Copper isotopes may prove to be a useful tool for investigating bacteria-metal interactions recorded in natural waters, soils, and rocks. However, experimental data which attempt to constrain Cu isotope fractionation in biologic systems are limited and unclear. In this study, we utilized Cu isotopes (δ65Cu) to investigate Cu-bacteria interactions, including surface adsorption and intracellular incorporation. Experiments were conducted with individual representative species of Gram-positive (Bacillus subtilis) and Gram-negative (Escherichia coli) bacteria, as well as with wild-type consortia of microorganisms from several natural environments. Ph-dependent adsorption experiments were conducted with live and dead cells over the pH range 2.5-6. Surface adsorption experiments of Cu onto live bacterial cells resulted in apparent separation factors (Δ65Cusolution-solid = δ65Cusolution − δ65Cusolid) ranging from +0.3‰ to +1.4‰ for B. subtilis and +0.2‰ to +2.6‰ for E. coli. However, because heat-killed bacterial cells did not exhibit this behavior, the preference of the lighter Cu isotope by the cells is probably not related to reversible surface adsorption, but instead is a metabolically-driven phenomenon. Adsorption experiments with heat-killed cells yielded apparent separation factors ranging from +0.3‰ to −0.69‰ which likely reflects fractionation from complexation with organic acid surface functional group sites. For intracellular incorporation experiments the lab strains and natural consortia preferentially incorporated the lighter Cu isotope with an apparent Δ65Cusolution-solid ranging from ∼+1.0‰ to +4.4‰. Our results indicate that live bacterial cells preferentially sequester the lighter Cu isotope regardless of the experimental conditions. The fractionation mechanisms involved are likely related to active cellular transport and regulation, including the reduction of Cu(II) to Cu(I). Because similar intracellular Cu machinery is shared by fungi, plants, and higher organisms, the influence of biological processes on the δ65Cu of natural waters and soils is probably considerable.

Zinc and Cu play important roles in the biogeochemistry of natural systems, and it is likely that these interactions result in mass-dependent fractionations of their stable isotopes. In this study, we examine the relative abundances of dissolved Zn and Cu isotopes in a variety of stream waters draining six historical mining districts located in the United States and Europe. Our goals were to (1) determine whether streams from different geologic settings have unique or similar Zn and Cu isotopic signatures and (2) to determine whether Zn and Cu isotopic signatures change in response to changes in dissolved metal concentrations over well-defined diel (24-h) cycles.Average δ66Zn and δ65Cu values for streams varied from +0.02‰ to +0.46‰ and −0.7‰ to +1.4‰, respectively, demonstrating that Zn and Cu isotopes are heterogeneous among the measured streams. Zinc or Cu isotopic changes were not detected within the resolution of our measurements over diel cycles for most streams. However, diel changes in Zn isotopes were recorded in one stream where the fluctuations of dissolved Zn were the largest. We calculate an apparent separation factor of ∼0.3‰ (66/64Zn) between the dissolved and solid Zn reservoirs in this stream with the solid taking up the lighter Zn isotope. The preference of the lighter isotope in the solid reservoir may reflect metabolic uptake of Zn by microorganisms. Additional field investigations must evaluate the contributions of soils, rocks, minerals, and anthropogenic components to Cu and Zn isotopic fluxes in natural waters. Moreover, rigorous experimental work is necessary to quantify fractionation factors for the biogeochemical reactions that are likely to impact Cu and Zn isotopes in hydrologic systems. This initial investigation of Cu and Zn isotopes in stream waters suggests that these isotopes may be powerful tools for probing biogeochemical processes in surface waters on a variety of temporal and spatial scales.

With the improvement of analytical methods and the development of multiple-collector inductively coupled plasma-mass spectrometry (MC-ICP/MS), research on non-traditional stable isotope (Cu, Zn, Fe, Se, Mo, Cr, Hg) in geochemistry has made tremendous progress in the past decade. Recent studies have demonstrated that both organic and inorganic reactions may cause Hg isotope fractionation, and variations of Hg isotopic composition in the environment have been successfully employed to explain Hg pollution history, Hg sources and tracking Hg pathways in nature. Furthermore, Hg isotopic fractionation studies can be a powerful tool in the calibration of global Hg cycling models. Stable isotope geochemistry of Hg is therefore becoming a new frontier subject in earth sciences. Based on summarizing previous research, this paper outlines the main advances in the study of Hg stable isotopes with particular emphasis placed on a brief explanation of Hg isotope analytical techniques, possible Hg isotope fractionation mechanisms observed in both natural and experimental processes, Hg isotope composition variations in different environmental matrices, and the application prospects of the Hg stable isotopes in environmental geosciences.

We have determined the extent of Se isotope fractionation induced by reduction of selenate by sulfate interlayered green rust (GRSO4), a Fe(II)-Fe(III) hydroxide-sulfate. This compound is known to reduce selenate to Se(0), and it is the only naturally relevant abiotic selenate reduction pathway documented to date. Se reduction reactions, when they occur in nature, greatly reduce Se mobility and bioavailability. Se stable isotope analysis shows promise as an indicator of Se reduction, and Se isotope fractionation by various Se reactions must be known in order to refine this tool. We measured the increase in the 80Se/76Se ratio of dissolved selenate as lighter isotopes were preferentially consumed during reduction by GRSO4. Six different experiments that used GRSO4 made by two methods, with varying solution compositions and pH, yielded identical isotopic fractionations. Regression of all the data yielded an instantaneous isotope fractionation of 7.36 ± 0.24‰. Selenate reduction by GRSO4 induces much greater isotopic fractionation than does bacterial selenate reduction. If selenate reduction by GRSO4 occurs in nature, it may be identifiable on the basis of its relatively large isotopic fractionation.

Transition metal stable isotope signatures can be useful for tracing both natural and anthropogenic signals in the environment, but only if the mechanisms responsible for fractionation are understood. To investigate isotope fractionations due to electrochemistry (or redox processes), we examine the stable isotope behavior of iron and zinc during the reduction reaction  + 2e = Mmetal as a function of electrochemical driving force, temperature, and time. In all cases light isotopes are preferentially electroplated, following a mass-dependent law. Generally, the extent of fractionation is larger for higher temperatures and lower driving forces, and is roughly insensitive to amount of charge delivered. The maximum fractionations are δ56/54Fe = −4.0‰ and δ66/64Zn = −5.5‰, larger than observed fractionations in the natural environment and larger than those predicted due to changes in speciation. All the observed fractionation trends are interpreted in terms of three distinct processes that occur during an electrochemical reaction: mass transport to the electrode, chemical speciation changes adjacent to the electrode, and electron transfer at the electrode. We show that a large isotope effect adjacent the electrode surface arises from the charge-transfer kinetics, but this effect is attenuated in cases where diffusion of ions to the electrode surface becomes the rate-limiting step. Thus while a general increase in fractionation is observed with increasing temperature, this appears to be a result of thermally enhanced mass transport to the reacting interface rather than an isotope effect associated with the charge-transfer kinetics. This study demonstrates that laboratory experiments can successfully distinguish isotopic signatures arising from mass transport, chemical speciation, and electron transfer. Understanding how these processes fractionate metal isotopes under laboratory conditions is the first step towards discovering what role these processes play in fractionating metal isotopes in natural systems.

Iron isotope fractionations produced during chemical and biological Fe(II) oxidation are sensitive to the proportions and nature of dissolved and solid-phase Fe species present, as well as the extent of isotopic exchange between precipitates and aqueous Fe. Iron isotopes therefore potentially constrain the mechanisms and pathways of Fe redox transformations in modern and ancient environments. In the present study, we followed in batch experiments Fe isotope fractionations between Fe(II)aq and Fe(III) oxide/hydroxide precipitates produced by the Fe(III) mineral encrusting, nitrate-reducing, Fe(II)-oxidizing Acidovorax sp. strain BoFeN1. Isotopic fractionation in 56Fe/54Fe approached that expected for equilibrium conditions, assuming an equilibrium Δ56FeFe(OH)3-Fe(II)aq fractionation factor of +3.0‰. Previous studies have shown that Fe(II) oxidation by this Acidovorax strain occurs in the periplasm, and we propose that Fe isotope equilibrium is maintained through redox cycling via coupled electron and atom exchange between Fe(II)aq and Fe(III) precipitates in the contained environment of the periplasm. In addition to the apparent equilibrium isotopic fractionation, these experiments also record the kinetic effects of initial rapid oxidation, and possible phase transformations of the Fe(III) precipitates. Attainment of Fe isotope equilibrium between Fe(III) oxide/hydroxide precipitates and Fe(II)aq by neutrophilic, Fe(II)-oxidizing bacteria or through abiologic Fe(II)aq oxidation is generally not expected or observed, because the poor solubility of their metabolic product, i.e. Fe(III), usually leads to rapid precipitation of Fe(III) minerals, and hence expression of a kinetic fractionation upon precipitation; in the absence of redox cycling between Fe(II)aq and precipitate, kinetic isotope fractionations are likely to be retained. These results highlight the distinct Fe isotope fractionations that are produced by different pathways of biological and abiological Fe(II) oxidation.

Sulfur isotope fractionation during dissimilatory sulfate reduction has been conceptually described by the widely accepted Rees model as related to the stepwise reduction of sulfate to sulfide within the cells of bacteria. The magnitude of isotope fractionation is determined by the interplay between different reduction steps in a chain of reactions. Here we present a revision of Rees’ model for bacterial sulfate reduction that includes revised fractionation factors for the sulfite-sulfide step and incorporates new forward and reverse steps in the reduction of sulfite to sulfide, as well as exchange of sulfide between the cell and ambient water. With this model we show that in contrast to the Rees model, isotope fractionations well in excess of −46‰ are possible. Therefore, some of the large sulfur isotope fractionations observed in nature can be explained without the need of alternate pathways involving the oxidative sulfur cycle. We use this model to predict that large fractionations should occur under hypersulfidic conditions and where electron acceptor concentrations are limiting.

Variations in the stable isotope abundances of transition metals have been observed in the geologic record and trying to understand and reconstruct the physical/environmental conditions that produced these signatures is an area of active research. It is clear that changes in oxidation state lead to large fractionations of the stable isotopes of many transition metals such as iron, suggesting that transition metal stable isotope signatures could be used as a paleo-redox proxy. However, the factors contributing to these observed stable isotope variations are poorly understood. Here we investigate how the kinetics of iron redox electrochemistry generates isotope fractionation. Through a combination of electrodeposition experiments and modeling of electrochemical processes including mass-transport, we show that electron transfer reactions are the cause of a large isotope separation, while mass transport-limited supply of reactant to the electrode attenuates the observed isotopic fractionation. Furthermore, the stable isotope composition of electroplated transition metals can be tuned in the laboratory by controlling parameters such as solution chemistry, reaction overpotential, and solution convection. These methods are potentially useful for generating isotopically-marked metal surfaces for tracking and forensic purposes. In addition, our studies will help interpret stable isotope data in terms of identifying underlying electron transfer processes in laboratory and natural samples.

Sulfur isotope effects produced by microbial dissimilatory sulfate reduction are used to reconstruct the coupled cycling of carbon and sulfur through geologic time, to constrain the evolution of sulfur-based metabolisms, and to track the oxygenation of Earth’s surface. In this study, we investigate how the coupling of carbon and sulfur metabolisms in batch and continuous cultures of a recently isolated marine sulfate reducing bacterium DMSS-1, a Desulfovibrio sp., influences the fractionation of sulfur isotopes.DMSS-1 grown in batch culture on seven different electron donors (ethanol, glycerol, fructose, glucose, lactate, malate and pyruvate) fractionates 34S/32S ratio from 6‰ to 44‰, demonstrating that the fractionations by an actively growing culture of a single incomplete oxidizing sulfate reducing microbe can span almost the entire range of previously reported values in defined cultures. The magnitude of isotope effect correlates well with cell specific sulfate reduction rates (from 0.7 to 26.1 fmol/cell/day). DMSS-1 grown on lactate in continuous culture produces a larger isotope effect (21-37‰) than the lactate-grown batch culture (6‰), indicating that the isotope effect also depends on the supply rate of the electron donor and microbial growth rate. The largest isotope effect in continuous culture is accompanied by measurable changes in cell length and cellular yield that suggest starvation. The use of multiple sulfur isotopes in the model of metabolic fluxes of sulfur shows that the loss of sulfate from the cell and the intracellular reoxidation of reduced sulfur species contribute to the increase in isotope effects in a correlated manner. Isotope fractionations produced during sulfate reduction in the pure culture of DMSS-1 expand the previously reported range of triple sulfur isotope effects (32S, 33S, and 34S) by marine sulfate reducing bacteria, implying that microbial sulfur disproportionation may have a smaller 33S isotopic fingerprint than previously thought.

We present a 3-year study of concentrations and sulfur isotope values (δ34S, Δ33S, and Δ36S) of sulfur compounds in the water column of Fayetteville Green Lake (NY, USA), a stratified (meromictic) euxinic lake with moderately high sulfate concentrations (12-16 mM). We utilize our results along with numerical models (including transport within the lake) to identify and quantify the major biological and abiotic processes contributing to sulfur cycling in the system. The isotope values of sulfide and zero-valent sulfur across the redox-interface (chemocline) change seasonally in response to changes in sulfide oxidation processes. In the fall, sulfide oxidation occurs primarily via abiotic reaction with oxygen, as reflected by an increase in sulfide δ34S at the redox interface. Interestingly, S isotope values for zero-valent sulfur sampled at this time still reflect production and recycling by phototrophic S-oxidation. In the spring, sulfide S isotope values suggest an increased input from phototrophic oxidation, consistent with a more pronounced phototroph population at the chemocline. This trend is associated with smaller fractionations between sulfide and zero-valent sulfur, suggesting a metabolic rate control on fractionation similar to that for sulfate reduction. Comparison of our data with previous studies indicates that the S isotope values of sulfate and sulfide in the deep waters are remarkably stable over long periods of time, with consistently large fractionations of up to 58‰ in δ34S. Models of the δ34S and Δ33S trends in the deep waters (considering mass transport via diffusion and advection along with biological processes) require that these fractionations are a consequence of sulfur compound disproportionation at and below the redox interface in addition to large fractionations during sulfate reduction. The large fractionations during sulfate reduction appear to be a consequence of the high sulfate concentrations and the distribution of organic matter in the water column. The occurrence of disproportionation in the lake is supported by profiles of intermediate sulfur compounds and by lake microbiology, but is not evident from the δ34S trends alone. These results illustrate the utility of including minor S isotopes in sulfur isotope studies to unravel complex sulfur cycling in natural systems.

Redox processes are ubiquitous in Earth science and are often associated with large isotope fractionations. In a previous study, voltage-dependent amplification of stable isotope fractionation was observed for an Fe reduction process. Here, we describe experiments showing a similar effect for a second transition metal, zinc. After electrochemical reduction, the composition of plated Zn metal is enriched in the light isotope (64Zn) with respect to the Zn2+ leftover in solution, with a voltage-dependent fractionation factor. Results from voltage-dependent electroplating experiments are in good agreement with a second data set following equilibrium fractional isotope evolution of Zn isotopes during an electroplating process which stepwise removes most of the Zn from the aqueous reservoir. Taken together, the results indicate a voltage-dependent isotope fractionation (in permil) of 66Zn with respect to 64Zn to be equal to −3.45 to 1.71 V. The negative slope trend is in contrast with previously published results on iron isotope fractionation during electroplating which shows a positive slope. These results are interpreted using an extension of Marcus theory, which predicts isotope fractionations as a function of driving force in an electrochemical system. Taken together with observations of natural fractionation of redox-sensitive and non redox-active elements, our modified Marcus theory provides a framework for quantitatively predicting transition metal isotope geochemical signatures during environmentally relevant redox processes in terms of simple energetic parameters.

The fractionation of H isotopes between the water in the growth medium and the organically bonded H from microalgae cultured under conditions, where light intensity and wavelength, temperature, nutrient availability, and the H isotope ratio of the water were controlled, is reproducible and light dependant. All studies were based either on the H isotope ratios of the total organic H or on the lipids, where most of the H is firmly bonded to C. H bonded into other macromolecules, proteins, carbohydrates and nucleic acids, does not exchange with water, when algae are incubated in water enriched with deuterium. Only after the destruction of quaternary H bonds are labile hydrogens in macromolecules free to exchange with water. By growing algae (18 strains), including blue-green algae, green algae and diatoms, in continuous light, the isotope fractionations in photosynthesis were reproducibly ?93 to ?178 %. depending on the organism tested. This fractionation was not temperature dependent. Microalgae grown in total darkness with an organic substrate did not show the isotope fractionation seen in cells grown in light. In both light- and dark-grown algae, however, additional depletion of deuterium (?30 to ?60%.) in cellular organic matter occurs during the metabolism of carbohydrates to form lipids. Plants from several natural populations also fractionated isotopes during photosynthesis by an average of ?90 to ?110%. In addition, the organically bonded H in nonsaponifiable lipids was further fractionated by ?80%. from that in saponifiable lipids, isolated from two geographically distinct populations of marsh plants. This difference between H isotope ratios of these two groups of lipids provides an endogenous isotopic marker.

We have studied the oxygen isotope signature of inorganic phosphate (Pi) generated by hydrolysis of nucleic acid phosphodiester (P-diester) compounds by cell-free enzymes (Deoxyribonuclease 1, Phosphodiesterase 1, Alkaline phosphatase) and microbial cultures at natural isotopic abundances. We demonstrate that the diesterase-catalyzed hydrolytic step leads to incorporation of at least one water O into released Pi for a total of two O atoms from water incorporated into Pi released from P-diesters. In the presence of Phosphodiesterase 1, 16O is preferentially incorporated into nucleotides released from DNA; whereas 18O is preferentially incorporated into nucleotides released from RNA. A strong consistency between predicted O-isotope regeneration signatures based on results of cell-free enzyme experiments and measured isotopic signatures from independent experiments with E. coli cultures was observed and confirms proposed models for phosphoester hydrolysis. Results from these studies made at natural 18O abundance levels provide a new tool, enzyme-specific O-isotope fractionation, for investigations of organophosphate metabolism and phosphorus cycling pathways in natural aquatic systems.

This study presents lithium (Li) and magnesium (Mg) isotope data from experiments designed to assess the effects of dissolution of primary phases and the formation of secondary minerals during the weathering of basalt. Basalt glass and olivine dissolution experiments were performed in mixed through-flow reactors under controlled equilibrium conditions, at low pH (2-4) in order to keep solutions undersaturated (i.e. far-from equilibrium) and inhibit the formation of secondary minerals. Combined dissolution-precipitation experiments were performed at high pH (10 and 11) increasing the saturation state of the solutions (moving the system closer to equilibrium) and thereby promoting the formation of secondary minerals. At conditions far from equilibrium saturation state modelling and solution stoichiometry suggest that little secondary mineral formation has occurred. This is supported by the similarity of the dissolution rates of basalt glass and olivine obtained here compared to those of previous experiments. The δ7Li isotope composition of the experimental solution is indistinguishable from that of the initial basalt glass or olivine indicating that little fractionation has occurred. In contrast, the same experimental solutions have light Mg isotope compositions relative to the primary phases, and the solution becomes progressively lighter with time. In the absence of any evidence for secondary mineral formation the most likely explanation for these light Mg isotope compositions is that there has been preferential loss of light Mg during primary phase dissolution. For the experiments undertaken at close to equilibrium conditions the results of saturation state modelling and changes in solution chemistry suggest that secondary mineral formation has occurred. X-ray diffraction (XRD) measurements of the reacted mineral products from these experiments confirm that the principal secondary phase that has formed is chrysotile. Lithium isotope ratios of the experimental fluid become increasingly heavy with time, consistent with previous experimental work and natural data indicating that 6Li is preferentially incorporated into secondary minerals, leaving the solution enriched in 7Li. The behaviour of Mg isotopes is different from that anticipated or observed in natural systems. Similar to the far from equilibrium experiments initially light Mg is lost during olivine dissolution, but with time the δ26Mg value of the solution becomes increasingly heavy. This suggests either preferential loss of light, and then heavy Mg from olivine, or that the secondary phase preferentially incorporates light Mg from solution. Assuming that the secondary phase is chrysotile, a Mg-silicate, the sense of Mg fractionation is opposite to that previously associated with silicate soils and implies that the fractionation of Mg isotopes during silicate precipitation may be mineral specific. If secondary silicates do preferentially remove light Mg from solution then this could be a possible mechanism for the relatively heavy δ26Mg value of seawater. This study highlights the utility of experimental studies to quantify the effects of natural weathering reactions on the Li and Mg geochemical cycles.

In recent years, a series of important progresses have been made in the aspect of magnesium isotopes behavior in weathering processes. These progresses are not only favorable to understand the change of the magnesium isotopic compositions in rivers, but also establish the foundation to further reveal the magnesium isotope geochemical cycle. The magnesium in rivers is both magnesium sink for weathering and magnesium source for the ocean. The Mg isotopic compositions in rivers are dominated by the magnesium sources and Mg isotope fractionations processes. The sources of magnesium in rivers originate mainly from draining rocks, as well as less contribution from the eolian deposition, groundwater, plant debris, and precipitation. The Mg isotope fractionations in rivers are mainly related to precipitation and dissolution of carbonate minerals, silicate mineral hydrolysis, adsorption on mineral or colloidal matter surface, and plant uptake. Generally, the contribution of carbonate minerals dissolution or precipitation is equal to add or reduce magnesium from carbonate endmember, which has a remarkably negative δ26Mg value. Based on the fact that most clay minerals are rich in 26Mg during nature silicate mineral hydrolysis, then it is possible to infer that residual weathering products enrich in 26Mg. However, there is no significant Mg isotope fractionation causing by the adsorption on mineral or colloidal matter surface during river water migration. For the plant uptake, the root prefers to have 26Mg, leading the plant itself rich in heavier Mg isotopic composition. In addition, formation of secondary minerals in rivers could also reflect the changes of chemical parameters in rivers (such as major elements, CO2 solubility, pH, etc.). Hence, Mg isotopic composition in rivers and associated isotope fractionations are not only the basis for the application of magnesium isotope to trace surface material cycle, but also have important significance for the further understanding the geochemical cycle of magnesium isotopes.

Stable iron isotope ratios in three soils (two Podzols and one Cambisol) were measured by MC-ICPMS to investigate iron isotope fractionation during pedogenic iron transformation and translocation processes under oxic conditions. Podzolization is a soil forming process in which iron oxides are dissolved and iron is translocated and enriched in the subsoil under the influence of organic ligands. The Cambisol was studied for comparison, representing a soil formed by chemical weathering without significant translocation of iron. A three-step sequential extraction procedure was used to separate operationally-defined iron mineral pools (i.e., poorly-crystalline iron oxides, crystalline iron oxides, silicate-bound iron) from the soil samples. Iron isotope ratios of total soil digests were compared with those of the separated iron mineral pools. Mass balance calculations demonstrated excellent agreement between results of sequential extractions and total soil digestions. Systematic variations in the iron isotope signature were found in the Podzol profiles. An enrichment of light iron isotopes of about 0.6‰ in δ57Fe was found in total soil digests of the illuvial Bh horizons which can be explained by preferential translocation of light iron isotopes. The separated iron mineral pools revealed a wide range of δ57Fe values spanning more than 3‰ in the Podzol profiles. Strong enrichments of heavy iron isotopes in silicate-bound iron constituting the residue of weathering processes, indicated the preferential transformation of light iron isotopes during weathering. Iron isotope fractionation during podzolization is probably linked to the ligand-controlled iron translocation processes. Comparison of iron isotope data from eluvial and illuvial horizons of the Podzol profiles revealed that some iron must have been leached out of the profile. However, uncertainties in the initial iron content and iron isotopic composition of the parent materials prevented thorough mass balance calculations of iron fluxes within the profiles. In contrast to the Podzol profiles, the Cambisol profile displayed uniform δ57Fe values across soil depth and showed only a small enrichment of light iron isotopes of about 0.4‰ in the poorly-crystalline iron oxide pool extracted by 0.5 M HCl. This work demonstrates that significant iron isotope fractionations can occur during pedogenesis in oxic environments under the influence of organic ligands. Our findings provide new insights into fractionation mechanisms of iron isotopes and will help in the development of stable iron isotopes as tracers for biogeochemical iron cycling in nature.
