Phaeodactylum tricornutum is a potential livestock for the combined production of eicosapentaenoic acid (EPA) and fucoxanthin. In this study, six marine diatom strains identified as P. tricornutum were cultured and their total lipid, fatty acid composition and major photosynthetic pigments determined. It was found that the cell dry weight concentration and mean growth rate ranged between 0.24–0.36 g/L and 0.31–0.33/d, respectively. Among the strains, SCSIO771 presented the highest total lipid content, followed by SCSIO828, and the prominent fatty acids in all strains were C16:0, C16:1, C18:1, and C20:5 (EPA). Polyunsaturated fatty acids, including C16:2, C18:2, and EPA, comprised a significant proportion of the total fatty acids. EPA was markedly high in all strains, with the highest in SCSIO828 at 25.65% of total fatty acids. Fucoxanthin was the most abundant pigment in all strains, with the highest in SCSIO828 as well, at 5.50 mg/g. The collective results suggested that strain SCSIO828 could be considered a good candidate for the concurrent production of EPA and fucoxanthin.  相似文献

Algal biotechnology has advanced greatly in the past three decades. Many microalgae are now cultivated to produce bioactive substances. Odontella aurita is a marine diatom industrially cultured in outdoor open ponds and used for human nutrition. For the first time, we have systematically investigated the effects of culture conditions in cylindrical glass columns and fiat-plate photobioreactors, including nutrients (nitrogen, phosphorus, silicon, and sulfur), light intensity and light path, on O. aurita cell growth and biochemical composition (protein, carbohydrate, β-1,3-glucan, lipids, and ash). The optimal medium for photoautotrophic cultivation of O. aurita contained 17.65 mmol/L nitrogen, 1.09 mmol/L phosphorus, 0.42 mmol/L silicon, and 24.51 mmol/L sulfur, yielding a maximum biomass production of 6.1-6.8 g/L and 6.7-7.8 g/L under low and high light, respectively. Scale-up experiments were conducted with fiat-plate photobioreactors using different light-paths, indicating that a short light path was more suitable for biomass production of O. aurita. Analyses of biochemical composition showed that protein content decreased while carbohydrate (mainly composed of 15-1,3-glucan) increased remarkably to about 50% of dry weight during the entire culture period. The highest lipid content (19.7% of dry weight) was obtained under 0.11 mmol/L silicon and high light conditions at harvest time. Fatty acid profiles revealed that 80% were Cx4, C~6, and C20, while arachidonic acid and eicosapentaenoic acid (EPA) accounted for 1.6%-5.6% and 9%-20% of total fatty acids, respectively. High biomass production and characteristic biochemical composition profiles make O. aurita a promising microalga for the production ofbioactive components, such as EPA and D-1,3-glucan.  相似文献

The inhibitory effects of methanol extracts from the tissues of three macroalgal species on the growths of three marine red tide microalgae were assessed under laboratory conditions.Extracts of Sargassum thunbergii(Mertens ex Roth) Kuntz tissue had stronger inhibitory effects than those of either Sargassum pallidum(Turner) C.Agardh or Sargassum kjellmanianum Yendo on the growths of Heterosigma akashiwo(Hada) Hada,Skeletonema costatum(Grev.) Grev,and Prorocentrum micans Ehrenberg.Methanol extracts of S.thunbergii were further divided into petroleum ether,ethyl acetate,butanol,and distilled water phases by liquid-liquid fractionation.The petroleum ether and ethyl acetate fractions had strong algicidal effects on the microalgae.Gas chromatography-mass spectrometry analyses of these two phases identified nine fatty acids,most of which were unsaturated fatty acids.In addition,pure compounds of four of the nine unsaturated fatty acids had effective concentrations below 5 mg/L.Therefore,unsaturated fatty acids are a component of the allelochemicals in S.thunbergii tissue.  相似文献

Lipid oxidation in salt-dried yellow croaker (Pseudosciaena polyactis) was evaluated during processing with commonly used analytical indices, such as the peroxide value (POV), the thiobarbituric acid reactive substances (TBARS) value, and oxidative-relative lipoxygenase (LOX) activity. Additionally, fatty acids were analyzed using gas chromatography-mass spectrometry. Both POV and TBARS increased significantly (P < 0.05) at the rinsing stage. POV reached its peak value of 3.63 meq O2 per kg sample at the drying stage, whereas TBARS constantly increased from 0.05 to 0.20 mg MDA per kg sample. Processing of salt-dried yellow croaker had an extremely significant (P < 0.01) effect on LOX activity. Twenty-six fatty acids were identified. Combined eicosapentaenoic acid (EPA; C20:5n3) and docosahexaenoic acid (DHA; C22:6n3) content varied between (19.20 ± 0.37) mg g?1 and (23.45 ± 1.05) mg g?1. The polyunsaturated fatty acid/saturated fatty acid (PUFA/SFA) ratio in yellow croaker was 0.73–1.10, and the n-6/n-3 PUFA ratio was approximately 0.13–0.20. The contents of most fatty acids varied significantly (P < 0.05) during the different processing stages, and these differences were caused by lipid oxidation. C18:0, C16:1n7, C19:0, and C22:6n3 showed clear changes in principle component one of a principle components analysis. These fatty acids are potential markers for evaluating lipid oxidation in fish muscle because there was a significant correlation between these markers and TBARS and LOX activity (P < 0.05) with Pearson’s coefficients > 0.931.  相似文献

In most bacteria,plants and algae,fatty acid biosynthesis is catalyzed by a group of freely dissociable proteins known as the type II fatty acid synthase(FAS II) system.In the FAS II system,enoylacyl carrier protein reductase(ENR) acts as a determinant for completing the cycles of fatty acid elongation.In this study,the cDNA sequence of ENR,designated as IgENR,was isolated from the microalga Isochrysis galbana CCMM5001.RACE(rapid amplification of cDNA ends) was used to isolate the full-length cDNA of IgENR(1 503 bp),which contains an open reading frame(ORF) of 1 044 bp and encodes a protein of 347 amino acids.The genomic DNA sequence of IgENR is interrupted by four introns.The putative amino acid sequence is homologous to the ENRs of seed plants and algae,and they contain common coenzymebinding sites and active site motifs.Under different stress conditions,real-time quantitative polymerase chain reaction(RT-qPCR) showed the expression of IgENR was upregulated by high temperature(35℃),and downregulated by depleted nitrogen(0 mol/L).To clarify the mechanism of lipids accumulating lipids,other genes involved in lipids accumulation should be studied.  相似文献

The seasonal variations in biomass, abundance, and species composition of plankton in relation to hydrography were studied in the saline Bange Lake, northern Tibet, China. Sampling was carried out between one to three times per month from May 2001 to July 2002. Salinity ranged from 14 to 146. The air and water temperature exhibited a clear seasonal pattern, and mean annual temperatures were approximately 4.8°C and 7.3°C, respectively. The lowest water temperature occurred in winter from December to March at-2°C and the highest in June and July at 17.7°C. Forty-one phytoplankton taxa, 21 zooplankton, and 5 benthic or facultative zooplankton were identifi ed. The predominant phytoplankton species were Gloeothece linearis, Oscillatoria tenuis, Gloeocapsa punctata, Ctenocladus circinnatus, Dunaliella salina, and Spirulina major. The predominant zooplankton species included H olophrya actra, Brachionus plicatilis, Daphniopsis tibetana, Cletocamptus dertersi, and A rctodiaptomus salinus. The mean annual total phytoplankton density and biomass for the entire lake were 4.52×10~7 cells/L and 1.60 mg/L, respectively. The annual mean zooplankton abundance was 52, 162, 322, and 57, 144 ind./L, in the three sublakes. The annual mean total zooplankton biomass in Lakes 1–3 was 1.23, 9.98, and 2.13 mg/L, respectively. The annual mean tychoplankton abundances in Bg1, 2, and 3 were 47, 67, and 654 ind./L. The annual mean tychoplankton biomass was 2.36, 0.16, and 2.03 mg/L, respectively. The zooplankton biomass(including tychoplankton) in the lake was 9.11 mg/L. The total number of plankton species in the salt lake was signifi cantly negatively correlated with salinity.  相似文献

Gracilaria asiatica,being highly efficient in nutrient absorption,is cultivated in sea cucumber ponds to remove nutrients such as nitrogen and phosphate.It was cultured in a laboratory simulating field conditions,and its nutrient absorption was measured to evaluate effects of environmental conditions.Ammonia nitrogen(AN),nitrate nitrogen(NN),total inorganic nitrogen(TIN),and soluble reactive phosphorus(SRP) uptake rate and removal efficiency were determined in a 4×2 factorial design experiment in water temperatures(T) at 15℃ and 25℃,algae biomass(AB) at 0.5 g/L and 1.0 g/L,total inorganic nitrogen(TIN) at 30 μmol/L and 60 μmol/L,and soluble reactive phosphorus(SRP) at 3 and 6 μmol/L.AB and ambient TIN or SRP levels significantly affected uptake rate and removal efficiency of AN,NN,TIN,and SRP(P<0.001).G.asiatica in AB of 0.5 g/L showed higher uptake rate and lower removal efficiency relative to that with AB of 1.0 g/L.Nitrogen and phosphorus uptake rate rose with increasing ambient nutrient concentrations;nutrient removal efficiency decreased at higher environmental nutrient concentrations.The algae preferred to absorb AN to NN.Uptake rates of AN,NN,and SRP were significantly affected by temperature(P<0.001);uptake rate was higher for the 25℃ group than for the 15℃ group at the initial experiment stage.Only the removal efficiency of AN and SRP showed a significant difference between the two temperature groups(P<0.01).The four factors had significant interactive effects on absorption of N and P,implying that G.asiatica has great bioremedial potential in sea cucumber culture ponds.  相似文献

Changes in the fungal and bacterial biomass and community structure in litter after the volcanic eruptions of Mount Usu, northern Japan were investigated using a chronosequence approach, which is widely used for analyzing vegetation succession. The vegetation changed from bare ground(10 years after the eruptions) with little plant cover and poor soil to monotonic grassland dominated by Polygonum sachalinense with undeveloped soil(33 years) and then to deciduous broad-leaved forest dominated by Populus maximowiczii with diverse species composition and well-developed soil(100 years). At three chronosequential sites, we evaluated the compositions of phospholipid fatty acids(PLFAs), carbon(C) and nitrogen(N) contents and the isotope ratios of C(δ13C) and N(δ15N) in the litter of two dominant species, Polygonum sachalinense and Populus maximowiczii. The C/N ratio, δ13C and δ15N in the litter of these two species were higher in the forest than that in the bare ground and grassland. The PLFAs gradually increased from the bare ground to the forest, showing that microbial biomass increased with the development of the soil and/or vegetation. The fungi-to-bacteria ratio of PLFA was constant at 5.3 ± 1.4 in all three sites, suggesting that fungi were predominant. A canonical correspondence analysis suggested that the PLFA composition was related tothe successional ages and the developing soil properties(P 0.05, ANOSIM). The chronosequential analysis effectively detected the successional changes in both microbial and plant communities.  相似文献

Nitrogen deficiency is an effective strategy for enhancing lipid production in microalgae. Close relationships exist among lipid production, microalgal species, and nitrogen sources. We report growth, lipid accumulation, and fatty acid composition in four microalgae (Chloroeoccum ellipsoideum UTEX972, Chlorococcum nivale LB2225, Chlorococcum tatrense UTEX2227, and Scenedesmus deserticola JNU19) under nitrate- and urea-nitrogen deficiencies. We found three patterns of response to nitrogen deficiency: Type-A (decrease in biomass and increase in lipid content), Type-B (reduction in both biomass and lipid content), and Type-C (enhancement of both biomass and lipid content). Type-C microalgae are potential candidates for large-scale oil production. Chlorococcum ellipsoideum, for example, exhibited a neutral lipid production of up to 239.6 mg/(L'd) under urea-nitrogen deficiency. In addition, nitrogen deficiency showed only a slight influence on lipid fractions and fatty acid composition. Our study provides useful information for further screening hyper-lipid microalgal strains for biofuel production.  相似文献

In this study, the proximate composition and the amino and fatty acid profi les of shrimp Litopenaeus vannamei(prey) and wild and cultured cuttlefi sh Sepia pharaonis(the latter fed the prey) were determined and compared with FAO/WHO recommendations. The resulting scores for isoleucine, phenylalanine+tyrosine, histidine, lysine, threonine, and tryptophan in cultured cuttlefi sh were ≥150. The ratio of EAA(essential amino acids)/nonessential amino acids in cultured cuttlefi sh(0.82) was higher than in the wild form(0.80). All EAA amino acid scores for cultured cuttlefi sh were higher than their wild counterparts, except for histidine and tryptophan. Both groups of cuttlefi sh possessed similar saturated fatty acid content, with the cultured containing much more total(Σ) monounsaturated fatty acids, Σ n-6 polyunsaturated fatty acids(PUFA), and eicosapentaenoic acid(C20:5 n-3) but less Σ n-3 PUFA, arachidonic acid(C20:4 n-6), and docosahexaenoic acid(C22:6 n-3) than their wild counterparts. Therefore, the present results suggest that these cultured cuttlefi sh were better than the wild form for human health. Notably, these results also indicate that the nutritional composition of these cuttlefi sh might have been signifi cantly affected by diet.  相似文献

Photo-protective functions were investigated in phytoplankton assemblages at Kongsfjorden, Svalbard in spring, using their UV-absorbing compounds (mycosporine-like amino acids (MAAs)), xanthophyll pigments (diadinoxanthin (DD) and diatoxanthin (DT)) and < beta >- dimethylsulphoniopropionate (< beta >-DMSP). The dominant phytoplankton species in the inner bay were dominated by Phaeocystis spp. and nanoflagellates, while the offshore waters were dominated by Thalassiosira spp. In the inner bay, UVabsorbing compounds and xanthophyll pigments exhibited higher ratios of MAA to chlorophyll a (MAA:chl a ratio), and both DD and DT to chlorophyll a (DD:chl a ratio and DT:chl a ratio), respectively. Thus, the photoprotective-pigments such as DD and DT appear to complement MAAs in the natural phytoplankton assemblage. However, the ratio of < beta >-DMSP to chlorophyll a (< beta >-DMSP:chl a ratio) did not show a distinct spatial distribution according to environmental factors or interspecies differences. In this study, we found that photoprotective compounds occurred in a manner dependent on the phytoplankton species composition in Kongsfjorden Bay, where Phaeocystis is the dominant species.  相似文献

Nannochloropsis oculata CS179, a unicellular marine microalga, is rich in long-chain polyunsaturated fatty acids (LCPUFAs). Elongase and desaturase play a key role in the biosynthesis of PUFAs. A new elongase gene, which encodes 322 amino acids, was identified via RT-PCR and 5′ and 3′ RACE. The sequence of the elongase gene was blast-searched in the NCBI GenBank and showed a similarity to those of the cryptosporidium. But the NJ-tree revealed that the N. oculata CS179 elongase clustered with those of the microalgae Phaeodactylum tricornutum, Ostreococcus tauri and Thalassiosira pseudonana.  相似文献

Illuminating conditions are crucial factors affecting the microalgal growth and biosynthesis.The effects of illuminating spectral quality on the growth and bio-component production of Nannochloris oculata were investigated.The results indicated that a high proportion of Red-light enhanced the pigments and carbohydrate production but reduced those of the biomass and lipid.Mono-chromatic Blue-light has advantageous effects on lipid production compared with Red-and White-light.The optimal light spectrum for the protein production was the combination of Red-and Blue-light at a ratio of 4:3 or 5:2.Among the seven fatty acids identified from N.oculata,the contents of C16:0,C18:0,and C18:3(n-3)in the lipid were inhibited by the increased proportion of Red-light while those of C18:2(n-9),C16:2(n-6),and C20:0 were inhibited by Blue-light.Monochromatic Red-and Blue-light and their com-binations were proposed as a promising illuminating strategy for the large-scale cultivation aiming for various bio-components.  相似文献

Shoot density, standing crop (above- and below-ground biomass) and habitat of salt marsh grass Porteresia coarctata were investigated along the coast of Bakkhali estuary, Cox’s Bazar, Bangladesh from January to December 2006. Shoot density of P. coarctata was influenced by season and was found to be higher (>2 500 shoots/m 2 ) in post-monsoon and minimal in monsoon season; plants were particularly active in vegetative propagation during pre-monsoon. Above-ground biomass was greater along the protected coast compared with the exposed one in this estuary. Below-ground biomass was higher (7.75-269.53 g DW/m 2 ) than that above ground (2.20-114.75 g DW/m 2 ). Standing crops of P. coarctata showed a negative relationship (R=-0.77; P<0.05) with sedimentation rate, while seasonal activity influenced sedimentation. The recorded sedimentation rate was lower (6.09 mg/(cm 2 ·d)) in pre-monsoon and highest (14.55 mg/(cm 2 ·d)) in monsoon season. The mean value of pore water salinity was higher (34.25±5.05) during post-monsoon and lowest (18.0±3.71) in monsoon season. The soil was sandy clay in this P. coarctata bed; it consisted of 86% sand, 13% clay and 1% silt. Soil organic matter dropped during the monsoon season (0.78%-0.67%) and was highest ((2.17±1.42)%-(2.3±1.47)%) during post-monsoon, probably owing to accumulation of decomposed peat on the marsh surface. The mean pore water NH 4 -N concentration ranged from 2.44±1.65 to 3.33±1.82 μg/L, with a minimum air temperature of 22.09°C in post-monsoon and a maximum of 31.16°C in pre-monsoon. Variations of physico-chemical parameters in the soil, water, and climate governed biological parameters of P. coarctata in the Bakkhali estuary, and were comparable with estuarine environments elsewhere.  相似文献

Soil microorganisms are sensitive indicator of soil health and quality. Understanding the effects of vegetation biomass and seasonal change on soil microorganisms is vital to evaluate the soil quality and implement vegetation restoration. This study analyzed the soil phospholipid fatty acids (PLFAs) in fresh and withered Kudzu (Pueraria montana var. lobata) vegetation conditions in different seasons. The results showed that vegetation biomass and seasonal change significantly affected microbial biomass and its community structure. Both fresh and withered Kudzu cover significantly increased soil microbial biomass, and the growth effect of microbes in the soil with fresh Kudzu cover was more obvious than that with withered Kudzu cover. Compared with the dry season, the rainy season significantly increased the microbial biomass and the B/F (the ratio of bacterial to fungal PLFAs) ratio but dramatically reduced the G+/G- (the ratio of gram-positive to gram-negative bacteria PLFAs). Kudzu cover and seasonal change had a significant effect on microbial structure in soil covered by higher vegetation biomass. Furthermore, soil temperature and moisture had different correlations with specific microbial biomass in the two seasons. Our findings highlight the effect of Kudzu vine cover on the soil microenvironment and soil microhabitat, enhancing the soil quality in the Dry-hot Valley of Jinsha River, Southwest China.  相似文献

The shell color of Pacific oyster (Crassostrea gigas) is a desirable trait; but the nutritional studies on C. gigas with different shell colors have not been conducted. Through successive selective breeding, five shell color strains of black (B), purple (P), orange (O), golden (G) and white (W) C. gigas have been developed. The aim of this study was to evaluate the chemical composition and nutritional value of five shell color strains and one commercial population with a common color. The biochemical composition including moisture, total protein, glycogen, ash, total fat, fatty acids (FA), amino acids and minerals was detected. The results indicated that the protein (50.76%–56.57%) was the major component. The content of glycogen showed a significant difference between orange shell and golden shell strains, as well as between commercial population and golden shell strain. In addition, all shell color strains contained a large amount of essential amino acids (12.20–14.15 g (100 g)?1), of them leucine (2.81–3.29 g (100 g)?1) and lysine (2.79–3.28 g (100 g)?1) were predominant. The oysters were rich in polyunsaturated fatty acids (42.26%–45.24% of total fatty acid) with high levels of DHA (18.53%–21.16% of total fatty acid) and EPA (17.23%–18.68% of total fatty acid). Significant differences of mineral contents (Mg, Zn, Fe and Cu) were identified among the six populations. These results indicated that C. gigas with different shell colors presented rich nutritional value with high protein, glycogen, essential amino acids and polyunsaturated fatty acids. The biochemical composition obtained in this study is useful for selective breeding of C. gigas with different shell colors.  相似文献

Trophic interaction among various biomass groups in a swimming crab Portunus trituberculatus polyculture pond was investigated using carbon and nitrogen stable isotope analysis. The polycultured animal species also included white shrimp Litopenaeus vannamei, short-necked clam Ruditapes philippinarum, and redlip mullet Liza haematochila. The mean δ13C value for all the biomass groups in polyculture ecosystem ranged from ?25.61‰ to ?16.60‰, and the mean δ15N value ranged from 6.80‰ to 13.09‰. Significant difference in the δ13C value was found between particulate organic matter (POM) and sediment organic matter (SOM) (P < 0.05), indicating that these two organic matter pools have different material sources. Assuming that a 13C-enrichment factor of 1.00‰ and a 15N-enrichment factor of 2.70‰ existed between consumer and prey, diets of the four cultured animals were estimated using a stable isotope mixing model. The estimated model results indicated that P. trituberculatus mainly feed on Aloidis laevis; L. vannamei mainly feed on shrimp feed; while A. laevis, R. philippinarum and L. haematochelia mainly feed on POM. Shrimp feed was also an important food source of R. philippinarum and L. haematochelia. The diets of P. trituberculatus, L. vannamei, R. philippinarum, and L. haematochila showed complementary effects in this polyculture ecosystem. Our finding indicated that the polyculture of these four organisms with suitable farming density could make an effective use of most of the food sources, which can make a highly efficient polyculture ecosystem.  相似文献

The ability of cell to modulate the fluidity of plasma membrane was crucial to the survival of microorganism at low temperature. Plasma membrane proteins, fatty acids and carotenoids profiles of Antarctic psychrophilc yeast Rhodotorula sp. NJ298 were investigated at -3 ℃, 0 ℃ and 8 ℃. The results showed that plasma membrane protein content was greater at -3 ℃ than that at 8 ℃, and a unique membrane polypeptide composition with an apparent molecular mass of 94.7 kDa was newly synthesized with SDS-PAGE analysis; GC analysis showed that the main changes of fatty acids were the percentage of unsaturated fatty acids (C18∶ 1 and C18∶ 2) and shorter chain saturated fatty acid (C10∶ 0) increased along with the decrease of the culture temperature from 8 ℃ to -3 ℃; HPLC analysis indicated that astaxanthin was the major functional carotenoids of the plasma membrane, percentage of which increased from 54.6±1.5% at 8 ℃ to 81.9±2.1% at -3 ℃. However the fluidity of plasma membrane which was determined by measuring fluorescence anisotropy was similar at -3 ℃, 0 ℃ and 8 ℃. Hence these changes in plasma membrane's characteristics were involved in the cellular cold-adaptation by which NJ298 could maintain normal plasma membrane fluidity at near-freezing temperature.  相似文献

Colony morphology is important for Microcystis to sustain a competitive advantage in eutrophic lakes. The mechanism of colony formation in Microcystis is currently unclear. Extracellular polysaccharide (EPS) has been reported to play an important role in cell aggregate formation of some phytoplankton. Microcystis aeruginosa was cultivated under varied abiotic conditions, including different nutrient, light, and temperature conditions, to investigate their effects on EPS production and morphological change. The results show that nutrient concentration and light intensity have great effects on EPS production in M. aeruginosa. There was a considerable increase in EPS production after M. aeruginosa was cultivated in adjusted culture conditions similar to those present in the field (28.9mg C/L, 1.98mg N/L, 0.65mg P/L, light intensity:100 μmol/(m2s)). These results indicate that abiotic factors might be one of the triggers for colony formation in Microcystis.  相似文献
