Immunohistochemical localization of enzymes is compared directly with localization of enzyme activity with (catalytic) enzyme histochemical methods. The two approaches demonstrate principally different aspects of an enzyme. The immunohistochemical method localizes the enzyme protein whether it is active or not whereas the catalytic method visualizes the functionally active enzyme only. Furthermore, enzyme histochemical methods appear to be highly sensitive in comparison with immunohistochemical methods. Finally, the catalytic methods are simple and last 5-60 min at room temp, whereas immunohistochemical methods take hours at least. It can be concluded that enzyme histochemical methods are the methods of choice to localize enzymes because they detect the functional activity of an enzyme, they are relatively simple and their sensitivity is high. 相似文献

Routine determination of dissolved organic nitrogen (DON) is performed in numerous laboratories around the world using one of three families of methods: UV oxidation (UV), persulfate oxidation (PO), or high temperature combustion (HTC). Essentially all routine methods measure total dissolved nitrogen (TDN) and calculate DON by subtracting the dissolved inorganic nitrogen (DIN). While there is currently no strong suggestion that any of these methods is inadequate, there are continuing suspicions of slight inaccuracy by UV methods.This is a report of a broad community methods comparison where 29 sets (7 UV, 13 PO, and 9 HTC) of TDN analyses were performed on five samples with varying TDN and DIN concentrations. Analyses were done in a “blind” procedure with results sent to the first author. With editing out one set of extreme outliers (representing 5 out of 145 ampoules analyzed), the community comparability for analyzing the TDN samples was in the 8–28% range (coefficient of variation representing one standard deviation for the five individual samples by 28 analyses). When DIN concentrations were subtracted uniformly (single DIN value for each sample), the comparability was obviously worse (19–46% cv). This comparison represents a larger and more diverse set of analyses, but the overall comparability is only marginally better than that of the Seattle workshop of a decade ago. Grouping methods, little difference was seen other than inconclusive evidence that the UV methods gave TDN values for several of the samples higher than HTC methods. Since there was much scatter for each of the groups of methods and for all analyses when grouped, it is thought that more uniformity in procedures is probably needed. An important unplanned observation is that variability in DIN analyses (used in determining the final analyte in most UV and PO methods) is essentially as large as the variability in the TDN analyses.This exercise should not be viewed as a qualification exercise for the analysts, but should instead be considered a broad preliminary test of the comparison of the families of methods being used in various laboratories around the world. Based on many independent analyses here, none of the routinely used methods appears to be grossly inaccurate, thus, most routine TDN analyses being reported in the literature are apparently accurate. However, it is not reassuring that the ability of the international community to determine DON in deep oceanic waters continues to be poor. It is suggested that as an outgrowth of this paper, analysts using UV and PO methods experiment and look more carefully at the completeness of DIN conversion to the final analyte and also at the accuracy of their analysis of the final analyte. HTC methods appear to be relatively easy and convenient and have potential for routine adoption. 粒度参数的计算方法主要有图解法和矩值法两种,为了比较不同方法计算结果的差异,利用新近在莱州湾南岸获取的LZ908孔岩心,在粒度分析的基础上,综合运用相关分析、回归分析、聚类分析等统计方法,从参数计算过程、沉积物粒度分布形态(多峰态粒度特征)等方面讨论了两种参数估计方法对计算结果的可能影响及其影响因素。结果表明:平均粒径、标准偏差相互替代的程度高,而偏度、峰度相互替代的程度低,或者不可替代;两者的差异是由各自本身的计算过程引起的,是沉积物粒度分布特征的一种反映,并且与沉积水动力条件密切相关。 相似文献

设计2种不同的固定方法,比较2种固定方法在刺参体腔细胞的扫描电镜制样和透射电镜制样中固定效果的优劣.选择能使体腔细胞充分分散的固定方法用于扫描电镜制样;而将能使体腔细胞凝聚成团的固定方法用于透射电镜制样.获得了令人满意的体腔细胞扫描和透射电镜照片.以多聚赖氨酸法、明胶法和聚乙烯醇缩甲醛法收集、黏附固定后的体腔细胞,实验结果显示:3种方法均可起到黏附体腔细胞的作用,通过探讨玻片黏附体腔细胞的有效方法,建立适用于海产无脊椎动物体腔细胞、血细胞、生殖细胞和原生动物、细菌等样品的扫描电镜制样方法. 相似文献

Risk analysis and assessment relating coastal structures has been one of the hot topics in the area of coastal protection recently. In this paper, from three aspects of joint return period of multiple loads, dike failure rate and dike continuous risk prevention respectively, three new risk analysis methods concerning overtopping of sea dikes are developed. It is worth noting that the factors of storm surge which leads to overtopping are also considered in the three methods. In order to verify and estimate the effectiveness and reliability of the newly developed methods, quantified mutual information is adopted. By means of case testing, it can be found that different prior variables might be selected dividedly, according to the requirement of special engineering application or the dominance of loads. Based on the selection of prior variables, the correlating risk analysis method can be successfully applied to practical engineering. 相似文献

An issue of concern for matched-field processing is the strong dependence between performance and precise knowledge about the environmental parameters. A robust matched-field processor based on minimax robust filtering methods was developed. Here, simulation methods are employed to evaluate the performance of the minimax robust method as well as other robust methods for a range-independent shallow water environment. The performance of the robust methods is compared with that of the nominal processor, that is, the processor based on a single set of environmental parameters thought to be closest to the actual. The matched-field processing performance is evaluated in terms of the peak-to-sidelobe ratio. The simulation results indicate that the robust methods provide significant performance improvements over the nominal processor in the presence of uncertainty in water column sound speed, channel depth, and sound speed in the bottom 相似文献

Environmental contours are often used in design of marine structures to identify extreme environmental conditions that may give rise to extreme loads and responses. Recently, attention has been given to the fact that different methods exist for establishing such contours, and that in some cases significant differences may be obtained from the various methods.In this study, another aspect of the uncertainty related to the calculation of environmental contours is addressed, namely the uncertainty due to sampling variability when environmental contours are constructed based on metocean data of finite sample size. The uncertainty of environmental contours for the joint distribution of significant wave height and wave period for different sample sizes (10, 25 and 100 years of data) are investigated considering different underlying datasets and for different estimation methods for the joint distribution. Both cases where samples are drawn from a known joint distribution of wave height and periods and cases where samples are drawn from a real hindcast dataset and fitted to the joint distribution are considered. The uncertainty of the estimated contours is quantified and discussed in light of differences that can be anticipated from the different methods used to calculate the contours.
