基于网络药理学探讨生脉散治疗病毒性心肌炎的作用机制

摘要:
目的:基于网络药理学探讨生脉散治疗病毒性心肌炎(VMC)的主要活性成分及其潜在作用机制。方法:运用本草组鉴数据库(HERB)初步获取生脉散的活性成分,结合SwissADME和SwissTargetPrediction平台筛选其中的主要活性成分,并预测其靶点。通过GeneCards及DisGeNET数据库筛选VMC相关靶点。取药物靶点与疾病靶点交集,获得生脉散治疗VMC的潜在治疗靶点。运用STRING数据库构建潜在治疗靶点蛋白质-蛋白质相互作用(PPI)网络图,并通过Cytoscape 3.7.1软件挖掘PPI网络中的核心靶点。运用Cytoscape 3.7.1软件构建“中药-活性成分-靶点-疾病”相互作用网络图,将潜在治疗靶点导入DAVID 6.8数据库进行基因本体(GO)功能及京都基因与基因组百科全书(KEGG)通路富集分析。结果:本研究共筛选出生脉散治疗VMC的有效活性成分151个,涉及55个潜在治疗靶点,405个生物过程,36个细胞组分,55个分子功能和147条代谢通路。结论:生脉散可通过调控细胞因子、蛋白激酶、天然免疫基因等多靶点、多途径干预VMC的进程,为其临床应用提供了理论基础。

Abstract:
Objective:To investigate the main active components and potential mechanism of action of Shengmai powder in the treatment of viral myocarditis (VMC) based on network pharmacology.Methods:HERB database was used to obtain the active components of Shengmai powder,and SwissADME and SwissTargetPrediction platforms were used to obtain its main active components and predict their targets.GeneCards and DisGeNET databases were used to obtain the targets associated with VMC.Drug targets and disease targets were intersected to obtain the potential therapeutic targets for Shengmai powder in the treatment of VMC.STRING database was used to construct a protein-protein interaction (PPI) network of the potential therapeutic targets,and Cytoscape 3.7.1 software was used to identify the core targets in the PPI network.Cytoscape 3.7.1 software was used to construct a “TCM drug-active component-.target-disease” network,and the potential therapeutic targets were imported into DAVID 6.8 database to perform gene ontology (Go) functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis.Results:This study obtained 151 effective active components of Shengmai powder in the treatment of VMC,involving 55 potential therapeutic targets,405 biological processes,36 cellular components,55 molecular functions,and 147 metabolic pathways.Conclusion:Shengmai powder can intervene against the progression of VMC by regulating multiple targets (including cytokines,protein kinase,and innate immune genes) and pathways,which provides a theoretical basis for the clinical application of Shengmai powder.
