采集虾蟹混养池塘沉积物,利用特定饵料培养基,并通过定期增加培养基中饵料添加量的方式,对沉积物中的复合菌进行了6个周期(7d/周期)的富集驯化培养。在COD含量约为1 000mg/L、菌接种量为10%的饵料降解溶液中,后期复合菌在5d内对COD的最大去除率分别达到了64%、70%和88%。在6个周期富集驯化结束后,从复合菌中分离出16株菌,经16S rRNA分析,其中8株属于弧菌属(vibrio sp.),1株属于芽孢杆菌目(Bacillales sp.),2株属于芽孢杆菌属(Bacillus sp.),2株属于交替假单胞菌属(Pseudoalteromonas sp.),1株属于气单胞菌属(Aeromonas sp.),1株属于希瓦氏菌属(Shewanella sp.)。经血平板初步筛选出4株不具潜在致病性的菌,1株属于芽孢杆菌目,1株为巨大芽孢杆菌(Bacillus megaterium),2株为交替假单胞菌,在菌接种量为1.0×10~7cfu/mL的降解溶液中,各株菌在5d内对COD的最大去除率分别达到了40%、40%、56%和46%。将这4株菌按一定比例组合后,在5d内对COD的最大去除率可达到72%。研究表明,经富集驯化后的复合菌及从中分离筛选出的4株不具潜在致病性的细菌及按一定比例重组后的菌在调控养殖环境有机物污染方面都具有潜在应用价值。  相似文献

磺胺类、四环素、喹诺酮类和多粘菌素抗生素是医学与养殖业中的常用抗生素,其中多粘菌素在畜牧养殖业中常作为促长剂添加。调查分析了这4类抗生素的常见耐药基因在东海温州近海水域的分布情况。利用抗性平板筛选温州近海水域对磺胺类、四环素类及喹诺酮类抗生素耐药的可培养细菌分离株,对耐药分离株通过PCR方法进行目标耐药基因检测;对目标耐药基因阳性的耐药分离株进行多粘菌素耐药基因MCR检测以及采用微量二倍稀释法测定对上述4类抗生素的耐药性。结果表明,利用抗性平板筛选获得可培养耐药分离株1 605株,从中筛选到目标耐药基因阳性的51株,检出率为3.18%,包括含磺胺类耐药基因35株、含四环素耐药基因36株、含喹诺酮耐药基因17株、含多粘菌素耐药基因2株; 51株耐药菌归于7个菌属,其中气单胞菌属(Aeromonas sp.) 43株,占比84.31%,枸橼酸杆菌属(Citrobacter sp.)和希瓦氏菌属(Shewanella sp.)各2株,埃希氏菌属(Escherichia sp.)、类芽孢杆菌属(Paenibacillus sp.)、Thalassobius sp.及Amphritea sp.各1株;上述2株非常见分离株不统计在内,常见的49株均对四环素耐药, 41株对多粘菌素耐药,对磺胺类和喹诺酮类耐药的分离株均为34株;同时,发现23株对4类抗生素均产生耐药, 16株对多达3种抗生素耐药,多重耐药现象普遍存在。同时,鉴定出MCR-3阳性的条件致病菌维氏气单胞菌和豚鼠气单胞菌各1株。综上,温州市近海水域中含有常见耐药基因的耐药分离株检出率较低,但同一耐药基因的细菌分布多样,对磺胺类、四环素类、喹诺酮类及多粘菌素类等4类抗生素表现出较强的耐药性,多重耐药现象明显。研究结果将为评估温州近海水域的细菌耐药性风险,规范抗生素的使用提供科学依据。  相似文献

为了解仿刺参采苗板时期细菌的多样性,对分离自仿刺参采苗板上的细菌进行了分离和鉴定。应用常规生理生化鉴定、16S rDNA序列分析和构建系统发生树对其进行分子鉴定。结果表明:共分离得到19株海洋细菌,鉴定为7个属,包括短芽孢杆菌属(Brevibacillus sp.)、假交替单胞菌属(Pseudoalteromonas sp.)、Cobetia属、肠杆菌属(Enterobacter sp.)、芽孢杆菌属(Bacillus sp.)、普罗威登斯菌属(Providencia sp.)和海洋螺菌属(Oceanospirillaceae sp.)。从海洋环境中分离筛选细菌对海洋资源的开发与利用具有重要意义,此外对仿刺参育苗时期进行细菌种类的鉴定有助于疾病的预防和治疗,对仿刺参的水产养殖具有一定的指导意义。  相似文献

以龙须菜为唯一的营养源对深海沉积物进行富集和筛选,获得一个可降解龙须菜(Gracilaria lemaneiformis)、紫菜(Porphyra umbilicalis)和海带(Laminaria japonica)产生还原性寡糖的菌群.通过16S rRNA序列及16S rRNA-RFLP分析对该菌群中的细菌多样性进行了研究.结果表明,降解菌群中的细菌组成主要为弧菌属(Vibrio,8株)、火色杆菌属(Flammeovirga,7株)和希瓦氏菌属(Shewanella,5株),其中希瓦氏菌属和火色杆菌属的细菌在菌群中的丰度较高.采用龙须菜为唯一营养源的筛选培养基对菌群中的细菌进行分离,获得4株具有琼胶酶活力的细菌,包括2株火色杆菌属和2株希瓦氏菌属细菌.培养和未培养的结果均表明火色杆菌和希瓦氏菌这2个属为所研究的深海沉积物主要的龙须菜降解菌.对原始降解菌群和所分离的关键菌株进行了龙须菜酶解产糖能力的比较,结果发现菌群中的弧菌属菌株虽然自身没有酶解龙须菜的能力,但可能可以协助关键菌株,提高菌群对龙须菜的降解效率.因此本研究中所获得的菌群和菌株有望在琼胶寡糖的绿色生产中得到广泛应用.  相似文献

2002年5月从汕尾健生鲍鱼场养殖水体和成鲍肠道中分离筛选到26株异养细菌,其中11株来自成鲍肠道,15株来自养殖水体。对它们产胞外酶的能力进行了比较分析。结果表明肠道异养细菌中有72.7%能分泌蛋白酶、淀粉酶或明胶酶,9.1%分泌脂肪酶,45.5%分泌卵磷脂酶;水体异养细菌中则有46.7%能分泌蛋白酶或脂肪酶,66.7%分泌卵磷脂酶、淀粉酶或明胶酶。比较发现,肠道中脂肪酶或卵磷脂酶生产菌的比例小于水体异养细菌的比例;而蛋白酶、淀粉酶或明胶酶生产菌的比例却高于水体中异养细菌的比例。另外,水体异养细菌分泌多种胞外酶的能力强于肠道异养细菌。由此揭示现有鲍鱼肠道菌群结构的不完善,进而为通过添加有益菌而提高鲍鱼饲料利用率提供了可能。  相似文献

高效降解有机物和促藻生长菌株的分离和筛选   总被引:3,自引:0,他引:3  
本实验采用对虾饲料浸出液培养基从对虾健康养殖池中富集并分离出60株优势菌.通过对蛋白质、淀粉和CODMn的降解能力及对小球藻(Chlorella sp.)生长的影响等指标的检测,筛选出5^#、9^#和24^#共3株能高效降解有机物和促藻生长的菌株.10d内,其对CODMn的去除率均在40%以上.在有机培养液中,这3株菌对小球藻生长均有显著促进作用(p〈0.05),经过7d培养,藻细胞密度分别比对照菌提高了92.3%、98.1%、32.7%.在无机培养液中,9^#菌对小球藻生长有明显的促进作用(P〈0.05),经过7d培养,藻细胞密度比对照菌提高了77.3%.经初步鉴定5^#、9^#和24^#这3株菌分别属于弧菌属(Vibrio)、假单胞菌属(Pseudomonas)和芽孢杆菌属(Bacillus)的细菌.  相似文献

通过纯培养和16SrRNA基因序列的相似性比对,并构建系统发育树对威海海域柄海鞘共附生细菌的多样性进行了研究。用2216E培养基从柄海鞘样品中分离到78株细菌,通过形态学特征和TCBS培养基选择性筛选后,选取30株具有代表性的菌株进行了16SrRNA基因测序和基于16SrRNA基因序列的系统发育多样性分析。结果表明,30株菌株分别属于细菌域的4个系统发育类群(Actinobacteria,Bacteroidetes,Firmicutes,Gro-teobacteria)的11个科,11个属。根据16SrRNA基因序列,大多数菌株与其系统发育关系最密切的模式菌株之间存在一定的遗传差异(16SrRNA基因序列相似性为91.82%~98.93%)。其中菌株HQW7,HQYD1,HQWD4,HQE1和HQE2与相关模式菌株的16SrRNA基因最高相似度仅分别为91.82%,92.51%,92.99%,93.52%,93.87%,这5株菌可能代表新的分类单元。威海海域柄海鞘共附生细菌存在着较为丰富的物种多样性和系统发育多样性,并可能存在新的物种。  相似文献

研究了饲料中添加中草药和芽孢杆菌对凡纳滨对虾Litopenaeus vannamei生长和肠道细菌的影响.以基础饲料为对照(C),通过添加中草药和芽孢杆菌Bacillus spp.配制成4种实验饲料:0.2%中草药(M),0.20%中草药制剂+0.30%芽孢杆菌(BMI),0.10%中草药制剂+0.15%芽孢杆菌(BM2),0.30%芽孢杆菌(B).实验分成5组,分别投喂上述饲料,共进行了56d.结果表明,各组对虾的成活率95.83%-98.33%,各处理间无显著差异(P>0.05);投喂实验饲料的各组对虾终末体重均高于对照组(C),其中BM2与C差异性显著(P<0.05);M、BM1和BM2的增重率和特定增长率显著高于C和B;M组的对虾肠道异养菌总数和弧菌数均高于其他各组,而共同添加组BM1和BM2的异养菌和弧菌数则最少.从各组选取菌落数在50-100,分布均匀的平板,随机挑取30株细菌进行分类鉴定,结果C、M、BM1、BM2和B组细菌属数分别为11、8、6、5和7个.饲料中添加中草药和芽孢杆菌促进了对虾的生长,且共同添加的效果好于单独添加,它们的使用改变了肠道细菌的数量和组成.  相似文献

对青岛近海海水中琼胶降解细菌进行系统的分离,筛选得到87株海洋琼胶降解细菌。从中选出15株有代表性的菌株,克隆其16SrDNA序列,进行分子鉴定。结果表明,这些细菌主要分布在Cellulophaga,Cytophaga,Microbulbifer,Glaciecola,Pseudoalteromonas,Pseudomonas,Alteromonas和Agarivorans共8个属中。其中QM5,QM21,QM36鉴定为Cellulophaga lytica,QM15和QM28鉴定为Glaciecola mesophila,QM11,QM35,QM47,QM65分别鉴定为Cytophaga fuci-cola,Pseudoalteromonas atlantica,Pseudoalteromonas haloplanktis,Alteromonas addita。其余5株菌能够鉴定到属的水平。其中1株细菌QM42还不能确立分类地位。  相似文献

为研究南太平洋环流区底层海水可培养细菌的多样性,通过IODP 329航次获得了该区域7个站点的底层海水样品,利用传统的分离培养法获得菌株后,进行16SrDNA测序及系统发生分析。结果表明,从南太平洋环流区7个站点的底层水中分离出174株深海细菌,这些菌株属于4个门,30个属,78个种。其中γ-变形菌纲(γ-Proteobacteria)有143株,在数量和种类方面均占主导地位;α-变形菌纲(α-Proteobacteria)7株,β-变形菌纲(β-Proteobacteria)2株,厚壁菌门(Firmicutes)11株,放线菌门(Actinobacteria)6株,拟杆菌门(Bacteroidetes)5株。优势属有假交替单胞菌属(Pseudoalteromonas)、交替单胞菌属(Alteromonas)、弧菌属(Vibrio)、盐单胞菌属(Halomonas)等10个属;优势种有居珊瑚假交替单胞杆菌(Pseudoalteromonas paragorgicola)、子午盐单胞菌(Halomonas meridiana)、坎氏弧菌(Vibrio campbellii)、西班牙交替单胞菌(Alteromonas hispanica)、河豚毒素假交替单胞菌(Pseudoalteromonas tetraodonis)、琼氏不动杆菌(Acinetobacter junii)等12个。在7个站点中,位于环流边缘的U1371站点分离出的菌株数量明显多于其他站点,且是7个站点中唯一1个包含了分离出的所有6个门类的站点,多样性最高;而U1369和U1370站点都只分离出γ-变形菌纲1个门类。此外,9株细菌可能为海洋细菌新属或新种。  相似文献

对人工饲料饲养的凡纳滨对虾Litopenaeus vannamei成虾肠道菌群组成及其水体细菌区系组成进行了研究.分离纯化后的革兰氏阴性菌鉴定到种的水平.对虾肠道菌群主要由弧菌Vibrio spp.、希瓦氏菌属Shewnella spp.、嗜氢菌属Hydrogenophaga spp.、伯克霍尔德氏菌Burkholderia spp.、气单胞菌Aeromonas spp.、食酸菌Acidovorax spp.和芽孢杆菌Bacillus spp.组成.水体细菌区系主要包括弧菌Vibrio spp.、希瓦氏菌属Shewnella spp.、艾肯菌Empedobacter spp.、无色杆菌Achromobacter spp.和芽孢杆菌Bacillus spp..肠道菌群和水体细菌区系的优势菌是副溶血弧菌Vibrio parahaemolyticus和芽孢杆菌Bacillus spp..利用Shannon-Wiener指数进行多样性分析,发现虾肠道多样性指数(H)和均匀度(J)高于水体细菌区系,而丰度(D)较低.  相似文献

Species belonging to the genus Bysmatrum are peridinoid, thecate, photosynthetic dinoflagellates. The plate formula of Bysmatrum spp., arranged in a Kofoidian series, is almost identical to that of Scrippsiella spp. Bysmatrum spp., which were originally classified as Scrippsiella spp., but were transferred to the genus Bysmatrum spp. because of separation of the intercalary plates 2a and 3a by plate 3??. Whether this transfer from Scrippsiella spp. to Bysmatrum spp. is reasonable should be genetically confirmed. Dinoflagellates were isolated from 2 solar saltons located in western Korea in 2009?C2010 and 3 clonal cultures from Sooseong solar saltons and 2 clonal cultures from Garolim solar saltons were successfully established. All of these dinoflagellates were identified as Bysmatrum caponii based on morphology analysis by light and electron microscopy. The plates of all Korean strains of B. caponii were arranged in a Kofoidian series of Po, X, 4??, 3a, 7??, 6c, 4s, 5?, 0 (p), and 24??. When properly aligned, the ribosomal DNA (rDNA) sequences of the 3 Sooseong strains of B. caponii were identical, as were those of the 2 Garolim strains. Furthermore, the sequences of the 3 Sooseong strains were 0.01% different from those of the Garolim strains. However, the sequences of SSU rDNA of these Korean B. caponii strains were 9% different from that of Bysmatrum subsalsum and > 10% from that of any other dinoflagellate thus far reported. In the phylogenetic trees generated using SSU and LSU rDNA sequences, these Korean B. caponii strains formed a clade with B. subsalsum which was clearly divergent from the Scrippsiella clade. However, this Bysmatrum clade was phylogenetically close to the Protoperidinium and/or Peridinium clades. The results of the present study suggest that Bysmatrum spp. are markedly different genetically from Scrippsiella spp..  相似文献

海洋微生物抗肿瘤活性菌株的分级组合筛选   总被引:10,自引:2,他引:8  
为探索如何快速获取活性菌株的筛选模式 ,组合使用海虾生物致死法和tsFT2 10细胞的流式细胞术筛选模型 ,对从青岛前海和胶州湾潮间带海泥、动植物样品中分离的 162株海洋微生物 ,进行了抗肿瘤活性的分级组合筛选。经海虾生物致死法一级初筛 ,得到活性菌株 5 8株 (LC50 <5 0 0 μg/mL) ,阳性率为 3 5 .8%。对初筛有效的 5 8株菌利用流式细胞术筛选模型进行二级复筛 ,得到对tsFT2 10细胞有各种活性的活性菌株共 2 5株 ,阳性率为复筛菌株数的 43 .1%、初筛菌株总数的 15 .4% ,其中 ,诱发细胞坏死伴有凋亡诱导活性的菌株 7株 (占总菌株的 4.3 % ) ,具有细胞周期抑制活性的菌株 18株 (占总菌株的 11.1% )。该分级组合筛选模式与流式细胞术筛选模型的单独筛选模式相比 ,无漏筛 ,具有成本低、速度快、宜于大规模筛选等特点。  相似文献

对野生健康中国对虾成虾肠道微生物区系进行了研究.从其肠道中分离出47株菌,它们分别属于弧菌属、发光杆菌属、不动杆菌属、假单胞菌属、黄杆菌属、气单胞菌属、屈挠杆菌属和色杆菌属8个属.其中弧菌属和发光杆菌属在整个肠道中为优势菌属,不动杆菌属和假单胞菌属为次优势菌属,黄杆菌属、气单胞菌属、屈挠杆菌属和色杆菌属为非优势菌属.在对虾的整个肠道中,前肠和中肠的优势菌属为弧菌属,而后肠的优势菌属为发光杆菌属.在弧菌属中溶藻胶弧菌、漂浮弧菌和坎贝氏弧菌为优势菌,哈唯氏弧菌为非优势菌.前肠、中肠和后肠的菌量分别为1.3×105、2.8×105和1.1×104cfu/虾体.  相似文献

Over 400 yeast strains from seawater and sediments were obtained, but only five strains named HN2 -3, N13d, N13C, Mb5 and HN3 - 2 among them could form clear zones around their colonies on the double plates with 2.0% casein. Peptides in the hydrolysate produced by the proteases from strains HN2 -3 and N13d had higher angiotensin I-converting-enzyme (ACE)-inhibitory activity. The two marine yeast strains were identified to be Aureobasidium pullulans according to the results of routine yeast identification and molecular methods. After purification of the proteases from the two marine yeast strains, it was found that the optimal pH for them was both 9.0, both of them were serine alkaline protease. However, the optimal temperature for the protease from the strain HN2 -3 was 52℃ while that from strain N13d was 48℃. ACE-inhibitory activity of the peptides in the hydrolysate of shrimp protein produced by the purified protease from the strain HN2 -3 was the highest while antioxidant activity in the hydrolysate of spirulina protein produced by the purified protease from the strain N13d was the highest.  相似文献

Seventeen lactic acid bacterial strains (LAB) were isolated using MRS agar medium from Jeotgal, a Korean fermented food, purchased at the Jukdo market of Pohang. To identify the strains isolated, they were tested by examining their cell morphologies, gram-staining, catalase activity, arginine hydrolase activity, D-L lactate form and carbohydrate fermentation. According to the phenotypic characteristics, three strains were tent atively identified asLactobacillus spp., ten wereEnterococcus spp. (orStreptococcus spp., orPediococcus spp.) and the rest were Leuconostoc spp. (orWeissella spp.). Five strains among 17 were chosen by preliminary bacteriocin activity test. Four bacterial strains which inhibited both indicator microorganisms were identified by 16S rRNA sequencing. The results are as follows; Leuconostoc mesenteroides (HK 4),Leuconostoc mesenteroides (HK 5),Leuconostoc mesenteroides(HK 11),Streptococcus salivarius(HK 8). In order to check LAB which are showing a high survival rate in gut, we investigated three strains inhibiting both indicator microorganisms in artificial gastric acid and bile juice -all except HK8. The three strains mentioned above grew in extreme low acid conditions.  相似文献

This study provides new insights for the hatchery released Chinese shrimp (Fenneropenaeus chinensis), including proportion, dynamic migration route, after they were released into nature for stock enhancement using a new strategy quite different than ever. Chinese shrimp were sampled at 22 survey stations during two investigation voyages acrossing 74 survey stations in the Bohai Sea from July 16 to August 9 in 2015. Among 289 sampled individuals during the second voyage, totally 155 shrimps were identified as hatchery shrimp released into the Laizhou Bay at mid-May in 2015 based on finger-print of eight SSR (simple sequence repeats) markers, and the proportion of hatchery released shrimp in recapture samples were from 41.30%–85.71% in each station with an average value 53.63%, which verified a previous view point that up to 90% of autumn season Chinese shrimp landing in the Bohai Sea were composed of hatchery released. Meanwhile, the dynamic migration route of hatchery released shrimp revealed that part of released shrimp migrated heading northwest along the west coast of the Bohai Sea up to the Bohai Bay but just remained at the Laizhou Bay until over-wintering migration at mid-October when they initiate over-wintering migration. Present unnatural spring season shrimp fishing model cut the throat of spawner shrimp chance to swim back to their respective spawning plants at each spring, it still no chance to clarify whether the hatchery released shrimp could replenish to the reproduce population and complete a whole life cycle as same as their natural relatives.  相似文献

用添加不同剂量L-精氨酸(L-arg)的饲料投喂凡纳滨对虾(Litopenaeus vannanei),测定其血淋巴液中一氧化氮(NO)含量以及一氧化氮合成酶(NOS)、酸性磷酸酶(ACP)、碱性磷酸酶(AKP)和超氧化物歧化酶(SOD)活力,并检测其对病原体的抗病力.结果显示:L-arg显著提高血淋巴液中NO含量,且变化趋势与NOS、AKP活性升高趋势相一致;中等剂量(质量分数为1%)L-arg显著提高血淋巴液中SOD和ACP活性,但高剂量(质量分数为1.5%)、低剂量(质量分数为0.5%)L-arg对血淋巴液中SOD(高剂量除外)和ACP活性影响不显著.副溶血弧菌人工感染后,中剂量L-arg组(1%)对虾死亡率最低,高剂量(1.5%)、低剂量组(0.5%)次之,对照组死亡率最高,各剂量L-arg组死亡率与对照组相比差异显著.表明饲料中添加适量L-精氨酸可使对虾体液免疫因子总体活力达到较高水平,L-精氨酸含量过高对对虾非特异免疫力的作用效果反而不佳.  相似文献

从浙江宁波海域的滩涂泥样中分离到54株海洋放线菌,用卤虫液体测定法进行了杀虫活性菌株的筛选,结果显示多株放线菌具有较高的杀虫活性,菌株XS902,XS904,BL803,BL807对卤虫的ID50高达100以上。同时研究了XS904活性菌株的形态培养特征和生理生化特征,初步鉴定该菌株属于链霉菌属。  相似文献
