Two strains H2-410 and H2-419 were obtained from the chemically mutated survivors of wild Haematococcus pluvialis 2 by using ethyl methanesulphonate (EMS). Strains H2-410 and H2-419 showed a fast cell growth with 13% and 20% increase in biomass compared to wild type, respectively. Then H2-419-4, a fast cell growth and high astaxanthin accumulation strain, was obtained by exposing the strain H2-419 to ultraviolet radiation (UV) further. The total biomass, the astaxanthin content per cell, astaxanthin production of H2-419-4 showed 68%, 28%, and 120% increase compared to wild H. pluvialis 2, respectively. HPLC (High Performance Liquid Chromatography) data showed also an obvious proportional variation of different carotenoid compositions in the extracts of H2-419-4 and the wild type, although no peak of carotenoids appeared or disappeared. Therefore, the main compositions in strain H2-419-4, like its wild one, were free of astaxanthin, monoester, and diester of astaxanthin. The asexual reproduction in survivors after exposed to UV was not synchronous, and different from the normal synchronous asexual reproduction as the mother cells were motile instead of non-motile. Interestingly, some survivors from UV irradiation produced many mini-spores (or gamete?), the spores moved away from the mother cell gradually 4 or 5 days later. This is quite similar to sexual reproduction described by Elliot in 1934. However, whether this was sexual reproduction remains questionable, as no mating process has been observed.  相似文献

Cells of Haematococcus pluvialis Flot. et Will were collected in four different growth phases. We quantified the initial and total enzyme activity of ribulose-1,5-bisphosphate carboxylase (Rubisco) in crude extracts, and the relative expression of large-subunit ribulose-1,5-bisphosphate caboxylase / oxygenase (rbcL) mRNA. We measured the ratio of photosynthetic rate to respiration rate (P/R), maximal effective quantum yield of photosystem II (Fv/Fm), electron transport rate (ETR), actual photochemical efficiency of PSII in the light (ΦPSII), and non-photochemical quenching (NPQ). Green vegetative cells were found to be in the most active state, with a relatively higher P/R ratio. These cells also displayed the lowest NPQ and the highest Fv/Fm, ETR, and ΦPSII, indicating the most effective PSII. However, both Rubisco activity and rbcL mRNA expression were the lowest measured. In orange resting cysts with relatively lower P/R and NPQ, Rubisco activity and rbcL expression reached a peak, while Fv/Fm, ETR, and ΦPSII were the lowest measured. Taking into account the methods of astaxanthin induction used in industry, we suggest that Rubisco may participate in astaxanthin accumulation in H. pluvialis. A continuous and sufficient supply of a carbon source such as CO2 may therefore aid the large scale production of astaxanthin.  相似文献

The antioxidative capacity of astaxanthin and enzyme activity of reactive oxygen eliminating enzymes such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX) were studied in three cell types of Haematococcus pluvialis exposed to high concentrations of a superoxide anion radical (O2). The results show that defensive enzymes and astaxanthin-related mechanisms were both active in H. pluvialis during exposure to reactive oxygen species (ROS) such as O2. Astaxanthin reacted with ROS much faster than did the protective enzymes, and had the strongest antioxidative capacity to protect against lipid peroxidation. The defensive mechanisms varied significantly between the three cell types and were related to the level of astaxanthin that had accumulated in those cells. Astaxanthin-enriched red cells had the strongest antioxidative capacity, followed by brown cells, and astaxanthin-deficient green cells. Although there was no significant increase in expression of protective enzymes, the malondialdehyde (MDA) content in red cells was sustained at a low level because of the antioxidative effect of astaxanthin, which quenched O2 before the protective enzymes could act. In green cells, astaxanthin is very low or absent; therefore, scavenging of ROS is inevitably reliant on antioxidative enzymes. Accordingly, in green cells, these enzymes play the leading role in scavenging ROS, and the expression of these enzymes is rapidly increased to reduce excessive ROS. However, because ROS were constantly increased in this study, the enhance enzyme activity in the green cells was not able to repair the ROS damage, leading to elevated MDA content. Of the four defensive enzymes measured in astaxanthin-deficient green cells, SOD eliminates O2, POD eliminates H2O2, which is a by-product of SOD activity, and APX and CAT are then initiated to scavenge excessive ROS.  相似文献

Most studies on Haematococcus pluvialis have been focused on cell growth and astaxanthin accumulation;far less attention has been paid to cell cycles and proliferation patterns.The purpose of this study was to clarify cell cycles and proliferation patterns in H.pluvialis microscopically using a camera and video recorder system.The complicated life history of H.pluvial i s can be divided into two stages:the motile stage and the non-motile stage.All the cells can be classifi ed into forms as follows:motile cell,nonmotile cell,zoospore and aplanospore.The main cell proliferation,both in the motile phase and non-motile phase in H.pluvialis,is by asexual reproduction.Under normal growth conditions,a motile cell usually produces two,sometimes four,and exceptionally eight zoospores.Under unfavorable conditions,the motile cell loses its fl agella and transforms into a non-motile cell,and the non-motile cell usually produces 2,4 or 8 aplanospores,and occasionally 20–32 aplanospores,which further develop into non-motile cells.Under suitable conditions,the non-motile cell is also able to release zoospores.The larger non-motile cells produce more than 16 zoospores,and the smaller ones produce 4 or 8 zoospores.Vegetative reproduction is by direct cell division in the motile phase and by occasional cell budding in the non-motile phase.There is,as yet,no convincing direct evidence for sexual reproduction.  相似文献

To increase the cell concentration and the accumulation of astaxanthin in the cultivation of Haematococcus pluvialis, effects of different iron electrovalencies (Fe2+-EDTA and Fe3+-EDTA) and species (Fe-EDTA, Fe(OH)x32x and FeC6H5O7) addition on cell growth and accumulation of astaxanthin were studied. Results show that different iron electrovalencies have various effects on cell growth and astaxanthin accumulation of H. pluvialis. Compared with Fe3+-EDTA, Fe2+-EDTA stimulate more effectively the formation of astaxanthin. The maximum astaxanthin content (30.70 mg/g biomass cell) was obtained under conditions of 18 μmol/L Fe2+-EDTA, despite the lower cell density (2.3×105 cell/ml) in such condition. Fe3+-EDTA is more effective than Fe2+-EDTA in improving the cell growth. Especially, the maximal steady-state cell density, 2.9×105 cell/ml was obtained at 18 μmol/L Fe3+-EDTA addition. On the other hand, all the various species of iron (EDTA-Fe, Fe(OH)x32x, FeC6H5O7) are capable to improve the growth of the algae and astaxanthin production. Among the three iron species, FeC6H5O7 performed the best. Under the condition of a higher concentration (36 μmol/L) of FeC6H5O7, the cell density and astaxanthin production is 2 and 7 times higher than those of iron-limited group, respectively. The present study demonstrates that the effects of the stimulation with different iron species increased in the order of FeC6H5O7, Fe(OH)x32x and EDTA-Fe. In recent years, the immune-modulatory role of all-trans astaxanthin from different pigment sources has been studied. It was reported that all-trans astaxanthin might exist as three stereoisomers, and the composition of all-trans stereoisomers in natural materials differs from that of synthetic products. However, the different biological effects of various all-trans stereoisomers still remain unclear. In the present study, we evaluated the bioactivity of three astaxanthin stereoisomers, (3S,3’S)-trans-, (3R,3’R)-trans-and meso-trans-astaxanthin, in regulating cell-mediated immune response using mice lymphocytes and peritoneal exudates cells (PECs) systems. After the treatment with three astaxanthin stereoisomers (20 μmol L?1), the lymphocyte proliferation capacity, neutral red phagocytosis of PECs and natural killer (NK) cell cytotoxic activity were comparatively assessed. The results showed that all three astaxanthin stereoisomers significantly promoted lymphocyte proliferation, phagocytic capacity of PECs, and cytotoxic activity of NK cells. Moreover, the (3S,3’S)-trans-astaxanthin exhibited a much higher response than others.  相似文献

In this study, we examined the effect of elevated temperature on the expression patterns of genes, i.e., nacrein, irr, n16, n19, and hsp70 in the pearl oyster Pinctada fucata. The experiment was carried out at 4 temperatures, i.e., 20℃(ambient, control), 24, 28℃, and 32℃. The expression levels of target genes in P. fucata were assayed at 0, 6, 24, 48, and 96 h via real-time polymerase chain reaction. Results showed that the expression levels of nacrein and irr had no significant variations among different time points below 28℃, but significantly increased over time at 32℃. The expression levels of n16 and n19 did not change markedly at 20℃. The former increased significantly at 6 h and 24 h while the latter substantially decreased during 6–96 h at 24, 28 and 32℃. Among different temperatures, the level of n16 was significantly lower at 20℃ than at other temperatures during 6–96 h, and the level of n19 significantly varied among different temperatures at 48 h and 96 h. The expression level of hsp70 was significantly higher at 32℃ than at 20, 24 and 28℃ at 24 h. These results demonstrated that elevated temperature impacted the physiological processes of P. fucata and potentially influenced its adaptability to thermal stress.  相似文献

Euphausia pacific is an important source of natural astaxanthin.Studies were carried out to assess the extractability of astaxanthin from E.pacific using subcritical 1,1,1,2-tetrafluoroethane(R134a).To examine the effects of multiple process variables on the extraction yield,astaxanthin was extracted under various conditions of pressure(30-150 bar),temperature(303-343 K),time(10-50 min),flow rate(2-10 g min-1),moisture content(5.5%-63.61%),and particle size(0.25-0.109 mm).The results showed that the extraction yield increased with temperature,pressure,time and flow rate,but decreased with moisture content and particle size.A maximum yield of 87.74% was obtained under conditions of 100 bar,333 K,and 30 min with a flow rate of 6 g min-1 and a moisture content of 5.5%.The substantial astaxanthin yield obtained under low-pressure conditions demonstrates that subcritical R134a is a good alternative to CO 2 for extraction of astaxanthin from E.pacific.  相似文献

Fishery biology of jumbo flying squid Dosidicus gigas off Costa Rica Dome   总被引:1,自引:0,他引:1  
The jumbo flying squid(Dosidicus gigas) population was surveyed with the help of Chinese squid jigging vessels off the Costa Rica Dome(4°–11°N, 90°–100°W) in 2009 and 2010. The daily catch of D. gigas in the two survey cruises ranged from 0 to 5.5 t and was mostly obtained from the areas bounded by 6°–9°N and 91°–94°W and by 6°30′–7°30′N and 96°–97°W. The sea surface temperature in the areas yielding the most catch ranged from 27.5 to 29℃. The sex ratio of the total catch was 3.75:1(female: male). The mantle length of the squid ranged from 211 to 355 mm(male) and from 204 to 429 mm(female) with an average of 297.9 and 306.7 mm, respectively. In the relationship of the mantle length(mm) and body weight(g) of the squid, there was no significant difference between sexes. The female and male were at a similar maturity, and most individuals are maturing or have matured with a few females being spent. The size(mantle length) and age at the first sexual maturity were 297 mm and 195 d in females, and less than 211 mm and 130 d in males, respectively. Most of the sampled stomachs(70.6%) had no food remains. The major preys of the squids were fish, cephalopods and crustaceans, with the most abundant Myctophum orientale and D. gigas. The preys in more than 65% of the non-empty sampled stomachs evidenced the cannibalism of D. gigas. The results improved current understanding of the fishery biology of D. gigas off the Costa Rica Dome, which may facilitate the assessment and management of relative fishery resources.  相似文献

Marine macroalgae can absorb carbon and play an important role in carbon sequestration. As an important economic macroalga, Gracilariopsis lemaneiformis has the potential to significantly affect carbon absorption and storage in wave-sheltered intertidal reef systems. However, detailed knowledge on seasonal biomass changes and carbon storage of G. lemaneiformis is lacking, especially in many small and scattered ecosystems. Considering the influence of human activities on wild distribution of G. lemaneiformis, the understanding of seasonal dynamics of an economically important species in nature is necessary. In this study, we first investigated seasonal variations in biomass, coverage area, and carbon storage during low tide from August 2011 to July 2012 in Zhanshan Bay, Qingdao, China. Furthermore, we estimated the carbon storage potential of wild G. lemaneiformis using light use efficiency(LUE). The results show that the standing biomass and coverage area changed significantly with season. However, seasonal variations in carbon content and water content were not obvious, with an average content of 35.1% and 83.64%, respectively. Moreover, carbon storage in individual months varied between 0.67 and 47.03 g C/m 2, and the value of carbon storage was the highest in August and June and the lowest in February. In Zhanshan Bay, LUE of G. lemaneiformis was only 0.23%. If it is increased to the theoretical maximum(5%–6%), the carbon storage will have an increase of at least 21 times compared with the current, which suggested that carbon storage of wild G. lemaneiformis had a high enhancement potential. The study will help to assess a potential role of G. lemaneiformis in reducing atmospheric CO2.  相似文献

By using field survey data from the sixth forest inventory of Jiangxi Province in 2003, the biomass and carbon storage for three studied species (Pinus massoniana, Cunninghamia lanceolata, and Pinus elliottii) were estimated in Taihe and Xingguo counties of Boyang Lake Basin, Jiangxi Province, China. The relationship between carbon density and forest age was analyzed by logistic equations. Spatio-temporal dynamics of forest biomass and carbon storage in 1985-2003 were also described. The results show that total stand area of the three forest species was 3.10 × 10^5 ha, total biomass 22.20 Tg, vegetation carbon storage 13.07 Tg C, and average carbon density 42.36 Mg C/ha in the study area in 2003. Carbon storage by forest type in descending order was: P. massoniana, C. lanceolata and P. elliottii. Carbon storage by forest age group in descending order was: middle stand, young stand, near-mature stand and mature stand. Carbon storage by plantation forests was 1.89 times higher than that by natural forests. Carbon density of the three species increased 8.58 Mg C/ha during the study period. The carbon density of Taihe County was higher in the east and west, and lower in the middle. The carbon density of Xingguo County was higher in the northeast and lower in the middle. In general, the carbon density increased with altitude and gradient. Afforestation projects contribute significantly to increasing stand area and carbon storage. Appropriate forest management may improve the carbon sequestration capacity of forest ecosystems.  相似文献

The influence of alkali on astaxanthin and the optimal working wave length for measurement of astaxanthin from Haematococcus crude extract were investigated, and a spectrophotometric method for precise quantification of the astaxanthin based on the method of Boussiba et al. was established. According to Boussiba's method, alkali treatment destroys chlorophyll. However, we found that: 1) carotenoid content declined for about 25% in Haematococcus fresh cysts and up to 30% in dry powder of Haematococcus broken cysts after alkali treatment; and 2) dimethyl sulfoxide (DMSO)-extracted chlorophyll of green Haematococcus bares little absorption at 520-550 nm. Interestingly, a good linear relationship existed between absorbance at 530 nm and astaxanthin content, while an unknown interference at 540-550 nm was detected in our study. Therefore, with 530 nm as working wavelength, the alkali treatment to destroy chlorophyll was not necessary and the influence of chlorophyll, other carotenoids, and the unknown interference could be avoided. The astaxanthin contents of two samples were measured at 492 nm and 530 nm; the measured values at 530 nm were 2.617 g/100 g and 1.811 g/100 g. When compared with the measured values at 492 nm, the measured values at 530 nm decreased by 6.93% and 11.96%, respectively. The measured values at 530 nm are closer to the true astaxanthin contents in the samples. The data show that 530 nm is the most suitable wave length for spectrophotometric determination to the astaxanthin in Haematococcus crude extract.  相似文献

A Multi-channel Oceanographic Fluorescence Lidar (MOFL), with a UV excitation at 355 nm and multiple receiving channels at typical wavelengths of fluorescence from oil spills and chlorophyll-a (Chl-a), has been developed using the Laser-induced Fluorescence (LIF) technique. The sketch of the MOFL system equipped with a compact multi-channel photomultiplier tube (MPMT) is introduced in the paper. The methods of differentiating the oil fluorescence from the background water fluorescence and evaluating the Chl-a concentration are described. Two field experiments were carried out to investigate the field performance of the system, i.e., an experiment in coastal areas for oil pollution detection and an experiment over the Yellow Sea for Chl-a monitoring. In the coastal experiment, several oil samples and other fluorescence substances were used to analyze the fluorescence spectral characteristics for oil identification, and to estimate the thickness of oil films at the water surface. The experiment shows that both the spectral shape of fluorescence induced from surface water and the intensity ratio of two channels (I 495/I 405) are essential to determine oil-spill occurrence. In the airborne experiment, MOFL was applied to measure relative Chl-a concentrations in the upper layer of the ocean. A comparison of relative Chl-a concentration measurements by MOFL and the Moderate Resolution Imaging Spectroradiometer (MODIS) indicates that the two datasets are in good agreement. The results show that the MOFL system is capable of monitoring oil spills and Chl-a in the upper layer of ocean water.  相似文献

This study on dynamic changes of culture color,astaxanthin and chlorophylls,inorganic N including N-NO3^-,N-NO2^- and N-NH4^ in batch culture of Haematococcus pluvialis exposed to different additive nitrate concentration showed(1)ast/chl ratio was over 0.8 for brown and red algae,but was usually less than 0.5 for green and yellow algae;(2)N-NO3^-,in general,was unstable and decreased,except for a small unexpected increase in nitrate enriched treatment groups;(3)measurable amounts of N-NO2^- and N-NH4^ were observed respectively with three change modes although no extemal nitrite and ammonia were added into the culture;(4)a non-linear correlation between ast/chl ratio(or color)changes and the levels of N-NO3^-,N-NO2^-,N-NH4^ in H.pluvialis culture;(5)up and down variation of the ast/chl ratio occurred simultaneously with a perceptible color change from yellow to brown(or red)when N-NO3^-,N-NO2^- and N-NH4^ fluctuated around 30,5,5μmol/L respectively;(6)existence of three dynamic modes of N-NO3^-,N-NO2^- and N-NH4^ changes,obviously associated with initial extemal nitrate;(7)the key level of total inorganic N concentration regulating the above physiological changes during indoor cultivation was about 50 μmol/L;and(8)0.5-10mmol/L of nitrate was theoretically conducive to cell growth in batch culture.  相似文献

This paper presents the experimental results of composition changes of heavy fuel oil by simulating weathering in static seawater under natural environmental conditions. The results indicate: n-C10 to n-C15 were lost gradually in 24 weeks and the relative abundance of alkanes with long chains (n-C19) increased markedly. The aromatic compounds with less than two rings (except C4N) were completely lost in 24 weeks and CnP and CnD became the main aromatics in the heavy fuel oil after 24 weeks. The ratios of n-C17/Pristane (Pr) and n-C18/Phytane (Ph) were suitable for identifying lightly weathered (3 weeks) heavy fuel oil. The ratios of n-C17/n-C18 and Pr/Ph were suitable for identifying moderately weathered heavy fuel oil (12 weeks); the ratios of C2D/C2P and C3D/C3P did not change significantly in 24 weeks and were more suitable for identifying moderately weathered heavy fuel oil (24 weeks).  相似文献

Compositional data on the sterol and alcohol fractions isolated from deep-sea marine sediments from the Okinawa Trough were obtained to determine the relative contribution from marine and terrestrial inputs. Following extraction, the sterol plus alcohol fraction was isolated by layer chromatography, derivatized with BSTFA and then analysed by capillary GC and GC—MS. A suite of C26−C29 stenols and stanols and C30−C32 keto—alcohols were identified in the sediments. The thermal stability of the compounds in these sediments was studied by heating portions of the surface sediment in glass tubes for 16 hours at temperatures from 50°C to 200°C. The C27 stanol/stenol ratio increased when temperatures went up to 175°C, but the distribution of C30−C32 Keto—alcohols remained unaffected. Afforestation and reforestation are effective and ecological ways of mitigating elevated atmospheric carbon dioxide(CO2) concentration and increasing carbon(C) storage in terrestrial ecosystems. In this study, we measured the above-ground(tree, herbaceous plants and litter) and below-ground(root and soil) C storage in an aspen plantation(Populus davidiana) monoculture(PD), a larch plantation(Larix pincipis-rupprechtii) monoculture(LP), a pine plantation(Pinus tabulaeformis) monoculture(PT), a larch and birch mixed plantation(L. pincipis-rupprechtii and Betula platyphlla mixed)(MLB), and an apricot plantation(Armeniaca sibirica) monoculture(AS) under the Desertification Combating Program in Hebei Province, the northern China. The objective was to assess the effect of afforestation species on ecosystem C pools of different plantation types. Results showed that C storage of LP stand(258.0 Mg/ha) and MLB(163.4 Mg/ha) were significantly higher than the C storage in PD(45.5 Mg/ha), PT(58.9 Mg/ha) and AS(49.4 Mg/ha), respectively. Soil C was the main carbon pool of the ecosystem C storage in the five plantation stands, ranging from 31.4 Mg/ha to 232.5 Mg/ha, which accounted for 69.0%–90.1% of the total ecosystem C storage. The C storage in tree layer was about 5.2%–23.2% of ecosystem C storage. The herbaceous plants and litter layers contained 1.0%–6.0% and 1.5%–3.3% of ecosystem C storage, respectively. Our results suggest that tree species should be incorporated to accurately develop regional C budget of afforestation program, and also imply that substantial differences in ecosystem C stocks among plantation types can facilitate decision making on C management.  相似文献

The effects of 60-day starvation on survival rate. condition index (CI) , changes ofnutrient composition of different tissues, respiration and excretion of scallop ChlartnCs farreri were studiedin laboratory from Oct. 17 to Dec. 15 ,1997. Two groups (control and starvation with 200 individualseach) were cultured in two 2 m3 tanks, with 31 to 32 salinity water at 1 7 “C . Starvation effects were mea-sured alter 1 0, 20, 40 and 60 days. There was no mass mortality of scallops cf the two tanks and survivalrates of the control and starvation groups were 93 .5 % and 92.0 %, respectively. Starvation had strongeffect on the meat condition of the scallops, especially after 10 days; when relative lipid percentagedropped sharply while relative protein percentage increased. The impact of starvation on the oxygen consumption mtc (OCR) and the ammonia-N cxcretion mtc (AER) was obvious.  相似文献

The aim of this study was to investigate the distribution, density, community structure and biodiversity characteristics of benthic diatoms, and to analyze whether differences in species composition and abundance exist in different regions of the Niyang River, Tibet. Among the 157 taxa observed in 15 sampling sites in the main river and tributary, most were casual species (>100), the relative abundance of the genera Achnanthes and Fragilaria was 67% of the total relative abundance. Achnanthes minutissima was the most important species and dominated the whole river reaches (average relative abundance was 30%); the average diatom densities were 7.4 × 105 cell/cm2 at all sites, and increased slowly from the upper section to downriver. The significant indicator taxa with higher relative abundance were Achnanthes biasolettiana (18.0%) and Fragilaria arcus (18.2%), Fragilaria capucina var. vaucheriae (31.2%), Fragilaria construens var. venter (11.3%) and Cymbella affinis (11.0%) in the upper, tributary and mid-river sections, respectively. Achnanthes minutissima was the most abundant species (56%) in the downriver section. Biodiversity indices showed a gradual decrease from the up- to down-river section, and dominant species were more abundant in the upper and mid-river sections than in the downriver section. A two-way indictor species analysis (TWINSPAN) of diatom composition clearly showed four different groups, namely the upper, mid, lower and tributary sections. Detrended correspondence analysis (DCA) supported the results of TWINSPAN, and the characteristics of site distribution and species composition in the Niyang River supported the spatial structure of diatom assemblages. This study indicates that bio-assessment programs utilizing benthic diatoms could clearly benefit lotic water with regional stratification.  相似文献
