Activities of hexokinase (HK), pyruvate kinase (PK), superoxide dismutase (SOD) and catalase (CAT) and Hsp70 level were measured to evaluate the response of the commercially important sea cucumber (Apostichopus japonicus Selenka) to rapid temperature changes in laboratory. Animals were subjected to a higher temperature (from 10 to 20℃) (Tinc treatment) or to a lower temperature (from 20 to 10℃) (Tddec treatment) for 72 h. At 1, 3, 12, 24, 72 h of exposure, animals were removed and prepared for further analysis. Results showed that the effect of acute temperature changes on enzyme activities was significant. In Tinc treatment, activities of SOD and CAT increased immediately. The significant enhancement of SOD and CAT activities suggested that oxidative stress increases significantly when ambient temperature increasing from 10 to 20℃. The up-regulation of Flsp70 in Tinc and Tdec treatments indicated that Hsp70 was a bioindicator of thermal stress in the sea cucumber, and the expression pattern depended on the thermal treatment.  相似文献

Effects of temperatures and salinities on oxygen consumption and ammonia-N excretion rate of clamMeretrix meretrix were studied in laboratory from Oct. 2003 to Jan. 2004. Two schemes were designed in incremented temperature at 10, 15, 20, 25°C at 31.5 salinity and in incremented salinity at 16.0, 21.0, 26.0, 31.5, 36.0, and 41.0 at 20°C, all for 8–10 days. From 10 to 25°C, both respiration and excretion rate were increased. One-way ANOVA analysis demonstrated significant difference (P<0.01) in physiological parameters in this temperature range except between 15 and 20°C. The highestQ 10 thermal coefficient value (12.27) was acquired between 10 and 15°C, and about 1 between 15 and 20°C, indicatingM. meretrix could well acclimate to temperature changes in this range. Salinity also had significant effects on respiration and excretion rate (P<0.05). The highest values of respiration and excretion rate ofM. meretrix were recorded at 16.0 salinity (20°C). These two physiological parameters decreased as salinity increased until reached the minimumQ 10 value at 31.5 (20°C), then again, these parameters increased with increasing salinity from 31.5 to 41.0.M. meretrix can catabolize body protein to cope with osmotic pressure stress when environmental salinity is away from its optimal range. No significant difference was observed between 26.0 and 36.0 in salinity (P>0.05), suggesting that a best metabolic salinity range for this species is between 26.0 and 36.0. Physiological responses of Chinese longsnout catfish to water temperature   总被引:1,自引:0,他引:1
We evaluated the effect of water temperature on the growth and physiology of the Chinese longsnout catfish (Leiocassis longirostris Günther). The fish were reared at four temperatures (20, 25, 30, and 35°C) and sampled on days 7, 20, and 30. We measured plasma levels of insulin, free thyroxine (FT4), free 3,5,3′-triiodothyronine (FT3), lysozyme and leukocyte phagocytic activity. The optimum water temperature for growth was 27.7°C. The plasma levels of insulin and FT4 declined significantly (P<0.05) on day 30 at temperatures above 20°C. Lysozyme activity was significantly (P<0.05) lower at 25°C than at other temperatures. We conclude that final weight, insulin, FT4, and lysozyme were significantly affected by water temperature.  相似文献

1 Introduction As a result of persistent increase in carbon dioxide in the atmosphere since the 1950s, global and regional climate features, such as temperature and precipitation, have ob- viously changed (Yu et al., 2002). The General Circulation Models (GCMs) provide potential climate scenarios by studying the effects of carbon dioxide on the temperature. Tickell (1993) predicted that the mean temperature will increase by 1℃ till the year 2050 and by 3℃ at the end of the 22th century. S…  相似文献

Estivation, hibernation, and starvation are indispensable inactive states of sea cucumbers Apostichopus japonicus in nature and in culture ponds. Generally, temperature is the principal factor that induces estivation or hibernation in the sea cucumber. The present study provided insight into the physiological adaptations of A. japonicus during the three types of inactivity (hibernation, estivation, and starvation) by measuring the oxygen consumption rates (Vo2) and biochemical compositions under laboratory conditions of low (3°C), normal (17°C) and high (24°C) temperature. The results show that the characteristics of A. japonicus in dormancy (hibernation and estivation) states were quite different from higher animals, such as fishes, amphibians, reptiles, and mammals, but more closely resembled a semi-dormant state. It was observed that the shift in the A. japonicus physiological state from normal to dormancy was a chronic rather than acute process, indicated by the gradual depression of metabolic rate. While metabolic rates declined 44.9% for the estivation group and 71.7% for the hibernation group, relative to initial rates, during the 36 d culture period, metabolic rates were not maintained at constant levels during these states. The metabolic depression processes for sea cucumbers in hibernation and estivation appeared to be a passive and an active metabolic suppression, respectively. In contrast, the metabolic rates (128.90±11.70 μg/g h) of estivating sea cucumbers were notably higher (107.85±6.31 μg/g h) than in starving sea cucumbers at 17°C, which indicated that the dormancy mechanism here, as a physiological inhibition, was not as efficient as in higher animals. Finally, the principle metabolic substrate or energy source of sea cucumbers in hibernation was lipid, whereas in estivation they mainly consumed protein in the early times and both protein and lipid thereafter.  相似文献

Effects of water temperature (17, 21, 25, 30 and 35℃) and body size (14.75-281.41 g initial body weight) on food consumption, growth, feed conversion, and dry matter content in orange-spotted grouper fed to satiation were investigated. The combined effect of temperature (T, ℃) and body weight (W, g) on maximum food consumption (Cmax, g/day) was described as: InCmax=-7.411+0.828 InW+0.317T4).004 7T2, and the optimum feeding temperature was 33.9℃. The combined effect of temperature and body weight on growth (G) was described as: lnG=-4.461-0.2081nW+0.394T-0.006 3T^2. The optimum growth temperature was 31.4℃, whereas overall growth rates were high at 25, 30 and 35 ℃. Feed conversion efficiencies (FCE, %), increasing first and then decreasing with increasing temperature, averaged from 1.8 to 2.1 in terms of dry weight of food fish. The optimum temperature for FCE tended to be lower than that for growth or feeding. Dry matter content increased with both increasing water temperature (17, 25, 30 and 35℃) and body weight, and the combined effect of temperature and body weight on dry matter content (DM, %) was described as: lnDM =3.232+0.01 4 lnW-0.004 4T+0.001 2TInW.  相似文献

Wetland stores substantial amount of carbon and may contribute greatly to global climate change debate. However, few researches have focused on the effects of global climate change on carbon mineralization in Zoigê al-pine wetland, Qinghai-Tibet Plateau, which is one of the most important peatlands in China. Through incubation ex-periment, this paper studied the effects of temperature, soil moisture, soil type (marsh soil and peat soil) and their in-teractions on CO2 and CH4 emission rates in Zoigê alpine wetland. Results show that when the temperature rises from 5℃ to 35℃, CO2 emission rates increase by 3.3-3.7 times and 2.4-2.6 times under non-inundation treatment, and by 2.2-2.3 times and 4.1-4.3 times under inundation treatment in marsh soil and peat soil, respectively. Compared with non-inundation treatment, CO2 emission rates decrease by 6%-44%, 20%-60% in marsh soil and peat soil, respec-tively, under inundation treatment. CO2 emission rate is significantly affected by the combined effects of the tempera-ture and soil type (p < 0.001), and soil moisture and soil type (p < 0.001), and CH4 emission rate was significantly af-fected by the interaction of the temperature and soil moisture (p < 0.001). Q10 values for CO2 emission rate are higher at the range of 5℃-25℃ than 25℃-35℃, indicating that carbon mineralization is more sensitive at low temperature in Zoigê alpine wetland.  相似文献

The growth, metabolism and physiological response of the sea cucumber, Apostichopus japonicus, were investigated during periods of inactivity. The body weight, oxygen consumption rate (OCR), activities of acidic phosphatase (ACP), alkaline phosphatase (AKP), catalase (CAT) and superoxide dismutase (SOD), and content of heat shock protein 70 (Hsp70) in the body wall and coelomic fluid of A. japonicus were measured during starvation, experimental aestivation and aestivation. The results showed that the body weight of sea cucumber in the three treatments decreased significantly during the experimental period (P < 0.05). The OCR of sea cucumber reduced in starvation and experimental aestivation treatments, but increased gradually in natural aestivation treatment. The activities of ACP and AKP of sea cucumber decreased gradually in all treatments, whereas those of SOD and CAT as well as Hsp70 content decreased in the starvation and experimental aestivation treatments and increased in natural aestivation treatment. The sea cucumber entered a state of aestivation at 24℃. To some extent, the animals in experimental aestivation were different from those in natural aestivation in metabolism and physiological response. These findings suggested that the aestivation mechanism of A. japonicus is complex and may not be attributed to the elevated temperature only.  相似文献

Seagrass restoration as part of ocean ecosystem protection has been launched for many years all over the world, but intensive research on this subject in China has just begun in recent years. Seed broadcasting has been widely accepted as the most potentially useful method for seagrass restoration over large areas. We examined the influence of key environmental factors on seed germination to help promote eelgrass bed restoration. Under anoxic conditions, the influence of temperature and salinity on the germination rate of eelgrass (Zostera marina L.) seeds was examined at different combinations of four temperatures (4, 9, 14, and 24°C) and nine salinities (5 to 45, increment of 5). The effect of significant interaction of temperature and salinity on germination rate was observed (ANOVA) (P<0.001). The highest germination rate (83.3 ± 3.5)% was reached in 8 weeks at 14°C and salinity 5. Higher temperature significantly increased the germination rate at salinity 5 (P<0.001) during the whole observation period except for 24°C, while lower salinity significantly increased the germination rate at 14°C (P<0.001). Although significant interaction was found between temperature and salinity (P<0.001), the influence of salinity was stronger than that of temperature for the germination of eelgrass seeds. These results provide useful information for the propagation of artificial seedlings for seagrass restoration in China.  相似文献

In laboratory conditions, effects of rearing temperature and stocking density were examined on hatching of fertilized egg and growth of auricularia larvae of Apostichopus japonicus respectively. Data series like larval length and density, metamorphic time, and survival rate of the larvae were recorded. Statistics showed that for A. japonicus, survival rate (from fertilized egg to late auricularia) decreased significantly with the increasing rearing temperature (P<0.05). At different temperatures SGR was statistically significant as well (P<0.05) from day 1, and maximal SGR was found on day 9 at 24°C (159.26±3.28). This study clearly indicated that at low temperature (<24°C), metamorphic rate was remarkably higher than at higher temperature (>26°C). Hatching rate was significantly different between 0.2–5 ind./ml groups and 20–50 ind./ml groups. Rearing larvae at the higher density had the smaller maximal-length, whereas needed longer time to complete metamorphosis. This study suggested that 21°C and 0.4 ind./ml can be used as the most suitable rearing temperature and stocking density for large -scale artificial breeding of A. japonicus’s larvae.  相似文献

Maximum rate of food consumption ( Cmax ) was determined for juvenile Sebastodes fuscescens (Houttuyn) at water temperature of 10, 15, 20 and 25℃. The relationships of Cmax to the body weight (W) at each temperature were described by a power equation: lnCmax = a b lnW. Covariance analysis revealed significant interaction of the temperature and body weight. The relationship of adjusted Cmax to water temperature (T) was described by a quadratic equation: Cmax = -0.369 0.456T - 0.0117T^2. The optimal feeding temperature calculated from this equation was 19.5 ℃. The coefficients of the multiple regression estimation relating Cmax to body weight (W) and water temperature (T) were given in the Table 2.  相似文献

Collagen of squid (Ommastrephes bartrami) skin was examined in the present study. Histology showed that collagen fiber in the skin was partially cross-linked with muscle fiber. Acid-solubilized collagen (ASC) and pepsin-solubilized collagen (PSC) were extracted from the skin and characterized. The results of amino acid composition and electrophoretic patterns revealed that ASC and PSC were both type I collagen, containing α1 and α2 chains. FTIR (fourier transform infrared spectroscopy) investigations confirmed the existence of helical arrangements in PSC of squid skin. The denaturation temperature (Td) and shrinkage temperature (Ts) of PSC were 29.4°C and 52.8°C, respectively.  相似文献

Reproductive rate of a marine planktonic copepodLabidocera euchaeta Giesbrecht in Xiamen Harbor bor was studied during the period from April 1986 to August 1987. Results showed that rate of egg production (F, eggs/(♀, d)) was positively correlated with temperature (T,oC) and body weight of spawning female (Wc, μgC) as:F=0.0637 (T−7)0.7445 Wc. Each month a maximum rate of egg production occurred at one of the five experimental temperatures (10°, 15°, 25°, and 30°C) near the monthly mean field temperature. The available food density (P, μgC/ml) had significant effect on specific rate of egg production (Fc, %C ♀/d) as:Fc=0.1732 (P-0.0179)/P. The gross efficiency of egg production of ca 62% in carbon was independent of food density snd ingestion rate. Observations on in situ egg production and hatching rate demonstrated that this species reproduces continuously all the year round, with summer as its high reproductive season and winter a low one. Ten generations a year is also suggested based on the number of the peaks of average population egg production rate. This agreed well with the result obtained from the generation-time equations. Egg-laying pattern was also observed and discussed.  相似文献

Effects of temperature on oxygen consumption rate (OCR) and ammonia—N excretion rate of scallopChlamys farreri (1.7–6.2 cm in shell height) were studied in laboratory from Dec. 30,1996 to Jan. 28,1997. Under the controlled conditions of ambient water temperature 10–31°C and salinity 32, the concentrations of dissolved oxygen and ammonia—N were determined by the Winkle method and the hypobromite method, respectively. Results showed that the OCR ranged from 1.20 mg/g (DW) · h to 5.76 mg/g (DW) · h. The OCR increased with temperature from 10°C to 23°C, but at 28°C the OCR of mature individuals decreased, and that of different size scallops reduced at 31°C. The ammonia—N excretion rate ranged from 113.03 μg NH4-N/g (DW) · h to 486.63 μg NH4-N/g (DW) · h, and increased with temperature from 10°C to 31°C. To study the effect of low temperature stress on hematological parameters and HSP gene expression in the turbot (Scophthalmus maximus), water temperature was lowered rapidly from 18 to 1°C. During the cooling process, three individuals were removed from culture tanks at 18, 13, 8, 5, 3, and 1°C. Blood samples and tissues were taken from each individual, hematological indices and HSP gene expression in tissues were measured. The red blood cell count, white blood cell count, and hemoglobin concentration decreased significantly (P < 0.05) as temperature decreased. Enzyme activities of plasma alanine transaminase and creatine kinase increased as temperature decreased, whereas aspartic transaminase and γ-glutamyl transpeptidase activities displayed no obvious changes above 1°C and lactate dehydrogenase activity increased first and then decreased. Blood urea nitrogen and uric acid levels were highest at 8°C, and creatinine concentration was highest at 3°C. The concentrations of plasma cortisol, cholesterol, and triglyceride all increased significantly (P < 0.05) as temperature decreased. The serum glucose concentration increased first and then decreased to the initial level. The HSP70 mRNA expression showed various patterns in different tissues, whereas HSP90 mRNA expression showed the same tendency in all tissues. Overall, these results indicate that temperature decreases in the range of 8 to 5°C may induce a stress response in S. maximus and that temperature should be kept above 8°C in the aquaculture setting to avoid damage to the fish.  相似文献

Gracilaria asiatica, being highly efficient in nutrient absorption,is cultivated in sea cucumber ponds to remove nutrients such as nitrogen and phosphate. It was cultured in a laboratory simulating field conditions, and its nutrient absorption was measured to evaluate effects of environmental conditions.Ammonia nitrogen(AN),nitrate nitrogen(NN),total inorganic nitrogen(TIN),and soluble reactive phosphorus(SRP) uptake rate and removal efficiency were determined in a 4×2 factorial design experiment in water temperatures(T) at 15℃ and 25℃,algae biomass(AB) at 0.5 g/L and 1.0 g/L,total inorganic nitrogen(TIN) at 30 μmol/L and 60 μmol/L,and soluble reactive phosphorus(SRP) at 3 and 6 μmol/L.AB and ambient TIN or SRP levels significantly affected uptake rate and removal efficiency of AN,NN,TIN,and SRP(P<0.001).G.asiatica in AB of 0.5 g/L showed higher uptake rate and lower removal efficiency relative to that with AB of 1.0 g/L.Nitrogen and phosphorus uptake rate rose with increasing ambient nutrient concentrations;nutrient removal efficiency decreased at higher environmental nutrient concentrations. The algae preferred to absorb AN to NN.Uptake rates of AN,NN,and SRP were significantly affected by temperature(P<0.001);uptake rate was higher for the 25℃ group than for the 15℃ group at the initial experiment stage.Only the removal efficiency of AN and SRP showed a significant difference between the two temperature groups(P<0.01).The four factors had significant interactive effects on absorption of N and P,implying that G.asiatica has great bioremedial potential in sea cucumber culture ponds.  相似文献

We measured the organic content and sinking velocities of biodeposits from two scallop species (Chlamys farreri, Patinopecten yessoensis) and abalone (Haliotis discus hannai) that were cultured on suspended long-lines. Measurements were conducted every two months from April 2010 to February 2011. The shellfish were divided into three size groups (small, middle, and big sizes). At each sample point, we assessed biodeposit organic content, average sinking velocity, the frequency distribution of sinking velocities, and the correlation between organic content and sinking velocity. The organic content of biodeposits varied significantly among months (P<0.05) and the pattern of change varied among species. Sinking velocities varied significantly, ranging from <0.5 cm/s to >1.9 cm/s. The sinking velocities of biodeposits from C. farreri and P. yessoensis were 0.5–1.5 cm/s and from H. discus hannai were <0.7 cm/s. The organic content was significantly negatively correlated to the sinking velocity of biodeposits in C. farreri (P<0.001) and P. yessoensis (P<0.05).  相似文献

Two hundred and thirty-five samples collected from Weinan loess section (L2 – S0) have been analyzed by TOC (total organic carbon) and pyrolysis hydrocarbons. The curve of TOC with depth has been set up and compared well with other climatic markers. The main periods of TOC variation of the last 0.14 Ma are 67.08 ka and 18.29 ka, which is near with the periods of the variation of sunshine, showing that the TOC variation may be controlled by the variation of 65°N sun-shine. The distribution of pyrolysis hydrocarbon and its parameters shows that there are two kinds of hydrocarbon associated with climatic change. One is called hydrocarbon pyrolysted directly from organism (pHC2–1) which is in the range of T max350°C – 500°C, and associated with atmospheric water. The other is called carbonate inclusion hydrocarbon (pHC2–2) which is in the range of T max 500°C – 600°C and formed by evaporation and associated with dry climate. The formula calculating humidity has been deduced Og=pHC2–1/(pHC2–1 + pHC2–2). The range of humidity of the section is 47% – 77% and the reducing order is S1–3, S1–2, S0, S1–1, S1–4, S1–1, L1–1, L1–3, and L1–5. And the trend of the humidity is still increasing today.  相似文献
