,获得了最大干重量0.28 g/L。最高油脂含量0.21 g/g是在缺氮条件下获得,同时得到干重0.24 g/L。在多不饱和脂肪酸的产出方面,NH4NO3和NH4Cl为氮源培养基时要好于缺氮和KNO3培养基,在NH4NO3和NH4Cl为氮源的培养条件下ICE-L胞内C18:3和C20:5的含量高。比较而言,缺氮和KNO3培养基时C16:0、C18:1和C18:2的含量要高。  相似文献

Abstract. During the austral summer 1993 – 94, microalgal density and biomass were investigated in the sea ice, in the underlying water column, in the melt water and during the formation of the sea ice. Of the 96 taxa identified, 59 were diatoms and 32 were dinoflagellates. Among the remaining five species, Pseudopleurochloris antarctica  was very abundant both in the sea ice and in the ice-free waters. Cell densities and biomass of microalgae were higher in the bottom of the sea ice and during the formation of the ice than in the seawater, and diatoms were the dominant group at higher microalgal biomass. Among these, Entomoneis kufferathii , Chaetoceros dichaeta and Fragilariopsis species were the most common taxa.  相似文献

Two measures of microbial activity were used to characterise a variety of sediment habitats in three intertidal inlets in the Nelson region, South Island, New Zealand. Rates of microbial mineralisation potential and epibenthic microalgal production were compared with sediment textures, concentrations of photosynthetic pigments, ATP concentrations, and organic and inorganic nutrients. Baseline ranges for these parameters were established for relatively undisturbed estuarine sites for assessing future environmental deterioration and for comparison with sites affected by organic enrichment. Sediment mineralisation rates were increased more than 1000‐fold by enrichment from a fruit processing plant and microalgal production was enhanced by more than 50‐fold at a site exposed to slaughterhouse wastes. The remaining variables, although often strongly correlated with activity measurements, were not as sensitive as measures of enrichment. Sediment microbial activity measurements are proposed as a means of detecting changes in nutrient status of estuarine environments.  相似文献
